        Effect of Tea Polysaccharides on Thermal and Pasting Properties of Wheat Starch

        The thermal and pasting properties of wheat starch (WS)/tea polysaccharides (TPS) composite system were investigated. Significant increases of transition temperatures (To, Tp, Tc) and the enthalpy of gelatinization (ΔH1) of WS/TPS (2 wet% and 4 wet%) were found as TPS content increased. The lower ΔH2 (endothermic enthalpy under reheating) and R2 (degree of retrogradation of WS) were also detected in WS/TPS in storage of 3,7,14 days when compared with those of WS. The agreements were shown by RVA with the lower total setback and by Texture Analyzer with lower gel firmness. The results of TPS affecting the thermal and pasting properties could be generally explained with Avrami equation of WS/TPS. The Avrami equation exposed that TPS affect the primary nucleation in early stage and the decreased nucleation rate announced by the constant k. The overall results demonstrated that the TPS has an inhibitory effect on the recrystallization of pasting WS in storage.

        The mechanical design and fabrication of 162.5 MHz buncher for China accelerator driven sub-critical system injector II

        A buncher is one of the main pieces of equipment in the medium energy beam transport line (MEBT) for China accelerator driven sub-critical system (C-ADS) Injector II. To focus the beam longitudinally and match the beam for the acceptance of the superconducting linac section, two room temperature quarter wave resonator (QWR) bunchers with frequency of 162.5 MHz have been designed as parts of the MEBT. According to the beam transmission matching of the MEBT and the geometric parameters requirements of bunchers, the unique mechanical structure and the main processing technology of buncher cavities and their couplers and tuners are described in this paper. The fabrication of bunchers and their parts have been completed and tested at high power, the test results agree well with the design requirements. These bunchers work well for about two years in Institute of Modern Physics, Chinese Academy of Sciences.

        A Modified Extraction Method of Circulating Free DNA for Epidermal Growth Factor Receptor Mutation Analysis

        Purpose: Circulating free DNA (cfDNA) in plasma is promising to be a surrogate for tumor tissue DNA. However, not all epidermal growth factor receptor (EGFR) mutations in tumor tissue DNA has been detected in matched cfDNA, at least partly due to inefficient cfDNA extraction method. The purpose of this study was to establish an efficient plasma cfDNA extraction protocol. Materials and Methods: The yield of plasma cfDNA extracted by our modified phenol-chloroform (MPC) method from non-small-cell lung cancer (NSCLC) patients was compared with that by QIAamp MinElute Virus Spin kit (Qiagen kit) as control, using the Wilcoxon rank-sum test. TaqMan quantitative polymerase chain reaction (qPCR) assays were used to quantify the plasma cfDNA extracted. Both Mutant-enriched PCR (ME-PCR) coupled sequencing and DxS EGFR mutation test kit were used to evaluate the impact of extraction method on EGFR mutation analysis. Results: MPC method extracted more plasma cfDNA than Qiagen kit method (p=0.011). The proportion of longer fragment (≥202 bp) in cfDNA extracted by MPC method was significantly higher than by Qiagen kit method (p=0.002). In the sequencing maps of ME-PCR products, a higher mutant peak was observed on plasma cfDNA extracted by MPC method than by Qiagen kit method. In DxS EGFR mutation test kit results, plasma cfDNA extracted by MPC method contained more tumor-origin DNA than by Qiagen kit method. Conclusion: An improved plasma cfDNA extraction method of MPC is provided, which will be beneficial for EGFR mutation analysis for patients with NSCLC.

        Ampelopsin alleviates sevoflurane-induced cognitive dysfunction by mediating NF-κB pathway in aged rats

        Background Cancer-induced bone pain (CIBP) is the pain caused by bone metastasis from malignant tumors, and the largest source of pain for cancer patients. miR-300 is an important miRNA in cancer. It has been shown that miR-300 regulates tumorigenesis of various tumors. Purpose This study aims to investigate the role of miR-300 in CIBP and its underlying molecular mechanisms in vitro and in vivo. Methods We constructed CIBP model in rats and investigated the mechanism through which miR-300 affects CIBP. We first examined expression level of miR-300 in CIBP rats and then tested the effect of its overexpression. Next, we identified the target of miR-300 using TargetScan analysis and double luciferase assay. Finally, we studied genetic interactions between miR-300 and its target and their roles in CIBP. Results We found that miR-300 was downregulated in CIBP rats. Overexpression of miR-300 significantly attenuated cancerinduced neuropathic pain (p < 0.01). Furthermore, TargetScan analysis and double luciferase assay show High Mobility Group Box 1 (HMGB1) is a target of miR-300. Notably, HMGB1 is overexpressed in CIBP rats, while up-regulation of miR300 significantly suppresses expression of HMGB1 (p < 0.01). Moreover, knockdown of HMGB1 by siRNA significantly relieves cancer-induced neuropathic pain in rats (p < 0.01). On the other hand, HMGB1 overexpression partially blocked the effect of miR-300 on cancer-induced nerve pain. Conclusion miR-300 relieves cancer-induced neuropathic pain by inhibiting HMGB1 expression. These results may be beneficial for the treatment of CIBP in clinical practice.

        ZnO@Ni–Co–S Core–Shell Nanorods-Decorated Carbon Fibers as Advanced Electrodes for High-Performance Supercapacitors

        The interconnected three-dimensional Ni–Co–S nanosheets were successfully deposited on ZnO nanorods by a one-step potentiostatic electrodeposition. The Ni–Co–S nanosheets provide a large electrode/electrolyte interfacial area which has adequate electroactive sites for redox reactions. Electrochemical characterization of the ZnO@Ni–Co–S core–shell nanorods presents high specifc capacitance (1302.5 F/g and 1085 F/g at a current density of 1 A/g and 20 A/g), excellent rate capabilities (83.3% retention at 20 A/g) and great cycling stability (65% retention after 5000 cycles at a current density of 30 A/g). The outstanding electrochemical performance of the as-prepared electrode material also can be ascribed to these reasons that the special structure improved electrical conductivity and allowed the fast diffusion of electrolyte ions.

        miR-300 mitigates cancer-induced bone pain through targeting HMGB1 in rat models

        Quantitative Evaluation Index for Analysis of Assembly Effect on Shield Tunnel Segment Structures

        The local force and deformation of shield tunnel will be transferred and redistributed owing to the discontinuous segmental structure. This study develops quantitative evaluation index to explore the influence of the axial compression ratio, eccentricity, and longitudinal force on the assembly effect. A full-scale test is conducted on a single segment and staggered assembled segmental linings. First, the results show that the assembly effect has an obvious redistribution influence on the force and deformation of the segment. Second, the influence of eccentricity on the bending moment is much larger than that of the axial force. The stiffness between rings varied at different positions and the larger the stiffness difference, the more obvious the assembly effect. The results demonstrate that the design and mechanical performance of the shield tunnel under different embedded conditions can be optimized by increasing the longitudinal force, which moderates the assembly effect and ensures the structure reaches the highest bearing capacity.

        Long-term Durability Study on Mechanical and Dynamic Mechanical Performance of a High-performance Carbon Fiberpolymer Laminate

        This paper reports a detailed study on the long-term mechanical and dynamic mechanical performance of carbonfiber (CF) plain-woven fabric-reinforced epoxy (CF/epoxy) laminates fabricated by the hot-compression molding technique. Water-sorption experiments, mechanical property tests, and dynamic mechanical analysis (DMA) tests were performed afterimmersion in de-ionized water at different temperatures (i.e., 25 oC, 60 oC, and 80 oC) for times up to 120 days. The resultsshow that weight-gain curves for the composites at elevated temperature (i.e., 80 oC) fit the theoretical Fickian diffusion curveduring the initial immersion time, but deviate substantially as time prolongs. The flexural properties and dynamic mechanicalproperties are highly susceptible to the effects of the hygrothermal environment, and the performance for both degrade withincreasing exposure temperatures and durations. The dynamic mechanical properties partly recover after the dryingtreatment, which is caused by the irreversible degradation in the interfacial region (i.e., fiber/matrix debonding) as evidencedby morphological images from scanning electron microscopy (SEM). Finally, the long-term degradation of the compositeswas estimated and the service life of the materials was also predicted using the Arrhenius degradation theory. The outputs ofthe research show that CF/epoxy laminates have flexural strength retention of 60 % for more than 2 years at the annualtemperature of Kyoto.

        MicroRNA-206 Protects against Myocardial Ischaemia-Reperfusion Injury in Rats by Targeting Gadd45β

        MicroRNAs are widely involved in the pathogenesis of cardiovascular diseases through regulating gene expression via translational inhibition or degradation of their target mRNAs. Recent studies have indicated a critical role of microRNA-206 in myocardial ischaemia-reperfusion (I/R) injury. However, the function of miR-206 in myocardial I/R injury is currently un-clear. The present study was aimed to identify the specific role of miR-206 in myocardial I/R injury and explore the underlying molecular mechanism. Our results revealed that the expres-sion level of miR-206 was significantly decreased both in rat I/R group and H9c2 cells subjected to hypoxia/reoxygenation (H/R) compared with the corresponding control. Overexpression of miR-206 observably decreased infarct size and inhibited the cardiomyocyte apoptosis induced by I/R injury. Furthermore, bioinformatics analysis, luciferase activity and western blot assay proved that Gadd45beta (growth arrest DNA damage-inducible gene 45beta) was a direct target gene of miR-206. In addition, the expression of pro-apoptotic-related genes, such as p53, Bax and cleaved caspase3, was decreased in association with the down-regulation of Gadd45beta. In summary, this study demonstrates that miR-206 could protect against myocardial I/R injury by targeting Gadd45beta.

        Comparison of the main compounds in Fuding white tea infusions from various tea types

        The purpose of this study was to compare the main components, and particularly catechins, caffeine, theanine, free amino acids, and water extracts, in Bai Hao Yin Zhen, Bai Mu Dan, and Shou Mei Fuding white tea infusions brewed at different temperatures, and their contributions to taste were estimated by dose-over-threshold value. Infusion temperature had a distinct effect on the main components extracted, and 100 C was found to be optimal for extracting catechins and caffeine. However, the effect of temperature on theanine, free amino acids, and water extracts varied with tea type. Bai Mu Dan and Shou Mei yielded a higher content of the major compounds than did Bai Hao Yin Zhen. Thus, infusion temperature had a large effect on extracting the main compounds, and the differences in content between the three white teas presumably reflected differences in the harvest time, processing method or leaf shape.
