        Effect of Apoptosis Induction of Ailanthus altissima on Human Lung Carcinoma Cells

        We investigated the inhibitory effects of solvent extracts from Ailanthus altissima in A549 human lung cancer cell. A. altissima has been recognized as a traditional healthy food due to its various biological activities against hypertension, strokes, fever, pain, neuralgia, inflammation, and cancer effects. Recently, it has been reported that the extracts of various wild vegetables show strong anti-cancer properties by induction of apoptosis. However, the mechanisms of their cytotoxicity in human lung cancer cells have been poorly understood. The present study was investigated the effects of solvent extracts from A. altissima on cell growth and apoptosis on A549 human lung cancer cells. A treatment of A. altissima inhibited the growth of A549 cells in a dose-dependent manner by inducing apoptosis. Especially, the chloroform fraction showed the highest anti-cancer effect among five kinds of fractions. And also, induction of apoptosis by chloroform fraction were associated with down-regulation of Bcl-2, and up-regulation of pro-apoptotic Bax expression. From these results, A. altissima may have a therapeutic potential in human lung cancer cells and as a functional food.

        자궁내막암 환자의 재발여부 확인에 있어 FDG-PET scan 의 유용성에 대한 고찰

        목적 : Inhibin은 α-subunit와 β-subunit로 구성된 이질 이량체의 당단백으로 β-subunit의 차이에 의해 inhibin A (α-βA)와 inhibin B (α-βA)로 구분된다. 여성에서 inhibin은 주로 난소의 과립막세포 및 황체에서 생산되어 뇌하수체의 FSH 분비를 억제한다고 알려져 왔으나, 임신중 태반과 남성의 고환에서도 생산된다는 사실이 밝혀져 다양한 생리적 작용이 추정되고 있으며 최근에 inhibin A와 B를 분리 측정할 수 있는 방법이 개발되어 inhibin에 대한 연구가 활발히 진행되고 있다. 이에 저자들은 정상 월경주기를 가진 가임기 여성, 폐경이행기 여성, 폐경후기 여성에서 inhibin A와 inhibin B의 농도를 측정하여 월경주기에 따른 정상치의 변화와 노화에 따른 농도 변화를 관찰하고, 난소에서 면역조직화학적 염색법으로 inhibin A와 inhibin B의 발현 양상과 변화를 비교 관찰하여 inhibin의 정확한 생리작용, 폐경이행기에서 inhibin의 역할 및 폐경이행기와 폐경의 예측 지표의 사용 가능성을 알아보고자 하였다. 연구 방법 : 월경주기와 난소기능이 정상이면서 난소종양이 없는 정상 가임기 여성 160명, 폐경이행기 여성 60명, 폐경후기 여성 20명에서 정맥혈을 채혈하여 혈중 inhibin A와 inhibin B 농도를 ELISA로 측정하였고, 그중 정상 가임기 여성 35명, 폐경이행기 여성 20명, 폐경기 여성 5명의 난소 조직을 inhibin α, βA 및 βB subunit에 대한 단클론 항체를 이용하여 면역조직화학적 염식을 시행하였다. 결과 : 1. 정상 가임기 여성에서 혈중 inhibin A 농도는 중증식기까지 낮은 농도를 유지하다 후증식기 (16.53±1.57 pg/ml)부터 증가하기 시작하여, 중분비기 (45.85±2.08 pg/ml)에 최고치에 이른 후 서서히 감소하는 경향을 보였다. Inhibin B 농도는 전증식기 (65.40±4.08 pg/ml)부터 증가하기 시작하여 증식기 동안 계속 높은 농도를 유지하다가 배란기 (110.74±9.83 pg/ml)에 최고치에 이른 다음 분비기에 급격히 감소하였다. 2. 폐경이행기 여성에서 혈중 inhibin A 농도는 증식기에 6.68±0.53 pg/ml, 분비기에 21.78±3.61 pg/ml로 모두 정상 가임기 여성과 비교하여 유의하게 낮았다 (P<0.01). Inhibin B의 농도는 증식기에 52.16±7.46 pg/ml, 분비기에 22.41±6.73 pg/ml로 모두 정상 가임기 여성과 비교하여 유의하게 낮았다 (각각 P<0.01, P=0.025). 3. 폐경후기 여성에서 inhibin A와 inhibin B 농도는 모두 검출되어 않았다. 4. 정상 가임기 여성에서 inhibin α subunit의 면역염색반응은 과립막세포, 난포막세포 및 황체에서 강하게 발현되었다. Inhibin βA의 면역염색반응은 난포에서는 거의 발현되지 않았으나 황체에서는 관찰되었다. Inhibin βB의 면역염색반응은 일차난포에서 처음 발현되어 성장난포와 성숙난포의 과립막세포와 난포막세포에서 관찰되었으나 Inhibin α보다는 약했고 황체에서는 거의 관찰되지 않았다. 5. 폐경이행기 여성에서 inhibin subunit들의 면역염색반응은, 정상 가임기 여성과 같은 양상으로 발현되었으나, 발현정도는 약했다. 모든 면역염색반응은 과립막세포보다 난포막세포에서 더 강하게 나타났다. 6. 폐경후기 여성에서 inhibin subunit들의 면역염색반응은 전혀 관찰되지 않았다. 결론 : Inhibin A는 후증식기 성숙난포에서 처음 생산되기 시작하여 배란 후 황체에서 주로 생산되며, Inhibin B는 일차난포에서 생성되기 시작하여 배란 시까지 분비되므로 inhibin A는 황체기능을, inhibin B는 난포의 기능을 반영한다. 폐경이행기가 되면 inhibin A와 B의 분비는 급속히 감소하게 되어 폐경 후에는 난소에서 생성되지 않는다. 따라서 inhibin A와 B는 여성의 월경주기에 따라 생산과 분비가 서로 다른 호르몬으로 난자 성숙과 난포발달에 관여하며, 난소의 노화 정도를 측정하는 폐경이행기의 지표로 이용될 수 있을 것이다. Objective : To understand the physiologic effects and secretion pattern of inhibin A and inhibin B during menstrual cycle and menopausal transition, inhibin A and inhibin B levels were measured. And to detect any changes in expression of inhibins in human ovary with age, we examined immunohistochemical staining of α, βA, and βB subunits of inhibin in ovarian tissues. This study was also designed to investigate whether or not inhibin is an early marker for menopausal transition. Methods : Inhibin A and inhibin B levels were measured in 320 samples from normal reproductive women, in 60 from perimenopausal women, and in 20 from menopausal women by ELISA. And we examined the immunohistochemical staining of α, βA, and βB subunits of inhibin in ovarian tissues of 35 normal reproductive, 20 perimenopausal, and 5 menopausal women, respectively. Results : In the normal reproductive women, inhibin A begins to increase in the late proliferative phase (16.53±1.57 pg/ml), reaches the peak in the mid-secretory phase (45.85±2.08 pg/ml), reaches the peak in the ovulatory phase (110.74±9.83 pg/ml), reaches the peak in the ovulatory phase (110.74±9.83 pg/ml), and thereafter declines rapidly. In the perimenopausal women, mean inhibin A serum concentration was 6.68±0.53 pg/ml during proliferative phase and 21.78±3.61 pg/ml during secretory phase, which were significantly lower than that of the same phase in the normal reproductive women (P<0.01). Mean inhibin B serum concentration was 52.16±7.46 pg/ml during proliferative phase and 22.41±6.73 pg/ml during secretory phase, which were significantly lower than that of the same phase in the normal reproductive women (P<0.01, P=0.025). In the menopausal women, both inhibin A and inhibin B were not detected. In the normal reproductive women, we observed strong immunostaining for α subunit in granulosa cells, theca cells, and corpus luteum. Immunostaining for βA subunit was observed in corpus luteum, but not in growing follicles. Immunostaining for βB subunit was observed in primary follicle, granulosa and theca cells of growing follicle, and mature follicle, but less strong than immunostaining for α subunit. No staining for βB subunit was observed in the corpus luteum. In the perimenopausal women, immunostaining for inhibin subunits were observed in the same pattern as that of the normal reproductive women, but weaker. Stronger immunostaining was observed in theca cells than granulosa cells. In the menopausal women, none of the immunostaining of inhibin subunits were observed.

      • Characterization and anti-tumor activity of saponin-rich fractions of South Korean sea cucumbers (Apostichopus japonicus)

        In this study, the saponin-rich fractions of five individual (two Red and three Black) sea cucumbers (Apostichopus japonicus) in South Korea were investigated for their antiproliferative effect against HL-60, B16F10, MCF-7, and Hep3B tumor cell lines. The red sea cucumber saponin-rich fraction (SSC) from Jeju Island (JRe) decreased the growth of HL-60 with an IC50 value of 23.55 ± 3.40 lg/mL, which represented the strongest anticancer activity among the extracts. Further, SSC downregulated B-cell lymphoma extra-large (Bcl-xL), while upregulating, to different degrees, Bcl-2-associated X protein (Bax), caspase-9, caspase-3, PARP cleavage, and apoptotic bodies in cancer cells. Evidence for SSC inducing apoptosis via the mitochondria-mediated pathway was found. The contents of SSCs were determined using ultra high-performance liquid chromatography coupled with a quadrupole orbitrap mass spectrometry to comparatively evaluate the regional influence. In West Sea, the total SSC content of A. japonicus was 15.5 mg/g, representing the highest content, while A. japonicus in the South Sea yielded the lowest content at 8 mg/g. The major saponin constituent in SSC was identified as Holotoxin A1, which may the anti-tumor compound in A. japonicus.

        <P>A(3) adenosine receptor (AR) ligands including A(3) AR agonist, N-6-(3-iodobenzyl)adenosine-5'-N-methyluronamide (1a, IB-MEGA) were examined for adiponectin production in human bone marrow mesenchymal stem cells (hBM-MSCs). In this model, 1a significantly increased adiponectin production, which is associated with improved insulin sensitivity. However, A(3) AR antagonists also promoted adiponectin production in hBM-MSCs, indicating that the A(3) AR pathway may not be directly involved in the adiponectin promoting activity. In a target deconvolution study, their adiponectin-promoting activity was significantly correlated to their binding activity to both peroxisome proliferator activated receptor (PPAR) gamma and PPAR delta. They functioned as both PPAR gamma partial agonists and PPAR delta antagonists. In the diabetic mouse model, la and its structural analogues A(3) AR antagonists significantly decreased the serum levels of glucose and triglyceride, supporting their antidiabetic potential. These findings indicate that the polypharmacophore of these compounds may provide therapeutic insight into their multipotent efficacy against various human diseases.</P>

        The Action of Triterpenoidal Glycosides of Dammarane Series and Their Aglycones on $K^{+}$ and $H^{-}$ Fluxes in Erythrocytes, Induced by lonophore $A_{23187}$ and Divalent ions

        Ginsenoside Rb,, at a concentration of 10 $\mu\textrm{g}$/ml and over, initiated the cycle of oscillation of ion flux in erythrocytes after the cells had been treated with a protonophore, carbonyl cyanide p-trifluoro-methoxyphenyl hydrazone (FCCP) and then with a $Ca^{2+}$ ionophore, A23,3,. Its action was similar to the additional portion of $Ca^{2+}$-ionophore or $Ca^{2+}$ ion to the erythrocytes. Effects of $Rg_1$ and Rf were different from that of Rb,. They did not induce the oscillation. They, however, increased the extracellular $K^{+}$ concentration and pH without returning to the initial state in the erythrocytes processed with FCCP and $A_{23187}$. We established that ginsenosides from 20-(5)-panaxatriol family induced the membrane hyperpolarization in erythrocytes, which was attenuated by the pretreatment of $Rb_1$, a major component of 20-(5)-panaxadiol.

        무당벌레(H. axyridis)는 세계적으로 진딧물의 포식자로 널리 알려지면서 농생태계내에서 진딧물을 방제하기 위한 방제인자로 많이 사용하고 있다. 또한 국내에서는 농약의 사용을 억제하는 정부정책과 소비자의 요구 등으로 인하여 친환경농업이 확대되면서 친환경농자재들이 많이 사용되고 있다. 따라서 친환경농자재의 사용은 무당벌레에 직간접적으로 영향을 미칠 수가 있다. 살충성친환경농자재(IEFAMs)의 경우 무당벌레의 각 발육단계에 대하여 대부분 안전한 경향을 나타내고 있다. 살균성친환경농자재(FEFAM)의 경우에도 FEFAM A를 제외한 다른 종류들은 무당벌레에 대하여 영향이 적다. 토양미생물친환경농자재(EFAMSM)인 EFAMSM C와 H는 무당벌레 알의 부화율을 떨어뜨릴 수가 있으며, EFAMSM A와 F는 1령 유충에 피해를 줄 수 있는 것으로 조사되었다. 식물성추출물친환경농자재(EFAMPE)는 EFAMPE A와 D는 무당벌레 알에 치명적인 독성을 보이고 있는 것을 알 수 있다. 그 이외의 다른 조사된 농자재들은 무당벌레에 비교적 독성이 약한 것으로 평가되었다. The multicolored Asian ladybird beetle (Harmonia axyridis) has been commonly used with biological control agents for control of several kinds of aphids in agroecosystems. Also, environment friendly agricultural materials have been commonly applied in crop fields because the government held down pesticide application and environment friendly agricultures are gradually increased with consumer's desires. The multicolored Asian ladybird beetles may be directly or indirectly under the influence of environment friendly agricultural materials In crop fields. The insecticidal environment friendly agricultural materials (IEFAMs) might be saff against each developmental stage of multicolored Asian ladybird beetle. Fungicidal environment friendly agricultural materials (FEFAMs) had a miner effect to each developmental stage of multicolored Asian ladybird beetle with the exception of FEFAM A. Environment friendly agricultural materials contained useful soil microorganisms (EFAMSMs) C and H might be down the hatching rate of eggs, and EFAMSM A and F had a killing effect to 1st instar of lady beetles. Environment friendly agricultural materials contained plant extracts (EFAMPEs) A and D might be suffered effect a deathblow of egg hatching with lady beetles. Otherwise, there was a miner effect to lady beetles with the rest of tested environment friendly agricultural materials.

        A case of subdural hematoma in patient with chronic myeloid leukemia treated with high-dose imatinib mesylate

        Imatinib mesylate (IM) is used to treat a wide range of diseases, including Philadelphia chromosome-positive chronic myeloid leukemia (CML), on account of its high tolerability and low incidence of minor adverse events. Hemorrhage is thought to be a rare complication of IM. Recently, IM has been associated with reduced a2-plasmin inhibitor and platelet dysfunction. We report here the case of a 33-year-old female patient with CML who experienced subdural hematoma after an incremental increase in IM dosage due to a loss of complete molecular response. This case indicates that physicians should be alert to this atypical cause of headache in patients taking high-dose IM.

        Inappropriate Gonadotropin Secretion in Polycystic Ovary Syndrome: The Relationship with Clinical, Hormonal and Metabolic Characteristics

        Objective To evaluate the clinical, hormonal and metabolic characteristics according to luteinizing hormone (LH)/follicle stimulating hormone (FSH) ratio in women with polycystic ovary syndrome (PCOS). Methods A total of 225 women with PCOS were included in this study. They were divided into two groups according to LH/FSH ratio; group A (LH/FSH < 2, n=160) and group B (LH/FSH > 2, n=65). We compared clinical, hormonal and metabolic characteristics including age at menarche, body mass index (BMI), blood pressures, ovarian volume, and serum androgen levels between the two groups. Serum glucose and insulin levels, fasting glucose/insulin ratio, homeostatic model assessment of insulin resistance (HOMA-IR), and lipid profi les were also compared between the two groups. Pearson correlation coeffi cients were used to evaluate correlation between the LH/FSH ratio and various parameters. Results Age of menarche in the group B was signifi cantly later than that in the group A. BMI, waist-hip ratio and blood pressures were signifi cantly lower in the group B compared to those of the group A. Ovarian volume and serum levels of LH, estradiol, total testosterone, sex-hormone binding globulin, 17-hydroxyprogesterone (OHP), and high density lipoprotein were signifi cantly higher in the group B. Postprandial 2 hours glucose, insulin and HOMA-IR were signifi cantly higher in the group A. After adjustment of BMI, the LH/FSH ratio was signifi cantly positively correlated with age at menarche, ovarian volume, total testosterone levels, and 17-OHP levels. Conclusion The inappropriate gonadotropin secretion may be negatively correlated with BMI, and positively with age of menarche and increased ovarian androgen production irrespective of BMI in women with PCOS. However, it may not be associated with metabolic characteristics.

        남한 내 북한이탈주민의 3년간 사회적응 추적 연구 : 2001년부터 2004년까지 생활과 교육을 중심으로 With prioty given to life of North Korea defectors from 2001 to 2004

        Purpose of this study : In 2001, this study that investigate actual conditions about adaptation degree of Korean social life of the North Korean defectors was carried out to 151 people that succeed in chases among 200 the North Korea defectors answer in study. Purpose of this study is as following. First, we wished to search change aspect of actual conditions about South Korea society adaptation of the North Korean defectors who take part in the first investigation for past 3 years. Second, we wished to search difference of adaptation aspect by populational and social element. Third, We wished to suggest things that can be beaconed to efficient support policy of government or private organization and successful South Korea social adaptation method which support the North Korean defectors on the basis of study finding about change of adaptation aspect of the North Korea defectors. Methods of Study : We designated 200 Panels to study and survey actual adaptation conditions of the North Korean defectors on South Korea society synthetically and for long-term. So, chasing about 151 people was possible in 2004. We are fertilized and supplement the questionnaire forming in 2001 and composed the new questionnaires to part of a life, mental state, Body and health. This study among them investigated a standard of living of the North Korean defectors. In this process, interviewer directly asked and recorded questions to reduce individual variation of people who receive interview. Study Results : Government's residence support policy about North Korean secession inhabitants was generally attained as effective and suitable for North Korean inhabitants' early South Korea settlement. Only, government need to discuss about North Korea secession inhabitants' residence support more variously for long-term. Second, education should be supported more actively and variously. Suitable point of education for their needs should make a effect of vocational training or education maximize. This is a very important part. Third, they need guidance and education that can receive more easily law support. Fourth, medical supports and health cares should be achieved continuously and systematically to them since they arrived in South Korea. To supports and studies North Korean secession inhabitants has the large significance to gain a foothold about reunification of North and south. Their South Korea society adaptation is becoming topic of conversation among South Korean. An atmosphere of unity is elated. And interest degrees about North Korean are actually rising as exchanges between South and North Korean is activated. Therefore, the adaptation study about North Korean secession inhabitants will have to be expanded in studies for South Korean, not to be limited in studies for North Korean.

        목적 : 학령전기 아동의 글씨 쓰기 명료도 및 속도와 지각-운동 기술의 상관관계를 확인하고, 글씨 쓰기에 가장 영 향을 주는 요인을 알아보고자 하였다. 연구방법 : 덴버 발달 선별 검사를 통하여 정상 발달로 선별된 23명의 학령 전기 아동(평균 연령=68.61개월, SD=2.04)을 대상으로 글씨 쓰기 명료도 및 속도 측정을 위하여 한글 자모 쓰기 검사지를 사용하여 자음과 모음을 쓰도록 하였다. 시지각과 시각-운동 통합 수행 능력 측정을 위하여 시지각 발달 검사를 사용하였고, 소운동 협응 수행 능력 측정을 위하여 Grooved pegboard test를 사용하였다. 결과 : 글씨 쓰기 명료도와 전반적인 시지각, 비운동성 시지각, 시각-운동 통합, 우세손 소동작 협응의 상관관계는 없었으며, 글씨 쓰기의 속도는 전반적 시지각과 우세손 소동작 협응 항목에서 유의한 상관관계를 보였다. 회귀 분 석 결과 글씨 쓰기 속도에 가장 큰 영향을 주는 요인은 전반적 시지각 항목이었다. 결론 : 만 5~6세 학령 전기 아동의 전반적 시지각 능력의 발달을 향상시키기 위한 중재를 제공하는 것이 글씨 쓰기 의 속도 능력을 향상 시킬 수 있다는 것을 알 수 있다. Objective : This study investigated the relationships of handwriting legibility and perceptual-motor skills, and handwriting speed and perceptual-motor skills. And identified the predictors that most affect the handwriting of preschool children. Methods : Twenty-three typically developing preschool aged children (mean age: 68.61 months, SD=2.04) were selected through the Korean-Denver Developmental Screening Test-2(K-DDST-2). The children were tested with regard to handwriting legibility, visual perception, visual-motor integration and fine-motor coordination. Results : First, a significant relationship was not found among handwriting legibility, visual perception, visualmotor integration and fine-motor coordination. Second, a significant relationship was found among handwriting speed, visual perception and fine-motor coordination. Third, stepwise multiple regression analyses showed that general visual perception were significant predictors for handwriting speed. Conclusion : Occupational therapists should evaluate children's visual perception levels utilizing a standardized test, and focus on general visual perception in order to improve handwriting skill(speed). Also, occupational therapists are expected to play an important role in the management and treatment of children's handwriting skills.
