        인간포배기 배아의 효과적인 유리화 동결법의 개발을 위한 연구

        Objective: The purpose of this study was to evaluate the survival rate of vitrified blastocyst according to the freezing vessels, equilibration time in cryoprotectant and artificial dehydration method. Methods: Human blastocysts were vitrified after loading onto the plastic straw, open-pulled straw (OPS), electron microscopy grid (EM grid) for 1.5 min or 3 min. They also were directly plunged into LN2 within 30sec. For artificial shrinkage of blastocysts, 36 gauge fine needle was pushed at the cellular junction of the trophectoderm into the blstocoele cavity until it shrank without damage of inner cell mass. Results: The survival rate of vitrified blastocysts on plastic straw, OPS, EM grid as freezing vessels were 26.7, 13.0 and 60.5%, respectively. The survival rate of EM grid was significantly higher than that of plastic straw and OPS (p<0.05). For 1.5 min equilibrium, the survival rates of early blastocyst (EB), middle blastocyst (MB) and late blastocyst (LB) were 64.4, 81.0, and 20.0% respectively. For 3 min equilibrium, the survival rates of EB, MB, and LB were 69.9, 50.0 and 57.5% respectively. The survival rates of EB and MB were significantly higher than that of LB in 1.5 min equilibrium group (p<0.05), however, the significance was not observed in 3 min equilibrium groups. In cytoplasmic shrinkage before vitrification, the survival rates of EB, MB and LB were 92.9, 100 and 75.9% respectively. The survival rate of MB was significantly higher than that of LB (p<0.05). The survival rates of vitrified blastocysts by artificial dehydration and slow-frozen blastocysts were not significantly different as 88.9 and 66.7%, respectively. Conclusion: This study showed that the vitrification of human blastocysts using EM grid and artificial dehydration is an effective method. Therefore, these methods would be an useful techniques for blastocyst cryopreservation.

        가압가열 및 microwave에 의한 중력분 반죽 gliadin의 항원성 변화

        본 연구에서는 가압가열 및 microwave 처리에 의한 중력분 반죽 추출물 내의 gliadin 단백질의 항원성 변화에 대해 살펴보았다. 중력분 반죽에 가압가열과 microwave를 단독 또는 병행으로 처리하여 ci-ELISA, SDS-PAGE 및 immunoblotting을 실시하였으며, 가압가열 처리에 의해서 anti-gliadin IgG 항체와 gliadin과의 결합력이 다소 감소한 것을 확인하였다. 특히 30 min 이상 처리시 더욱 감소한 것으로 나타났으며, SDS-PAGE와 immunoblotting 결과에서도 gliadin band의 강도가 약해지고 항체와의 반응도 나타나지 않았다. Microwave 처리의 경우, 5 min 이상 처리시 일부 gliadin 단백질의 소실이 관찰되었으나, 항원성에는 큰 변화가 없었다. 또한 가압가열 및 microwave 병행 처리에 의해 항원-항체 결합력이 더욱 감소되었으며, 특히 가압가열 50 min, microwave 10 min 처리시 약 35.0%로 감소되었다. 이상의 결과를 통해 가압가열 처리에 의해 중력분 반죽 추출물 내 gliadin의 항원성이 감소되는 것을 확인하였으며, microwave와 병행 처리하는 경우, 더욱 감소하는 것을 확인하였다. The aim of this study was to determine the optimal physical treatment to reduce the antigenicity of gliadin in wheat dough. Medium wheat dough was treated with an autoclave (5, 10, 30, and 50 min at 121℃, 1 atm), a microwave (1, 5, and 10 min) or both (10, 30, and 50 min/5, 10 min). The proteins in the dough extracts were analyzed by SDS-PAGE and the binding ability of anti-gliadin IgG to gliadin was examined by ci-ELISA and immunoblotting. Results showed that the ability of anti-gliadin IgG to bind to gliadin in wheat dough treated with an autoclave alone or in combination with a microwave was decreased. Especially, it declined to ~77% after autoclaving for 30 min and 35% after both autoclaving for 50 min and microwaving for 5 min. In addition, the intensity of gliadin bands in SDS-PAGE were weakened and anti-gliadin IgG did not recognize gliadin in immunoblotting. However, microwaving alone did not affect the antigenicity of gliadin in wheat dough. These results indicate that autoclaving may affect the reduction of the antigenicity of gliadin in medium wheat dough. Moreover, autoclaving in combination with microwaving is more effective for reducing the antigenicity of wheat dough.

        Preparation and characterization of ferroelectric Hf<sub>0.5</sub>Zr<sub>0.5</sub>O<sub>2</sub> thin films grown by reactive sputtering

        <P>HfO<SUB>2</SUB>-ZrO<SUB>2</SUB> solid-solution films were prepared by radio frequency sputtering, and the subsequent annealing process was optimized to render enhanced ferroelectric behavior. The target power, working pressure and O<SUB>2</SUB> partial pressure ratios were varied, along with the annealing gas, time and temperature. Then, the film’s structural and electrical properties were carefully scrutinized. Oxygen-deficient conditions were necessary during the sputter deposition to suppress grain growth, while annealing by O<SUB>2</SUB> gas was critical to avoid defects and leakage problems. It is expected that the grain size difference under various deposition conditions combined with the degree of TiN top and bottom electrode oxidation by O<SUB>2</SUB> gas will result in different ferroelectric behaviors. As a result, Hf<SUB>0.5</SUB>Zr<SUB>0.5</SUB>O<SUB>2</SUB> prepared by radio frequency sputtering showed optimized ferroelectricity at 0% of O<SUB>2</SUB> reactive gas, with a doubled remnant polarization value of ∼20 <I>μ</I>C cm<SUP>−2</SUP> at a thickness of 11 nm. Film growth conditions with a high growth rate (4–5 nm min<SUP>−1</SUP>) were favorable for achieving the ferroelectric phase film, which feasibly suppressed both the grain growth and accompanying monoclinic phase formation.</P>

        Palm prints have been used for personal identification because they are unique to each individual. Palm prints have also been used for diagnosing genetic disorders and for revealing physical anthropological characteristics, due to their specific appearance characteristic of genetic factors and nationality. Therefore, several analytical methods of palm prints have been developed and applied for the purpose described. However, in the previous reports, the analytical methods of palm prints and the numbers of subjects were insufficient to find Standard values for Korean palm prints. In this study, in order to determine the Standard value for Korean palm prints, we examined palm prints of 3216 Korean adults (2095 males and 1121 females), analyzed the data according to sex and side of hand, and compared with those of foreigners. The results are s follows: 1) The incidence of 7, 9, and 11 of D was 41.8%, 33.5%, and 20.4%, respectively. Comparing the incidences 017, 9, and 11 of D in Koreans with those of foreigners, the Korean belong to the Asians including the Japanese, the Chinese, and the Philippine, however, the Korean was doser to the Blacks and distant from the Whites than the other Asians. 2) The palm prints were classified into the open type in which B terminates at the ulnar border of hand, and the dosed type in which B does not. Open type, 7-5-5 (male 37.6%, female 39.5%) and 9-7-5 (male 23.9%, female 25.5%), was more frequent in female than in male, while dosed type, 11-9-7 (male 9.8%, female 9.1%), was more frequent in male than in female. Also, open type, 7-5-5 (right 28.9%, left 47.6%), was more frequent in left hand than in right hand, while dosed type, 11-9-7 (right 17.1%, left 2.1%), was more frequent in right hand than in left hand. Consequently, open type was more frequent in the weak hand, whereas dosed type was more frequent in strong hand. 3) Because a-b (39.2) and c-d (35.0) were more numerous than b-c (27.8), distance between a and b and between c and d were longer than that between c and d. The long distance of a, b and c, d could be resulted by wide moving range of second finger and fifth finger, respectively. Also, there was a tendency that the palmar ridge counts of right hand was more numerous than those of left hand, which could be resulted by wide moving range of right fingers in the right-handed persons who were more frequent than the left-handed persons. 4) O, in which there was no triradius, and X, in which dermal ridge from a triradius was blocked and terminated at the other dermal ridge, were frequent in C (O 7.8%, × 7.2%) than in D (O 0.1%, × 0.1%) or in B (O 0.4%, X 1.2%). The frequency of 0 and X in C was thought to be related with the location of ein the narrow space between d and b. The narrow space between d and b was known because c-d and b-c was less numerous than a-b. 5) The distances between adjacent triradii were longer in male than in female, however, the palmar ridge counts for a-b, b-c, and c-d were less numerous in male than in female. Consequently, dermal ridges were thought to be thicker in male than in female. The Standard values of Korean palm prints obtained from this study are expected to be used for both diagnosing genetic disorders and for revealing physical anthropological characteristics in Korea. Further studies to clarify the relationship between the palm prints and the power of hand as well as the moving range of fingers are in need.

        치면열구전색제의 조건에 따른 법랑질 결합강도에 관한 연구

        In this study, the shear bond strengths and accompanying elastic modulus ratio, displacement at bond failure and work up to fracture of pit and fissure sealants to toothe enamel were analyzed by the following variables, those were acid etching, moisture content of the surface of enamel, dentin bonding agent(species and method), thermocycling, saliva contamination. One kind of chemically cured resin based pit and fissure sealant, two kinds of light cured resin based pit and fissure sealants and one kind of glass ionomer based pit and fissure sealant, and two kinds of dentin bonding agents were studied. The buccal surface of extracted premolars were ground flat with # 600 silicone carbide papers, and the ground surface was treated depending on the experimental conditions. After each treatment, each pit and fissure sealant was filled into plastic tube of 3mm diameter, which were laid on the surface of ground enamel surface, and were cured. Sealant bonded teeth specimens were immersed in 37℃ distilled water form 24 hours. The shear bond strength of sealant to enamel surface was measured with Universal testing machine with the cross head speed of 1mm/min. From the obtained load-deflection curves, modulus ratio, bond strength, displacement at bond failure and work up to fracture were analyzed. From this experiment, the following results were obtained : 1. Comparing the results of standard condition(i. e. acid etching, dry surface, no dentin bonding agent and 300 thermocyling), the elastic modulus ratio of chemically cured resinbased pit and fissure sealant was significantly higher than those of other materials(P<0.05), and the bond strength, displacement at bond failure and work up to fracture of glass ionomer based pit and fissure sealant were significantly lower than those of other materials(P<0.05). 2. The results of ANOVA with the variables of type of material, acid etching, moisture condition of the enamel surface, dentin bonding agents were as follows ; 1) variables showed significant main effect on the shear bond strength of pit and fissure sealant to enamel(P<0.01), 2) type of material, acid etching and moisture condition of enamel influenced the bond strength significantly(P<0.01), 3) type and application methods of dentin bonding agents influenced on the bond strength significantly(P<0.05). 3. There were statistically significant correlation between the work up to fracture and the bond strength, and between the work up to fracture and the displacement at bond failure(P<0.01), and in these cases the correlation coefficient was 0.87.

        Effect of Forging Type on the Deformation Heterogeneities in Multi‑Axial Diagonal Forged AA1100

        The present study investigated the effects that different types of forging exert on the deformation heterogeneities developedin AA1100 during multi-axial diagonal forging. To measure the deformation heterogeneities of deformed workpieces, thevalues for hardness and Kernel average misorientation were measured at the center section following each forging process. Type-D forging that consists of diagonal forging and return-diagonal forging was relatively advantageous compared withType-P forging that includes plane forging and return-plane forging for minimizing the non-uniformity of deformationdeveloped in workpieces. The effective strain developed in a workpiece during the 2 types of forging was simulated using3-D FEA. FEA revealed that the positions and degrees of occurrence for soft and hard-zones in workpieces vary greatlydepending on the forging type. Type-D forging was relatively advantageous compared with Type-P forging for minimizingthe non-uniformity of effective strain developed in workpieces.

        Since the changes of Na^+-Li^+ countertransport (SLC) Vmax in essential hypertension have been reported. it has been known that hyperlipidemia could affect SLC independently of blood pressure. To reveal the relation between SLC Vmax and RBC membrane fatty acid conposition in the hypertensive patients, we divided patients into two groups according to their levels of SLC Vmax of erythrocytes(elevated SLC Vmax: n=4, normal SLC Vamx: n=6). The results were as follows: 1) There was no difference in body mate index, waist-hip ratio, blood pressure, serum cholesterol level between two groups. Triglyceride were higher in elevated group (278.0±180.0 ㎎/㎗) than normal group (124.3±60.7 ㎎/㎗) but there was no statistical significance. HDL-cholesterol levels were significantly lower in elevated group (32.0±3.6 vs. 44.8±8.9 ㎎/㎗) and free fatty acid concentrations, intracellular (RBC) Na^+ concentrations were similar between two groups. 2) The results of oral glucose tolerance test showed no differences in the glucose and insulin area, however the incremental insulin areas of elevated group (108.8±31.3, μU. min/㎖×10²) were significantly higher compared with those of normal group (56.3±20.7 μU. min/㎖×10²). 3) In the elevated group, palmitic acid proportions of erythrocytes (22.1±0.7%) were higher than those of normal group (20.1±0.7%) and SLC Vmax showed significant correlation (r=0.81, p$lt;0.01) with palmitic acid proportion. These results suggest that the abnormal lipid metabolism may change the RBC Na^+ transport system through the alterations of RBC membrane fatty acid composition.

        Recently, the robustness against daily impact is important according to portablization and downsizing of mobile electronics. Especially, almost parts of cellular phone should undergo drop test when they fall 1.5 m above ground. This test simulates the case when cellular phone slips through user's fingers while he is talking on the phone. This paper studies a drop test of auto focusing (AF) actuator for camera module in cellular phone. This component is composed of voice coil motor (VCM) as an actuator and leaf spring as a guide and suspension. The leaf spring's deformation is essential for the test because its permanent distortion disables the focusing, and then, a high quality photograph cannot be obtained. Up to now the drop test has been carried out after fabricating real AF actuator. We propose a dropt test model which simulates the drop test based on finite element analysis. This model makes us enable investigate the stress acting on the clamping and curved parts of leaf spring. Stress over Von Mises criterion lets the spring deformed permanently, and then AF actuator malfunctioned, It helps us to design and modify AF actuator without manufacturing the real product. And also, it saves the time and cost for the development of new products.

        Characterizations of Modified Silica Nanoparticles(I)

        (3-mercaptopropyl)trimethoxysilane (MPTMS) was used as a silylation agent, and modified silica nanoparticles were prepared by solution polymerization. 2.0 g of silica nanoparticles, 150 ml of toluene, and 20 ml of MPTMS were put into a 300 ml flask, and these mixtures were dispersed with ultrasonic vibration for 60 min. 0.2 g of hydroquinone as an inhibitor and 1 to 2 drops of 2,6-dimethylpyridine as a catalyst were added into the mixture. The mixture was then stirred with a magnetic stirrer for 8 hrs. at room temperature. After the reaction, the mixture was centrifuged for 1 hr. at 6000rpm. After precipitation, 150 ml of ethanol was added, and ultrasonic vibration was applied for 30 min. After the ultrasonic vibration, centrifugation was carried out again for 1 hr. at 6000rpm. Organo-modification of silica nanoparticles with a ${\gamma}$-methacryloxypropyl functional group was successfully achieved by solution polymerization in the ethanol solution. The characteristics of the ${\gamma}$-mercaptopropyl modified silica nanoparticles (MPSN) were examined using X-ray photoelectron spectroscopy (XPS, THERMO VG SCIENTIFIC, MultiLab 2000), a laser scattering system (LSS, TOPCON Co., GLS-1000), Fourier transform infrared spectroscopy (FTIR, JASCO INTERNATIONL CO., FT/IR-4200), scanning electron microscopy (SEM, HITACHI, S-2400), an elemental analysis (EA, Elementar, Vario macro/micro) and a thermogravimetric analysis (TGA, Perkin Elmer, TGA 7, Pyris 1). From the analysis results, the content of the methacryloxypropyl group was 0.98 mmol/g and the conversion rate of acrylamide monomer was 93%. SEM analysis results showed that the organo-modification of ultra-fine particles effectively prevented their agglomeration and improved their dispensability.

        A comparison of Min’s glasses and conventional occlusion therapy in the treatment of amblyopic children: a prospective study

        Min’s glasses are specially manufactured in order to enhance the satisfaction of the wearer and increase treatment effectiveness. We compared the effectiveness of Min’s glasses with that of conventional occlusion therapy in amblyopic pediatric patients. We prospectively studied 60 amblyopic patients. For four to 30 months, 24 of the patients were treated with conventional patch occlusion (patch group) and 36 of them were treated with Min’s glasses (glasses group). We compared the improvement in visual acuity and the treatment compliance between both groups, according to age (over six and less than six), sex, type of amblyopia, and the duration of treatment. Sixteen patients (66.7%) in the patch group and 32 patients (88.9%) in glasses group showed improvement in visual acuity (p=0.002). The pre-treatment average log MAR acuity was 0.56±0.25 in the patch group, and 0.59 ±0.25 in the glasses group. The average improvement in visual acuity was a 0.17 log MAR score in the patch group, and a 0.31 log MAR score in the glasses group (p=0.004). Compliance was 59.54% in the patch group and 83.44% in the glasses group (p=0.012). The pre-treatment average log MAR acuity was 0.49 ± 0.23 for children over six years of age in the patch group, and 0.58±0.28 for children over six years of age in the glasses group. For children over six years of age in the glasses group the improvement in visual acuity (0.29 log MAR score) was greater than for children over six years of age in the patch group (0.06 log MAR score) (p=0.0003). The pre-treatment average log MAR acuity was 0.55±0.22 for female patients in the patch group, and 0.60±0.25 for female patients in the glasses group. Female patients in the glasses group also showed a greater visual acuity improvement (0.29 log MAR score) than female patients in the patch group (0.14 log MAR score) (p=0.0028). However, there were no differences between the groups in patients less than six years of age and in male patients. In conclusion, Min’s glasses were more effective than conventional treatment with a patch in improving visual acuity and encouraging compliance in pediatric amblyopic patients, especially in children over six years of age and in girls.
