        Ziwuliuzhu Acupuncture Modulates Clock mRNA, Bmal1 mRNA and Melatonin in Insomnia Rats

        Background: In clinics, Ziwuliuzhu acupuncture is widely considered an effective method of treating insomnia; however, there is currently limited information available regarding its possible mechanisms. Although the method of Ziwuliuzhu acupuncture possesses a unique rhythmic pattern. Objectives: In this study, we have creatively combined the traditional Chinese medicine of Ziwuliuzhu with a modern biological rhythm to investigate the internal mechanism of insomnia. Methods: Pathological tissue from the hypothalamus was analyzed using hematoxylin– eosin staining. The level of TNF (tumor necrosis factor)-α in the SCN (suprachiasmatic nucleus) area of the hypothalamus was detected in situ using the TUNEL fluorescence staining assay. The concentration of hypothalamic melatonin was detected using the enzyme-linked immunosorbent assay (ELISA). The mRNA expression of Clock and Bmal1 was measured using RT-qPCR. Results: In the Ziwuliuzhu acupuncture groups, the structural damage in the hypothalamic neurons was alleviated compared to the model group and the expression of inflammatory factors was reduced. The mRNA expression levels of Clock and Bmal1 were significantly increased (p < 0.05). The concentration of melatonin was significantly increased (p < 0.001). Although there were no significant differences between the treatment groups (diazepam group, Nazi group, Najia group, and routine group) (p > 0.05). Conclusion: Ziwuliuzhu acupuncture alleviated neuronal damage and modulated the inflammatory reaction in the hypothalamus of rats with insomnia.         Covered Stenting Is an Effective Option for Traumatic Carotid Pseudoaneurysm with Promising Long-Term Outcome

        Objective : Covered stenting is an optional strategy for traumatic carotid pseudoaneurysm, especially in malignant conditions of potential rupture, but the long-term outcomes are not clear. Our aim was to determine if covered stenting is an effective option for traumatic carotid pseudoaneurysm with promising long-term outcomes. Methods : Self-expanding Viabahn and balloon-expandable Willis covered stents were separately implanted for extra- and intracranial traumatic carotid pseudoaneurysm. The covered stent was placed across the distal and proximal pseudoaneurysm leakage under roadmap guidance. Procedural success was defined as technical success (complete exclusion of the pseudoaneurysm and patency of the parent artery) without a primary end point (any stroke or death within 30 days after the procedure). Long-term outcomes were evaluated as ischemic stroke in the territory of the qualifying artery by clinical follow-up through outpatient or telephone consultation and as the exclusion of the pseudoaneurysm and patency of the parent artery by imaging follow-up through angiography. Results : Five patients with traumatic carotid pseudoaneurysm who underwent covered stenting were enrolled. The procedural success rate was 100%. No ischemic stroke in the territory of the qualifying artery was recorded in any of the five patients during a mean clinical follow-up of 44±16 months. Complete exclusion of the pseudoaneurysm and patency of the parent artery were maintained in all five patients during a mean imaging follow-up of 39±16 months. Conclusion : Satisfactory procedural and long-term outcomes were obtained, suggesting that covered stenting is an effective option for traumatic carotid pseudoaneurysm.

        Clusterin negatively modulates mechanical stress-mediated ligamentum flavum hypertrophy through TGF-β1 signaling

        Ligamentum flavum hypertrophy (LFH) is a major cause of lumbar spinal canal stenosis (LSCS). The pathomechanisms for LFH have not been fully elucidated. Isobaric tags for relative and absolute quantitation (iTRAQ) technology, proteomics assessments of human ligamentum flavum (LF), and successive assays were performed to explore the effect of clusterin (CLU) upregulation on LFH pathogenesis. LFH samples exhibited higher cell positive rates of the CLU, TGF-β1, α-SMA, ALK5 and p-SMAD3 proteins than non-LFH samples. Mechanical stress and TGF-β1 initiated CLU expression in LF cells. Notably, CLU inhibited the expression of mechanical stress-stimulated and TGF-β1-stimulated COL1A2 and α-SMA. Mechanistic studies showed that CLU inhibited mechanical stress-stimulated and TGF-β1-induced SMAD3 activities through suppression of the phosphorylation of SMAD3 and by inhibiting its nuclear translocation by competitively binding to ALK5. PRKD3 stabilized CLU protein by inhibiting lysosomal distribution and degradation of CLU. CLU attenuated mechanical stress-induced LFH in vivo. In summary, the findings showed that CLU attenuates mechanical stress-induced LFH by modulating the TGF-β1 pathways in vitro and in vivo. These findings imply that CLU is induced by mechanical stress and TGF-β1 and inhibits LF fibrotic responses via negative feedback regulation of the TGF-β1 pathway. These findings indicate that CLU is a potential treatment target for LFH.

        Growth of monolayer and bilayer MoS2 through the solution precursor for high-performance photodetectors

        Various studies suggest that the performances of TMDs are largely thickness dependent. In this paper, we develop a chemical vapor deposition method to synthesis monolayer and bilayer MoS2 flakes with a solution precursor. The MoS2 phototransistors were prepared to investigate their optoelectronic performance. The MoS2 photodetectors exhibit high detectivity of 2.44 × 1011 and a fast response/recovery time of 97 ms/291 ms. The photoresponsivity of bilayer MoS2 flakes was found up to 7160 A W−1. Our research will pave a pathway to control the layer numbers of other TMDs nanostructures, expand the application of high performance 2D materials.

        Effects of different culture systems on the culture of buffalo (Bubalus bubalis) spermatogonia stem cell-like cells in vitro

        Currently, the systems for culturing buffalo spermatogonial stem cells (SSCs) in vitro are varied, and their effects are still inconclusive. In this study, we compared the effects of culture systems with undefined (foetal bovine serum) and defined (KnockOut Serum Replacement) materials on the in vitro culture of buffalo SSC-like cells. Significantly more DDX4- and UCHL1-positive cells (cultured for 2 days at passage 2) were observed in the defined materials culture system than in the undefined materials system (p < 0.01), and these cells were maintained for a longer period than those in the culture system with undefined materials (10 days vs. 6 days). Furthermore, NANOS2 (p < 0.05), DDX4 (p < 0.01) and UCHL1 (p < 0.05) were expressed at significantly higher levels in the culture system with defined materials than in that with undefined materials. Induction with retinoic acid was used to verify that the cultured cells maintained SSC characteristics, revealing an SCP3+ subset in the cells cultured in the defined materials system. The expression levels of Stra8 (p < 0.05) and Rec8 (p < 0.01) were significantly increased, and the expression levels of ZBTB16 (p < 0.01) and DDX4 (p < 0.05) were significantly decreased. These findings provided a clearer research platform for exploring the mechanism of buffalo SSCs in vitro.

        Preparation and property evaluation of a temperature-resistant Zr-crosslinked fracturing fluid

        Compared with the traditional water-based fracturingfluid thickener guanidine gum, the price ofsynthetic polymer is relatively low, and the temperature resistance, shear resistance and viscoelasticityof the formed fracturingfluid are better. Therefore, the fracturingfluid formed by the crosslinking ofHPAM and organic metal crosslinker will become one of the hot spots in the research of water-basedfracturingfluid. In this paper, zirconium oxychloride, acetic acid and triethanolamine were used as rawmaterials to prepare organic Zr-crosslinked agent. The polymer gel was formed by crosslinking theorganic Zr-crosslinked agent with the acrylamide copolymer KY-5S solution, and the viscosity of the gelwith different concentrations was investigated. The performance of the fracturingfluid was evaluatedthrough laboratory experiments. The results showed that the viscosity of the fracturingfluid remainedabove 100 mPa s at the shear rate of 170 s 1 and after 2 h of shear at 140 C. When the temperature was20 C, the shear stress was 0.1 Pa, and the frequency f varied from 0.1 to 10 Hz, the elastic modulus offracturingfluid was at least 11.6 Pa, and the viscous modulus was at least 8.54 Pa. At the temperature of90 C, when the addition of glue breaker was 0.01 wt%, the longest breaking time was 3 h, and themaximum viscosity of breaking liquid was 4.05 mPa s. The damage rate of core permeability of gluebreaker was about 13%. When the temperature was 25 C and the shear rate was 0–170 s-1, the viscosity offracturingfluid remained above 200 mPa s.

      • Kinetic Resolution of Allylic Alcohol with Chiral BINOL-Based Alkoxides: A Combination of Experimental and Theoretical Studies

        <P>The development and characterization of enantioselective catalytic kinetic resolution of allylic alcohols through asymmetric isomerization with chiral BINOL derivatives-based alkoxides as bifunctional Brønsted base catalysts were described in the study. A number of chiral BINOL derivatives-based alkoxides were synthesized, and their structure-enantioselectivity correlation study in asymmetric isomerization identified a promising chiral Brønsted base catalyst, which afforded various chiral secondary allylic alcohols (ee up to 99%, <I>S</I> factor up to >200). In the mechanistic study, alkoxide species were identified as active species and the phenol group of BINOL largely affected the high reactivity and enantioselectivity via hydrogen bonding between the chiral Brønsted base catalyst and substrates. The strategy is the first successful synthesis strategy of various chiral secondary allylic alcohols through enantioselective transition-metal-free base-catalyzed isomerization. The applicability of the strategy had been demonstrated by the synthesis of the bioactive natural product (+)-veraguensin.</P> [FIG OMISSION]</BR>

        Investigation on the interactions of scutellarin and scutellarein with bovine serum albumin using spectroscopic and molecular docking techniques

        The binding abilities of scutellarin (Scu) andscutellarein (Scue) with bovine serum albumin (BSA) wereinvestigated using equilibrium dialysis, high performanceliquid chromatography, fluorescence spectroscopy, competitivesite marker and molecular docking. The resultsshowed that the average protein binding ratios of Scu andScue with BSA were (79.85 ± 1.83) and (85.49 ± 1.21) %respectively. Under simulated physiological conditions, thefluorescence data indicated that Scu and Scue bound withBSA through a static mechanism. The thermodynamicparameters indicated that the interactions of Scu-BSA andScue-BSA mainly occurred by van der Waals forces andhydrogen bonds and it was easier for Scue to bind withBSA than Scu, indicating that the glucuronic acid moleculein Scu decreased the binding affinity. Site competitivemarker experiments showed that the binding sites of Scuand Scue mainly located within the sub-domain IIA ofBSA. Furthermore, molecular docking studies indicatedthat one BSA could bind three Scue, while one BSA couldcarry only two Scu. All these results clearly indicated theinteractions of Scu and Scue with BSA, which will lay thefoundation for further research to determine the pharmacologyand pharmacodynamics of Scu and Scue for treatingischemic cerebrovascular disease.
