        A Comparison of Lyophilized Amniotic Membrane with Cryopreserved Amniotic Membrane for the Reconstruction of Rabbit Corneal Epithelium

        Many researchers have employed cryopreserved amniotic membrane (CAM) in the treatment of a severely damaged cornea, using corneal epithelial cells cultured on an amniotic membrane (AM). In this study, two Teflon rings were made for culturing the cells on the LAM and CAM, and were then used to support the AM, which is referred to in this paper as an Ahn's AM supporter. The primary corneal epithelial cells were obtained from the limbus, using an ex-plantation method. The corneal epithelium could be reconstructed by culturing the third­passage corneal epithelial cells on the AM. A lyophilized amniotic membrane (LAM) has a higher rate of graft take, a longer shelf life, is easier to store, and safer, due to gamma irradiation, than a (AM. The corneal epithelium reconstructed on the LAM and (AM, supported by the two­Teflon rings, was similar to normal corneal epithelium. However, the advantages of the LAM over that of the (AM make the former more useful. The reconstruction model of the corneal epithelium, using AM, is considered as a good in vitro model for transplantation of cornel epithelium into patients with a severely damaged cornea.

        Ecological characteristics of areas naturally inhabited by Dolbae trees(Pyrus pyrifolia) on Mt. Kaya

        The ecological characteristics of Dolbae trees, a valuable genetic resource for native Pyrus fruit trees, were investigated at four naturally inhabited areas on Mount Kaya, Gyeongsangnam-Do. Most Dolbae trees were found on the southwest side of an old hiking path to a mountain valley at altitudes of 610~670 m. The surrounding flora consisted of 46 families, 69 genera, 75 species, and 10 varieties. The natural habitat of the Dolbae trees was basically a deciduous broad-leaved forest with a predominance of Quercus serrata and ligneous plants(xylophytes) from the Fagaceae and Betulaceae families. The habitat was also found to be in a second transition resulting from forest disturbance and exhibited a degree of diversity of 2.901-5.065, based on Shannon-Weiner's index. Examination of the Dolbae trees found in Mount Kaya revealed diameter breast heights(DBHs) ranging from 10 to 60 cm, including six old and large Dolbae trees with a DBH of 50~60 cm, which will be particularly valuable as a genetic resource. Accordingly, the current results suggest that the study areas are a secondary forest in a typical mid-temperate zone resulting from forest damage about 10 years earlier.

        Reconstruction of Rabbit Corneal Epithelium on Lyophilized Amniotic Membrane Using the Tilting Dynamic Culture Method

        <P>Abstract: </P><P>Rabbit corneal epithelium was reconstructed using tilting dynamic culture with a self-manufactured, amniotic membrane (AM) supporter and a lyophilized amniotic membrane (LAM). Rabbit corneal epithelial (RCE) cells were cultured and cryopreserved after isolation from the limbus. The second- and third-passage RCE cells were plated onto the epithelial side of the LAM of Ahn's AM supporter. Two days later, the air–liquid interface culture was maintained with third-passage RCE cells for 6 days and second-passage corneal epithelial cells for 9 days. The average viability of thawed RCE cells, assessed using trypan blue dye exclusion, was 77.42%. The reconstructed corneal epithelium was characterized by histological (hematoxylin and eosin) and immunohistochemical staining (proliferating cell nuclear antigen) for light microscopy, and by reverse transcriptase-polymerase chain reaction, glucose assay, and transmission electron microscopy. The basal layer of the reconstructed corneal epithelium was well formed, and the epithelium was tightly constructed due to the increase in cell proliferation and differentiation caused by the tilting dynamic culture, as opposed to static culture. Tilting dynamic culture was useful for the reconstruction of the epithelium using easily damaged epithelial cells and resulted in more stratum cell layers. Moreover, cytokeratin (CK3) mRNA expression in tilting dynamic cultured third-passage RCE cells seeded onto AM was greater than in static cultured third-passage RCE cells. The morphology of the reconstructed corneal epithelium on LAM by tilting dynamic culture for 9 days resembled that of the skin epidermis. This was thought to be because the tilting dynamic culture not only accelerated the proliferation and differentiation of cells by physical or mechanical stimulation, but also ensured that the supply of medium was delivered to the basal cells more efficiently. Thus, the reconstruction of the corneal epithelium using LAM and tilting dynamic culture was considered to be a good in vitro model for autologous or allogeneic transplantation of corneal epithelium and skin epidermis in patients with damaged epithelia. </P>

        The purpose of this research was to uncover the relative impact of children's temperament, mothers' characteristics, and parents' child-rearing agreement on mother's parenting stress. Subjects of this study were mothers of 293 preschoolers, being aged 3 to 5 years old from five kindergartens placed in Ahn-yang city and Em-sung gun, Chungbuk. Data were analyzed with Pearson's correlation and heirarchical Regression by using SPSS program. The results showed that children's emotional sensitivity and mother's neuroticism had a positive impact on mother's parenting stress and mother's extroversion had a negative impact on mother's parenting stress. Moreover, couple's child-rearing agreement showed a negative influence on mother's parenting stress.

        용해실리카의 기준경도를 이용한 나노 압입시스템의 실험적 교정

        Indirect calibration of a nanoindentation system was performed by deriving a reference loading curve. This reference load-depth relationship was accomplished by assuming a perfect nanoindentation test on an ideal reference material; if all components in a nanoindentation system are well calibrated and the reference material has unique hardness and Young’s modulus, the indentation depth can be formulated with a function of the indentation load having two parts of contact and surrounding elastic depths. Substituting both the hardness and Young’s modulus of a fused silica into the derived function, the reference loading curve was plotted and directly compared with experimental data of the fused silica measured by two commercial nanoindenters. All indentation loading curves showed leftward shifts compared to the reference loading curve, and this is discussed, offering two reasons for the result, system compliance and indenter apex geometry.(Received January 6, 2014)

        전통누룩 진균류를 이용한 입국의 제조 및 입국곰팡이의 동정

        다양한 향미를 가진 막걸리의 개발을 위해 전통누룩으로부터 분리한 곰팡이로 입국을 제조한 후 품질특성을 분석하여 입국의 규격에 적합하며 이취가 없고 관능이 우수한 9균주를 입국 제조용 우수균주로 최종 선발하였다. 선발된 균주는 Aspergillus oryzae(C1-5-2-2, C20-7-3, CN1.3.1-4, CN16.19.1-1, N152-1, N220-1), Mycocladus corymbiferus (N162-2), Rhizopus oryzae(N20), Lichtheimia corymbifera (N21)로 동정되었으며, 제조한 입국의 산도는 5.0~6.8, 당화력은 128~241sp이었다. Various koji were prepared by fungi isolated from traditional nuruk and their quality characteristics were investigated. Acidity and saccharification power of their koji were ranged in 5.0~6.8 and 128sp~241sp. Nine fungi which were showed good quality and sensory evaluation were identified by analysis of their nucleotide sequences with PCR-amplified 18S rDNA internal transcribed spacer-1(ITS-1) and ITS-4 genes. Among them, six strains were identified as Aspergillus oryzae and the other strains were identified as Mycocladus corymbiferus, Rhizopus oryzae, Lichtheimia corymbifera.

      • 몽고 홉스골 호수 주변의 식물유전자원 분포

        본 조사 결과, 교목류는 5과 11속 13종 1변종의 총 14 분류군으로 나타났다. 4속 5종이 조사된 소나무과 수목은 몽고에서 주요 목재자원으로 육성되고 있다. 버드나무과는 Salix속 1종과 Populus속 1종이 조사되었다. 주로 호수와 연결된 소하천 주변에 점상으로 분포하고 있었다. BEtula속은 1종이 조사되었는데 햇볕이 잘 드는 숲의 전면부에서 군락을 형성하고 있었다. 조사된 장미과의 3종은 모두 꽃과 열매가 아름다운 식물로 관상식물 및 과수작물로의 개발이 가능하다. 관목류는 9과 17속 24종으로 조사되었다. 그 가운데 관상 가치가 높은 진달래과 및 장미과의 낙엽성 관목이 각각 6종, 8종으로 나타났다. Ledum palustre 및 Rhododendron parvifolium은 향료 및 차로 이용가치가 높은 식물이다. Ribes altissimum과 Ribes nigrum은 열매의 식용가치가 높다. 꿀풀과의 Thymus quinquecostatus는 향료 및 최근에 각광받고 있는 허브작물로서의 이용 가능성도 높은 식물이다. 39과 108속 143종 1변종 총 144 분류군으로 조사된 초본성 식물은 약용, 관상용, 식용, 공예용 등으로 이용가치가 높은데 Allium diantum, Lilium pumilum, Iris uniflora, Angelica deccurens, Aconitum barbatum, Stellera rosae, Pulsatilla ambigua, Glycyrrhiza uralensis. 등이 대표적인 식물이다. 서양민들레(Taraxacum officinalis) 및 서양금혼초(Hypocaeris radicata) 등의 귀화식물도 높은 피도와 군도로 나타났다. The plant resources around the lake Hovsgol in Mongolia were botanically surveyed. The examination area was include the region that had various vegetations such as damp ground, dry land, wood land and grassland. The followings are representative plant, which found in examination areas ; the 13 taxa such as Larix sibirica, Abies sibirica, Salix rorida, Crataegus maximowiczii, Prunus sibirica and Sorbus sibirica, etc. were recorded as high tree, while the 24 taxa such as Ledum palustre, Rhododendron parvifolium, Arctous eryhocapa, Vaccinium uligonosum, Vaccinium vitis-idaea, Ribes altissimum, Ribes nigrum and Thymus quinquecostatus, etc. as shrub. And there are 144 taxa herbaceous plants, such as Lilium tenuifolium, Iris uniflora, Aconitum barbatum, Stellera rosae, Angelica deccurens, Pulsatilla ambigua, Taraxacum mongolicum, Aster aplinus, etc. Especially, plants like, Allium diantum, Lilium pumilum and Glycyrrhiza uralensis which have high values of ornamental, edible and medical use.

      • 불악산(경기도)의 식물상에 관한 연구

        Field surveys were carried out 3 times from 2000 5. 17 to 2000. 6. 4. Main results obtained from this study wet·e as follows : 1.The vascular Plants in Mt. Bull-ak were consisted of 221 taxa such as 71 families, 163 genera, 188 species, 30 varieties, 2 forma, 1 hybrid. 2.Family Compositae and Gramineae covers 8.6% and 7.2% of total number of naturalized plants, respectively. 3.Pteridophyte, Gymnosperm, Dicotyledon and Monocotyledon covers 2.5%, 2.7%, 75.9% and 18.1% of total number of naturalized plants, respectively. 4.The life cycle of the plants was annual 11.8%, biennial 11.3%, perennial 76.9%. As it compared with consistency ratio in the flora of Korea, the distribution of biennial appeared highly. 5.The highest peak of blooming month is May (49.3% among total number of naturalized plants). 6.In this area the Korean endemic plants are 5 taxa (2.3% among total number of naturalized plants). 7.Number of naturalized is 20 taxa (9.0% among total number of naturalized plant)

        The objectives of this study were 1) to examine the relationship between shopping value and shopping satisfaction.; 2) to examine the relationship between store type and shopping value. Self-administrated questionnaire was used to collect the data for this study after reviewing previous studies and conducting pre-test twice. The data was analyzed by using analysis, Multiple regression, T-test. The followings are the results of this study, First, There were significant relationships between Tourists' Shopping value and Shopping satisfaction. Second, Store type were significant differences according to Tourists' Shopping value. The result of this study will be helpful for the shopping industries which gave needs to establish the marketing strategies.
