韓國人에서 血友病 B 遺傳子(血液凝固因子 Ⅸ)의 構造 및 制限酵素 Dde Ⅰ, MnI Ⅰ 切片길이 多形性에 關한 分子 遺傳學的 考察

Hemophilia B, a human chromosome X-linked recessive disease, is a bleeding disorder resulting from defect or abnormality in blood coagulation factor IX. DNA-based prenatal diagnosis or carrier detection for hemophilia B in Korean has been developed by analyzing restriction fragment length polymorphisms(RFLPs). Two polymorphisms( Dde I and Mnl I) were investigated as follow. Genomic DNAs were extracted from blood of 50 females at the age of twenty. The primers were chemically synthesized by the method of phosphoamidite. Mnl I primers were derived from exon 6, while Dde I primers, from the flanking sequences of intron 1 of the factor IX gene. Genomic DNAs were amplified with Mnl I primers to generate 405 nts long fragments in all cases. They were digested with Mnl I to analyze the polymorphic site on agarose gel. No MnlI polymorphic site was found in all cases. Also, the genomic DNAs were amplified with Dde I primers to generate ca. 320 nts long fragments in all cases. These results are not correlated with those obtained from the Caucasian. It suggests that Korean could exhibit different patterns of Dde I and Mnl I polymorphisms in the gene for blood coagulation factor IX. Direct sequencing of the polymorpic sites will confirm the above sugestion.

누에의 行動半徑에 對한 系統間 差異에 관한 硏究

우리나라 장려 잠품종의 원종 중에서 14품종을 공시하여 주에의 행동반경에 대한 차이와 유전력 및 실용형질과의 관련성을 분석하였다. 행동반응은 유충의 각 시기에 따라서 품종간 차이가 있었으며 대체로 일본계통이 중국 계통에 비하여 민감한 경향이었고 각 품종 공히 행동반응이 가장 민감한 시기는 숙잡기였으며 다음은 개미누에였고 2령부토 5령까지는 상당히 둔한 경향이였다. 행동반경의 추정된 유전력은 유충의 시기별로 차이가 있었으며 h²(%)=52,121~88,411의 범위였으며, 행동발경과 타협질과의 관계에 있어서 행동반경과 생견 1 ㅣ 과수 및 행동반경과 최청부터 발아까지의 기간 사이에서는 "正"의 상관, 행동반경과 고치무게의 시이에서는 "負"의 상관이 인정되었다. The strain differences in larval walking behavior and their heredity in 14 varieties fo silk worm, Bombyx mori, has been investigate. The strain difference of walking response have been responsed in each larval stage and the bahavioral response of Japanse races was more sensitive than that of Chinese races, mostly. During matured silk worm larval stage the walking response was the highest and the next was the larvae hatched newly. Also very low sensitivity appeared from 2nd instar to 5th instar. Heritability of larvar walking distance were different depend on the larval instar the range of heritabilites(%) was 52.121-88.411. There was positive corelation between walking distance and period from incubation to emergence, but negative co relation between walking distance and single cocoon weight.

한국인에서 혈액응고인자 Ⅷ 유전자의 제한효소 절편길이 다형성에 관한 연구

Hemophilia A, an X chromosome-linked bleeding disorder affecting 1 in 5,000 males worldwide, due to the defect of blood coagulation factor Ⅷ. The wide range of clinical severity exhibited by haemophiliacs plus the hight incidence of sporadic cases suggest that hemophilia A be caused heterogeneous mutation of the gene. Restriction fragment length polymorphism(RFLP) is being used for prenatal diagnosis or carrier detection of the genetic disorder, However, the polymorphisms exhibits heterogeniety among races indicating that data from a people may not be applied to others. Two polymorphic sites(Bcl Ⅰ and Hind Ⅲ) of the factor Ⅷ gene were examined in Korean employing polymerase chain reaction(PCR). The PCR product of Bcl Ⅰ restriction was 948 nucleotides(nts) long, while that of Hind Ⅲ was 730 nts long. The former(Bcl Ⅰ) can generate three fragments(90nts, 100nts, and 480nts from 5' to 3' direction) with the polymorphic site, or two fragments(190nts and 480nts from 5' to 3' direction) wihout the polymorphic site. The latter(Hind Ⅲ) can generate three fragments(148nts, 286nts, and 514nts from 5' to 3' direction) with the polymorphic site, or two fragments(434nts and 514nts from 5' to 3' direction) without the polymorphic site. The heterozygote frequency calculated from the allele frequencies(0.754/0.246) of the Bcl Ⅰ (intron 18) polymorphism was 37.1%. The heterozygote frequency calculated from the allele frequencies(0.807/0.193) of the Hind Ⅲ polymorphism(intron 19) was 31.2%. Thus, the two intragenic polymorphisms predicted to be informative was 56.7% in these studies.

Expressed sequence tag(EST) has a good value to discover a new gene or to study its structure. Some 20 ESTs were generated to obtain new genetic resources of chinese cabbage(Brassian campestris L. var. pekinensis Makino). Poly A+ RNAs were isolated from 10-day-old seedlings grown at 25℃ under the day light. cDNA gene bank was constructed using λ ZAP /cDNA synthesis /Giga Pack Gold Packing kit. About a million clones were able to obtain from the library. All the clones examined so far had insert DNAs. Nucleotide compositions of randomly selected clones were determined by the Sanger mthod. The DNA sequences were compared with those deposited in the GenePept and GenBank database to figure out nucleotide homologies. Two ESTs showed significant similarities to the enlisted sequences. They are chloroplast GADPH subunit of Arabidopsis thaliana and carbonic anhydrase of A. thaliana. The full DNA sequences of the two clones are being determined. The cDNA gene bank constructed. in this experiment will being used to isolate more genes induced by the light in the plant.

In Korea, careinoma of the cervix is the most common malignancy in women annually affecting 25 per every 100,000 female. The widespread use of the Papanicolaou smear has resulted in earlier detection of the tumor and a reduction in the number of patients with advanced disease, but the incidence of cervix. carcinoma is still the fifth most common malignancy in the world. Cervical carcinoma spreads to the small bowel primarily by direct extension from involved lymph nodes, most commonly from peraaortic or mesenteric nodes to the serosa of the bowel. But, the involvement of small bowel by cervical carcinoma is so rare, and shows a maximum incidence of small bowel involvement of less than 1.5%. We report a patient with squamous cell carcinoma of the cervix metastases to the second portion of the duodenum in whom the diagnosis was made by endoscopic biopsy of the lesion.

Study on the Reproductive Function in Transgenic Pig Harboring Human Erythropoietin (hEPO) Gene

Our previous study showed that transgenic (TG) pigs harboring human EPO (hEPO) gene have been shown to have reproductive disorders, including low pregnancy rates, irregular estrus cycle and low little size. To investigate these reasons, we assessed estrus behavior (standing response) and plasma $17{\beta}$-estradiol ($E_2$) level, which partly reflect reproductive function, during the estrus cycles after synchronization and superovulation by hormone treatments. Then, we analysed blood composition and expression of hEPO gene in TG pigs. Pigs were injected with PG600. After 10 days, pigs were fed with Regumate porcine for 6 days. Blood samples were collected from jugular vein. Analysis of blood composition and $E_2$ level were measured by Hemavet 950 and $E_2$ ELISA kit, respectively. And, the expression of hEPO gene in reproductive organs was quantitated by real-time RT-PCR. The percentage of estrus behavior in TG was significantly decreased. Hematocrit (HCT), hemoglobin (Hb) concentration and red blood cell (RBC) number were significantly higher in TG than wild type (WT). On the other hand, high expression of hEPO gene in TG was observed in the mammary gland as well as in the uterus. Moreover, plasma $E_2$ level was significantly higher in TG than WT. These results suggest that nonspecific expression of hEPO gene in the other organs of TG may affect blood composition and plasma $E_2$ level, thereby causing reproductive disorders.

Effect of IRES Controlled Reporter Gene on Screening and Production of Recombinant Human EPO Proteins from Cultured CHO Cells

This study was conducted to examine the effect of IRES controlled reporter gene on screening and production of recombinant human erythropoietin (EPO) proteins from cultured CHO cells. The cDNA was cloned for EPO from human liver cDNA Using site-directed mutagenesis, we generated recombinant human EPO (rhEPO) with two additional N-glycosylations (Novel erythropoiesis-stimulating protein: NESP). Wild type hEPO and NESP were cloned into expression vectors with GFP reporter gene under regulatory control of CMV promoter and IRES so that the vectors could express both rhEPO and GFP. The expression vectors were transfected to cultured CHO-K1 cells. Under microscopy, expression of GFP was visible. Using supernatant of the culture, ELISA assay, immunocytochemistry and in vitro assay using EPO dependant cell line were performed to estimate biological activity to compare the production characteristics (secretion levels, etc.) between rhEPO and NESP. The activity of NESP protein, obtained by mutagenesis, was described and compared with its rhEPO counterpart produced under same conditions. Although NESP had less secretion level in CHO cell line, the biological activity of NESP was greater than that of rhEPO. These results are consistent with previous researches. We also demonstrated that rhEPO and GFP proteins expressed simultaneously from transfected CHO cell line. Therefore we conclude that use of GFP reporter gene under IRES control could be used to screen and produce rhEPO in cultured CHO cells.

태권도장의 관계마케팅 요인이 고객만족, 고객신뢰 및 고객충성도에 미치는 영향

본 연구는 태권도장의 관계마케팅 요인이 고객만족, 고객신뢰 및 고객충성도에 미치는 영향을 실증적으로 규명하는데 목적이 있다. 본 연구의 대상은 2009년 현재 경기도의 수원, 용인, 오산, 안성 등 4개 지역을 중심으로 각 1곳씩 총 4개 도장을 대상으로 편의표본추출법을 이용 279명을 표집하였다. 본 연구의 설문지 신뢰도를 검사한 결과 Cronbach`s α는 .608~.880으로 나타나, 내적일관성이 비교적 높음을 알 수 있다. 본 연구에서 사용한 통계기법은 빈도분석, 상관분석, 단순회귀분석 등이다. 이상과 같은 연구방법 및 자료분석 결과를 기초로 하여 본 연구에서 도출된 결과는 다음과 같다. 첫째, 관계마케팅 요인 중 고객만족의 하위요인인 경제적 만족에는 고객관계지향, 커뮤니케이션, 물리적 시설 요인이 영향을 미쳤으며, 비경제적 만족에는 유대관계, 커뮤니케이션, 전문성, 물리적 시설 요인 등이 영향을 미치는 것으로 나타났다. 둘째, 관계마케팅 요인 중 고객신뢰는 고객관계지향, 유대관계, 전문성, 물리적 시설 요인 등이 영향을 미치는 것으로 나타났다. 셋째, 관계마케팅 요인 중 고객충성도의 하위요인인 태도적충성에는 유대관계, 커뮤니케이션, 물리적 시설 요인 등이 영향을 미쳤으며, 행동적충성에는 고객관계지향, 커뮤니케이션, 전문성 요인 등이 영향을 미치는 것으로 나타났다. 넷째, 고객만족 요인인 경제적 만족, 비경제적 만족 모두 고객신뢰에 영향을 미치는 것으로 나타났다. 다섯째, 고객만족 요인인 경제적 만족과 비경제적 만족 모두 고객충성도의 하위요인인 태도적충성과 행동적 충성 모두에 영향을 미치는 것으로 나타났다. 여섯째, 고객신뢰는 고객충성도의 하위요인인 태도적 충성과 행동적 충성 모두에 영향을 미치는 것으로 나타났다. This research aims at identifying the effect of relationship marketing factors of Taekwondo gym on customer satisfaction, customer reliability, and customer loyalty. The research was conducted targeting total four Taekwondo gyms located in Suwon, Yongin, Osan, and Anseong in Gyeonggi-do as of 2009. And 279 subjects were sampled using Convenience Sampling. The statistical techniques used in this research were Frequency Analysis, Correlation Analysis, and Simple Regression Analysis, etc.. Based on above research methods and data analysis results, the conclusions drawn therefrom are as follows: First, customer relationship orientation, communication and physical factors of facility factors affected economic satisfaction which was sub factor of customer satisfaction among relationship marketing factors. And the factors in relation to ties with others, communication, expertise, physical factors of facility, etc. had influences on non-economic satisfaction. Second, customer relationship orientation, ties with others, expertise, and physical factors of facility, etc. had impacts on customer reliability among relationship marketing factors. Third, ties with others, communication, and physical factors of facility factors affected attitude loyalty which was sub factor of customer loyalty among relationship marketing factors, and that customer relationship orientation, communication, expertise, etc. had influences on behavioral loyalty. Fourth, economic satisfaction and non-economic satisfaction which were customer satisfaction factors all affected customer reliability. Fifth, both economic and non-economic satisfactions which were sub factors of customer satisfaction had impacts on both attitudinal and behavioral loyalty. Sixth, customer reliability affected both attitudinal and behavioral loyalty which were sub factors of customer loyalty.

Tea bag 포장한 멸치 복합 분말의 가공 및 그 추출물의 정미성분

This study was carried out to prepare anchovy based powder for instant soup packed in tea bag which can be used handily as a extractives, and to determine the taste compounds of extractives of anchovy based powder for instant soup. The anchovy based powder for instant soup was made by adding of 72% in the redried anchovy, 14% in the sea tangle, 7% in the mushroom and 7% in the katsuobushi to the total mixtures. And the anchovy based powder for instant soup was packed in tea bag. The desirable extraction time are 5 min in package in tea bag with air permeability 100 ㎥/㎡/min and over 20 min in package in tea bag with air permeability 65 ㎥/㎡/min, respectively. Judging from the result of extraction rate of total nitrogen, color and sensory evaluation in extractives of anchovy based powder for instant soup extracted by optimal extraction time, the quality in extractives of instant soup packed in tea bag with air permeability 100 ㎥/㎡/min was superior to those of instant soup unpacked in tea bag and of instant soup packed in tea bag of air permeability 65 ㎥/㎡/min. The principal taste compounds of extractives of anchovy based powder for instant soup packed in tea bag with air permeability 100 ㎥/㎡/min were IMP (9.26 ㎎/100 ㎖ in 5% solution) and free amino acids (24.30 ㎎/100 ㎖ in 5% solution) such as histidine, proline, lysine and taurine. Total creatinine, betaine and TMAO were seemed to act an auxiliary role in taste of extractives of anchovy based powder for instant soup packed in tea bag with air permeability 100 ㎥/㎡/min.
