HMGB1 is associated with human cancers and is an activator of autophagy which mediates chemotherapy resistance. We here show that the mRNA levels of HMGB1 are high in leukemia cells and it is involved in the progression of childhood chronic myeloid leukemia (CML). HMGB1 decreases the sensitivity of human myeloid leukemia cells K562 to anti-cancer drug induced death through up-regulating the autophagy pathway, which is confirmed by the observation with an increase in fusion of autophagosomes and autophagolysosomes. When overexpressing HMGB1, both mRNA levels of Beclin-1, VSP34 and UVRAG which are key genes involved in mammalian autophagy and protein levels of p-Bcl-2 and LC3-II are increased. Luciferase assays document that over-expression of HMGB1 increases the transcriptional activity of JNK and ERK, which may be silenced by siRNA. The results suggest that HMGB1 regulates JNK and ERK required for autophagy, which provides a potential drug target for therapeutic interventions in childhood CML. [BMB reports 2011; 44(9): 601-606]

To evaluate the relationship between abortions and risk of breast cancer, we conducted a case-control study with 669 cases and 682 population-based controls in Jiangsu Province of China. A structured questionnaire was used to elicit detailed information. Unconditional logistic regression analysis was performed to calculate odds ratios (ORs) and 95% confidence intervals (CIs). The results have revealed that induced abortion was related to increased risk of breast caner. Premenopausal women who had ${\geq}3$ times of induced abortion were at increased crude OR (2.41, 95%CI: 1.09-5.42) and adjusted-OR (1.55, 95%CI: 1.15-5.68). Postmenopausal women with a previous induced abortion were at increased crude OR (2.04, 95%CI: 1.48-2.81) and adjusted-OR (1.82, 95%CI: 1.30-2.54), and there was a significant increase trend in OR with number of induced abortions (p for trend: 0.0001). Overall, spontaneous abortion did not significantly alter the risk of breast cancer, but postmenopausal women who had history of spontaneous abortion were at increased OR. These results suggested that relationship between breast cancer and abortions may depend on menopausal status and induced abortion may played an important role in the development of breast cancer in Jiangsu' women of China.

The ancient Chinese wild rice (Zizania latifolia (Griseb) Turcz) (CWR) has valuable biological and medicinal functions. To assess the advantages lost in modern cultivated rice after domestication, we compared the composition of bioactive compounds and the results of proteomic analysis with those of Indica rice (N22). We used routine methods to determine the protein, total dietary fiber, amino acid, mineral substance, plant secondary metabolites, and amino acid composition of CWR and N22. The protein and mineral contents of CWR were two times that of N22, and the levels of calcium, potassium, magnesium, chromium, iron, and zinc were significantly higher than those of N22 (P < .05). There was ~7.6 times more dietary fiber in CWR than in N22, but fewer carbohydrates (P < .05). Anthocyanins and chlorophyll were detected in CWR, but were absent from N22. Compared with N22, CWR had 53, 19, and 5.4 times higher (P < .05) levels of saponins, flavonoids, and plant sterols, respectively. The amino acid score of CWR was 66.6, which was significantly higher than N22. Sodium dodecyl sulfate–polyacrylamide gel electrophoresis (SDS-PAGE) indicated that the main seed proteins of CWR were glutelins, including both acid and alkaline subunits, which were approximately twice those of N22. To investigate the differences in protein profiles between CWR and N22, we conducted two-dimensional electrophoresis (2-DE) analysis of the total proteins in the seeds of the two rice species. 2-DE gels revealed 19 differentially expressed proteins. Information obtained from peptide mass fingerprinting indicates that glutelin precursor caffeoyl coenzyme A (CoA) O-methyltransferase and putative bithoraxoid-like protein can provide good gene sources for improving rice quality.

A new actinomycete strain NA4 was isolated from a deep-sea sediment collected from the South China Sea and showed promising antifungal activities against soilborne fungal pathogens. It was identified as Streptomyces cavourensis by morphological, physiological, and phylogenetic analyses based on its 16S rRNA gene sequence. The main antifungal components were isolated and identified from the fermentation culture as bafilomycins B1 and C1. These compounds exhibited significant antifungal activities and a broad antifungal spectrum. The results suggest that the Streptomyces cavourensis NA4 and bafilomycins B1 and C1 could be used as potential biocontrol agents for soilborne fungal diseases of plants.

Design optimization of construction dewatering has important values in protecting the surrounding environment and reducing the engineering cost. The existing dewatering design methods are mostly based on the optimization of pumping rate needed to maintain a stable water level, while the drop and rise of water level are not considered in the estimation of pumping rate and dewatering time. The dynamics process of construction dewatering is taken into account in this article. The method for calculating the total water yield and dewatering time from the initial groundwater level to the target water level is established. Then, the design optimization of well group dewatering is provided with the consideration of total water yield and dewatering time in dynamic dewatering process. Finally, the method for the calculation of total water yield and dewatering time when dewatering regions change is derived. The proposed method can realize design optimization and dynamic control of dewatering as the dewatering regions and water level changes in construction. The reliability of this method is verified in on-site engineering applications.

The tazarotene-induced gene 1 (TIG1) protein is a retinoid-inducible growth regulator and is considered a tumor suppressor. Here, we show that DnaJ heat shock protein family member C8 (DNAJC8) is a TIG1 target that regulates glycolysis. Ectopic DNAJC8 expression induced the translocation of pyruvate kinase M2 (PKM2) into the nucleus, subsequently inducing glucose transporter 1 (GLUT1) expression to promote glucose uptake. Silencing either DNAJC8 or PKM2 alleviated the upregulation of GLUT1 expression and glucose uptake induced by ectopic DNAJC8 expression. TIG1 interacted with DNAJC8 in the cytosol, and this interaction completely blocked DNAJC8-mediated PKM2 translocation and inhibited glucose uptake. Furthermore, increased glycose uptake was observed in cells in which TIG1 was silenced. In conclusion, TIG1 acts as a pivotal repressor of DNAJC8 to enhance glucose uptake by partially regulating PKM2 translocation.

Preparation and Antioxidant Activities of High Fischer’s Ratio Oligopeptides from Goat Whey
Yusi Qin,Ming Cheng,Xiaoxue Fan,Xiaoqing Shao,Cunfang Wang,Hua Jiang,Xiaoning Zhang 한국축산식품학회 2022 한국축산식품학회지 Vol.42 No.5
This study aimed to obtain high Fischer’s ratio oligopeptides from goat whey (HFO) and investigate antioxidant property of it. Hydrolysis of goat whey was done with the approach of sequential digestion of pepsin and flavourzyme. With the adsorption of aromatic amino acids by activated carbon, HFO with a Fischer’s ratio of 27.070 and a molecular weight of 200–1,000 Da were obtained, and the branched-chain amino acids accounted for 22.87%. Then the antioxidant activity of HFO was evaluated. At the concentrations of 2.0 mg/mL and 0.50 mg/mL, HFO scavenged 77.27% and 99.63% of 1,1- diphenyl-2-picrylhydrazyl and 3-ethylbenzthiazoline-6-sulphonate free radicals respectively. The scavenging rate of HFO against hydroxyl radicals reached 92.31% at the concentration of 0.25 mg/mL. Animal experiments demonstrated that HFO could moderate the changes of malondialdehyde, superoxide dismutase and glutathione peroxidase caused by CCl4- induced oxidative stress in vivo. This study indicated that HFO from goat whey was capable of oxidation resistance both in vivo and in vitro, which provided a scientific basis for the high-value processing and application of goat milk whey.
