      • 알쯔하이머형 치매와 혈관성 치매환자에서의 ALDH2와 APOE 유전자의 다형성에 관한 연구

        연구목적: 본 연구는 국내 치매환자들을 대상으로 치매발병의 위험요소가 될 수있는 ALDH2와 APOE 유전자형이 어떤 양상으로 나타나는지 알아보기 위하여 시행되었다. 방 법: 충남 부여군에 위치한 부여노인병원에 입원중인 65세 이상의 알쯔하이머형 치매와 혈관성 또는 혼재성 치매환자 59명(남:19, 녀:40)을 대상으로 ALDH2와 APOE 유전자형을 제한효소법에 의하여 조사하였고 치매유형에 따른 유전자형과 대립유전자의 빈도를 비교하였다. 결 과: 1) 알쯔하이머형 치매환자군에서 혈관성 또는 혼재성 치매환자군보다 ALDH2*(-/2)유전자형의 빈도가 높았으나 통계학적으로 의미있는 차이는 없었다. 2) 알쯔하이머형 치매화자군에서 혈관성 또는 혼재성 치매환자군보다 APOE*(-/ε4) 유전자형의 빈도가 높았으나 통계학적으로 의미있는 차이는 없었다. 3) APOE*ε4와 ALDH2*2를 알쯔하이머병의 균등한 위험대립유전자로 가정하고 각 유전자형의 대립유전자의 수를 점수화하여 각 치매환자군의 위험유전자 전체점수를 비교한 결과 알쯔하이머형 치매환자군에서 점수가 더 높게 나왔으나 통계학적으로 의미있는 차이는 없었다. 결 론: 알쯔하이머형 치매환자군에서 혈관성 또는 혼재성치매치매환자군보다 ALDH2*2와 APOE*ε4 대립유전자의 빈도가 높게 나왔으나 통계학적으로 의미있는 차이는 없었다. A study on gene polymorphism of ALDH2 and APOE in patients with alzheimer's disease and vascular or mixed dementia. Objective : This study was designed to investigate gene polymorphism of ALDH2 and APOE, reported the risk factor of Alzheimer's disease, in korean dementia patients. Method : 59 Patients(male:19, female:40) with dementia, aged 65 and older in Buyeo geriatric hospital located in Buyeo-gun, chungnam, were finally included in this study. gene polymorphism of ALDH2 and APOE was typed with polymerase chain reaction in patients with Alzheimer's desease and vascular or mixed dementia. Results : 1) There was no difference in allelic frequency of ALDH2 gene polymorphism between patients with Alzheimer's disease and vascular or mixed dementia. 2) There was no difference in allelic frequency of APOE gene polymorphism between patients with Alzheimer's disease and vascular or mixed dementia. 3) Supposing APOE*ε4 and ALDH2*2 were equal risk allele of Alzheimer's disease, the sum of score by counting each risk allele was higher in patients with Alzheimer's disease than vascular mixed dementia, however there was no significant difference. Conclusion : The genotype frequency of ALDH2*(-/2) and APOE*(-/ε4) was higher in patients with Alsheimer's disease than vascular or mixed dementia, however there was no significant difference in allelic frequency of gene polymorphism of ALDH2 and APOE between patients with Alzheimer's disease and vascular or mixed dementia.

        Idiopathic hypereosinophilic syndrome is characterized by marked elevation of eosinophil count ( >1,500/㎣) in peripheral blood without explaining etiology. It shows tissue involvement in many organs such as liver, G-I tract, myocar dium, lung, skin, kidney and nerve. Some case of hypereosinophilic syndrome with hepatic involvement have been reported, but not much. We have recently experienced a case with idiopathic hypereosinophilic syndrome of a 40 year old man who presented with RUQ pain. By abdominal CT, ill- defined subcapsular hematoma was detected. Liver biopsy showed eosinophilic infiltration within the portal tract and sinusoid. Drain procedure and corticosteroid adminiiatbrought marked improvement of clinical manifestation, hematologic abnormality and radiologic abnormality.

        야뇨증 환아들의 심리사회적 특성에 대한 다기관 연구 : 행동 및 정서 문제를 중심으로

        Objectives : The aim of this study was to examine the behavioral and emotional problems associated with nocturnal enuresis in Korean children. Methods : Three hundred eighteen children with nocturnal enuresis, together with their parents, completed the Child Behavior Checklist (CBCL), Disruptive Behavior Disorder Scale according to DSM-IV (DBDS), Children's Depression Inventory (CDI), State-Trait Anxiety Inventory for Children (STAIC), and Piers-Harris Children's Self-Concept Scale (PHCSC). Ninety-three normal students were selected as the control group. Results : Compared to the normal control group, the mean scores with regard to the withdrawn, social problems, attention problems, delinquent behavior, aggressive behavior, externalizing problems and total problems profiles were significantly higher in the nocturnal enuresis group according to the CBCL results. The nocturnal enuresis group also scored significantly higher in the ADHD and ODD profiles of the DBDS. The nocturnal enuresis group was more depressed and anxious than the control group according to the results of the CDI and STAI. The mean score of the PHCSC was significantly lower in the nocturnal enuresis group when compared to the normal control group. Conclusion : The results of this study suggest that children with nocturnal enuresis in Korea have clinically relevant behavioral and emotional problems. The findings support the link between nocturnal enuresis and psychopathology in Korean children.

        The effect of alloy compositions on the microstructures of STS 410L has been studied after welding the specimens. Because of the interaction between the ferrite grains and ε-Cu precipitates in the matrix, the ferrite grains adjacent to HAZ of the specimen containing Cu became fine and uniform compared to those of the specimens containing B and W. It was mast effective to add 0.4% Cu in order to obtain fine and uniform matrix in the welds.

        The purpose of this study is to know the reason of softening deficiency for STS 420J2 cold strip, domestic material, after annealing. The obtained results and solution are as follows : 1) It takes more time for STS 420J2 to be recovered and recrystallized during annealing than JIS 420J2 because the contents of C and N for the one is lower than those of the other. 2) The hardness of STS 420J2 is basically higher than that of JIS 420J2 by strenghening effect of Si solid solution. 3) It is recommended to soften fully the STS 420J2 cold strip that the continous annealing temperature become 900℃ and the line speed of strip become appropriate.

        저혈청 배지에서 인간 전골수세포(HL-60)를 이용한 tPA 생산과 세포사멸기작에 관한 연구

        tPA 생산과 세포 사멸의 연구를 하기 위해서 인간 전골수세포가 배양되었다. 연속 관류 배양하에서 최대 tPA 생산량은 324 ng/ml이며 최대 세포 농도는 5.27×10 exp (6) cells/ml였다. Phorbol ester를 넣었을 때 tPA 생산량은 420 ng/ml까지 향상되었다. 형광 현미경으로 관찰했을 때, apoptosis는 세포 배양 초기부터 중반까지 우세했고 빠른 교반 속도에서도 우세했다. 그러나 necrosis는 세포배양 말기와 동결된 세포가 해동 되었을 때 우세했다. HL-60 was cultivated to produce tPA (tissue-type plasminogen activator) and study the mechanism of cell death. Maximum cell density and tPA production were obtained as 5.27×10 exp (6) cells/ml and 324 ng/ml, respectively under perfusion cultivation. tPA production was enhanced to 420 ng/ml in adding 160 nM of phorbol ester. The cells were gradually differentiated to granulocytes rather than proliferation. By Fluorescent microscope, apoptosis was prevailed except the death phase and in high agitation speed, but necrosis was prevailed in thawed cells and during the latter periods of the cultivation. It was also proved that tPA was most produced in apoptosis. To obtain higher tPA productivity, the cells must be maintained in apoptosis, not necrosis phase when the cells were dying.

      • 가열 온도에 의한 소나무와 잣나무 생엽과 생지의 연소 온도변화

        본 연구는 산불 발생시 연소대상 침엽수종인 소나무(Pinus densiflora)와 잣나무 (Plnus koraiensis)의 생엽과 생지를 가열온도에서 시료량에 따른 연소온도 변화와 연소열량의 관계를 조사한 것이다. 잣나무 시료의 함수량은 80℃ 에서 2시간이 경과될 때, 소나무 시료보다 9.91% 정도 많았으나 4 시간이 경과되면서 두 시료의 함수량은 5% 정도로 급격히 저하되어 강한 인화성 건엽으로 전환되었다. 소나무 생엽과 생지 시료의 착화는 400℃의 가영온도에서 각각 약 7분 10초와 13분 20초에 일어났으며, 발염은 각각 약 12분 20초와 15분경에 일어났다. 생엽과 생지 시료의 연소온도는 가열온도와 관계없이 각각 76℃, 106℃ 상승했다. 400℃의 가열온도에서, 소나무와 잣나무 생엽 시료의 고온 지속시간은 각각 0-07'16", 0-07'08" 였으며, 600℃에서는 각각 0-04'45", 0-02'13"였다. 따라서, 고온 지속시간은 고온일수록 짧았다. 각 가열온도에 따른 연소온도와 최고 온도의 관계는 가영온도가 낮으면 상승되는 온도는 낮았고, 높은 가열온도에서는 높은 온도가 상승했다. 생엽과 생지 시료의 전연소열량은 대체적으로 각각 1253.4㎉ (21.5%), 1290.6㎉ (14.3%) 였으며 생엽 시료가 생엽 시료보다 약 37.2 ㎉ (7.2%) 더 적었다. Present study aims at examining how the amount of sample and changes in combustible temperature of fresh branches and leaves treated with heating temperature are associated with combustible time of two coniferous trees, Pinus densiflora (D) and Pinus koraiensis (K), which are the main victims of forest fire. During the first two hours at 80℃, moisture content of K was higher than that of D by 9.9%, but after four hours, the moisture content was both lowered by 5% and turned into highly combustible leaves. At 400℃ of heating temperature, catch-fire of fresh leaves and branches of D occurred in 0-07'10" and 0-13'20", and also burning times were 0-12'20" and 0-15'03", respectively. Also, with fresh leaves (D,K), the duration of high temperature was as follows: 0-07'16", 0-07'8" at 400℃ and 0-04'45", 0-02'-13" at 600℃, respectively. With fresh leaves and branches, the maximum combustible temperatures, regardless of heating temperature, were 76℃ and 106℃. Concerning the relation between the amount of burnt fuel and maximum temperature, the more amount of the former was, the higher the latter was. The total amount of combustion-heat of fresh branches and leaves were 1253.4 kcal (21.5%) and 1290.6 kcal (14.3%), respectively. So, fresh leaves were about 37.2 kcal (7.2%) less than fresh branches.

      • 중합효소연쇄 반응법에 의한 인형거대세포바이러스(human cytomegalovirus)의 신속한 검출

        목 적 : 인형거대세포바이러스(HCMV)는 면역 저하 환자에서 폐렴, 망막염, 간염 등 치명적인 감염증의 원인이다. 전통적인 세포 배양법으로 HCMV를 분리하기까지는 1주에서 4주가 걸리므로 이 바이러스에 의한 질환이 의심되는 경우 신속한 진단법을 이용한 진단이 바람직하다. 저자들은 한국에서 분리되는 야생주 HCMV를 검출하는데 중합효소 연쇄 반응법을 이용할 경우의 진단적 유용성을 알기 위하여, 전통적인 세포배양법으로 HCMV가 분리된 검체를 이용하여 중합효소 연쇄 반응법의 특이도와 민감도를 평가하였다. 방 법 : 국내 야생주 HCMV 15주, 그리고 전통적인 바이러스 배양법으로 HCMV가 분리된 소변 15검체를 대상으로 중합효소 연쇄 반응을 실시하였다. 중합효소 연쇄 반응은 Towne주의 immediate early antigen의 유전자에서 유리된 primer MIE와 AD169주의 immediate early antigen의 유전자에서 유리된 primer IE를 이용하였다. 증폭 산물은 겔 전기영동 후 EtBR으로 염색하여 자외선 하에서 관찰하였다. Primer IE에 의한 증폭산물은 또한, DNA blot hybridization 방법으로 확인하였다. 결 과 : 1) 국내 HCMV 야생주 15주를 대상으로 중합효소 연쇄 반응을 한 결과 primer IE를 이용한 경우에는 100%(15/15)에서 177bp의 증폭 산물이 관찰되었고, 이들 증폭 산물은 모두 probe IE와 보합 결합되었다. primer MIE를 이용한 경우에는 93%(14/15)에서 435bp의 증폭 산물이 관찰되었다. 2) 세포 배양법으로 HCMV가 분리된 소변 15검체를 대상으로 primer IE를 이용하여 중합효소 연쇄 반응을 한 결과 direct gel analysis 법으로는 73%(11/15)에서, 보합 결합법으로는 87%(13/15)에서 관찰되었다. Primer MIE를 이용한 경우에는 direct gel analysis법으로 87%(13/15)에서 435bp의 증폭 산물이 검출되었다. 결 론 : Primer IE와 MIE를 이용한 중합효소 연쇄 반응법은 국내 야생주 HCMV를 신속히 검출하는데 유용한 검사법이다. Background : Human cytomegalovirus(HCMV) can cause pneumonitis, hepatitis, retinitis and other serious diseases in the immunocompromised patients. It takes 1 to 4 weeks to diagnose HCMV infection by conventional virus culture. Therefore, when HCMV diseases are suspected, a rapid diagnostic method such as polymerase chain reaction(PCR), antigen assay or shell vial culture is desirable. We evaluate the sensitivity and specificity of a PCR for the rapid detection of HCMV wild strains in Korea. Methods : We used 2 sets of primers ; primer IE and primer MIE derived from the sequence for immediate early gene of AD169 strain and Towne strain, respectively. Fifteen clinical isolates of HCMV, suspended in MRC-5 cells, were amplified by PCR. Fifteen urine specimens which were positive for HCMV by conventional virus culture were also amplified. Amplification products were analyzed by polyacrylamide gel electrophoresis. The products from PCR with primer IE were also identified by DNA blot hybridization. Results : PCR using primer IE gave the PCR products in all of the 15 HCMV wild strains. All of these were hybridized with probe IE. When primer MIE were used, 93%(14/15) of the wild strains showed amplified bands by direct gel analysis. When the urine specimens were amplified by PCR with primer IE, amplified bands were seen in 73%(11/15) by direct gel analysis ; 87%(13/15) by hybridization method. When primer MIE were used, 87%(13/15) of the urine specimens showed the PCR products by direct gel analysis. Conclusion : Polymerase chain reaction with primer IE and MIE may be a specific and sensitive diagnostic method for rapid detection of HCMV wild strains in Korea.

      • 소결밀도에 따른 분말 소결금속과 탄소강의 이종금속 레이저 용접성 고찰

        Sintered specimen which used for a blade of diamond tool was manufactured in order to verify CO_(2) laser weldability depend on sintered temperature. Five kind of specimen were prepared and the range of temperature is from 600℃ to 1000℃ at intervals of 100℃. As a result of the sintered density test, the porosity rate appeared in the range of 2.1%∼21.4%. After welding, the most segments had exceeds the minimum fracture stress (600MPa, The Standard Safety of Europe) at the welding strength test except on the sintered at 600℃. In case of the sintered at 700℃, even satisfied the safety allowable stress but cannot get the good quality for bead appearance because of humping defect. In the conclusion, we could know that it showed not only relatively soundness bead but also enough welding strength when the sintered blade of diamond tool is included less than 4% of porosity rate.
