        Investigation on Pharmacokinetics, Tissue Distribution and Excretion of a Novel Anticancer Platinum Compound by Inductively Coupled Plasma Mass Spectrometry after Intravenous Administration to Rats

        SM54111 [cis-3, 5-diisopropylsalylic cyclohexanodiaminoplatinum (Ⅱ), Saliplatin] is a novel platinum compound with encouraging anticancer effect. In order to get more useful information in multiple species for the security evaluation, dosage design, and drug delivery design, its pharmacokinetic behaviors in rats after intravenous administration were investigated in this study. Based on its pharmacokinetics in plasma, the distribution of SM54111 in heart, liver, spleen, lung, kidney, brain, adipose, testicle, or ovaries of rats sampled at 0.5 h, 1 h, and 3 h after intravenous administration were determined utilizing inductively coupled plasma mass spectrometry (ICP-MS). Its amounts in urine and feces were determined at 8 sampling times till 96 h as well. It was proved that SM54111 fitted open two-compartment model in rats, with wide distribution in tissues and slow excretion. Its reasonable pharmacokinetic properties in rats make this novel platinum compound worthy of further research and development.

      • Short Low Concentration Cisplatin Treatment Leads to an Epithelial Mesenchymal Transition-like Response in DU145 Prostate Cancer Cells

        Background: Prostate cancer is one of the main causes of cancer death, and drug resistance is the leading reason for therapy failure. However, how this occurs is largely unknown. We therrfore aimed to study the response of DU145 cells to cisplatin. Materials and Methods: Du145 prostate cancer cells were treated with a low dose of cisplatin for 24 h and cell viability and number were determined by MTT assay and trypan blue exclusion assay, respectively. The real time polymerase chain reaction (PCR) was used to assess responses to cisplatin treatment. Results: After 24h $2{\mu}g/ml$ treatment did not result in significant reduction in cell viability or number. However, it led to enhanced cancer cell invasiveness. E-cadherin mRNA was reduced, and vimentin, Snail, Slug, metalloproteinase 9 (MMP9) mRNA expression increased significantly, a feature of epithelial-mesenchymal transition (EMT). Conclusions: Short time low concentration cisplatin treatment leads to elevated invasiveness of DU145 cancer cells and this is possibly due to EMT.

        Flavonoid Glycosides and Potential Antivirus Activity of Isolated Compounds from the Leaves of Eucalyptus citriodora

        The extraction and solvent partition of the leaves ofEucalyptus citriodora, and repeated column chromatography for n-BuOH fraction yielded a new flavonoid glycoside, citrioside C (1),along with three known flavonoid glycosides (2-4). The latter wereidentified with kaempferol-3-O-β-D-glucopyranosyl (12)-α-L-rhamnoside(2), kaempferol-3-O-α-L-rhamnoside (3), and quercetin-3-O-α-Lrhamnoside(4). Their chemical structures were identified on thebasis of spectroscopic data analyses including NMR, MS, UV, andIR. All constitutents were isolated for the first time from the leavesof Eucalyptus citriodora. The potential antivirus activity of all theisolated compounds was evaluated. Compound 4 showed potentantiviral activity against respiratory syncytial virus with 50%inhibition concentration (IC50) value of 1.9 μg/mL and selectiveindex value of 9.8.

        An Aqueous Extract of Yunnan Baiyao Inhibits the Quorum-Sensing-Related Virulence of Pseudomonas aeruginosa

        Yunnan Baiyao is a famous Chinese medicine that has long been directly applied to wounds to reduce bleeding, pain, and swelling without causing infection. However, little is known about its ability to prevent infection. The present study aimed to assess in vitro the anti-virulence activity of an aqueous extract of Yunnan Baiyao (YBX) using Pseudomonas aeruginosa as a pathogenic model. We found that a sub-MIC (2.5 mg/ml) of YBX can efficiently interfere with the quorum-sensing (QS) signaling circuit. Real-time polymerase chain reaction analysis showed that a sub-MIC of YBX downregulated the transcriptions of lasR, lasI, rhlR, and rhlI,which resulted in global attenuation of QS-regulated virulence activities, such as biofilm formation, and secretion of LasA protease, LasB elastase and pyocyanin. Further, YBX reduced the motility of P. aeruginosa related to QS, and impaired the formation of biofilms. These results suggest that YBX may possess global inhibitory activity against the virulence of P. aeruginosa and that YBX may also exhibit antimicrobial activity in vivo. The present study suggests that Yunnan Baiyao represents a potential source for isolating novel, safe, and efficacious antimicrobial agents.

      • Cellular and ultrastructural changes in the seedling roots of upland rice (Oryza sativa) under the stress of two allelochemicals from Ageratina adenophora

        Two sesquiterpene-derivative compounds, 4,7-dimethyl-1-(propan-2-ylidene)-1,4,4a,8atetrahydronaphthalene-2,6(1H, 7H)-dione (DTD) and 6-hydroxy-5-isopropyl-3,8-dimethyl-4a,5,6,7,8,8a-hexahydronaphthalen-2(1H)-one (HHO), are the major putative allelochemicals of the aqueous leachates of Ageratina adenophora.A laboratory experiment was conducted, using the hydroponic method, to evaluate the cellular and ultrastructural changes in the seedling roots of upland rice under the stress of DTD and HHO. The subsequent changes were observed in the treated upland rice roots in comparison with their controls. The scanning electron microscopy results showed that the DTD-treated root tip cells turned into an irregular arrangement and shape and that most of them were wizened, with a poor cytoplasm. In the HHO treatment, the root tips had many irregularly shaped cells, with a greater number of sloughing cells, as well as short, wide cells that resulted in spherical and wider, but shorter, roots. At the ultrastructural level, DTD and HHO induced irregularly shaped and lobed nuclei, increased cytoplasmic vacuolation, reduced ribosome density and dictyosomes, and a reduced number of mitochondria in the cells, which indicated limited protein transportation and a reduced capability to export substances for cell development and growth in the upland rice seedling roots. The overall effect of HHO on the upland rice seedlings was more pronounced than that of DTD.

        『붉은 여왕』 에서 체현된 문화적 헤게모니와 오리엔탈리즘

        소설 『붉은 여왕(The Red Queen)』은 예술적으로나 사상적 측면에서 높은 평가를 받고 있다. 하지만 그 이면에 내포되어 있는 문화적 헤게모니와 오리엔탈리즘은 간과할 수 없는 문제로 대두된다. 이 소설의 저자는 수차례나 자신의 문화상대주의 입장을 표명하고 소설 속에서도 그 뜻을 분명히 드러냄으로, 표면적인 서술에 독자들이 쉽게 속는 경우가 있었다. 이 소설은 조선역사에서의 왕비- 홍옥련의 유령을 서술자로 설정하여 동양화된 새로운 오리엔탈리즘을 제시하는 책략으로 독자들을 기만하는 수법을 사용하였다. 때문에 독자들은 이 작품을 접할 때 그 속에 숨어 있는 문화적 헤게모니와 오리엔탈리즘에 대해서는 분명한 인식과 경계심을 갖고 좀 더 날카롭게 접근해야 한다. Whether in terms of artistry or of thought the novel The Red Queen has won high praise. However, under the aura of praise in its depth there is implied cultural hegemony and orientalism which should not be neglected. The author has declared her position of cultural relativism on many different occasions and the novel shows such a stand many times. Readers may be easily blinded by such endeavors. Since the choice of the late Korean princess, Lady Hyegyong as the narrator to realize self-orientalizing, a strategy of new orientalism, is more deceptive, readers should be more alert to such works so that they will not lose critical voices.

Compound 4 showed potent antiviral activity against respiratory syncytial virus with 50% inhibition concentration ($IC_{50}$) value of $1.9{\mu}g/mL$ and selective index value of 9.8.

      • Research on Aggregation and Propagation of Self-Similar Traffic in Satellite Network

        It has already been confirmed that the traffic in high-speed terrestrial network presents self-similarity, but there is little research on self-similarity of traffic in satellite network. Considering time-varying network topology and link status, this paper analyzes the aggregation and propagation of self-similar traffic between nodes in satellite network. Furthermore, a sort of special network node called ground gateway is modeled, based on which the characteristics of the output traffic that the input traffic from terrestrial network passes gateway into satellite network are analyzed. Theoretically analyses demonstrate that after aggregation and propagation between satellite nodes, traffic is still self-similar, and the self-similarity of the output traffic generated by gateway from terrestrial network to satellite network is more often than not weakened.

        Single Nucleotide Polymorphisms of the GnRHR Gene Associated with Reproductive Traits of Japanese Flounder (Paralichthys olivaceus)

        Gonadotropin-releasing hormone receptor (GnRHR) gene is expressed at the anterior pituitary gland and plays a key role in gonad development. This study aimed to investigate molecular genetic characteristics of the GnRHR gene and elucidate the effects of single nucleotide polymorphisms (SNPs) of GnRHR gene on sex steroid level in Japanese flounder (Paralichthys olivaceus). We used polymerase chain reaction single-strand conformation polymorphism (PCR-SSCP) and sequencing of the GnRHR gene in 75 individuals. We identified three SNPs in the GnRHR gene: P1 locus (C759A and C830T) in the coding region of exon2 which were both linked together and P2 locus (G984T) in the coding region of exon3, which added a new transcript factor (ADR1) and a new methylation site (CG). Only C830T of P1 leads to amino acid changes Thr266Ile. Statistical analysis showed that P1 was significantly associated with $17{\beta}$-estradiol ($E_2$) level (p<0.01) and gonadosomatic index (GSI) (p<0.05). Individuals with genotype BB of P1 had significantly higher serum $E_2$ levels (p<0.01) and GSI (p<0.05) than those of genotype AA or AB. Another SNP, P2, synonymous mutation, was significantly associated with GSI (p<0.05). Individuals with genotype AB of P2 had significantly higher GSI (p<0.05) than that of genotype AA. In addition, there was a significant association between one diplotype based on three SNPs and reproductive traits. The genetic effects for both serum $E_2$ level and GSI of diplotype D4 were super diplotypes (p<0.05). These results suggest that the SNPs in Japanese Flounder GnRHR are associated with $E_2$ level and GSI.

        Pesticide-induced changes in fecundity and rice stripe virus transmission ability in Laodelphax striatellus (Homoptera: Delphacidae)

        The widespread use of pesticides in rice has caused the resurgence of the small brown planthopper (SBPH), Laodelphax striatellus. In addition to inciting damage by feeding on phloem cells, SBPH also functions as a vector for rice stripe virus (RSV), which can cause serious yield losses. In general, studies focused on pesticide-induced stimulation of SBPH populations have focused on the insects; little information is available on the impact of pesticides on RSV transmission by SBPH. The present study investigated the effects of two pesticides (validamycin and triazophos) on the fecundity and RSV transmission ability of SBPH. Our results demonstrated that the fecundity of non-viruliferous (naïve) or viruliferous SBPH was enhanced by exposure to triazophos at the LC20 and validamycin at 200 mg L−1. Meanwhile, the increased number of eggs produced by viruliferous females treated with pesticides was larger than those from naïve females. Exposure to validamycin caused an increase in horizontal transmission of RSV; furthermore, vertical viral transmission rates of SBPH were significantly increased by exposure to triazophos. The present study provides valuable information for understanding the mechanisms underlying the resurgence of SBPH and subsequent outbreaks of RSV.
