Molecular analysis of TMC1 gene in the Korean patients with nonsyndromic hearing loss

Hereditary nonsyndromic hearing loss (NSHL) is a highly heterogeneous disorder in humans. Mutations of the transmembrane channel‐like (TMC1) gene have been identified as the genetic cause for both autosomal recessive (DFNB7/11)and autosomal dominant (DFNA36) nonsyndromic hearing loss. To evaluate the spectrum and frequency of mutation(s)caused by TMC1 gene in the Korean population, we have performed sequencing analysis of the PCR products amplified from genomic DNA of each proband in 193 unrelated families showing 30 autosomal dominant and 163 autosomal recessive inheritance patterns. As a result, we identified eight different novel sequence variations for the first time in this study,respectively. However, none of these showed co‐segregation of phenotype in the families. Therefore, our study suggests that the TMC1 gene is not the cause of nonsyndromic hearing loss in the Korean population.

Starting with the structural unit of 8-ring in supercage for dehydrated ?? and Ca-zeolite A, we studied chemical characterization and electronic structure and properties of monovalent ?? and divalent ?? cations with framework oxide ions of the above materials by semiempirical molecular orbital theory. On 8-ring in the supercage for dehydrated Tl and Ca-zeolite A, framework oxide ions are strongly bonded by chemical bonding. This results show structure stability of structural skeleton for z대lite A. When ?? or ?? cations are incorporated in 8-ring, framework oxide ions with ?? cation are bonded by chemical antibonding while framework oxide ions with ?? cation are weakly bonded by chemical antibonding. Charge transfer in ?? with framework oxide ions is moved ?? to 8-ring. Then, net charge of O atoms in 8-ring including ?? cation are increased. But, net charge of O atoms in 8-ring including ?? cation are decreased. With these results, we here indicate that ?? ion are located in the 8-ring and ?? ion are not located in the 8-ring. These results are supported by the recent X-ray experimental study by Kim et al..

Invasive pulmonary aspergillosis is known to have a poor prognosis and major cause of morbidity and motality in patients with hematologic disease. Without adequate therapy, invasive pulmonary aspergillosis will almost always progress to fatal pneumonia. Early administration of antifungal agents in patient with invasive pulmonary aspergillosis is most important. And surgical resection of aspergillous-infected tissue may be useful in patients with lesions that are contiguous with great vessel and lesions causing hemoptysis. We reported a rare case of surgical resection as curative treatment for invasive pulmonary aspergillosis patient in 58-year-old man who had the history of acute myeloid leukemia.

Ulcerative colitis is a chronic inflammatory bowel disease manifested by diffuse continuous mucosal and submucosal inflammation. Adenocarcinoma of the colon may be complicated in the long-standing, extensive ulcerative colitis. But the case of an atypical carcinoid tumor arising in the chronic ulcerative colitis has been very rarely reported. A case was experienced involving a 54-year-old woman with chronic ulcerative pancolitis that was complicated by an atypical carcinoid tumor of the sigmoid colon. A small, flat, elevated lesion was found at the sigmoid colon, 8 years after the initial diagnosis of ulcerative colitis had been made. The microscopic finding of the lesion showed the nest of moderately differentiated, medium sized cells with large nuclei. The immunohistochemical stains for cytokeratin, NSE, chromogranin A, and synaptophysin were positive.The patient was treated with a total colectomy accompanied with an ileostomy, and is now in a very favor able state of recovery.

The terms $quot;gastrointestinal stromal tumor$quot; (GIST) have been applied to mesenchymal tumos that represent neither typical leiomyoma nor schwannoma. The majority of GISTs are Located in the stomach and small intestine, and only 4% of GISTs are found in duodenum. The most important characteristic is their indolent, slow-growing nature, rendering the most common definitions of malignancy invalid and inapplicable. Clinical and pathological criteria to differentiate benign from malignant GISTs are not well established. Tumor size and mitoic activity are commonly considered as important features, allowing for the prediction of biological behaviour and outcome. The case of a 87-year-old male patient who was presented with melena and acute anemia is herein reported. Esophagogastroduodenoscopy and computed tomography of the abdomen revealed two submucosal tumors in the stomach and duodenum. Histological and immunohistochemical studies on the surgical resection specimen revealed a gastrointestinal stromal tumor of an uncommiteed type, with no evidence of increased mitotic activity. The patient was treated with local excision of the tumors and is now in a favorable state.
Catechins activate muscle stem cells by Myf5 induction and stimulate muscle regeneration

<P><B>Abstract</B></P> <P>Muscle weakness is one of the most common symptoms in aged individuals and increases risk of mortality. Thus, maintenance of muscle mass is important for inhibiting aging. In this study, we investigated the effect of catechins, polyphenol compounds in green tea, on muscle regeneration. We found that (−)-epicatechin gallate <B>(</B>ECG) and (−)-epigallocatechin-3-gallate (EGCG) activate satellite cells by induction of Myf5 transcription factors. For satellite cell activation, Akt kinase was significantly induced after ECG treatment and ECG-induced satellite cell activation was blocked in the presence of Akt inhibitor. ECG also promotes myogenic differentiation through the induction of myogenic markers, including Myogenin and Muscle creatine kinase (MCK), in satellite and C2C12 myoblast cells. Finally, EGCG administration to mice significantly increased muscle fiber size for regeneration. Taken together, the results suggest that catechins stimulate muscle stem cell activation and differentiation for muscle regeneration.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Catechins stimulate muscle regeneration after muscle damage. </LI> <LI> Catechins activate muscle satellite cells through Akt activation and Myf5 induction. </LI> <LI> Catechins stimulate myogenesis through the induction of Myogenin and MCK. </LI> </UL> </P>

<P>MicroRNAs (miRNAs) are small noncoding nucleotides that play critical roles in the regulation of diverse biological functions, including the response of host immune cells. Autophagy plays a key role in activating the antimicrobial host defense against <I>Mycobacterium tuberculosis</I>. Although the pathways associated with autophagy must be tightly regulated at a posttranscriptional level, the contribution of miRNAs and whether they specifically influence the activation of macrophage autophagy during <I>M. tuberculosis</I> infection are largely unknown. In this study, we demonstrate that <I>M. tuberculosis</I> infection of macrophages leads to increased expression of miRNA-125a-3p (miR-125a), which targets UV radiation resistance-associated gene (UVRAG), to inhibit autophagy activation and antimicrobial responses to <I>M. tuberculosis</I>. Forced expression of miR-125a significantly blocked <I>M. tuberculosis</I>–induced activation of autophagy and phagosomal maturation in macrophages, and inhibitors of miR-125a counteracted these effects. Both TLR2 and MyD88 were required for biogenesis of miR-125a during <I>M. tuberculosis</I> infection. Notably, activation of the AMP-activated protein kinase significantly inhibited the expression of miR-125a in <I>M. tuberculosis</I>–infected macrophages. Moreover, either overexpression of miR-125a or silencing of UVRAG significantly attenuated the antimicrobial effects of macrophages against <I>M. tuberculosis</I>. Taken together, these data indicate that miR-125a regulates the innate host defense by inhibiting the activation of autophagy and antimicrobial effects against <I>M. tuberculosis</I> through targeting UVRAG.</P>
