        Fibrothecoma are mesenchymal tumors deriving from the ovarian stroma and consisting of theca-like elements and fibrous tissues. The reported incidence is between 0.4 and 8.0% of all ovarian tumors and this side variability can be attributed to the difficulty in differentiation fibrothecoma from ovarian fibroma or hyperplastic stroma nodules. We experienced a rare case of ovarian fibrothecoma in woman at 38 weeks gestation with previous repeat cesarean section diagnosed after tertiary cesarean delivery and bilateral tubal ligation with a brief review of literature.

        The strutures of euojaponines, alkaloidal components from Euonymus japonica are composed of β-agarofuran type sesquiterpene and pyridic dibasic acid, and they are linked via cyclodiester bonds. The structures of these compounds were elucidated by the application of various two dimensional NMR methods. From the results of complete interpretation of NMR spectra, several correlating aspects between structure and spectral pattern of sesquiterpene moiety were found, which can be utilized for the structural determination of the same family compounds.

        Nitric Oxide (NO), derived from L-arginine, is produced by two types (constitutive and inducible) of nitric oxide synthase (NOS: cNOS and iNOS). The NO produced in large amounts by the iNOS is known to be responsible for the vasodilation and hypotension observed in septic shock and inflammation. The inhibitors of iNOS, thus, may be useful candidate for the treatment of inflammatory diseases accompanied by the overproduction of NO. We prepared alcoholic extracts of herbal drugs which have been used for the treatment of inflammation in oriental medicine. We have screened the inhibitory activity of NO production in lipopolysaccharide (LPS)-activated macrophages after the treatment of these extracts. Among the 81 kinds of extracts of herbal drugs, 34 extracts showed potent inhibitory activity of NO production above 50% at the concentration of 50 (μg/ml. The inhibitory activities of NO production were also evaluated for several solvent fractions at three different concentrations. Especially, hexane soluble fractions of Agrimonia pilosa, Hydrangea serrata, Machilus thunbergii, Prunella vulgaris, Saussurea lappa, Tussilago farfara, and ethyl acetate soluble fractions of Angelica gigas, Ostericum koreanum, Torilis japonica, Perilla frutescence showed moderate activity at 10 and/ or 25 (μg/ml. These fractions are promising candidates for the study of the activity-guided chromatographic purification of active compounds.

      • 요하지통 환자에서 두 개의 캐눌라를 이용한 박동성 고주파 신경근 응고술의 임상 연구

        Dorsal root ganglion(DRG) block by the local anesthetics and steroids which has done classically in low back pain(LBP) patients with radiculopathy has high incidence of recurrence rate and side effects of steroids. Recently a new technique of pulsed radiofrequency thermocoagulation(P-RFTC) was introduced and substituted for it because of the benefits such as relatively low recurrence rate, low risk of nerve injury and comfort during the procedure. We experienced 15 patients who had LBP with radiculopathy and no or little response to epidural steroid injection. When we performed DR ganglionotomy by P- RFTC under the C-arm guide in these patients, we tried to approach the exact DR ganglion using tow cannulas - the one for obtaining patient's subjective symptoms and injection of contrast dye, the other for making a RF lesioning after identifying the impedance of sensory and motor stimulation. In conclusion, DR ganglionotomy by P- RFTC using two cannulas in LBP patients is an easy and safe procedure with satisfactory results.

      • 질편모충(Trichomonas vaginalis)의 시험관내에서의 용혈능

        To evaluate the hemolytic activity of T. vagindlis from Korean women and fresh human RBC of A type were nsed for hemolytic assay. RBC(1×10 /ml) was added to RPMI-TPS media(RPMI 1840:TPS-1= 20 : 1) containing T. vaginalis(2×10 ) and incubated at 37℃, 5% CO₂incubator for 6hr and hemolysis was measured by spectrophotometer at 412m. We investigated the parasite to erythrocyte ratio and lemperature needed for hemotysis. Optimal erythrocyte lysis by T. vaginalis was achieved at ratio of 1 : 5 and optimol temperature 37℃. Minimal hemolytic activity was detected in lysates of T. vaginalis. No hemolysis occured when a membrane a 3㎛ pore size was nsed to prevent contact between parasites and erythrocytes. These data suggest a need for contact for trichomonol hemolysis. Hemolytic activity did not correlated with the production of subcutaneous abscess in mice.

      • 항혈청으로 처치된 질트리코모나스(Trichomonas vaginalis) 미세구조의 변화

        Trichomonas vaginalis is a parasitic flagellate found in the urogenital tract of humans. The present study was performed to evaluate the effect of antiserum from rat and rabbit on fine structure of T. vaginalis. Trophozoites were subcultured in TPS-1 medium(Diamond, 1968) at 37℃, CO₂incubator. T. vaginalis were inoculated in mixed medium containing of antiserum for 10 and 30 minutes, then observed with transmission and scanning electron microscope. The results are as follows: 1. T. vaginalis was ellipsoidal or ovoidal in shape with 4 flagella, an undulating membrane and an axostyle. The plasma membrane of body surface was smooth and furrowed. Pseudopodia(filopods) were often observed by SEM. Ultrastructure of trophozoites consisted of an elliptical shaped nucleus, RERs and Golgi complex around the nucleus, hydrogenosomes, glycogen granules, polysomes and vacuoles in cytoplasm. As the organelles for supporting and movement, an axostyle, kinetosomes, flagella, undulating membrane, costa and parabasal filaments were observed. 2. T. vaginalis treated with antiserum, its surface appeared destruction of plasma membrane, alteration and/or disappearance of axostyle, flagella and undulating membrane under the SEM and showed fine structural changes such as the nucleus surrounded by many vacuoles, decreased ribosomes, which attached to nuclear membrane and RER, vesiculated RER and Golgi complex. The area of filamentous cell coat and a number of coated vesicles were increased on plasma membrane with partial damage. According to going by the incubation time, the progressive destructions of cytoplasmic organelles, nuclear membrane, hydrogenosomal membrane, kinetosome and rootlet fiber were noted. With above results, it is presumed that specific antiserum induced the disturbance of protein systhesis and the damage of cytoplasm resulting lysis of T. vaginalis.

      • 면역시킨 마우스 흉선 및 비장세포에 대한 병원성 Naegleria의 세포병변 효과

        Naegleria fowleri is a causative agent of primary amoebic meningoencephalitis (PAM) in man and experimental animal. We investigated the interaction of immunized mouse thymocyte and splenocyte with N. fowleri trophozoites in vitro. BALb/c mouse was immunized with 150∼200×10⁴/ml N. fowleri trophozoites, intraperitoneally, once a week, for two times. Thymus and spleen cells from immunized or normal mouse cocultured with N.fowleri in 10% FCS+MEM, 37℃, 55%-CO₂incubator and cell: amoeba ratio was 200∼400:1. Thymus or spleen cells and N. fowleri were observed at 24, 48, 72 hrs after incubation by inverted microscope. Spleen cells from immunized mice showed moderate lysis aftre 72 hrs with round amoeba, but other group cells were completely lysed at 48 hrs after incubation with N. fowleri. It may be secreted a certain cytolytic amterial from N. fowleri because of cell lysis occured abruptly after 24 hrs culture. And amoebae showed better condition i.e., pseudopodaand vacuoles, after phagocytosis of lytic cell material. On the other hand, nonadherent cells from immunized spleen were completely lysed at 48 hrs with N. fowleri conculture and immunized thymus cell with aditive activated mactophage showed prolonged cell survival. And so, cooperative reaction of immunized T-lympocyte and macrophage are important to supperss N. fowleri in vitro.

      • 절편모충(Trichomonas vaginalis)에 대한 단일클론항체의 생산과 그 특성

        Trichomonas vaginalis, mucosal protozoan parasite of the urogenital tract, is responsible for the most common clinically recognized sexually transmitted disease. Surface antigen of parasitic protozoa play the important roles in disease pathogenesis by serving as attachment factors, direct virulence factors, or mediating evasion of the host immune response through direct suppressoin or antigenic variation. In this study seven monoclonal antibodies against plasma membrane of T. vaginalis HY-1 were prepared and were investigated the effector function of them. Seven monoclonal antibodies were produced and their isotypes were Ig G1 (Tv1, Tv7), Ig G2a (Tv4), Ig M(Tv2, Tv3, Tv5, Tv6). All of the seven monoclonal antibodies recognized the determinants located on the surface of live organism by IFAT. Although these surface reactive seven monoclonal antibodies agglutinated T. vaginalis trophozoites, especially MAb Tv1 and Tv4 could agglutinate eight isolates of T. vaginalis. In immunoblot assay, MAb Tv1 & Tv4 detected single peptide with molecular weight of 60 kDa, and Tv7 detected 80 kDa. On the other hand Tv2 and Tv6 detected two (40 kDa & 120 kDa) or more peptides (62 kDa, 64 kDa & 120 kDa), and Tv3 and Tv5 did not bind any pepetide. From these results, MAb Tv1 reacted with 60 kDa, major surface membrane protein of T. vaginalis and agglutinated trichomonads.
