<P>We investigated the effect of low dose radiation on diabetes induced suppression of neurogenesis in the hippocampal dentate gyrus of rat. After 0.01 Gy, 0.1 Gy, 1 Gy and 10 Gy radiation was delivered, the dentate gyrus of hippocampus of streptozotocin (STZ)-induced diabetic rats were evaluated using immunohistochemistry for 5-bromo-2-deoxyuridine (BrdU), caspase-3, and terminal deoxynucleotidyl transferase-mediated nick end-labeling (TUNEL) staining. The number of BrdU positive cells in the non-diabetic rats, diabetic rats without radiation, diabetic rats with 0.01 Gy radiation, diabetic rats with 0.1 Gy radiation, diabetic rats with 1 Gy radiation and diabetic rats with 10 Gy radiation were 55.4±8.5/mm<SUP>2</SUP>, 33.3±6.4/mm<SUP>2</SUP>, 67.7±10.5/mm<SUP>2</SUP>, 66.6±10.0/mm<SUP>2</SUP>, 23.5±6.3/mm<SUP>2</SUP>and 14.3±7.2/mm<SUP>2</SUP>, respectively. The number of caspase-3 positive cells was 132.6±37.4/mm<SUP>2</SUP>, 378.6±99.1/mm<SUP>2</SUP>, 15.0±2.8/mm<SUP>2</SUP>, 57.1±16.9/mm<SUP>2</SUP>, 191.8±44.8/mm<SUP>2</SUP>and 450.4±58.3/mm<SUP>2</SUP>, respectively. The number of TUNEL-positive cells was 24.5±2.0/mm<SUP>2</SUP>, 21.7±4.0/mm<SUP>2</SUP>, 20.4±2.0/mm<SUP>2</SUP>, 18.96±2.1/mm<SUP>2</SUP>, 58.3±7.9/mm<SUP>2</SUP>, and 106.0±9.8/mm<SUP>2</SUP>, respectively. These results suggest low doses of radiation paradoxically improved diabetes induced neuronal cell suppression in the hippocampal dentate gyrus of rat.</P>
비장, 골수세포 및 대식세포에서의 Macrophage Inflammatory Protein-1α (MIP-1α)에 관한 연구

The Macrophage inflammatory protein-1α(MIP-1α) from activated T cell or macrophage, which is small inducible cytokine of unknown biological function, has been shown to display inflammation chemokinetic activities, as well as myelosuppressive effect on more immature progenitor cells. In this paper we show the MIP-1α mRNA expression and the presence of MIP-1α binding sites from murine macrophage cell line RAW 264.7, but not inhibited by cyclosporin A treatment, and also was expressed from mouse splenocyte and bone marrow cell which were not increased by ferritin or lactoferrin treatment. The results of receptor binding assay showed that radiolabeled RAW 264.7 cell with kd value of 0.91 nM, and binding sites per cell of 378. Bone marrow cell and splenocyte also appealed to have MIP-1α binding sites 33 and 11 per cell, respectively.
Glucose Oxidase로 손상된 대뇌신경세포에 대한 Herba Epimedii의 영향에 관한 연구

To elucidate the neurotoxicity of glucose oxidase(GO) in cultured cerebral neurons derived from neonatal mouse cytotoxicity was measured by MTT assay after cultured cells were grown for 4 hours in the media containing 1∼40mU/ml concentrations of GO. In addition, the neuroprotective effect of Herba Epimedii was measured by MTT assay in these cultrures. Cell viability was positively decreased in a dose-dependent manner after exposure of cultured mouse cerebral neurons to 20mU/ml GO for 4 hours. In the neuroprofective effect of Herba Epimedii on GO-mediated toxicity, Herba Epimedii prevented the GO-induced neurotoxicity in these cultures. From these results, it suggests that Epimedii is effective in prevetion of the neurotoxicity induced by GO.
Effect of Rhizoma gastrodiae on oxidative stress in cultured mouse spinal motor neurons

To clarify the toxic effect of oxidative stress, hydrogen peroxide $(H_{2}O_{2})-induced$ neurotoxicity was examined in cultured newborn mouse spinal motor neurons after spinal motor neurons were grown in the media containing various concentrations of glucose oxidase (GO). And also, the protective effect of Rhizoma gastrodiae extract against GO-induced neurotoxicity was evaluated. Cytotoxicity was expressed as a cell viability by 3-(4,5-dimethylthiazol-2-yl) -2,5-diphenyltetrazolium bromide (MTT) assay. In this study, exposure of motor neurons to GO-induced cell death significantly, in a dose- and time-dependent manners in spinal motor neuron cultures. The decrease in cell viability of motor neurons damaged by GO was proventioned by Rhizoma gastrodiae extract. These results suggest that the neuroprotective effect of Rhizoma gastrodiae extract on GO-induced neurotoxicity may result from a attenuation of $H_{2}O_{2}-induced$ oxidative stress.

To clarify the cardiotoxicity of methotrexate(MTX) in cultured myocardial cells, cardiotoxicity was assessed by MTS assay. Myocardial cells were treated with 20∼80ug/ml MTX for 24 hours. and also, the cardioprotective effect of Humuli Herba was measured by MTS assay in these cultrures. Cell viability was remarkably decreased dose-dependently after myocardial cells were exposed to 60ug/ml MTX for 24 hours. In the cardioprotective effect of Humuli Herba on the cardiotoxicity induced by MTX, Herba Humuli prevented the cardiotoxicity induced by MTX in these cultures. From above the results, it suggests that MTX is toxic in cultured myocardial cells and herb extract, Humuli Herba has protective effect against the cardiotoxicity induced by MTX.
