      • Generation of Transgenic Pigs Carrying a Construct for Endothelial Specific Expression of CD73

        Despite of the absence of hyperacute rejection and acute humoral xenograft rejection, the organ graft of the a1,3-galactosyltransferase (GalT) gene knockouted (KO) and complement regulatory protein (CRP) expressing pig into a nonhuman primate is rejected by development of a thrombotic microangiopathy and/or a consumptive coagulopathy. Thus further introduction of genes to overcome the coagulation incompatibilities between pig and primate under GalT KO/CRP genetic background has been strongly suggested. CD73 (ecto-5'-nucelotidase) is an enzyme attached via a glycosyl phosphoinositol anchor to the extracellular membrane of endothelial cells, which catalyses the hydrolysis of adenosine triphosphate to adenosine. Loss of activity of CD73 results in activation and aggregation of platelets by a reduced capacity to convert nucleotides to adenosine. In previous study, we reported generation of GalT KO fibroblasts concurrently expressing membrane cofactor protein and produced cloned pigs by nuclear transfer of the fibroblast cells (1). In this study, we constructed a vector for expression of human CD73 under control of promoter of pig Icam2 gene expressed specifically at endothelial cells. This vector was introduced into porcine fibroblasts using the nucleofection technology, by which we had forty three fibroblasts clones carrying pIcam2- CD73 vector. Somatic cell nuclear transfer resulted in generation of two transgenic piglets survived.

        In this study, Artemisia capillaris, which has been used as oriental medicine area and folks remedy, was investigated for effective substance (scopoletin) analysis. As an attempt to develop new functional beverage by using medicinal herb, Artemisia capillaris. Therefore we investigate the effect of scopoletin in Artemisia capillaris on vessel and regional cerebral blood flow (rCBF) of rats. Artemisia capillaris extract was significantly decreased vessel constraction and significantly increased rCBF. Artemisia capillaris has often been cited in medical literature for its medicinal effects. Although the Korean Food and Drug Administration indicated Artemisia capillaris as the In-Jin-Sook which was allowed as a food stuff. From the view point of the subjects who eat In-Jin-Sook products, most of them ingested it for a certain pharmacological efficacy rather than as a beverage or a food. According to this facts, we need develop functional foods using In-Jin-Sook. Also scopoletin, get known to smooth muscle relaxation content was 326.13 ug/g. These results demonstrate that Artemisia capillaris has pharmaceutical effects and balanced nutritional contents. In according with Artemisia capillaris has a sufficiency of the terms use of food stuff in functional foods.

      • Identification of AFLP Marker Linked to a SCN Resistant Gene in Soybean

        The soybean cyst nematode (Heterodera glycines Inchinoe; SCN) is a devastating pest of soybean and is responsible for significant losses in yield. The use of resistant cultivars is the effective method to reduce or eliminate SCN damage. The objective of this research is to identify AFLP markers linked to the SCN resistant genes. Bulked genomic DNA was made from resistant and susceptible genotypes to SCN and a total of 19 primer combinations were used. About 31 fragments were detected per primer combination. The banding patterns were readily distinguished in resistant and susceptible bulked genotypes. Polymorphic fragments were detected between resistant and susceptible bulked genotypes in the primer combination of CGT/GGC, CAG/GTG and CTC/GAG. In primer combinations of CGT/GGC and CAG/GTG, bulked resistant genotype produced a polymorphic bands. However, in primer of CTC/GAG, bulked susceptible genotype produced a polymorphic fragments. Three AFLP markers identified as a polymorphic fragments between bulked genomic DNA were mapped in 85 F2 population. Among them, only two markers, CGT/GGC and CTC/GAG, was linked and was mapped. Broad application of AFLP marker would be possible for improving resistant cultivars to SCN.

        Distinct function of miR-17-92 cluster in the dorsal and ventral adult hippocampal neurogenesis

        <P><B>Abstract</B></P> <P>It has been known that the dorsal and ventral areas of the dentate gyrus in the hippocampus have distinct roles in memory and mood behaviors. We previously reported that microRNA <I>miR-17-92</I> regulates adult hippocampal neurogenesis and mood disorders. Here, we suggest that the <I>miR-17-92</I> cluster is highly expressed in the ventral than the dorsal dentate gyrus in the adult mouse hippocampus. Deletion of <I>miR-17-92</I> in the adult hippocampus only affects development of neural progenitors in the ventral dentate gyrus, and <I>miR-17-92</I> knockout mice have no defects in memory functions. Our results suggest that regional expression of <I>miR-17-92</I> in the dentate gyrus is associated with their distinct functions in hippocampal neurogenesis and related behaviors.</P> <P><B>Highlights</B></P> <P> <UL> <LI> miR-17-92 is highly related with hippocampal neurogenesis and mood behavior. </LI> <LI> The expression of miR-17-92 cluster is regionally different in hippocampus. </LI> <LI> miR-17-92 KO less affect development of neural progenitors in ventral hippocampus. </LI> <LI> Interestingly, memory function was not altered by miR-17-92 KO. </LI> <LI> These findings suggest the distinct function of regional expression of miR-17-92. </LI> </UL> </P> <P><B>Graphical abstract</B></P> <P>[DISPLAY OMISSION]</P>

        Jeong Jin-Kuy is a leading poet who has published 9 collection of poems and a faithful poet to the classical features of poetry. In his early works he tried to search invisible world rather than visible real world. But he could not the identity of onthology about such a invisible world. The more he passioned for a search for the invisible world, the more he sunk into a abyss of despair and doubt. [Identity], [The most beautiful dust of classic] are clearly showed that point. Therefore the poet turned to a daily world entering 1970's. But it dosen't mean a whole change to the daily world giving up the mentalism of invisible world. In his poem of forming a daily world, the mentalism of search for eternity and immortality was contained. The poet find out a paradoxical truth in daily world. The truth is that to make empty is to fill up and to reach to the world of eternity. [A writting with a pencil], [About a bone] are the good works expressed such a his poetic world. His mentalism has been deeply related to Taoism. Such a poetic attitude come near falling into a philsophical and aphoristical saying. But he shows the real and concrete world of his experience expressing that such a truth lies in just our 'body'. He is a uncompleted poet preparing for more writting works, we don't know how his poem is going for. But on the our study by this time, we can expect that his poem is still going to show the intensive mentalism in spite of making a new poetic world. His mentalistic poem until now has a important meaning of our modern poetic history. During his working period from 60's to early 90's, A sociaty-concerning poetry and A break-form poetry-are widely spread in our modern poetry. On this poetic mood he is evaluated to pursuit of original value of poetry on the ground of mentalism and to maintain a poetic dignity.

        역번역을 활용한 한국 시조 이해 교육 연구 - 영어권 한국어 고급 학습자를 대상으로 -

        This study aimed to present the availability and implications of back-translation learning methods in Korean sijo education as a foreign language. Results were based on the understanding of Korean sijo by advanced English-speaking Korean learners using back translation. In this study, the significance of back translation as a method of learning sijo is summarized as follows: First, learner-centered activities that encourage learners to participate exist; second, reading native texts can help obtain “advanced knowledge;” third, results are an exploratory process involving learners’ independent thinking; and fourth, awareness of diversity in interpretation encourages freedom to express views on texts. This study selected Hwang Jin-i’s sijo “A Long Night of Winter Solstice” as an experimental text and categorized the understanding of English-speaking advanced learners of the Korean language using back translation into “accepting the meaning of text through reading translations,” “interpretation of the meaning of text through back translations,” and “deeper understanding of the text by comparing back translations with source texts.” Advanced English-speaking Korean learners showed attitudes related to actively organizing the meaning of sijo while performing back translation; they continued to interpret the text by mobilizing their own experience and knowledge through the back translation process. The different order of words in the mother tongue and Korean also led to difficulty in switching to the target language. The following implications were derived based on the understanding of advanced English-speaking Korean learners using back translation. First, the teacher should effectively adjust the primary response the learner sees after reading the translation text. Second, proper educational measures from teachers are needed when learners spend a considerable amount of time on a single poem. Third, the cultural context of learners should be extended after comparing the back translated and source texts. (Seoul National University)

        제7차 교육과정과 중등 불교교육의 방향

        이 글은 7차 교육과정에 의거하여 시행될 우리나라 중등학교 종교교육과정의 개편현황을 종교교육의 성격, 종교교육의 내용, 종교교육의 교수 학습과 평가준거 및 방법의 측면에서 탐색하였다. 이러한 탐색에 기초하여 2002년부터 일선학교에서 시행될 '종교(불교)' 교육의 방향을 위의 교육과정 편제와 구조적 틀 속에서 논의하였다. 종교교육과정이 7차 교육과정을 계기로 국가단위의 완성된 편제를 갖추게 되었다는 사실은 '종교(불교)' 교과가 교육과정상의 공적기준을 갖추었을 뿐만 아니라, 이에 기초하여 종교교육의 책무성(accountability)에 대한 문제를 제기할 수 있는 조건이 마련되었다는 것을 의미한다. 따라서 이번 7차 교육과정 개정을 계기로 종교교육은 교육과정과 관련된 구조적 논의의 장에서 그 바람직한 방향을 체계적으로 모색해볼 필요가 있다. 우리나라의 중등학교에서 실시되어온 종교교육은 그 동한 종교학적 종교교육과 신앙적 종교교육간에 절충점과 타협점을 모색하기 위해 고민해왔다. 때로는 종교교육에 대한 이 두가지 성격규정이 상반되거나 모순된 것으로 인식되어, 긴장과 불일치를 야기하고, 일선 학교 교육과정 운영에 있어 실질적인 부담요인으로 작용되어온 측면도 있었다. 이번 7차 종교 교육과정 개정을 계기로 이러한 문제들이 교육과정의 구조와 틀 속에서 논의됨으로 보완 해소될 수 있는 여지를 마련할 필요가 있다. 이러한 한 논의에 기초해서 이 글에서는 중등학교 종교(불교)교육의 바람직한 방향을 탐색하고자 하였다. The purpose of this article is to explore the direction of Buddhistic Education in the view point of the religious education in secondary level which come in force on the 7th curriculum Revision in Korea. At 2002 years in 7th curriculum Revision, Buddistic Education as a form of religious education has a complete curriculum structure of national level which has a formal criteria and educational accountability. In the light of curriculum forms of instructional method about Buddhistic education, there are sharp contrasts between the science of religion approach and dogmatic approach. As many researchers has mentioned, the science of religion approach in Buddhistic education purpose to be educated a learner who can be only a intellectually understanding to the religion. But in contrast to the science of religion approach, dogmatic approach are purpose to made learner a man of religion who believes only a religion. And so there are many practical attempts which persuade to compromise the conflict of religious and dogmatic approach in the field of Buddhistic education. This article are willing to opportunities which seeks to solve this curriculum issues to has been generated tensions and burdens in Buddhistic Education which take the opportunity in the 7th curriculum revision which are planned to act in Korean Secondary schools at 2002 years

      • 세라믹 캔들 필터 주위의 분진 속도분포 측정

        본 연구는 독일 Schumacher의 1m 크기의 세라믹 캔들 필터가 1개 설치된 실험장치를 구성하여 집진시 단일 필터 주위에서의 분진 속도분포를 PDPA를 이용하여 측정하고, 분진층의 유무와 분진 투입 위치에 따른 분진 속도분포의 변화를 관찰하였다. 측정 결과 집진이 계속되어 분진층이 발달함에 따라 접진 속도가 감소됨을 알 수 있었다. 특히, 투입구가 위치하는 필터표면에서 집중적으로 분진층이 발달하여 이곳에서의 분진 속도 감소가 크게 나타났다. 그리고 필터내부와 압력용기부사이의 압력 차에 의한 필터 표면에서의 분진 속도 평균값은 0.28m/sec 이었다. 또한 분진 투입을 암력용기부 하단에 위치한 투입구로 투입하였을 때 필터 주위의 분진의 고른 속도분포를 관찰할 수 있었다. The experimental set-up with a 1m long ceranic candle filter of Schumacher(Germany) was built in this work. The dust velocity field around the filter was measured using PDPA. The effects of dust cake laver and dust inlet position on the dust velocity field were also analyzed. It was found that the filtration velocity decreases as the dust cake builds up on the filter. The filtration velocity largely decreases around the filters near the dust inlet since the dust cake tlevelops fast there. The average dust velocity on the filter due to the pressure difference through the filter was measured to be 0.28m/s. More uniform dust velocity field around the filter was obtained when feeding dust in a lower position of the pressure vessel.
