        The city's countryside is adjusting to change by urbanisation. As it, the recent phenomena in this area decline in rural landuse and damage of landscape which has local characteristics. But, no study think together landuse and landscape at present in this area. The aim of this study was to clarify causes and problems of change by investigating intersection of environments and causes, processes and results of landuse and landscape in city's countryside. The sites are 2 Up and 2 Myon belong to Yongin Gun closed by Seoul City. These site are rapidly changed and almost intermediate between city and rural at present. As methods, I exeminated not only various statistical data but also unstatistical data. This case study revealed the diversity of change in city's countryside. 1) The change of landuse is clarified by Local Main commercial district is Yongin Up, Development of Traffic and industry is Kihung Up, Bed-town closed by city is Suji Myon and Recreation development is Pogok Myon. Each landuse divided by landscape types as local diversity in several Ri. Also each case seperatied by its special shape from each Ri and have causes and problems. 2) From these results did I point out problems and make a proposal about national landuse plan and landscape management in city's countryside. It stressed the need of a long-ranged policy of development in countryside considering local characteristics and residential environments. This paper was the start of case study about landuse and landscape in city's countryside.

      • 人乳의 일반 성분 및 Vitamin 함량에 관한 연구

        Human milk samples taken from 78 healthy lactating women in Suweon area were analyzed to find out the changes of the general and vitamin contents during 5 different lactation period. The results were as follows; 1. The average contents of protein, fat, lactose, ash, and moisture in human milk were 1.25±0.05, 3.56±0.45, 6.82±0.58, 0.21±0.08, and 87.88±0.98%, respectively. 2. The protein and ash were significantly decreased from 1.86 and 0.26% to 1.18 and 0.18%, but the contents of fat, lactose, and total solid were gradually increased from 2.95, 6.18, and 11.87% to 3.72, 7.02, and 12.25% during lactation period, respectively. 3. Protein, fat, and lactose showed positive correlation with total solid. Ash showed positive correlation with protein but it showed negative correlation with other constituents. 4. Vitamin contents of colostrum were 270 IU/100g of vit. A, trace of vit. B_1, 0.03mg/l00g of vit. B_2, 8.5㎍/l00g of vit. B_6, 0.06㎍/100g of vit. B_12, 3.4mg/100g of vit. C, 0.7mg/l00g of vit. E, and 0.2㎍/100g of vit. K_1. vit. A, β-carotene. vit. E decreased, but vit. C and B groups increased during lactation period.

      • Bifidobacterium angulatum KCTC3236의 α-galactosidase에 의한 Oligo당 생성의 최적조건

        Bifidobacterium angulatum KCTC3236 showed the highest α-galactosidase activity among the tested strains of Bifidobacteria( data not shown). Optimum culture conditions for the maximum production of α-galactosidase were as follows. The culture media was MRS broth containing 1% galactose as a carbon source and its initial pH was 7.0. The incubation temperature and time were 37℃, 21 hours respectively. Melibiose have been used as substrate. The formation of oligosaccharides was maximum at 40℃ when the substrate concentration was 30%. And the unit of α-galactosidase was 40 unit. Finally, we could observe the maximum production of oligosaccharides after 14 hours' enzyme reaction.

      • 용인 자연농원 장미원의 이용후 평가에 관한 연구

        The POE (Post Occupancy Evaluation) was carried out to provide basic behavioral knowledge for its planning and management of the Rose garden at Yong-In Farmland. Through its users a total of 1545 questionnaire were completed. They were designed by four sets of variables, such as behavoir pattern, motivation for visting, post-use evaluation, and users demographic variables. The findings are as follows: Firstly, descriptive statistics were employed to analyze behavoir pattems and users' demographic varialbes. As a result, 86% of visitors lived in Kyoung-Ki Do and Seoul : most of the vistiors is in their twenties and thirties : most of visitors were accompanied by their families, relatives and lovers : 66% of vistiors came by cars to Yong-In Farmland : 47% of visitors stayed for 1-2 hours in average. Secondly, factor analysis was used in order to group satisfaction factors. Five factors are : 'manegement of the garden', 'rose appearance', 'design and aesthetics', 'lighting and sound', and 'theme display'. Thirdly, regression analysis was used to examine the sources of satisfaction. The results of regression analysis shows that 42% of total variances was explained by ten variables. The most effective variable is 'the aspect of rose condition and appearance'. The overall evaluation for the new rose garden was 'satisfied' though the level of the 'satisfied' varies according to their ages. Twenties and thirties prefer that image of fantastic European style which was frequently showed on T.V or movies. Forties and fifties like more abudant scenery of roses of an Europenan style garden but unsatisfied with shortage of roses.

      • Thermus caldophilus GK24로부터 내열성 alkaline phosphatase의 특성

        Thermostable alkaline phosphatase was found from Thermus caldophilus GK24, extreme thermophile. T caldophilus GK24 alkaline phosphatase was found to be inducible. When starved of inorganic phosphate, T caldophilus GK24 produces the enzyme alkaline phosphatase. The addition of inorganic phosphate to growth medium had a repressive effect on enzyme synthesis. T caldophilus GK24 was tested for the production of alkaline phosphatase by the addition of various concentrations of sodium glutamate, bactotryptone, glucose and yeast extract to basal salts. Sodium glutamate was found to be effective for the alkaline phosphatase induction. The enzyme was stable in the pH range of 7-12. The optimum temperature, pH of the enzyme were 75-85℃ and 11.0-11.5, respectively.

      • 韓國人에서 血友病 B 遺傳子(血液凝固因子 Ⅸ)의 構造 및 制限酵素 Dde Ⅰ, MnI Ⅰ 切片길이 多形性에 關한 分子 遺傳學的 考察

        Hemophilia B, a human chromosome X-linked recessive disease, is a bleeding disorder resulting from defect or abnormality in blood coagulation factor IX. DNA-based prenatal diagnosis or carrier detection for hemophilia B in Korean has been developed by analyzing restriction fragment length polymorphisms(RFLPs). Two polymorphisms( Dde I and Mnl I) were investigated as follow. Genomic DNAs were extracted from blood of 50 females at the age of twenty. The primers were chemically synthesized by the method of phosphoamidite. Mnl I primers were derived from exon 6, while Dde I primers, from the flanking sequences of intron 1 of the factor IX gene. Genomic DNAs were amplified with Mnl I primers to generate 405 nts long fragments in all cases. They were digested with Mnl I to analyze the polymorphic site on agarose gel. No MnlI polymorphic site was found in all cases. Also, the genomic DNAs were amplified with Dde I primers to generate ca. 320 nts long fragments in all cases. These results are not correlated with those obtained from the Caucasian. It suggests that Korean could exhibit different patterns of Dde I and Mnl I polymorphisms in the gene for blood coagulation factor IX. Direct sequencing of the polymorpic sites will confirm the above sugestion.

      • 도시 하수오니에 의한 고분자 물질의 생분해도 측정

        Factors influencing the biodegradability of polymer materials by municipal sewage sludge were studied using starch, polyethylene glycol(PEG) and polyvinyl alcohol(PVA). The degree of biodegradability was determined by measuring evolved carbon dioxide as a function of time, when the polymer material is exposed to activated sewage sludge. The biodegradability of starch reached around 70% and 40% after 30day incubation in summer and autumn, respectively. This is caused by the activity of microorganism and incubation temperature. The biodegradability of PEG 8000 and PVA was 1/7 and 1/14 of starch, respectively. Biodegradation of PEG 8000 after 30 day incubation in summer was slightly higher than that in spring while lag phase was significantly longer in spring. This shows that the activity of microorganism as well as incubation temperature is important for biodegradation.

      • 한국인에서 혈액응고인자 Ⅷ 유전자의 제한효소 절편길이 다형성에 관한 연구

        Hemophilia A, an X chromosome-linked bleeding disorder affecting 1 in 5,000 males worldwide, due to the defect of blood coagulation factor Ⅷ. The wide range of clinical severity exhibited by haemophiliacs plus the hight incidence of sporadic cases suggest that hemophilia A be caused heterogeneous mutation of the gene. Restriction fragment length polymorphism(RFLP) is being used for prenatal diagnosis or carrier detection of the genetic disorder, However, the polymorphisms exhibits heterogeniety among races indicating that data from a people may not be applied to others. Two polymorphic sites(Bcl Ⅰ and Hind Ⅲ) of the factor Ⅷ gene were examined in Korean employing polymerase chain reaction(PCR). The PCR product of Bcl Ⅰ restriction was 948 nucleotides(nts) long, while that of Hind Ⅲ was 730 nts long. The former(Bcl Ⅰ) can generate three fragments(90nts, 100nts, and 480nts from 5' to 3' direction) with the polymorphic site, or two fragments(190nts and 480nts from 5' to 3' direction) wihout the polymorphic site. The latter(Hind Ⅲ) can generate three fragments(148nts, 286nts, and 514nts from 5' to 3' direction) with the polymorphic site, or two fragments(434nts and 514nts from 5' to 3' direction) without the polymorphic site. The heterozygote frequency calculated from the allele frequencies(0.754/0.246) of the Bcl Ⅰ (intron 18) polymorphism was 37.1%. The heterozygote frequency calculated from the allele frequencies(0.807/0.193) of the Hind Ⅲ polymorphism(intron 19) was 31.2%. Thus, the two intragenic polymorphisms predicted to be informative was 56.7% in these studies.

        F9 murine embryonal carcinoma cells expresed gene for tissue plasminogen activator(tPA), upon differentiation in vitro induced by retinoic acid(5×10 exp(-6)M/1). A recombinant plasmid(pBTPA) capable of tPA sense and antisense RNAs was constructed, and used for the RNA protection assay for tPA gene transcript in the differentiated F9 cells.
