        Effect of aberrantly methylated androgen receptor target gene PCDH7 on the development of androgen-independent prostate cancer cells

        Background Androgen-independent prostate cancer (AIPC) is an extremely malignant tumor developed from the androgen dependent (ADPC). However, the mechanism of transition process from ADPC to AIPC remains unknown. Objective Here we aimed to identify the androgen receptor (AR) target gene and its roles in AIPC. Methods Target genes of AR were identified by ChIP-seq in AIPC cells. AR target gene PCDH7 was detected by real time PCR and western blot. Methylation of PCDH7 was measured by bisulfite sequencing and bisulfite amplicon sequencing. Cell growth, invasion and apoptosis were measured by CCK-8, transwell and flow cytometry, respectively. Results AR was significantly enriched in the upstream of PCDH7 gene. The expression of PCDH7 was significantly decreased, while the methylation of PCDH7 was increased in the AIPC cells compared to the ADPC cells. DNA methyltransferase inhibitor significantly suppressed the methylation and increased the mRNA and protein level of PCDH7. Moreover, overexpression of DNMT1 remarkably reduced the mRNA and protein level of PCDH7. DNA methyltransferase inhibitor decreased the cell growth and invasion while promote the cell apoptosis in the AIPC cells. AR significantly target PCDH7, whose hypermethylation may repress cell growth and invasion, and promote apoptosis in AIPC. Conclusions This study might provide a novel potential target for the treatment of AIPC.

        Selective recovery of valuable metals from partial silicated sphalerite at elevated temperature with sulfuric acid solution

        The purpose of this work was to study the feasibility at laboratory-scale of a hydrometallurgical process for the selective recovery of valuable metals from partial silicated sphalerite in an oxygen pressure acid leaching system. The factors influencing dissolution efficiency of the ore were investigated and optimized. Under optimum conditions (i.e., temperature of 433 K, sulfuric acid concentration of 41.2 g/L, leaching time of 2.5 h, liquid/solid ratio of 6 mL/g, and pressure of 1.6 MPa) over 97% Zn was extracted into the leach liquor together with 0.3% SiO2 and 2.9% Pb. The leaching slurry had good solid–liquid separation characteristics, and the filtration rate could be as high as 716 L/m2 h. About 96% oxidation of sulfide sulfur to sulfate was achieved under these conditions. Analysis of the content of elemental sulfur in the leaching residues indicated that the fraction of sulfide sulfur determined as elemental sulfur was about 10% at 393 K, and that it decreased with temperature down to 0.5% at 453 K. Ultimate solid residues, which have been concentrated in silica and lead, can be oriented toward the lead smelter after alkali roasting-water washing pretreatment for metal recovery.

        Coriolis Coupling Influence on the H+LiH Reaction

        We have reported the reaction probability, integral reaction cross section, and rate constant for the title system calculated with the aid of a time-dependent wave packet approach. The ab initio potential energy surface (PES) of Prudente et al. (Chem. Phys. Lett. 2009, 474, 18) is employed for the purpose. The calculations are carried out over the collision energy range of 0.05-1.4 eV for the two reaction channels of H + LiH → Li + H₂and H b + LiH a → H b + LH a. The Coriolis coupling (CC) effect are taken into account. The importance of including the Coriolis coupling quantum scattering calculations are revealed by the comparison between the Coriolis coupling and the centrifugal sudden (CS) approximation calculations.

        Safety and antifatigue effect of Korean Red Ginseng: a randomized, double-blind, and placebo-controlled clinical trial

        Background: Korean Red Ginseng (KRG) is widely used for strengthening the immune system andfighting fatigue, especially in people with deficiency syndrome. However, there is concern that the longtermapplication or a high dose of KRG can cause “fireness” (上火in Chinese) because of its “dryness” (燥性in Chinese). The aim of this study was to assess the safety and efficacy of a 4-week treatment with KRGin participants with deficiency syndrome. Methods: This was a 4-week, randomized, double-blind, placebo-controlled clinical trial. A total of 180Chinese participants were randomly allocated to three groups: placebo control group, participants weregiven a placebo, 3.6 g/d; KRG 1.8 g and 3.6 g groups. The primary outcomes were the changes in firenessand safety evaluation (adverse events, laboratory tests, and electrocardiogram). The secondary outcomeswere the efficacy of KRG on fatigue, which include the following: traditional Chinese medicine (TCM)symptom scale and fatigue self-assessment scale. Results: Of the 180 patients, 174 completed the full study. After 4 weeks of KRG treatment, the Fire-heatsymptoms score including Excess fire-heat score and Deficient fire-heat score showed no significantchange as compared with placebo treatment, and no clinically significant changes in any safetyparameter were observed. Based on the TCM syndrome score and fatigue self-assessment score, TCMsymptoms and fatigue were greatly improved after treatment with KRG, which showed a dose- and timedependenteffect. The total effective rate was also significantly increased in the KRG groups. Conclusion: Our study revealed that KRG has a potent antifatigue effect without significant adverse effectsin people with deficiency syndrome. Although a larger sample size and longer treatment may berequired for a more definite conclusion, this clinical trial is the first to disprove the common conceptionof “fireness” related to KRG.

        Transplantation of Bone Marrow Stromal Cells Enhances Nerve Regeneration of the Corticospinal Tract and Improves Recovery of Neurological Functions in a Collagenase-Induced Rat Model of Intracerebral Hemorrhage

        The reorganization of brain structures after intracerebral hemorrhage (ICH) insult is crucial to functional outcome. Although the pattern of neuronal rewiring is well-docu-mented after ischemic stroke, the study of brain plasticity after ICH has been focusing on the enhancement of dendritic complexity. Here we hypothesized that functional restoration after ICH involves brain reorganization which may be favorably modulated by stem cell transplantation. In this study, bone marrow stromal cells (BMSCs) were transplanted into the perilesional sites of collagenase-induced ICH in adult rats one day after ICH injury. Forelimb functional recovery was monitored with modified limb placing and vibrissae-elicited forelimb placement tests. Anterograde and retrograde tracing were used to assess the reorganization of bilateral forelimb areas of the sen-sorimotor cortex. We found that in rats transplanted with BMSCs after ICH injury, axonal sprouting occurred in the contralateral caudal forelimb area of the cortex, and was significantly higher than in ICH rat models that received only the vehicle (P < 0.01). The number of positive neurons in the ipsilateral rostral forelimb area of the cortex of the BMSC group was 1.5- to 4.5-fold greater than in the vehicle group (P < 0.05). No difference was found between the BMSC and vehicle groups in hemispheric atrophy or labeled neurons in the ipsilateral caudal forelimb area (P = 0.193). Scores for improved functional behavior in the BMSC group were in accord with the results from histology. Neuronal plasticity of the denervated corticospinal tract at bilateral forelimb areas of the cortex in the collagenase-induced ICH rat models was significantly enhanced by BMSC transplantation. BMSC transplantation may facilitate functional recovery after ICH injury.

        <P><B>Abstract</B></P><P>The dynamics of a tachyon attached to a Dvali, Gabadadze and Porrati (DGP) brane is investigated. Exponential potential and inverse power law potential are explored, respectively. The quasi-attractor behavior, for which the universe will eventually go into a phase similar to the slow-roll inflation, is discovered in both cases of exponential potential and inverse power law potential. The equation of state (EOS) of the virtual dark energy for a single scalar can cross the phantom divide in the branch θ=−1 for both potentials, while the EOS of the virtual dark energy for a single scalar cannot cross this divide in the branch θ=1.</P>
