<P>Transfection with either hypoxia-inducible factor-1α (HIF-1α) or heme oxygenase-1 (HO-1) gene can induce neovascularization in ischemic tissues. Although expression of transfected HIF-1α gene occurs rapidly, the expressed HIF-1α protein degrades quickly, limiting its therapeutic efficacy. Meanwhile, expressed HO-1 protein does not rapidly undergo degradation, but gene expression occurs a couple of days after transfection, resulting in apoptosis and a delay in angiogenesis in ischemic tissues at the incipient period of HO-1 gene transfection. We hypothesize that combined delivery of HIF-1α and HO-1 gene will enhance antiapoptosis and neovascularization in ischemic tissue compared with HIF-1α or HO-1 single-gene therapy. To test this hypothesis, ischemic mouse hindlimbs were treated with HIF-1α and/or HO-1 gene therapy. The combined gene therapy proved superior to both single-gene therapies, resulting in rapid expression of HIF-1α gene and long-term maintenance of expressed HO-1 protein. The apoptosis in the ischemic region was significantly less, and angiogenic growth factor secretion and angiogenesis were greater in the combined gene therapy than in either of the single-gene therapies. Our results suggest that a combined gene therapy of HIF-1α and HO-1 enhances the transfection of both genes and improves angiogenesis compared with either single-gene therapy.</P>

The package of secondary battery is made of material, which stands acid-resistivity because it contains strong acid electrolyte in it. The characteristics of package in secondary battery have been investigated using peel strength test and electrolyte dipping test. When designing the battery cell package, three layer(Nylon66, aluminum, PP + PE copolymer) are used. In peel strength test, experiment results of Crome surface treatment have a superior advantage of adhesives force and anti-leakage characteristics. Also, it is found that polyolefin and polypropylene have good quality in electrolyte dipping test.
광조사 강도가 복합레진의 중합반응속도에 미치는 영향에 관한 실시간 체적측정법을 이용한 연구

Objectives The aim of this study is to evaluate the effect of light intensity variation on the polymerization rate of composite resin using IB system (the experimental equipment designed by DR. IB Lee) by which real-time volumetric change of composite can be measured. Methods Three commercial composite resins [Z100(Z!), AeliteFil(AF), SureFil(SF)] were photopolymerized with Variable Intensity Polymerizer unit (Bisco, U.S.A.) under the variable light intensity (75/150/225/300/375/450mW^2) during 20 sec. Polymerization shrinkage of samples was detected continuously by IB system during 110 sec and the rate of polymerization shrinkage was obtained by its shrinkage data. Peak time(P.T.) showing the maximum rate of polymerization shrinkage was used to compare the polymerization rate. Results Peak time decreased with increasing light intensity(p<0.05). Maximum rate of polymerization shrinkage increased with increasing light intensity(p<0.05). Statistical analysis revealed a significant positive correlation between peak time and inverse square root of the light intensity (AF:R=0.965, Z1:R=0.974, SF:R=0.927). Statistical analysis revealed a significant negative correlation between the maximum rate of polymerization shrinkage and peak time(AF:R=-0.933, Z1:R=-0.892, SF:R=-0.883), and a significant positive correlation between the maximum rate of polymerization shrinkage and square root of the light intensity(AF:R=0.988, Z1:R=0.974, SF:R=0.946). Discussion and Conclusions The polymerization rate of composite resins used in this study was proportional to the square root of light intensity. Maximum rate of polymerization shrinkage as well as peak time can be used to compare the polymerization rate. Real-time volume method using IB system can be a simple, alternative method to obtain the polymerization rate of composite resins.

Despite the use of aggressive local and systemic treatment including autologous stem cell transplantation in multiple myeloma, extramedullary recurrences are common and the prognosis of these patients is poor. Many novel drugs such as thalidomide, lenalidomide and bortezomib improve the response of treatment of multiple myeloma, but some reports failed to describe thalidomide has effect in extramedullary plasmacytoma. Recent data report on the successful treatment plasmacytomas with bortezomib in patients with advanced multiple myeloma. We treated 4 relapsed or refractory extramedullary plasmacytomas with bortezomib at our institution. We recognized all these extramedullary plasmacytomas decreased and showed more than partial response. This report lends support to the efficacy of bortezomib in the treatment of plasmacytoma and describes the safe use of bortezomib. Responses may, however, be of short duration. Therefore, despite our limited experience, we propose that bortezomib may be considered a therapeutic option for such patients who have risk of radiation therapy

At present, the standard for installing evacuation equipment that enables occupants to evacuate on their own is determined by the use of a building and its floor area. However, according to the “Installation Maintenance and Safety Control of Fire-Fighting Systems Act” and “Fire Safety Standards for evacuation equipment”, the installation standard for evacuation equipment mentions only the minimum number of equipment to be installed based on the floor area. Thus, this installation standard fails to reflect the corresponding number of occupants in a building and the effectiveness of the evacuation equipment. Thus, there is a high probability that most of the evacuation equipment installed based on the “one-size-fits-all” legal regulation will not guarantee the safety of occupants in case of a fire. To address this, the performance evaluation of the effectiveness of evacuation and the distribution ratio of equipment per occupant needs to be prioritized. Therefore, we conduct mock experiments on the descending lifeline and elevating evacuation equipment to analyze their efficiencies. Moreover, we propose a model for calculating the evacuation time for the descending lifeline and elevating evacuation equipment based on the experimental results. Furthermore, we conduct effectiveness evaluation by using the evacuation equipment. The results show that most people cannot be evacuated using the evacuation equipment. That is, evacuation equipment was determined to be insufficient for ensuring safety as per the current standard. Therefore, the installation of evacuation equipment according to its efficiency and capacity is considered necessary.

The Efficacy of Bone Morphogenetic Protein‐2 Depends on Its Mode of Delivery

<P><B>Abstract</B></P><P>Bone morphogenetic protein‐2 (BMP‐2) induces bone regeneration in a dose‐dependent manner, with higher doses of BMP‐2 inducing greater bone formation. Previously, we showed that long‐term delivery of BMP‐2 provides better ectopic bone formation than short‐term delivery of an equivalent dose. In the present study, we investigated the efficacy of orthotopic bone formation over a range of BMP‐2 doses, using different delivery modes. Heparin‐conjugated poly(lactic‐co‐glycolic acid) nanospheres suspended in fibrin gel were used as a long‐term delivery system, and fibrin gel was used as a short‐term delivery system. Different doses of BMP‐2 were delivered to mouse calvarial defects using either long‐term or short‐term delivery systems. Eight weeks after treatment, bone regeneration was evaluated by histomorphometry. For both delivery systems, bone regeneration increased as the BMP‐2 dose increased up to 1 µg and did not increase beyond this dose. Importantly, at BMP‐2 doses higher than 1 µg, long‐term delivery resulted in much greater bone formation than short‐term delivery. This study shows that long‐term delivery of BMP‐2 is more effective at enhancing orthotopic bone formation than short‐term delivery over a range of doses.</P>

Objective : To determine the presence of specific immune recognition of type II collagen (CII) and its immunodominant epitope CII (255-274) in patients with rheumatoid arthritis(RA). Methods : T cell proliferative responses to bovine CII and synthetic peptide encompassing CII (255-274), by peripheral blood mononuclear cells (PBMC) and synovial fluid mononuclear cells (SFMC) from RA patients, and PBMC from osteoarthritis (OA) patients and healthy controls were assayed by mixed lymphocyte culture. Results : Stimulation index (SI) and the number of positive T cell responses (SI≥2) to CII were higher in RA patients (n=106) than in OA patients(n=26) and healthy controls (n=34). T cell responses to CII (255-274) were also enhanced in RA and correlated well with those to CII. In SFMC, positive responses to CII using SFMC were stronger and more prevalent than peripheral responses. SI and positive responses to CII were higher in early RA than in late RA. IgG antibodies to CㆍII in SF inversely correlated with T cell responses to CII. Conclusion : T cell responses to CII or CII(255-274) were enhanced in RA, especially in early disease. Synthetic peptide CII (255-274) as well as CII could be recognized as immunogenic antigens by T cells, particularly those in SF. These observations suggest that CII reactive T cells play an important role in the pathogenesis of RA. Peripheral tolerance induction using CII(255-274) might be useful in the treatment of RA.
