Variation of sexual dimorphism and asymmetry in disease expression of inflammatory arthritis among laboratory mouse models with different genomic backgrounds

        Wei Dong,Cheng Tian,Z. Galvin Li,David Brand,Yanhong Cao,Xiaoyun Liu,Jiamin Ma,Andy Chai,LindaK.myers,Jian Yan,Karen Hasty,John Stuart,Yan Jiao,Weikuan Gu,Xiaojun Cai 한국실험동물학회 2023 Laboratory Animal Research Vol.39 No.4

Sex difference has shown in the arthritis diseases in human population and animal models. We investigate how the sex and symmetry vary among mouse models with different genomic backgrounds. Disease data of sex and limbs accumulated in the past more than two decades from four unique populations of murine arthritis models were analyzed. They are (1) interleukin-1 receptor antagonist (IL-1ra) deficient mice under Balb/c background (Balb/c KO); (2) Mice with collagen II induced arthritis under DBA/1 background; (3) Mice with collagen II induced arthritis under C57BL/6 (B6) background and (4) A F2 generation population created by Balb/c KO X DBA/1 KO. Our data shows that there is a great variation in sexual dimorphism for arthritis incidence and severity of arthritis in mice harboring specific genetic modifications. For a F2 population, the incidence of arthritis was 57.1% in female mice and 75.6% in male mice. There was a difference in severity related to sex in two populations: B6.DR1/ B6.DR4 (P < 0.001) and F2 (P = 0.023) There was no difference Balb/c parental strain or in collagen-induced arthritis (CIA) in DBA/1 mice. Among these populations, the right hindlimbs are significantly higher than the scores for the left hindlimbs in males (P < 0.05). However, when examining disease expression using the collagen induced arthritis model with DBA/1 mice, sex-dimorphism did not reach statistical significance, while left hindlimbs showed a tendency toward greater disease expression over the right. Sexual dimorphism in disease expression in mouse models is strain and genomic background dependent. It sets an alarm that potential variation in sexual dimorphism among different racial and ethnic groups in human populations may exist. It is important to not only include both sexes and but also pay attention to possible variations caused by disease expression and response to treatment in all the studies of arthritis in animal models and human populations.

Preparation of silver Nano-Crystal patterns in oxide glasses under electric field accompanied by heat treatment

        Jian Lin,Wen-Hai Huang,Shu-Hua Lei,Chang-Cheng Liu,Qi Bian,Li-Na Ma,Dong-Wei Li 한양대학교 세라믹연구소 2008 Journal of Ceramic Processing Research Vol.9 No.4

Nano-crystals embedded in glass always have interesting quantum effects, while nano-crystal patterns may have wide potential applications in preparing PIC, OEIC and other types of electronic and optoelectronic devices. A direct current electric field, accompanied by heat treatment, was applied on tellurite or silicate glass sheets. The silver ions in a diffused film were introduced into the glass matrix under a high-voltage electric field, instead of supplied during glass melting. The direct diffusion of metal ions, the nucleation and crystallization of nano-crystals in the glass were well controlled by the electric field accompanied by heat treatment. It was found that silver nano-crystal patterns can be transfer printed into tellurite and silicate glass sheets with both patterned diffusion and patterned crystallization. CircularRNA_104670 plays a critical role in intervertebral disc degeneration by functioning as a ceRNA

        Jian Son,Hong-Li Wang,Ke-Han Song,Zhi-Wen Ding,Hai-Lian Wang,Xiao-Sheng Ma,Fei-Zhou Lu,Xin-Lei Xia,Ying-Wei Wang,Fei-Zou,Jian-Yuan Jiang 생화학분자생물학회 2018 Experimental and molecular medicine Vol.50 No.-

This study was carried out to explore the roles of circular RNAs (circRNAs) in nucleus pulposus (NP) tissues in intervertebral disc degeneration (IDD). Differentially expressed circRNAs in IDD and normal NP tissues were identified based on the results of microarray analysis. Bioinformatics techniques were employed to predict the direct interactions of selected circRNAs, microRNAs (miR), and mRNAs. CircRNA_104670 was selected as the target circRNA due to its large multiplier expression in IDD tissues. After luciferase reporter and EGFP/RFP reporter assays, we confirmed that circRNA_104670 directly bound to miR-17-3p, while MMP-2 was the direct target of miR-17-3p. The receiver-operating characteristic (ROC) curve showed that circRNA_104670 and miR-17-3p had good diagnostic significance for IDD (AUC circRNA_104670 = 0.96; AUC miRNA-17-3p = 0.91). A significant correlation was detected between the Pfirrmann grade and expression of circRNA_104670 (r = 0.63; p = 0.00) and miR-17-3p (r = −0.62; p = 0.00). Flow-cytometric analysis and the MTT assay showed that interfering with circRNA_104670 using small interfering RNA (siRNA) inhibited NP cell apoptosis (p < 0.01), and this inhibition was reduced by interfering with miR-17-3p. Interfering with circRNA_104670 suppressed MMP-2 expression and increased extracellular matrix (ECM) formation, which were also reduced by interfering with miR-17-3p. Finally, an MRI evaluation showed that circRNA_104670 inhibition mice had a lower IDD grade compared with control mice (p < 0.01), whereas circRNA_104670 and miRNA-17-3p inhibition mice had a higher IDD grade compared with circRNA_104670 inhibition mice (p < 0.05). CircRNA_104670 is highly expressed in the NP tissues of IDD and acts as a ceRNA during NP degradation.

Molecular characterization and comparative analysis of two waxy alleles in barley

        Jian Ma,Qian-Tao Jiang,Laroche Andre,Zhen-Xiang Lu,Guo-Yue Chen,Ya-Xi Liu,You-Liang Zheng,Yu-Ming Wei 한국유전학회 2010 Genes & Genomics Vol.32 No.6

Two alleles of the barley waxy locus were characterized from non-waxy cultivar Bowman and waxy cultivar CDC Candle, respectively. Their nucleotide and protein sequences were compared with other known waxy genes. The comparison results indicated that there were 100 polymorphic sites, among which 69 were in the non-coding region and 31 were in the coding region. Out of 100 polymorphic sites, 45 were transversion,35 were transition and 20 were indels. A 397 bp deletion and a 193 bp insertion in the promoter region and a 15bp insertion in the coding region were found in CDC Candle, but not in Bowman. A deletion (11 bp) was detected in Bowman, which exhibited no effects on normal waxy expression. In summary, the 397 bp deletion was supposed to account for the reduction of GBSS I, resulting in the low amylose in CDC Candle; whereas other polymorphic sites might be not correlated with amylose synthesis.

        Wei, Guo,Nie, Ming-Ming,Shen, Xiao-Jun,Xue, Xu-Chao,Ma, Li-Ye,Du, Cheng-Hui,Wang, Shi-Liang,Bi, Jian-Wei Asian Pacific Journal of Cancer Prevention 2014 Asian Pacific journal of cancer prevention Vol.15 No.1

Objective: To observe local and systemic toxicity after sustained-release 5-fluorouracil (5-Fu) implantation in canine peritoneum and para-aortic abdominalis and the changes of drug concentration in the local implanted tissue with time. Methods: 300 mg sustained-release 5-Fu was implanted into canine peritoneum and para-aorta abdominalis. Samples were taken 3, 5, 7 and 10 days after implantation for assessment of changes and systemic reactions. High performance liquid chromatography was applied to detect the drug concentrations of peritoneal tissue at different distances from the implanted site, lymphatic tissue of para-aortic abdominalis, peripheral blood and portal venous blood. Results: 10 days after implantation, the drug concentrations in the peritoneum, lymphatic tissue and portal vein remained relatively high within 5 cm of the implanted site. There appeared inflammatory reaction in the local implanted tissue, but no visible pathological changes such as cell degeneration and necrosis, and systemic reaction like anorexia, nausea, vomiting and fever. Conclusions: Sustained-release 5-Fu implantation in canine peritoneum and para-aortic abdominalis can maintain a relatively high tumour-inhibiting concentration for a longer time in the local implanted area and portal vein, and has mild local and systemic reactions. Besides, it is safe and effective to prevent or treat recurrence of gastrointestinal tumours and liver metastasis.

Robust Nonlinear Feedback Control of a Chaotic Permanent-magnet Synchronous Motor with a Load Torque Disturbance

Jian Hu,Long Liu,Da-wei Ma 한국물리학회 2014 THE JOURNAL OF THE KOREAN PHYSICAL SOCIETY Vol.65 No.12

The permanent-magnet synchronous motor (PMSM) system, which is a nonlinear dynamic system, will demonstrate a variety of chaotic phenomena when its parameters or external inputs fallinto a certain area, which will lead to a deterioration of its performance. Thus, chaos should besuppressed or eliminated. In this paper, the property of equilibrium points is analyzed, and thecondition for the occurrence of a Hopf bifurcation in a PMSM system is given based on a mathematicalmodel of the PMSM system with a bifurcation diagram, a Lyapunov exponent map and phaseplane diagrams given. After the drawbacks of the existing control methods have been analyzed, arobust nonlinear feedback controller is designed to control the chaos in the PMSM system with aload torque disturbance. The object is to eliminate the chaos and to drive the system speed to adesired value, Numerical simulation proves the validity of this control method.

Tetrandrine Exerts a Radiosensitization Effect on Human Glioma through Inhibiting Proliferation by Attenuating ERK Phosphorylation

        Ma, Ji-wei,Zhang, Yong,Ye, Ji-cheng,Li, Ru,Wen, Yu-Lin,Huang, Jian-xian,Zhong, Xue-yun The Korean Society of Applied Pharmacology 2017 Biomolecules & Therapeutics(구 응용약물학회지) Vol.25 No.2

Tetrandrine (Tet), a bisbenzylisoquinoline alkaloid, has been reported to have a radiosensitization effect on tumors. However, its effects on human glioma and the specific molecular mechanisms of these effects remain unknown. In this study, we demonstrated that Tet has a radiosensitization effect on human glioma cells. It has been hypothesized that Tet has a radiosensitization effect on glioma cells by affecting the glioma cell cycle and DNA repair mechanism and that ERK mediates these activities. Therefore, we conducted detailed analyses of the effects of Tet on the cell cycle by performing flow cytometric analysis and on DNA repair by detecting the expression of phosphorylated H2AX by immunofluorescence. We used western blot analysis to investigate the role of ERK in the effect of Tet on the cell cycle and DNA repair. The results revealed that Tet exerts its radiosensitization effect on glioma cells by inhibiting proliferation and decreasing the expression of phosphorylated ERK and its downstream proteins. In summary, our data indicate that ERK is involved in Tet-induced radiosensitization of glioma cells via inhibition of glioma cell proliferation or of the cell cycle at G0/G1 phase.

      • Transmembrane Protein 166 Expression in Esophageal Squamous Cell Carcinoma in Xinjiang, China

        Sun, Wei,Ma, Xiu-Min,Bai, Jing-Ping,Zhang, Guo-Qing,Zhu, Yue-Jie,Ma, Hai-Mei,Guo, Hui,Chen, Ying-Yu,Ding, Jian-Bing Asian Pacific Journal of Cancer Prevention 2012 Asian Pacific journal of cancer prevention Vol.13 No.8

Objective: Transmembrane protein 166 (TMEM166) expression in esophageal squamous cell carcinoma (ESCC) and remote normal esophageal tissues was examined to assess any role in tumour biology. Methods: TMEM166 mRNA expression in 36 cases with ESCC (36 tumour samples, 36 remote normal esophageal tissue samples) was detected by RT-PCR. TMEM166 protein expression was analysed in paraffin-embedded tissue samples from the same cases by immunohistochemistry. Results: Semi-quantitative analysis showed TMEM166 mRNA expression in ESCCs to be significantly lower than in remote normal esophageal tissues ($0.759{\pm}0.713$ vs. $2.622{\pm}1.690$, P=0.014). TMEM166 protein expression was also significantly reduced (69.4% vs. 94.4%, P<0.01). Conclusion: TMEM166 mRNA and protein expression demonstrated significant reduction in ESCCs compared with remote esophageal tissues, albeit with no correlation with tumour size, differentiation, stage, and lymph node metastasis, suggesting a role in regulating autophagic and apoptotic processes in the ESCC.
