        The Prolonging Effect of Natural Plant Extracts on the Storage Period of Postharvest Litchi

        This work was conducted to determine the preservation effects of essential oil to prolong the postharvest storage and freshness period of ‘Feizixiao’ litchi fruit. Essential oil was extracted from Lysimachi foenum-graecum Hance, Ginkgo biloba extract was obtained from the leaves, and anthocyanin was obtained from eggplant peel. Aspects of quality such as appearance, nutritional value, and enzyme activity were studied in postharvest litchi fruits. The results showed that the weight loss, decay rate, and malondialdehyde (MDA) content of litchi gradually increased in the four treatments and controls, while chroma L<SUP>*</SUP> and Vc, soluble solids, and titratable acid contents decreased with the extension of the storage time, while the activities of polyphenol oxidase (PPO) and peroxidase (POD) increased at first and then decreased slowly. Compared with room temperature preservation, the treatments with 0.5% essential oil extract, 0.5% ginkgo biloba extract, 0.5% anthocyanin extract, and 0.05% potassium sorbate reduced PPO and POD activity, membrane lipid peroxidation, decay, and weight loss, while providing better color protection and higher Vc, soluble solids, and titratable acid contents. The extracts delayed fruit browning, protected the nutrients and flavour quality of litchi pulp to a certain extent, and extended shelf life of postharvest litchi. Above all, the effect of 0.5% essential oil extract on the postharvest preservation of litchi was significantly better than other treatments. In addition, that of 0.5% ginkgo biloba extract was similar to that of 0.05% potassium sorbate but better than that of 0.5% anthocyanin extract. These results indicated that essential oil from Lysimachi foenum-graecum Hance prevented the decline of postharvest litchi quality, delayed browning and aging, and had the potential to be further developed as a natural fruit and vegetable preservative.

      • CCNG2 Suppressor Biological Effects on Thyroid Cancer Cell through Promotion of CDK2 Degradation

        This study aimed to analyze the expression and clinical significance of cyclin G2 (CCNG2) in thyroid carcinoma and the biological effects of CCNG2 overexpression in a cell line. Immunohistochemistry and Western blotting were used to analyze CCNG2 protein expression in 63 cases of thyroid cancer and normal tissues to allow the relationship with clinical factors to be assessed. CCNG2 lentiviral and empty vectors were transfected into the thyroid cancer K1 cell line. Reverse transcription-polymerase chain reaction (RT-PCR) and Western blotting were applied to detect the mRNA and protein levels of CCNG2. MTT assay and cell cycle were also conducted to assess the influence of up-regulated expression of CCNG2 on K1 cell biology. The level of CCNG2 protein expression was found to be significantly lower in thyroid cancer tissue than normal tissues (P<0.05). Western blot: The relative amount of CCNG2 protein in thyroid cancer tissue was respectively found to be significantly lower than in normal tissues (P<0.05), correlating with lymph node metastasis, clinic stage and histological grade (P<0.05), but not gender, age or tumor size (P>0.05). Loss of CCNG2 expression correlated significantly with poor overall survival time on Kaplan-Meier analysis (P<0.05). The results for biological functions showed that K1 cell transfected CCNG2 had a lower survival fraction, a greater percentage in the G0/G1 phases, and lower cyclin-dependent kinase 2 (CDK2) protein expression compared with K1 cells non-transfected with CCNG2 (P<0.05). CCNG2 expression decreased in thyroid cancer and correlated significantly lymph node metastasis, clinic stage, histological grade and poor overall survival, suggesting that CCNG2 may play important roles as a negative regulator in thyroid cancer K1 cells by promoting degradation of CDK2.

        Realgar bioleaching solution suppress ras excessive activation by increasing ROS in Caenorhabditis elegans

        Although realgar bioleaching solution (RBS) hasbeen proved to be a potential candidate for cancer therapy, themechanisms of RBS anticancer are still far from beingcompletely understood. Dosed with RBS in C. elegans, themultivulva phenotype resulting from oncogenic ras gain-offunctionwasinhibited in a dose dependentmanner. It could beabrogated by concurrent treatment C. elegans with RBS andthe radical scavengerDMSO. However, RBS could not induceDAF-16 nuclear translocation in TJ356 or the increase of HSP16.2 expression in CL2070, which both could be arousedvisible GFP fluorescent variation to represent for oxidativestress generation. Treatment C. elegans with superoxide aniongenerator paraquat, similar results were also obtained. Ourresults indicated that RBS suppress excessive activated ras byincreasing reactive oxygen species (ROS) in C. elegans. Secondly, ROS induced by RBS significantly accumulated ona higher level inC. elegans with amutational ras than that withwild ras, thus leading to oxidative stress on ras gain-of-functionbackground rather than on normal ras context. Our resultsfirstly demonstrated that using C. elegans as amodel organismfor evaluating prooxidant drug candidates for cancer therapy.

      • Silencing of COX-2 by RNAi Modulates Epithelial-Mesenchymal Transition in Breast Cancer Cells Partially Dependent on the PGE<sub>2</sub> Cascade

        In order to prove whether downregulation of COX-2 (Cyclooxygenase-2) could modulate the epithelial-mesenchymal transition (EMT) of breast cancer, celecoxib and siRNA were respectively used to inhibit COX-2 function and expression in MDA-MB-231 cells. The EMT reversal effect in the RNAi treated group was better than that of the celecoxib group while there were no obvious differences in the medium $PGE_2$ levels between the two groups. The results show that COX-2 pathways may contribute considerably to EMT of breast cancer cells, partially dependent on the PGE2 cascade. Akt2, ZEB2 and Snail were measured to clarify the underlying mechanisms of COX-2 on EMT; COX-2 may modulate EMT of breast cancer by regulating these factors. This finding may be helpful to elucidate the mechanisms of selective COX-2 inhibitor action in EMT modulation in breast cancer.

      • Preventive and Therapeutic Effects of Quercetin on Experimental Radiation Induced Lung Injury in Mice

        Objective: To investigate the protective effect of quercetin on radiation induced lung injury (RILI) and related mechanisms. Materials and Methods: Mice treated with radiation and/or quercetin were sacrificed at 1-8 weeks after irradiation under anesthesia. Lung tissues were collected for histological examination. Immunohistochemistry (IHC) and Western blotting were performed to detect the protein expression of nuclear factor-${\kappa}B$ ($NF-{\kappa}B$) and Mitogen-activated protein kinases (MAPK) pathway. Results: Hematoxylin and eosin (HE) staining showed that radiation controls displayed more severe lung damage than quercetin groups, either high or low dose. Results of IHC and Western blotting demonstrated the expression level of $NF-{\kappa}B$ to be decreased and that of an inhibitor of $NF-{\kappa}B$ ($I{\kappa}b-{\alpha}$) to be increased by the quercetin intervention compared with the radiation control group. Numbers of JNK/SAPK, p38 and p44/p42 positive inflammatory cells were decreased in the radiation+quercetin injection group (P<0.05). Conclusions: Quercetin may play a radio-protective role in mice lung via suppression of $NF-{\kappa}B$ and MAPK pathways.

        Photo-induced antifouling polyvinylidene fluoride ultrafiltration membrane driven by weak visible light

        To enhance the performance of ultrafiltration membranes, a polyvinylidenefluoride (PVDF) blendingmembrane with photo-induced antifouling performance was fabricated. La3+- and Yb3+-doped titaniumdioxide-reduced graphene oxide (TiO2-RGO) composites were initially prepared by adsorption-phasesynthesis coupled with solvothermal reduction. The doped TiO2-RGO composites were then blendedwith the PVDF materials. The results showed that adding well-distributed, doped, and hydrophilic TiO2-RGO improved the pore structures and surface polarity of the blending membranes, thus improving theirpermeation and antifouling performance. After addition of doped TiO2-RGO with photocatalytic activity,the antifouling performance of the resulting blending membranes increased under weak visible-lightirradiation. The agglomerated doped TiO2-RGO in the polymeric matrix reduced the hydrophilicity andporosity of the resulting membranes, thereby decreasing their water permeability and antifoulingperformance. Solvothermal reduction through ethylene glycol also improved the photocatalytic activityof Yb-doped TiO2-RGO and enhanced the affinity between doped TiO2-RGO and PVDF. These two effectssimultaneously increased the water permeability and photo-induced antifouling performance of theblending membranes. Theflux recovery rate of the blending membranes containing Yb-doped TiO2-RGOtreated by ethylene glycol exceeded 69% after illumination. In particular, theflux recovery rate ofmembranes reached about 96% after illumination coupling with washing.

        Optimized BD-ZF Precoder for Multiuser MIMO-VFDM Cognitive Transmission

        In this paper, we study an optimized block-diagonal zero-forcing (BD-ZF) precoder in a two-tiered cognitive network consisting of a macro cell (MC) and a small cell (SC). By exploiting multiuser multiple-input and multiple-output Vandermonde-subspace frequency-division multiplexing (VFDM) transmission, a cognitive SC can coexist with an MC. We first devise a cross-tier precoder based on the idea of VFDM to cancel the interference from the SC to the MC. Then, we propose an optimized BD-ZF intra-tier precoder (ITP) to suppress multiuser interference and maximize the throughput in the SC. In the case where the dimension of a provided null space is larger than that required by the BD-ZF ITP, the optimized BD-ZF ITP can collect all limited channel gain by optimizing rotating and selecting matrices. Otherwise, the optimized BD-ZF ITP is validated to be equivalent to the conventional BD-ZF ITP in terms of throughput. Numerical results are presented to demonstrate the throughput improvement of the proposed optimized BD-ZF ITP and to discover the impact of imperfect channel state information.
