        Mean-square Exponential Stability of Impulsive Stochastic Time-delay Systems with Delayed Impulse Effects

        This paper is concerned with the mean-square exponential stability problem for a class of impulsivestochastic systems with delayed impulses. The delays exhibit in both continuous subsystem and discrete subsystem. By constructing piecewise time-varying Lyapunov functions and Razumikhin technique, sufficient conditions arederived which guarantee the mean-square exponential stability for impulsive stochastic delay system. It is shownthat the obtained stability conditions depend both on the lower bound and the upper bound of impulsive intervals,and the stability of system is robust with regard to sufficiently small impulse input delays. Finally, two examplesare proposed to verify the efficiency of the proposed results.

        miR-1301/TRIAP1 Axis Participates in Epirubicin-Mediated Anti-Proliferation and Pro-Apoptosis in Osteosarcoma

        Purpose: Epirubicin is one of the most effective drugs against osteosarcoma. miR-1301 is involved in the occurrence and developmentof osteosarcoma. Whether miR-1301 is responsible for the chemosensitivity of osteosarcoma cells to epirubicin remainslargely unknown. Materials and Methods: U2OS and SAOS-2 cells were treated with various concentrations of epirubicin. Flow cytometry was employedto evaluate cell apoptotic rate. Cell proliferation was measured by Cell Counting Kit-8 assay. Western blot and quantitativereal-time polymerase chain reaction were utilized to detect the expressions of B-cell lymphoma-2 (Bcl-2), Bcl-2 assaciated X protein(Bax), cleaved-caspase-3, cleaved-poly (ADP-ribose) polymerases (PARP1), TP53-regulated inhibitor of apoptosis 1 (TRIAP1),and microRNA-1301 (miR-1301). The relationship between miR-1301 and TRIAP1 was determined by luciferase reporter assay. Results: Epirubicin inhibited proliferation in a dose-dependent manner, induced apoptosis, decreased the expression of Bcl-2,and increased the expressions of Bax, cleaved-caspase-3, and cleaved-PARP1 in osteosarcoma cells. miR-1301 was downregulatedin U2OS and SAOS-2 cells. Importantly, epirubicin significantly increased the levels of miR-1301. Overexpression of miR-1301suppressed proliferation and promoted apoptosis. Interestingly, those effects were enhanced by epirubicin. In contrast, miR-1301depletion attenuated the epirubicin-mediated anti-osteosarcoma effect. miR-1301 negatively regulated the expression of TRIAP1in U2OS and SAOS-2 cells. Furthermore, epirubicin inhibited the mRNA and protein levels of TRIAP1 by upregulating miR-1301levels. Epirubicin suppressed cell proliferation by downregulating TRIAP1. Conclusion: miR-1301 was implicated in the chemosensitivity of osteosarcoma to epirubicin by modulating TRIAP1.

      • Development of ligustrazine-loaded lipid emulsion: Formulation optimizatiom, characterization and biodistribution

        Ligustrazine is a traditional Chinese medicine used to treat various cardiovascular and neurovascular complications. However, this compound exhibits rapid first-pass metabolism, a short biological half-life, low stability and potential vascular irritation that restrict its use for long-term therapy. The use of a lipid emulsion as a carrier for intravenous administration of ligustrazine might provide sustained and prolonged release, thereby reducing the frequency of administration and improving patient compliance. The main purpose of our study was to develop a highly stable and sterile optimal formulation of a ligustrazine lipid emulsion (LLE) and to evaluate its pharmacokinetic behavior and tissue distribution in rats. The final optimal formulation consisted of soybean oil (12.0%), oleic acid (0.6%), lecithin (1.0%), poloxamer 188 (0.6%) and glycerol (2.25%). The average particle size, polydispersity index (PDI), zeta-potential and pH of the final product were 215.0±2.5 nm, 0.076±0.033, -40.4±5.3 mV and 7.25±0.05, respectively. The LLE was stable for at least three months at room temperature. In vitro drug release studies of the LLE suggested a sustained release profile, which was further confirmed by in vivo pharmacokinetic studies in rats. The area under the drug concentration-time curve from 0 h to 10 h (AUC(0-10h)) for LLE was increased by 1.6-fold compared with that of the commercially available ligustrazine injection (LI), suggesting enhanced bioavailability from the lipid-based emulsion. Furthermore, a tissue distribution study showed significant improvement in the distribution pattern of ligustrazine with a higher AUC(0-180 min) observed in all tissues for LLE than for LI. In conclusion, LLE, with excellent stability, improved pharmacokinetics and tissue distribution, demonstrates great potential for the delivery of ligustrazine for clinical applications.ⓒ2012 Elsevier B.V.All rights reserved.

        A Demand Side Management Strategy to Reduce the Impact of High Power Pulse Load Integration

        High pulsed power loads (HPPLs) are highly non-linear and can cause signifi cant stability and power quality issues in a microgrid. Developing alternative methods for controlling and analyzing the microgrid with multiple HPPLs will provide insight into tradeoff s that can be made during the design phase. In this paper, a demand side management strategy of HPPLs in a microgrid is formulated. Firstly, the HPPLs are divided into energy storage connected high power pulse loads (EHPPLs) and direct connected high power pulse loads (DHPPLs) according to the power use of the back-end loads, and the energy management models of both the EHPPLs and the DHPPLs are built. Then the demand side management strategy to minimize the impacts of running HPPLs is proposed. Finally, three typical cases are studied to verify the fl exibility and eff ectiveness of the proposed method. The simulation and experiment results indicate that the proposed strategy can eff ectively reduce the peak power demand of the HPPLs, thus reducing the impact on the microgrid

        Lipid Emulsion as a Drug Delivery System for Breviscapine: Formulation Development and Optimization

        In this study, we developed an optimized formulation of a breviscapine lipid emulsion (BLE) and evaluated the physicochemical properties and in vivo pharmacokinetics of BLE in rats. For the preparation of the lipid emulsion, soybean oil and oleic acid were used as the oil phase, lecithin and poloxamer 188 as surfactants and glycerol as co-surfactant. An optimized formulation consisting of soybean oil (10.0%), oleic acid (0.9%), lecithin (1.5%), poloxamer 188 (0.4%), and glycerol (2.25%) was selected. The results showed that the average particle size, polydispersity index, and zeta potential of the optimized formulation were 183.5 ± 5.5 nm, 0.098 ± 0.046, and −35.0 ± 2.5 mV, respectively. The BLE was stable for at least three month at room temperature. After a single intravenous dose of 4 mg/kg to rats, the AUC of scutellarin from the lipid emulsion was about 1.5-fold higher than that of the commercial product (breviscapine injection). In conclusion, the optimized formulation of BLE showed positive results over the commercial product in terms of the physicochemical properties and pharmacokinetics of BLE in rats.

        Identifying long non-coding RNAs and characterizing their functional roles in swine mammary gland from colostrogenesis to lactogenesis

        Objective: This study was conducted to identify the functional long non-coding RNAs (lncRNAs) for swine lactation by RNA-seq data of mammary gland. Methods: According to the RNA-seq data of swine mammary gland, we screened lncRNAs, performed differential expression analysis, and confirmed the functional lncRNAs for swine lactation by validation of genome wide association study (GWAS) signals, functional annotation and weighted gene co-expression network analysis (WGCNA). Results: We totally identified 286 differentially expressed (DE) lncRNAs in mammary gland at different stages from 14 days prior to (-) parturition to day 1 after (+) parturition, and the expressions of most of lncRNAs were strongly changed from day –2 to day +1. Further, the GWAS signals of sow milk ability trait were significantly enriched in DE lncRNAs. Functional annotation revealed that these DE lncRNAs were mainly involved in mammary gland and lactation developing, milk composition metabolism and colostrum function. By performing weighted WGCNA, we identified 7 out of 12 lncRNA-mRNA modules that were highly associated with the mammary gland at day –14, day –2, and day +1, in which, 35 lncRNAs and 319 mRNAs were involved. Conclusion: This study suggested that 18 lncRNAs and their 20 target genes were promising candidates for swine parturition and colostrum occurrence processes. Our research provided new insights into lncRNA profiles and their regulating mechanisms from colostrogenesis to lactogenesis in swine.         Endophytic Fungi from Dalbergia odorifera T. Chen Producing Naringenin Inhibit the Growth of Staphylococcus aureus by Interfering with Cell Membrane, DNA, and Protein

        This study focused on the antibacterial effects of the endophytic fungi producing naringenin from Dalbergia odorifera T. Chen against Staphylococcus aureus. The antibacterial activity was measured by the inhibition diameters, minimum inhibitory concentration (MIC), and minimum bactericidal concentration (MBC). The time-killing curve was also used to evaluate its antibacterial efficacy. The results of antibacterial activity determinations showed that endophytic fungi secondary metabolites can inhibit the growth of five pathogenic bacteria (S. aureus, Escherichia coli, Salmonella enteritidis, Pseudomonas aeruginosa, and Bacillus subtilis) and the most sensitive strain was S. aureus that had the MIC and MBC values of 0.13 and 0.50 mg/mL, respectively. The membrane permeability study was measured by a DNA leakage assay and electrical conductivity assay. Furthermore, the whole-cell protein lysates and DNA fragmentation assay was evaluated. The morphology of S. aureus treated with the endophytic fungi products was observed by scanning electron microscopy (SEM). The probable antibacterial mechanism of endophytic fungi secondary metabolites was the increased membrane permeability that leads to leaks of nucleic acids and proteins. SEM results further confirmed that the extracts can interfere with the integrity of S. aureus cell membrane and further inhibit the growth of bacteria, resulting in the death of bacteria. This study provides a new perspective for the antibacterial functions of endophytic fungi secondary metabolites for biomedical applications.

        SOC balance‑based decentralized control strategy for hybrid energy storage in integrated power systems

        The hybrid energy storage systems (HESSs) in vessel integrated power systems can support pulse load and improve system stability. However, the unbalanced SOC of different energy storage devices can cause over-charge and over-discharge which damages the energy storage devices and affects the stable operation of the entire system, especially when there are multiple groups of HESSs. Therefore, a decentralized control strategy for the HESSs in integrated power systems (IPSs) based on extended droop control combined with SOC balance control is proposed in this paper. In the proposed strategy, SOC recovery control is introduced to the virtual capacitance droop coefficient of the supercapacitor, and SOC equalization control is introduced to the virtual resistance droop coefficient of the lithium battery to adjust the output characteristics of the HESS according to the SOC. Then the frequency response characteristics are analyzed and the stability of the system is calculated. Finally, the hardware in loop simulation is conducted to verify the effectiveness of the proposed strategy. Comparisons show that the proposed strategy can compensate for the pulse load according to the responding characteristics of the HESS and achieve SOC balance between different energy storage devices under various working conditions.

        Failure Mechanisms and Modes of Tunnels in Monoclinic and Soft-Hard Interbedded Rocks: A Case Study

        The issue of large deformation mechanism in soft rock tunnels has puzzled tunnel scholars for decades. Previous studies have not evolved a clear and common understanding. Therefore, detailed on-site measurement, full investigation and statistical analysis have been conducted on the instability and failure of Muzhailing Tunnel since its construction, whose length is beyond 15 km. The study aims at systematically analyzing the failure mechanisms and modes of Muzhailing Tunnel in monoclinic and soft-hard interbedded rock strata. Study results show that the angle between strata strike and tunnel axis greatly determines the magnitude of deformation, the dip direction significantly controls the bias direction and maximum deformation direction, and the dip angle deeply affects the deformation form. The failure modes of surrounding rock mainly include four types: spalling and overturning failure, bending failure, shear slip failure and buckling failure. Large deformation characteristics are summarized from six aspects: failure form, groundwater, sensitivity to influencing factors, deformation degree, deformation speed and deformation duration. The instability modes of primary lining include in-plane (transverse) instability and out-plane (longitudinal) instability. Finally, the causes of large deformation are analyzed from geological, structural, engineering and human factors.
