        Rational design of porous NiCo2S4 nanotubes for hybrid supercapacitor

        The nanotube-consisted flower-like NiCo2S4 is successfully fabricated by a novel two-step hydrothermal technique. X-ray diffraction (XRD) identifies the spinel structure, scanning electron microscopy (SEM) and transmission electron microscopy (TEM) imply the flower-like morphology of the synthesized NiCo2S4. The electrochemical behaviors are studied by cyclic voltammetry and galvanostatic charge-discharge measurements. The NiCo2S4 nanotubes demonstrate enhanced pseudocapacitive performance of 429.5 C g− 1 at current density of 0.5 A g− 1 . The NiCo2S4//AC device delivers high energy density of 37.69 Wh kg− 1 , maximum power density of 4000.6 W kg− 1 and satisfied cycle property of 96% capacitance retention after over 7000 cycles. The results show that the NiCo2S4 nanotubes are promising electrode material for high performance supercapacitor applications.

        A Pilot Study of Clinical Evaluation and Formation Mechanism of Irritable Bowel Syndrome-like Symptoms in Inflammatory Bowel Disease Patients in Remission

        Background/Aims Some inflammatory bowel disease (IBD) patients in remission suffer from irritable bowel syndrome (IBS)-like symptoms (IBD-IBS). The pathogenesis has not yet been elucidated. The study aim is to evaluate relationships among quality of life (QOL), psychological status, and visceral sensitivity, and explore the formation mechanism of IBD-IBS. Methods Forty-seven patients with Crohn’s disease in remission, 24 ulcerative colitis in remission, 26 IBS, and 20 healthy controls were included in the study. The abdominal pain, QOL, anxiety, and depression were evaluated through questionnaires. Visceral sensitivity was measured by rectal balloon distension. The serum levels of 5-hydroxytryptamine (5-HT) and nerve growth factor (NGF) were measured by enzyme-linked immunosorbent assay. The expressions of tryptase, 5-HT, NGF, and related receptors in colonic tissues were detected by immunohistochemistry and western blot. Results Prevalence of IBS-like symptoms in Crohn’s disease and ulcerative colitis patients in clinical remission was 29.8% and 50.0%, respectively. The QOL was lower, the anxiety/depression scores were higher in IBD-IBS patients than those without IBS-like symptoms. Additionally, patients with IBD-IBS existed visceral hypersensitivity. Besides, abdominal pain was associated with poor QOL, visceral hypersensitivity, anxiety, and depression in IBD-IBS patients. The number of mast cells (MCs) and expressions of 5-HT, NGF, and related receptors were higher in IBD-IBS patients than those with no such symptoms. The serum levels of 5-HT and NGF positively correlated with abdominal pain and visceral hypersensitivity. Conclusion IBD-IBS patients may have low QOL and psychological abnormalities, as wells as visceral hypersensitivity which may be related to increased 5-HT and NGF levels released from activated mast cells.         Transcriptome analysis reveals critical genes and key pathways involved in early phyllotaxy development in maize

        Integrated networks of gene expression, hormonal signaling and metabolite sensing regulate phyllotaxy pattern development. In this study, we characterized differentially expressed genes (DEGs) between maize plants with alternate and opposite phyllotaxies. Gene ontology and Kyoto Encyclopedia of Genes and Genomes analyses showed that 2432 DEGs were involved in biological processes, molecular functions, cellular components and many pathways. Importantly, we identified 19 DEGs related to plant hormone signal transduction. Additionally, we identified four main alternative splicing types: skipped exons, retained introns, alternative 50-splice sites, and alternative 30-splice sites, which exhibited different characteristics in the alternate and opposite phyllotaxy libraries. The reliability of the sequencing data was verified through using quantitative real-time reverse transcription PCR analysis of the 19 genes: 15 were validated to play a role in phytohormone signal transduction pathways. Taken together, our data provide new insight into the mechanisms of phyllotaxy pattern development, and will increase our understanding of how relative changes in gene expression determine alternate/opposite phyllotaxy in maize.

        An efficient Agrobacterium-mediated transformation method for aflatoxin generation fungus Aspergillus flavus

        Aspergillus flavus often invade many important corps and produce harmful aflatoxins both in preharvest and during storage stages. The regulation mechanism of aflatoxin biosynthesis in this fungus has not been well explored mainly due to the lack of an efficient transformation method for constructing a genome-wide gene mutant library. This challenge was resolved in this study, where a reliable and efficient Agrobacterium tumefaciens-mediated transformation (ATMT) protocol for A. flavus NRRL 3357 was established. The results showed that removal of multinucleate conidia, to collect a homogenous sample of uninucleate conidia for use as the transformation material, is the key step in this procedure. A. tumefaciens strain AGL-1 harboring the ble gene for zeocin resistance under the control of the gpdA promoter from A. nidulans is suitable for genetic transformation of this fungus. We successfully generated A. flavus transformants with an efficiency of ~ 60 positive transformants per 106 conidia using our protocol. A small-scale insertional mutant library (~ 1,000 mutants) was constructed using this method and the resulting several mutants lacked both production of conidia and aflatoxin biosynthesis capacity. Southern blotting analysis demonstrated that the majority of the transformants contained a single T-DNA insert on the genome. To the best of our knowledge, this is the first report of genetic transformation of A. flavus via ATMT and our protocol provides an effective tool for construction of genome-wide gene mutant libraries for functional analysis of important genes in A. flavus.

        Expression profile and estrogenic regulation of Amh during gonadal sex differentiation in northern snakehead (Channa argus)

        Background Anti-Müllerian hormone (Amh) plays a critical role in both early sex determination and later gonad development in vertebrate species. However, it remains unknown in northern snakehead (Channa argus), which is economically important freshwater fish with sexual dimorphism. Objective This study aimed to identify the expression profiles and estrogenic regulation of CaAmh during gonadal sex differentiation in C. argus. Methods The cDNA and genomic DNA sequences of CaAmh were identified by PCR and RACE techniques. The expression patterns of CaAmh were detected by qRT-PCR during the gonadal sex differentiation and after 17α-ethinyloestradiol (EE2) treatments. Results CaAmh is composed of seven exons and six introns, and its full-length cDNA is 2413 bp in length, with 1635 bp open reading frame (ORF) that encodes a 544 amino acid protein. Tissues expression patterns revealed that CaAmh display the highest expression in testis of XY males (40.36 folds, p < 0.01). The spatio-temporal expression patterns during gonadal sex differentiation indicated that CaAmh expression differed between XX females and XY males at 30 day after hatching (dah), and reached to the peak (36.03 folds, p < 0.01) at 90 dah in XY gonads. However, CaAmh expression in XX gonads remained low throughout the sampling period. Furthermore, CaAmh expression in the gonads (ovaries) of the sex-reversed XY fish (XY-F) by the administration of estrogen EE2 was downregulated to low level, similar to that in ovaries of normal XX females (XX-F). Conclusions These results show that Amh plays a critical role in testicular differentiation of C. argus and it is apparently modulated by estrogens in this species.

        Preparation and Investigation of Ulex Europaeus Agglutinin IConjugated Liposomes as Potential Oral Vaccine Carriers

        We prepared and optimized Ulex europaeus agglutinin I (UEAI)-modified Bovine serum albumin (BSA)-encapsulating liposomes (UEAI-LIP) as oral vaccine carriers and examined the feasibility of inducing systemic and mucosal immune responses by oral administration of UEAILIP. The prepared systems were characterized in vitro for their average size, zeta potential,encapsulation efficiency (EE%) and conjugation efficiency (CE%). In vitro release studies indicated that the presence of UEAI around the optimized liposomes was able to prevent a burst release of loaded BSA and provide sustained release of the encapsulated protein. In vivo immune-stimulating results in KM mice showed that BSA given intramuscularly generated systemic response only but both systemic and mucosal immune responses could be induced simultaneously in the groups in which BSA-loaded liposomes (LIP) and UEAI-LIP were administered intragastrically. Furthermore, the modification of UEAI on the surface of liposomes could further enhance the IgA and IgG levels obviously. In conclusion, this study demonstrated the high potential of lectin-modified liposomes containing the antigen as carriers for oral vaccine.

        Research on the Influence of Inter-turn Short Circuit Fault on the Temperature Field of Permanent Magnet Synchronous Motor

        When the inter-turn short circuit (ITSC) fault occurs, the distortion of the magnetic field is serious. The motor loss variations of each part are obvious, and the motor temperature field is also affected. In order to obtain the influence of the ITSC fault on the motor temperature distribution, firstly, the normal and the fault finite element models of the permanent magnet synchronous motor (PMSM) were established. The magnetic density distribution and the eddy current density distribution were analyzed, and the mechanism of loss change was revealed. The effects of different forms and degrees of the fault on the loss were obtained. Based on the loss analysis, the motor temperature field calculation model was established, and the motor temperature change considering the loop current was analyzed. The influence of the fault on the motor temperature distribution was revealed. The sensitivity factors that limit the motor continuous operation were obtained. Finally, the correctness of the simulation was verified by experiments. The conclusions obtained are of great significance for the fault and high temperature demagnetization of the permanent magnet analysis.

        Functional analysis of the HD-Zip I gene ZmHDZ1 in ABA-mediated salt tolerance in rice

        The homeodomain-leucine zipper I (HD-Zip I) transcription factors play crucial roles in the regulation of abiotic stress responses, whereas, up to now, most of them are still functionally unknown. In present study, we identified a maize HD-Zip I gene, designing as ZmHDZ1, and investigated its potential roles in salt stress response. The qRT-PCR results showed that expression of ZmHDZ1 was induced by ABA and salt stress. Transactivation assay and transient expression analyses indicated that ZmHDZ1 protein was located in the nucleus in tobacco leaf cells and had the transactivation activity in yeast. Over-expression of ZmHDZ1 in rice reduced tolerance to salt stress, thus, led to greater accumulation of malondialdehyde (MDA) as well as higher level of relative electrolyte leakage (REL) compared to WT plants. In addition, the transgenic seedlings also increased sensitivity to exogenous ABA. Furthermore, the expression levels of four salt responsive genes were different between transgenic and WT rice under normal or NaCl treatment. These results suggested that ZmHDZ1 function as a negative regulator in response to salt stress through the ABAmediated signal transduction pathways.
