        김영근(金泳根)의 <금강산졀긔 동유록>에 나타난 서술방법(敍述方法)과 의식세계(意識世界) 연구(硏究)

        조선 시대에도 많은 사람들이 여행을 하였는데, 그 여행지를 대표하는 공간이 금강산이다. 금강산을 유람한 유람객은 산수유람을 목적으로 하였으나, 강원도 관찰사는 관할 지역을 순력(巡歷)하는 여정에 금강산에 이르는 노정을 포함하기도 했다. 근대이전의 금강산 유람객은 대부분 당대의 지식인이었고, 자신의 여행 체험과 견문을 한문으로 기록하였다. 1863년(철종 14) 강원도 관찰사로 부임한 김영근(金泳根)은 순력과 근친(覲親)하러 오는 아들과의 만남을 목적으로 8월 6일 원주 감영을 출발한 후 9월 4일 감영으로 복귀하였다. 이때의 견문과 체험을 기록한 것이 <금강산졀긔 동유록>이다. 이 기록은 한글로 기록한 일기체 유산록(遊山錄)으로 19세기 유람록이 지닌 특징을 보여준다. 문학적 표현(表現)보다는 사실적 재현(再現)에 집중하고 있으며, 잡록(雜錄)의 글쓰기가 주를 이루고 있다. 김영근은 순력, 제명, 전설을 통해 자신의 인식을 드러내었다. 관할 지역을 순시하면서 지방관으로서의 소명의식, 제명을 하면서 가족의식을 드러내었다. 지역에 대한 정보를 기록하고, 설화를 기록하는 등 금강산의 장소적 특성과 의미를 전달하고자 했다. Many people traveled even during the Joseon Dynasty, and the space representing the travel destination was Mt. Geumgang. The tourists who visited Mt. Geumgang aimed at landscape sightseeing, but the Gangwon-do governor also included a route to Mt. Geumgang in his supervisory journey throughout his jurisdiction. Before the modern era, most of the tourists visiting Mt. Geumgang were intellectuals of the era, and they recorded their travel experiences and knowledge in Chinese characters. Kim Young-geun, who was appointed as the governor in Gangwon-do in 1863 (14th year of King Cheoljong), left his Wonju government office on August 6 for the purpose of his supervisory journey throughout his jurisdiction and also meeting his son visiting him for filial piety, and returned to his office on September 4. The record of his knowledge and experiences of this time is < Geumgangsan-jyeolgwi Dongyurok >. < Dongyurok > is a diary type record of playing in the mountain written in Korean language and has an incomplete composition, but it shows the characteristics of the 19th century excursion record. It focuses on realistic representation rather than literary expression, and is mainly composed of miscellaneous written records. Kim Young-geun revealed his own perception through supervisory journey, writing his name on scenic spots, and old story. As he toured his jurisdiction, he revealed his sense of vocation as a local government official, his family and family consciousness by writing his name on scenic spots. While recording the old story, he newly recognized the place value and meaning of Mt. Geumgang.

        벼 생육 및 수집에 대한 대체농자재의 영향

        본 시험은 환경농업기술을 직접 농가에 파급시킬 목적으로 3년간('96~'98)수행하였던 연구결과를 토대로 1999년에 환경농업 시범마을인 남원시 대강면 방산리의 일반농가포장에서 수행한 결과이다. 계분 발효퇴비와 깻묵을 시용한 환경농업과 절충형농업 시험구의 벼 생육은 화학비료를 시용한 일반관행농업 시험구에 비하여 저조하였으며, 왕우렁이(Ampullarius insularus)를 이용한 잡초방제 효과는 제초제 2회(초·중기)살포효과보다 우수하였다. 목초액의 병해충 방제효과는 저조하였고 또한 환경농업과 절충형농업 시험구의 정조수량은 일반관행농업 시험구에 비하여 3~5% 감소 되였다. 수질은 환경농업과 절충형농업 시험구가 일반관행농업 시험구에 비하여 COD, NO₃-N, SO₄^(2-), Cl^(-)이 낮았으며, 물벼룩의 밀도는 높았다. The aim of this study was to apply the new agricultural technique developed for three years(96-98) by demonstration at an model farm of environmental agriculture located in Bangsan-ri, Dagang-myun, Namwon-city. Rice growth of environmental farming plot, which supplied with oil cake and compost fermented with chicken manure was reduced compared to that of conventional farming plot, which supplied with chemical fertilizers. Weed control by using of Ampullarius insularus in the rice paddy field was effective compared to that of control, which treated with chemical herbicide twice. The yield of rice was decreased of 3-5% in both environmental and eclectic farming good compared to that of conventional farming plot. The water quality of environmental not and eclectic farming plot were good compared to conventional farming plot. Density of water flea was high in environmental and eclectic farming plot compared to that of the conventional farming plot.

      • 食用油脂類에 對한 免疫生物學的 硏究

        食用油脂는 健康維持에 必須的인 營養素이다. 그러나 食用油脂는 脂肪을 構成하는 脂肪酸의 種類와 攝取하는 量에 따라 健康을 保衛하는 必須的인 食品의 成分이 되기도 하고 有害한 影響을 끼칠 수도 있어서 脂質代謝와 關連된 疾病 뿐만아니라 老化를 促進하고, 乳房癌, 大腸癌等의 癌發生과도 關連된 것으로 밝혀져 있다. 本 硏究는 우리生活環境에서 食用으로 接할 수 있는 10種의 油脂에 對한 免疫生物學的인 活性에서 究明한 結果 對照群인 참기름食餌群에 比해 올리브유食用群은 全般的으로 體液性 및 細胞性免疫을 有意性있게 增加시켰으나, 大食細胞의 活性과 末梢循環白血球數는 有意性없는 減少를 보였고, 動物性 油脂인 牛脂食餌群과 植物性 油脂인 옥수수기름食餌群, 들기름食餌群, 米糠油食餌群, 大豆油食餌群, 菜種油食餌群 및 고추씨기름食餌群에서는 全般的으로 體液性 및 細胞性免疫, 大食細胞의 活性 및 末梢循環白血球數를 有意性있게 減少시켰다. This study was performed to investigate the effects of edible oils and fats on the immunobiological responses in ICR male mice. Ten groups of experimental diets, such as sesame oil diet, beef tallow diet, lard oil diet, olive oil diet, corn oil diet, perilla oil diet, rice polishing oil diet, soybeen oil diet, rape seed oil diet, and red pepper seed oil diet were fed adlibitum to the ICR male mice for 27 days. The results of this study were summarized as followings. 1. Both humoral and cellular immune responses, phagocyte activity, and number of leukocytes in sesame oil group were increased. However, the increasing rate of body weight and the weight retios of spleen and thymus to body were generally decreased. 2. Hemagglutination titer(HA) and rosette forming cell(RFC) of the beef tallow group and the lard group were significantly lower than the sesame oil group(P<0.01), but the lard group significantly increased Arthus reaction and delayed type hypersensitivity(DTH)(P<0.01). 3. Arthus reaction, DTH, and RFC of the olive oil group were significantly higher than the sesame oil group(P<0.01). 4. DTH of the corn oil group was significantly higher than the sesame oil group(P<0.01) whereas phagochyte activity was significantly lower(P<0.01). But no statistically significant differences were found between the two groups in the humoral immune response. 5. HA, PFC, DTH, RFC, phagocyte activity, and number of leukocytes of the perilla oil group and the rice polishing oil group were significantly lower than the sesame oil group(P<0.01). 6. Both humoral and cellular immune responses, phagocyte activity, and number of leukocytes of the soybeen oil group, the rape seed oil group, and the red pepper oil group were significantly lower than the sesame oil group(P<0.01).

      • 분리 핵을 이용한 Trichoderma koningii의 형질전환

        Trichoderma koningii의 영양요구성 돌연변이주인 CUT 121로부터 얻은 원형질체를 야생형인 ATCC 26113의 핵과 혼합하여 PEG 용액을 처리한 결과, 독립영양형의 형질전화체가 30% 이상의 빈도로 생성되었다. 이 독립영양형 군체로부터 얻은 분리체 중의 하나는 xylanase의 활성이 야생형보다 3배 가량 증진되었으며, 다른 세포외 다당류 분해효소능도 야생형과 유사한 수준을 나타내었다. 분리체의 DNA 함량, 인위적인 분리양상 및 동위효소 양상 등을 조사 분석한 결과, 독립영양형의 형질전환체는 실험에 사용된 두 핵간의 유전물질교환에 의하여 생성된 형질전환체로 판명되었다. 이상의 결과로부터 Trichoderma속 균류의 균주개량법으로, 핵전이법이 통상의 원형질체 융합법보다 효율적인 것으로 판명되었다. When protoplasts from auxotrophic mutant of Trichoderma koningii CUT121(Lys^-, Met^-) were mixed with isolated nuclei of wild type T. koningii ATCC 26113 and treated with PEG solution, protrophic colonies were produced with frequency of more than 30 percent. One of segregants from prototrophic colonies showed increased xylanase activity and other polysaccharide-hydrolyzing activities comparable to those of wild type strain. Through measurement of DNA contents, induced segregation, and analysis of isozyme patterns, it was revealed that the prototrophic colonies were transformants resulted from exchange of genetic materials between the two kinds of nuclei used. These results suggest that nuclei transfer technique is more efficient than conventional protoplast fusion technique for strain improvement of Trichoderma species.

      • Coacervation-상분리법으로 만든 Ethyl Cellulose 마이크로캅셀의 Glibenclamide 방출특성 및 수용성 첨가제의 영향

        Glibenclamide (GBC) is a well-known antidiabetic agent, which stimulates the secretion of endogenous insulin by pancreatic βcells. However, administration of GBC at a high dose can occasionally induced the fatal hypoglycemia, especially in the patients with impaired liver function. Therefore, in order to develop an oral sustained-release preparation which reduces the side effect, microcapsulation of GBC has been carried out in this experiment. Microcapsules of GBC were prepared by coacervation-phase separation method with ethylcellulose as a wall-forming material in cyclohexane, using polyisobutylene as a coacervation-inducing agent. Different amounts of hydrophilic additives. L-arginine(ARG) and polyethylene glycol 4000 (PEG), were added to the microcapsule wall, in order to alter the porosity of the wall and hence to enhance the release of the core material. The microcapsules prepared were examined for physical properties by scanning electron microscopy (SEM) and particle size analysis. The release of the poorly water-soluble GBC was found to be very slow from the microcapsules and strongly dependent on the core to wall ratio of the microcapsules, but it was accelerated considerably with increasing amount of PEG or ARG. In general, on the basis of dissolution kinetics, GBC releases from microcapsules were followed apparent first-order kinetics, while the dissolution data of GBC powder conformed to Hixon-Crowell's cube root law. Therefore, in conclusion, it might be possible to design the sustained-release formulation by the combination of both GBC powder and microcapsules of different core wall ratio with or without hydrophilic additives.

        Soil Microbial Communities and Growth of Sweet Potato(Ipomoea batatas L.) in Paddy and Upland Soils

        The soil microbial communities and growth of sweet potato are often affected by soil type. This study evaluated the variations in microbial communities of paddy and upland soils used for two sweet potato cultivars (Jinhongmi and Sinjami) cultivation by their fatty acid methyl ester (FAME). The pH, available P2O5, exchangeable K, Ca, and Mg of upland soil at the harvesting stage were significantly higher than those of paddy soil. The moisture content of paddy soil (28.5%) has increased by 6.7% as compared to upland soil (p < 0.001). The average microbial biomasses in paddy soil were approximately 2.8 times larger for arbuscular mycorrhizal fungi (AMF) and 1.4 - 1.6 times larger for fungi, total bacteria, Gram-positive bacteria, total FAMEs, Gram-negative bacteria, and actinomycetes. In addition, The total FAMEs, Gram-positive bacteria, fungi, total bacteria, and AMF in Jinhongmi cultivated soils were significantly larger than those in Sinjami cultivated soils. AMF communities in paddy soil showed significantly larger than that in upland soil (p < 0.001) indicating AMF are potentially responsible for the microbial community differentiation between paddy and upland field. The tuber yield of sweet potato was 28% higher, respectively, in the paddy soil and Sinjami treatment than in the upland soil and Jinhongmi treatment. It was confirmed that the paddy field was more effective on the productivity of sweet potato and soil microbial biomass.

      • Effects of Chiropractic Manipulative Therapy and Low Back Exercise on Lumbar Lodortic Angle, Gravity Line, ROM, and Pain Degree in University Students using a Commuter Bus

        The purpose of the present study was to investigate the effects of chiropractic manipulative therapy and low back exercise on lumbar lodortic angle, gravity line, ROM, and pain degree in university students using a commuter bus. In the present study, thirty participants who use a commuter bus and complain of low back pain were randomly divided into the chiropractic group and the exercise group. We provided chiropractic care for fifteen participants in the chiropractic group twice per week, and low back exercise for fifteen participants in the exercise group three times per week for 8 weeks. After 8 weeks, we compared the lumbar lodortic angle, gravity line, lumbar range of motions (ROMs), and pain degree scale between pre- and post-tests. In the present results, both chiropractic manipulative therapy and low back exercise significantly improved lumbar ROMs, and alleviated low back pain. From these results, it can be inferred that chiropractic manipulative therapy and low back exercise may be beneficial for university students with low back pain using a commuter bus.

        요석환자에서 유로시트라®의 생리학적 효과

        PURPOSE: To investigate the biochemical change in serum and 24-hour urine after therapy with Urocitra® in patients affected by urolithiasis, who had hypocitraturia alone or associated with other metabolic disorder.<br> MATERIALS AND METHODS: One hundred eighteen patients with evidence of 1 or more stone attacks within the last 3 years participated in the present study. They were 78 men and 40 women (6 to 78 years old, with a mean age of 47.01±12.95 years). All of the patients received 15 to 20㎖ of Urocitra®-solution or 5 g of Urocitra®-C powder, three or four times daily for 3 months. Before treatment, 24-hour urine and venous blood samples were obtained, while patients were maintained on a random diet, and analyzed for various stone risk factors. After 1 week, 1 month and 3 months of treatment, samples were again obtained and analyzed in the same manner. Thereafter, we compared the biochemical values before and after treatment.<br> RESULTS: In all three follow-up periods Urocitra® induced a significant increase in urinary citrate (p<0.001) level. Urinary potassium (p<0.001), pH (p<0.001) and total volume (p<0.05) also increased significantly after 1 and 3 months of therapy, as did urinary citrate excretion in patients with hypocitraturia and normocitraturia. Urocitra® did not alter calcium, sodium or phosphorus urinary excretion. There was no significant change of serum chemistry after administration.<br> CONCLUSIONS: Urocitra® was effective in increasing urinary pH and citrate. Furthermore, it was relatively free of side effects, except for minor gastrointestinal distress. Thus, our study provides physiological and clinical validation for the use of Urocitra® in patients affected by urolithiasis, who have hypocitraturia alone or associated with another metabolic disorder.

        전북 남원지역 육계에서 살모넬라증에 대한 역학적 조사

        This survey was performed to investigate avian salmonellosis from broiler farms in Namwon area for 4 years. The whole samples from farms were 541 and chicken samples were 294. Out of 294, 83 cases were identified with avian salmonellosis for 4 years by various serological and biochemical tests. There was no outbreak of pullorum disease, but fowl typhoid occurred in 36 farms. The outbreak of avian salmonellosis occurred continuously regardless of seasons throughout the year. Avian salmonellosis mostly happened within 10 days of age which were 52 cases(63%). The selection of susceptible antibiotics has changed from Quinoline group in 2000 to amoxacillin+clavulanic acid(AmC).
