      • Expression of PGDH Correlates with Cell Growth in Both Esophageal Squamous Cell Carcinoma and Adenocarcinoma

        Esophageal cancer represents the fourth most common gastrointestinal cancer and generally confers a poor prognosis. Prostaglandin-producing cyclo-oxygenase has been implicated in the pathogenesis of esophageal cancer growth. Here we report that prostaglandin dehydrogenase, the major enzyme responsible for prostaglandin degradation, is significantly reduced in expression in esophageal cancer in comparison to normal esophageal tissue. Reconstitution of PGDH expression in esophageal cancer cells suppresses cancer cell growth, at least in part through preventing cell proliferation and promoting cell apoptosis. The tumor suppressive role of PGDH applies equally to both squamous cell carcinoma and adenocarcinoma, which enriches our understanding of the pathogenesis of esophageal cancer and may provide an important therapeutic target.

        A Spodoptera exigua serpin is involved in the immunity of hemolymph induced by Heliothis virescens ascovirus 3h

        Serine protease inhibitors (serpins) are a broadly distributed protease superfamily in various organisms, which participate in the negative regulation of melanization in insects. Our pervious study demonstrated that induced melanization by Heliothis virescens ascovirus 3h is helpful for HvAV-3h viral replication. In this study, the serpin-2.1 like gene from Spodoptera exigua (Seserpin-2.1 like) which is a specific serpin after HvAV-3h infection was identified. Seserpin-2.1 like has the typical characteristics of serpin proteins family. Seserpin-2.1 like widely distributed in various tissues (including hemolymph, fat body, midgut, cuticle and head), and expressed at a higher level in fat body, hemolymph and head. At different developmental stages, it tends to express at eggs and 5th instar larvae. Seserpin-2.1 like shows special phenomenon for HvAV-3h infection. On the one hand, the transcription level of Seserpin-2.1 like in hemolymph was significantly upregulated after HvAV-3h compared to other immune challenges Gram-positive bacteria (Bacillus thuringiensis), Gram-negative bacteria (Escherichia coli), and Baculovirus (Autographa californica nucleopolyhedrovirus). On the other hand, its expression disappeared during 72 to 120 h post infection. In summary, our results suggest that Seserpin-2.1 like may be involved in innate immunity of S. exigua after infection with HvAV-3h. And HvAV-3h may inhibit the expression of Seserpin-2.1 like to promote the melanization of host larvae for viral replication.

        Morphology and ultrastructure of the male reproductive system of the jewel wasp, Nasonia vitripennis (Walker) (Hymenoptera: Pteromalidae)

        The genital systems of insects are undoubtedly of great key for the success of mating and population reproduction. In the meantime,morphological and ultrastructural characteristics of the reproductive systems also provide valuable evidences for the studies of taxonomy, reproductive biology and evolutionary biology.More previously, the reproductive systemof female Nasonia vitripennis was already described. To complement the information of reproductive systems from this species, the male reproductive system was here dissected and characterized by light microscopy and transmission electron microscopy. Results showed that the system follows the pattern of most Hymenoptera species: paired testes with one follicle, deferent ducts, seminal vesicles, accessory glands and a fused ejaculatory duct ultimately connected to the external aedeagus. Histologically, the testes, seminal vesicles and accessory glands are all composed of three portions, all of which include epithelial and luminal regions. In addition, both lipidic inclusions and nuclei are presented in the three organs with variable sizes and shapes. Spermatozoa are observed in the testes and seminal vesicles, and also vary considerably in size and shape, suggesting different phases of spermatogenesis.Mitochondria are abundant in seminal vesicles and accessory glands. Notably, membranous inclusions and Golgi complexes are found only in seminal vesicles, whereas secretory granules are presented only in accessory glands, being indicative of organ or age specificity. Together, this study complements the information of the reproductive systems from N. vitripennis, and provides an extensive resource for taxonomy, reproductive biology and evolutionary biology.

      • On/off Switch Mediated by Exo+ Polymerases: Experimental Analysis for Its Physiological and Technological Implications

        The potential physiological role and technological application of the premature termination of DNA polymerization through the off-switch of exo+ polymerases were studied using 3' phosphorothioate-modified or unmodified primers with single base mismatch distal to the 3' terminus. With exonuclease-digestible unmodified primers, a gradient premature termination of DNA polymerization was observed when amplified with exo+ polymerases. With 3' allele specific phosphorothioate-modified primers, an efficient off-switch effect occurred in the discrimination of a single nucleotide polymorphism when directly using genomic DNA. Clearly, the off-switch of exo+ polymerases is useful in biomedical research.

      • Let-7c Inhibits NSCLC Cell Proliferation by Targeting HOXA1

        Objective: The aim of the present study was to explore mechanisms by which let-7c suppresses NSCLC cell proliferation. Methods: The expression level of let-7c was quantified by qRT-PCR. A549 and H1299 cells were transfected with let-7c mimics to restore the expression of let-7c. The effects of let-7c were then assessed by cell proliferation, colony formation and cell cycle assay. Mouse experiments were used to confirm the effect of let-7c on tumorigenicity in vivo. Luciferase reporter assays and Western blotting were performed to identify target genes for let-7c. Results: HOXA1 was identified as a novel target of let-7c. MTS, colony formation and flow cytometry assays demonstrated that forced expression of let-7c inhibited NSCLC cell proliferation by inducing G1 arrest in vitro, consistent with inhibitory effects induced by knockdown of HOXA1. Mouse experiments demonstrated that let-7c expression suppressed tumorigenesis. Furthermore, we found that let-7c could regulate the expression of HOXA1 downstream effectors CCND1, CDC25A and CDK2. Conclusions: Collectively, these results demonstrate let-7c inhibits NSCLC cell proliferation and tumorigenesis by partial direct targeting of the HOXA1 pathway, which suggests that restoration of let-7c expression may thus offer a potential therapeutic intervention strategy for NSCLC.

        High-Definition Computed Tomography for Coronary Artery Stent Imaging: a Phantom Study

        Objective: To assess the performance of a high-definition CT (HDCT) for imaging small caliber coronary stents (≤ 3 mm) by comparing different scan modes of a conventional 64-row standard-definition CT (SDCT). Materials and Methods: A cardiac phantom with twelve stents (2.5 mm and 3.0 mm in diameter) was scanned by HDCT and SDCT. The scan modes were retrospective electrocardiography (ECG)-gated helical and prospective ECG-triggered axial with tube voltages of 120 kVp and 100 kVp, respectively. The inner stent diameters (ISD) and the in-stent attenuation value (AVin-stent) and the in-vessel extra-stent attenuation value (AVin-vessel) were measured by two observers. The artificial lumen narrowing (ALN = [ISD - ISDmeasured]/ISD) and artificial attenuation increase between in-stent and in-vessel (AAI = AVin-stent - AVin-vessel) were calculated. All data was analyzed by intraclass correlation and ANOVA-test. Results: The correlation coefficient of ISD, AVin-vessel and AVin-stent between the two observers was good. The ALNs of HDCT were statistically lower than that of SDCT (30 ± 5.7% versus 35 ± 5.4%, p < 0.05). HDCT had statistically lower AAI values than SDCT (15.7 ± 81.4 HU versus 71.4 ± 90.5 HU, p < 0.05). The prospective axial dataset demonstrated smaller ALN than the retrospective helical dataset on both HDCT and SDCT (p < 0.05). Additionally, there were no differences in ALN between the 120 kVp and 100 kVp tube voltages on HDCT (p = 0.05). Conclusion: High-definition CT helps improve measurement accuracy for imaging coronary stents compared to SDCT. HDCT with 100 kVp and the prospective ECG-triggered axial technique, with a lower radiation dose than 120 kVp application, may be advantageous in evaluating coronary stents with smaller calibers (≤ 3 mm).         Flavonoid Glycosides and Potential Antivirus Activity of Isolated Compounds from the Leaves of Eucalyptus citriodora

        The extraction and solvent partition of the leaves ofEucalyptus citriodora, and repeated column chromatography for n-BuOH fraction yielded a new flavonoid glycoside, citrioside C (1),along with three known flavonoid glycosides (2-4). The latter wereidentified with kaempferol-3-O-β-D-glucopyranosyl (12)-α-L-rhamnoside(2), kaempferol-3-O-α-L-rhamnoside (3), and quercetin-3-O-α-Lrhamnoside(4). Their chemical structures were identified on thebasis of spectroscopic data analyses including NMR, MS, UV, andIR. All constitutents were isolated for the first time from the leavesof Eucalyptus citriodora. The potential antivirus activity of all theisolated compounds was evaluated. Compound 4 showed potentantiviral activity against respiratory syncytial virus with 50%inhibition concentration (IC50) value of 1.9 μg/mL and selectiveindex value of 9.8.

        Gamma and X-ray irradiation as a phytosanitary treatment against various stages of Planococcus lilacinus (Hemiptera: Pseudococcidae)

        The cacao mealybug, Planococcus lilacinus Cock, is an important quarantine pest. Infested commodity should be subject to appropriate phytosanitary treatment, while irradiation is recommended for the cacao mealybug. Radio-tolerance comparison tests were conducted on the crawler, nymphs, and adult females of P. lilacinus at the X-ray radiation doses of 40, 80, and 120 Gy, respectively. The results showed that irradiation had a strong effect on preventing of development and reproduction; the adult female stage was identified as the most tolerant. During the following dose–response tests, among young and late females X-ray-irradiation (20–100 Gy), the late females were most tolerant when preventing F 1 generation 2nd instars emergence was used as the evaluation criterion. Minimum absorbed dose and its 95 % fiducial limits to provide probit 9 efficacy at 95 % confidence level (100 % mortality/inhibition in an estimated population of 93,616 individuals) were 131.5 Gy (122.5, 142.6 Gy) and 144.4 Gy (132.7, 159.4 Gy), estimating from the probit analysis on dose-mortality data of 1–30 and 1–10-day-old neonates laid by late females, respectively. In the large-scale confirmatory tests, a total of estimating 97,384 late females of P. lilacinus rearing on the pumpkins fruits were irradiated with gamma-ray at the target dose of 135 or 145 Gy (measured doses 126.1–163.0 Gy), which resulted in no F 1 generation 2nd nymphs developing during a 6-week post-treatment period. The treatment efficacy calculated is 99.9969 % at the 95 % confidence level. Therefore, a minimum absorbed dose of 163.0 Gy is recommended for phytosanitary treatment of P. lilacinus in infested commodity.
