        Construction of a High-Quality Yeast Two-Hybrid Library and Its Application in Identification of Interacting Proteins with Brn1 in Curvularia lunata

        Curvularia lunata is an important maize foliar fungal pathogen that distributes widely in maize growing area in China, and several key pathogenic factors have been isolated. An yeast two-hybrid (Y2H) library is a very useful platform to further unravel novel pathogenic factors in C. lunata. To construct a high-quality full length-expression cDNA library from the C. lunata for application to pathogenesis-related protein-protein interaction screening, total RNA was extracted. The SMART (Switching Mechanism At 5' end of the RNA Transcript) technique was used for cDNA synthesis. Double-stranded cDNA was ligated into the pGADT7-Rec vector with Herring Testes Carrier DNA using homologous recombination method. The ligation mixture was transformed into competent yeast AH109 cells to construct the primary cDNA library. Eventually, a high qualitative library was successfully established according to an evaluation on quality. The transformation efficiency was about $6.39{\times}10^5$ transformants/$3{\mu}g$ pGADT7-Rec. The titer of the primary cDNA library was $2.5{\times}10^8cfu/mL$. The numbers for the cDNA library was $2.46{\times}10^5$. Randomly picked clones show that the recombination rate was 88.24%. Gel electrophoresis results indicated that the fragments ranged from 0.4 kb to 3.0 kb. Melanin synthesis protein Brn1 (1,3,8-hydroxynaphthalene reductase) was used as a "bait" to test the sufficiency of the Y2H library. As a result, a cDNA clone encoding VelB protein that was known to be involved in the regulation of diverse cellular processes, including control of secondary metabolism containing melanin and toxin production in many filamentous fungi was identified. Further study on the exact role of the VelB gene is underway.

        Involvement of a Polyketide Synthetase ClPKS18 in the Regulation of Vegetative Growth, Melanin and Toxin Synthesis, and Virulence in Curvularia lunata

        The clpks18 gene was first cloned and identified in Curvularia lunata. It contains 6571 base pairs (bp) and an 6276 bp open reading frame encoding 2091 amino acids. The ClPKS18 deletion mutant displayed an albino phenotype, and almost lost the ability to product 5-(hydroxymethyl) furan-2-carboxylate (M5HF2C) toxin, implying that clpks18 gene in C. lunata is not only involved in 1,8-dihydroxynaphthalene melanin synthesis, but also relatively associated with M5HF2C toxin biosynthesis of the pathogen. The pathogenicity assays revealed that ${\Delta}ClPKS18$ was impaired in colonizing the maize leaves, which corresponds to the finding that ClPKS18 controls the production of melanin and M5HF2C in C. lunata. Results indicate that ClPKS18 plays a vital role in regulating pathogenicity of in C. lunata.

        The Genotype-Based Morphology of AldosteroneProducing Adrenocortical Disorders and Their Association with Aging

        Primary aldosteronism (PA) is the most common cause of secondary hypertension, and is associated with an increased incidence of cardiovascular events. PA itself is clinically classified into the following two types: unilateral PA, mostly composed of aldosteroneproducing adenoma (APA); and bilateral hyperaldosteronism, consisting of multiple aldosterone-producing micronodules (APMs) and aldosterone-producing diffuse hyperplasia. Histopathologically, those disorders above are all composed of compact and clear cells. The cellular morphology in the above-mentioned aldosterone-producing disorders has been recently reported to be closely correlated with patterns of somatic mutations of ion channels including KCNJ5, CACNA1D, ATP1A1, ATP2B3, and others. In addition,in non-pathological adrenal glands, APMs are frequently detected regardless of the status of the renin-angiotensin-aldosterone system (RAAS). Aldosterone-producing nodules have been also proposed as non-neoplastic nodules that can be identified by hematoxylin and eosin staining. These non-neoplastic CYP11B2-positive nodules could represent possible precursors of APAs possibly due to the presence of somatic mutations. On the other hand, aging itself also plays a pivotal role in the development of aldosterone-producing lesions. For instance, the number of APMs was also reported to increase with aging. Therefore, recent studies indicated the novel classification of PA into normotensive PA (RAAS-independent APM) and clinically overt PA.

        Analysis on the Production Efficiency of Private Industrial Enterprises in 31 Provinces of China

        Purpose: The purpose of this study is to understand the status quo of production efficiency in private industrial enterprises above designated scale in China s 31 provinces (including municipalities directly under the central government, autonomous regions) (hereinafter referred to as China s 31 provinces). Research design, data and methodology: Find out the factors affecting the development of production efficiency in private industrial enterprises, using DEA, Data Envelopment Analysis and Malmquist index analysis, build the evaluation model of production efficiency in private industrial enterprises, and analyze the data of China s 31 provinces private industrial enterprises in 2015-2019. Results: The research results show that the production efficiency of private industrial enterprises in China is improving on the whole. Although the total factor productivity has decreased slightly, the overall efficiency and pure technical efficiency have increased significantly. Conclusions: The conclusion of this study can provide reference for Chinese private industrial enterprises to improve production efficiency and make development plan. The limitation of this paper lies in the fact that the private industrial enterprises in inefficient provinces have not been given specific improvement plans.

        Research on Indoor Localization Algorithm Based on SVM

        In order to solve the problem of a large positioning error, low refresh rate and high-power consumption of UWB in an indoor high-density environment. This paper proposes a UWB indoor positioning method based on SVM (support vector machine), which transforms the positioning problem into the solution of the multi-classification problem. The SVM model based on TDOA (time difference of arrival) is established, and the one-to-one classification model is used to solve the coordinate value quickly. Experimental results show that the proposed algorithm has higher accuracy and less time in indoor positioning. The algorithm proposed in this paper has more application prospects.

        Dominant failure modes identification and structural system reliability analysis for a long-span arch bridge

        Failure of a redundant long-span bridge is often described by innumerable failure modes, which make the structural system reliability analysis become a computationally intractable work. In this paper, an innovative procedure is proposed to efficiently identify the dominant failure modes and quantify the structural reliability for a long-span bridge system. The procedure is programmed by ANSYS and MATLAB. Considering the correlation between failure paths, a new branch and bound operation criteria is applied to the traditional stage critical strength branch and bound algorithm. Computational effort can be saved by ignoring the redundant failure paths as early as possible. The reliability of dominant failure mode is computed by FORM, since the limit state function of failure mode can be expressed by the final stage critical strength. PNET method and FORM for system are suggested to be the suitable calculation method for the bridge system reliability. By applying the procedure to a CFST arch bridge, the proposed method is demonstrated suitable to the system reliability analysis for long-span bridge structure.

        Novel RGO and Concave Cube Cu2O Co-modified BiVO4 Nanosheets with Enhanced Photocatalytic and Surface Adsorption Performances of Tetracycline

        A novel ternary Cu2O/BiVO4/RGO photocatalyst is successfully constructed by hydrothermal and evaporation-induced method, and it exhibits superior photocatalytic performance for degradation tetracycline (TC). Meanwhile, the visible light absorption range of composite photocatalyst is effectively broadened by the formation of heterojunction with narrow band gap semiconductor Cu2O. And the separation efficiency of the photogenerated electron–hole pairs is significantly enhanced by the synergistic effect of Cu2O and RGO. More importantly, the adsorption of TC by ternary Cu2O/BiVO4/RGO possesses high adsorption capacity, which is 23.73 times higher than that of pure BiVO4. Additionally, the possible reaction mechanism is clearly revealed by radical trapping experiment, electron spin-resonance (ESR) spectroscopy. This work provides a new insight to design a photocatalyst with excellent adsorption to remove organic contaminants in water.

        The Effects of Omni-channel Characteristics on Chinese Consumers' Perceived Cost and Purchase Intention

        Purpose: As a new retail model, omni-channel is different from the previous single-channel, multi-channel and cross-channel retail models, researching and exploring the impact of its characteristics on consumers' omni-channel shopping intention is beneficial to the industry and enterprise, which can better meet customer needs and improve customers satisfaction and corporate competitiveness. Based on the characteristic of omni-channel, the literature and theory on the intention of purchase through the perceived cost and omni-channel, this paper established the conceptual model of the influence mechanism of Chinese consumer on the intention of purchasing in omni-Channel. Research design, data, and methodology: A total of 293 valid samples were obtained through online questionnaire and offline face-to-face reply, taking consumers who had experienced shopping through omni-channel as subjects, and model reliability and validity test, fit test of the model, model hypothesis verification and path coefficient, mediation effect analysis were conducted by SPSS 20.0 and AMOS 20.0. Results: The empirical analysis shows that the omni-channel characteristics such as immediacy, interactivity and consistency were the main drivers that directly affected consumers' purchase intention. Immediacy and consistency not only directly but also indirectly affected consumers’ omni-channel purchase intentions, while interactivity only directly affected consumers’ omni-channel purchase intention. Among the factors affecting consumers’ omni-channel purchase intention, the effect of consistency was the largest (0.42), followed by immediacy (0.38), and the effect of interactivity was the least (0.17). Perceived cost acted as a mediating variable, and perceived cost negatively affected consumers' purchase intention of omni-channel. Immediacy and consistency of omni-channel affected purchase intention through perceived cost. Interactivity had no indirect impact on the shopping intention, mainly because consumers were used to resort to channels, so perceived cost was not sensitive to omni-channel interactivity. Conclusions: According to the research results, this paper puts forward some countermeasures and Suggestions for the retail enterprises' omni-channel strategy from the three aspects. Increasing the purchase intention by improving consistent products, services, information and experiences. Improving the immediacy level through big data and information technology to meet the needs of customers anytime and anywhere. Reducing consumer perceived costs through improved omni-channel characteristics and improving logistics levels.

        Intelligent identification of mortar void in ballastless slab track using the wheelset acceleration combined with CNN-SVM

        Mortar void is a hidden defect in ballastless slab track difficult to be efficiently identified by traditional detection methods. This paper is dedicated to proposing a new detection method to identify the mortar void position and length using the vehicle response combined with the hybrid convolutional neural network-support vector machine (CNN-SVM) classifier. The vertical wheelset accelerations with different mortar void conditions are collected from a vehicle-track coupled dynamics simulation model. The first components decomposed from wheelset accelerations by local mean decomposition and their envelopes are utilized as the training data due to their sensitivity to mortar void. To improve the identification precision, the scope descent method is proposed to determine the range influenced by mortar void (IMVR) and samples are labeled according to IMVR. Meanwhile, identification results are post processed based on the mortar void characteristics. The results show that over 90 % mortar void conditions with the length of 0.65 m are detected correctly and the identification has a higher precision with the mortar void length greater than 0.95 m. The proposed technology of mortar void detection using the wheelset accelerations with the hybrid CNN-SVM classifier provides reference for engineering application, which is of great significance to relieve the pressure of health monitoring of railway track.
