      • CHEMISTRY : Protective effect of arabinose and sugar beet pulp against high glucose-induced oxidative stress in LLC-PK1 cells

        The protective effects of L-arabinose and sugar beet (SBP) on high glucose-induced oxidative stress were investigated using LLC-PK cells. Under the high glucose-induced cellular oxdative model, the treatment of 45 mM of glucose led to the decrease in cell viability and increase in lipod peroxidation_However. L-arabinose and SBP significantly inhibited the high glucose-induced cytotoxicity and lipid per-oxidation. In addition, the formation of nitric oxide(NO) was increased by the treatment of 45mM glu-cose. while the treatment of L-arabinose or SBP inhibited signigicantly the NO formation compared with high glucose-treated control. The superoxide anion production of groups treated whth L-arabinose or SBP was significantly lower than that of the control treated with high glucose. Futhermore. L-arabinose and SBP elevated the glucose uptake. resulting in lower glucose concentration compared with non-treated control. High glucose levels induced the overexpressions of bax. inducible NO synthase and cyclooxygen-ase-2. but L-arabinose or SBP treatrnent down-regulated the expressions of these genes. Arabinose and SBP also inhibited the expression of nuclear factor-kappa B induced by 45mM glucose in LLC-PK cells. In particular, arabinose exhibited stronger inhibitory activities on high glucose-induced oxdative stress than SBP did. These findings indicate that L-arabinose and SBP are promising antioxidative agents with protective activities against hyperglycemia.

      • 가압볶음 무말랭이 열수 추출물의 항산화 효과

        The antiradical property of hot water extract from dried radish (DR) or dried radish roasted with pressure (DRRP) was investigated in vitro and in LLC-PK1 cell system. The contents of total free amino acid and reducing sugar in DR were decreased by 72.86% and 3.17%, respectively, after pressurized roasting. In vitro test, IC50 for DR and DRRP for DPPH radical scavenging activity were 646.70 and 135.45 μg/mL, 896.10 and 566.98 μg/mL for superoxide anion radical, and 722.26 and 531.84 μg/mL for hydroxy radical, respectively. The radical scavenging effects of DRRP was significantly greater than those for DR (p<0.001). These radical scavenging effects of DR and DRRP were confirmed in LLC-PK1 at which oxidative stresses were induced by superoxide, nitric oxide and peroxynitrite generated in the treatment of pyrogallol, SNP, and SIN-1, respectively. Cell viability was increased in the presence of DR or DRRP, dose dependently (p<0.05), and TBARS formation was decreased. The protective effects of DRRP against oxidative damage in LLC-PK1 were greater than those of DR at the same concentration tested (p<0.05). This superior antiradical activity of DRRP might be due to the products produced during the pressurized roasting in addition to the anti oxidative compounds originally present in the radish. 5-hydroxyl methyl furfural (5-HMF) known as an intermediate product of the maillard reaction was detected in DRRP (0.57 mg/g), but not from DR. In conclusion, daily consumption of DRRP may prevent oxidative damage by retarding oxidative stress.

        가압볶음 무말랭이 열수 추출물의 항산화 효과

        무말랭이 및 가압볶음 무말랭이 열수 추출물의 항산화 효과를 in vitro와 LLC-PK₁ cellular system에서 살펴보았다. 무말랭이 열수 추출물과 가압볶음 무말랭이 열수 추출물의 라디칼 소거능을 IC??로 비교해 보았을 때 DPPH(646.70 vs 135.45 ㎍/mL), superoxide anion(896.10 vs 566.98 ㎍/mL) 및 hydroxyl radical(722.26 vs 531.84 ㎍/mL)에 대한 가압볶음 열수 추출물의 효과가 유의적으로 높은 것으로 나타났다(p<0.001). 이러한 유리기 소거효과는 LLC-PK₁ cell에서 pyrogallol, SNP 및 SIN-1 처리로 superoxide, nitric oxide 및 peroxynitrite를 생성하여 산화스트레스를 유발한 다음 무말랭이 및 가압볶음 무말랭이 열수추출물을 첨가하였을 때 농도 의존적으로 세포생존율이 증가하고, 과산화물 생성량이 감소하여 세포손상을 보호하는 효과가 관찰되었다. 가압볶음 무말랭이 열수 추출물의 산화손상에 대한 보호 효과는 무말랭이 열수추출물에 비해 모든 유리기에서 유의적으로 높았다(p<0.05). 이러한 무말랭이 열수추출물의 항산화효과는 무에 함유되어 있는 함황 물질, 유리아미노산, 배당체 등에 의한 것으로 생각되며 가압볶음 무말랭이 열수 추출물의 효과가 더 높은 이유는 무말랭이에 함유된 유효성분이 가압볶음에 의해 증가되고 더불어 볶음과정 중에서 생성된 maillard 생성물의 항산화성 때문으로 생각된다. 무말랭이의 환원당 및 유리아미노산 함량은 볶음 후 유의적으로 감소하였으며(p<0.05), 이에 반해 maillard 생성물의 중간산물인 5-hydroxymethyl-2-furfural(5-HMF) 함량은 무말랭이 열수 추출물에서는 검출되지 않았던 것이 가압볶음 무말랭이 열수 추출물에서는 0.57 ㎎/g이 측정되었다. 본 연구 결과가압볶음 무말랭이 차의 섭취는 체내 유리기에 의한 산화적 손상을 보호하는 효과가 높은 것으로 사료된다.         홍채 이미지를 위해 사용되는 카메라는 현재 고가이며 사용적인 부분에서 제한점이 많고 소프트웨어와의 연동에 불편함을 가지고 있다. 또한 홍채를 촬영 시 발생하는 빛 반사는 이미지 분석에 정확한 ROI추출에 어려움을 주고 있고, 혼자서 스스로 홍채 이미지 촬영시 영상획득에 많은 어려움을 가지고 있다. 따라서 본 논문에서는 스스로 홍채 촬영이 가능한 안경형 홍채 카메라 구조를 제안한다. 제안 방식은 소프트웨어와 밀접한 기능성을 제공하는 일체형 홍채카메라를 개발하여 혼자서도 홍채 촬영시 흔들림이나 초점 불일치 등 문제를 해결 할 수 있어 영상 분석시 분석결과의 높은 정확도를 보인다. Now day cameras used for iris image are some problem theat a expensive, have many limitations in their use and uncomfortable with software connections. Also, light reflection that occurs when taking iris poses difficulty in image analysis, and it is difficult to obtain images when taking iris image alone. Therefore, in this paper, an eyeglass-type iris camera structure that can be filmed by itself is proposed. The proposed method can solve problems such as shaking or focusing inconsistencies when filming iris by developing an all-inclusive iris camera that provides functionality close to the software, thus showing high accuracy in analyzing the results of video analysis.

        A 22-year-old male with severe atopic dermatitis presented with fever and myalgia. He was diagnosed with active infective endocarditis causing multiple cerebral infarction, splenic infarction, and septic shoulder requiring synovectomy. Blood culture proved methicillin-resistant Staphylococcal aureus bacteremia. After treated with intravenous nafcillin and vancomycin, patient was improved. Another 42-year-old female with severe atopic dermatitis who presented with fever and chilling was hospitalized due to acute infective endocarditis. She also had left flank pain and visual disturbance, due to splenic infarction and acute cerebral infarction, respectively. As blood culture revealed methicillin-resistant Staphylococcal aureus bacteremia, she treated with intravenous vancomycin. Infective endocarditis can arise in the context of acute exacerbations of atopic dermatitis. Dermatologists need to pay attention to this risk and actively manage such acute flares in order to decrease the risk of infective endocarditis arising from skin lesions in atopic patients. For these reasons, we herein report two cases of infective endocarditis in patients with atopic dermatitis.

        Increasing Prevalence of the Sensitization to Cat/Dog Allergens in Korea

        Background: Recently, the number of domestic pets has increased. As a consequence, sensitization to animal allergens, such as cat or dog allergens, has become a problem. Objective: We studied the annual trends of sensitization to cats or dogs, and the characteristics of the patients. Methods: We retrospectively analyzed the medical records of 7,469 patients who visited a dermatology clinic and underwent an allergic profile test, from January 2011 to December 2015. Specific immunoglobulin E (IgE) levels to cat or dog antibody greater than 3.50 IU/ml were regarded as positive results. Results: In all, 274 patients showed significant increase in levels of specific IgE antibody to dog, and 307 revealed increase in levels of of specific IgE antibody to cat. The prevalence of these specific IgEs increased from 2011 to 2015. Independent risks for sensitization to cat allergens were sensitization to dog, but not to house dust, Dermatophagoides pteronyssinus, and D. farinae. Independent risks for sensitization to dog allergens were sensitization to cat, but not to house dust, D. pteronyssinus, and D. farinae. Total IgE level was not related to specific IgE level against either cats or dogs. Conclusion: In conclusion, the prevalence of sensitization to cat or dog has increased. Sensitization to cat or dog is related to each other, but is irrelevant to the total IgE level. (Ann Dermatol 30(6) 662∼667, 2018)

        Expression and Efficient One-Step Chromatographic Purification of a Soluble Antagonist for Human Leukemia Inhibitory Factor Receptor in Escherichia coli

        Leukemia inhibitory factor (LIF) is a member of the IL-6 cytokine family, having pleiotropic actions such as maintaining stem cell pluripotency and enabling blastocyst implantation. Because the action of LIF is mediated by a ligand-receptor interaction with the LIF receptor (LIF-R), an antagonist for LIF-R has been developed to inhibit LIF-induced signaling. In this study, we present a novel method for the production and purification of an antagonist to human LIF-R (hLA). His-tagged hLA was expressed in E. coli, and simple purification methods without any endopeptidase cleavage were designed. In addition, we determined the optimal temperature conditions for enhancing the production of soluble hLA. Finally, the bioactivity of His-tagged hLA was examined using STAT3 phosphorylation and receptivity of human endometrial ECC-1 cells. Our strategy provides a rapid and efficient method to produce biologically active recombinant hLA.

        지역 농부의 일상생활 활동 시 통증 개선을 위한 스트레칭과 탄성밴드 운동의 효과

        Purpose This paper aims at investigating the pain managementin the activities of daily living by using stretching and elastic band exercise. 20 community-dwelling farmers participated in the program divided into two groups: Exercise group (n=10, 61.29±±4.07 year-old) and Control group (n=10, 62.13±±10.91 year-old). Methods This research employed structuralized survey method from July to August, 2014, by applying 6 kinds of activities of daily living causing the inconveniencies from pain and used Visual Analog Scale to see the most painful intensity in a day. Results In the exercise group, 12 times of stretching and elastic band exercise were implemented for 4 weeks. As a result, no significant differences appeared between the two groups in the demographic characteristics and pain characteristics. In 4 weeks after the exercise application, the tests of long walking (≥≥10 min), squatting and pick-up, and sitting to floor on the exercise group respectively showed more improved effects than those prior to the treatment. And the degree of pain in a day also showed significant improved effects compared with them. Conclusion The stretching and elastic band exercise is considered not only to decrease the pain but also to be an effective intervention method to improve the activities of daily living of regional farmers.
