        카드뮴과 아연이 사람 단핵구성 세포주(THP-1)에서 IL-6의 분비에 미치는 영향

        Objectives : This study was performed to investigate effects of cadmium and zinc on IL-6 secretion using human monocyte in a culture system. Methods : We have used trypan blue dye exclusion methods to examine the effect of cytotoxicity of CdCl₂, ZnCl₂in THP-1 cells. IL-6 was measured by the ELISA method in the cell culture supernatants and the expression of IL-6 mRNA was examined by reverse transcription-polymerase chain reaction. Results : IL-6 production by THP-1 cells were decreased at 0.1 mM cadmium concentration (p<0.05) and increased as zinc concentration increased. Zinc prevented cadmium-induced suppression of IL-6, and the addition of a chelating agent, EDTA, restores IL-6 secretion. The expression of IL-6 mRNA decreased at 0.08 mM and 0.1 mM cadmium concentrations. Conclusion : This results suggest that cadmium depressed production and gene expression of IL-6 in stimulated human monocytes, and zinc prevented cadmium-induced suppression of IL-6.

      • Lineage Differentiation Program of Invariant Natural Killer T Cells

        <P>Invariant natural killer T (iNKT) cells are innate T cells restricted by CD1d molecules. They are positively selected in the thymic cortex and migrate to the medullary area, in which they differentiate into 3 different lineages. Promyelocytic leukemia zinc finger (PLZF) modulates this process, and PLZF<SUP>high</SUP>, PLZF<SUP>intermediate</SUP>, and PLZF<SUP>low</SUP> iNKT cells are designated as NKT2, NKT17, and NKT1 cells, respectively. Analogous to conventional helper CD4 T cells, each subset expresses distinct combinations of transcription factors and produces different cytokines. In lymphoid organs, iNKT subsets have unique localizations, which determine their cytokine responses upon antigenic challenge. The lineage differentiation programs of iNKT cells are differentially regulated in various mice strains in a cell-intrinsic manner, and BALB/c mice contain a high frequency of NKT2 cells. In the thymic medulla, steady state IL-4 from NKT2 cells directly conditions CD8 T cells to become memory-like cells expressing Eomesodermin, which function as premade memory effectors. The genetic signature of iNKT cells is more similar to that of γδ T cells and innate lymphoid cells (ILCs) than of conventional helper T cells, suggesting that ILCs and innate T cells share common developmental programs.</P>

        Improvement of interlaminar properties of carbon fiber‑reinforced epoxy composites using aluminum trihydroxide

        This study provides an economical and effective method to improve the interlaminar properties of carbon fiber-reinforced polymers (CFRPs) using aluminum trihydroxide (ATH) microparticles. ATH microparticles are cheap and are expected to show good affinity to epoxies in the matrix and sizing agents of the carbon fibers owing to the presence of three hydroxyl groups. In addition, ATH particles are reported to improve the mechanical properties of polymers when used as the reinforcement. In this study, ATH microparticles of various sizes, 1.5, 10, and 20 μm, were used to improve the interlaminar properties of the CFRPs. ATH particles with a size of 1.5 μm improved the tensile properties of the ATH/epoxy resin and did not significantly alter the curing behavior. The interfacial adhesion between the carbon fiber and the epoxy resin was also improved, and the impregnation of the resin mixture remained similar to that of the neat resin, resulting in no significant void and defect formation. Considering the above results, the resulting 1.5 μm ATH-reinforced CFRP showed improved interlaminar properties compared to CFRP without ATH. However, 10 and 20 μm ATH-reinforced CFRPs showed deteriorated interlaminar properties due to the diminished tensile properties of the resin itself and resin impregnation, which resulted in more voids and defects, despite the interfacial adhesion between the fiber and the matrix resin.

        금은화(金銀花) 및 금은화전초(金銀花全草)가 Raw 264.7 cell에서 LPS로 유도된 NO의 생성, iNOS, COX-2 및 cytokine에 미치는 영향

        Lonicerae Flos has antibacterial effects against Staphylococcus aureus, streptococci, pneumococci, Bacillus dysenterii, Salmonella typhi, and paratyphoid. It is an antiviral agent. The herb has a cytoprotective effect against $CCl_{4}-induced$ hepatic injury. It has antilipemic action, interfering with lipid absorption from the gut. Nowadays this herb is used mainly in the treatment of upper respiratory infections, such as tonsillitis and acute laryngitis. It is also used in the treatment of skin suppurations, such as carbuncles, and to treat viral conjunctivitis, influenza, pneumonia, and mastitis. Lonicerae Flos is dried flower buds of Lonicera japonica, L. hypoglauca, L. confusa, or L. dasystyla. But, for the most part, we use whole plant of Lonicera japonica, as a flower bud of it. And, little is known of the original copy of effects of whole plant, except for the 'Bon-Cho-Gang-Mok', which is written the effects of flower of Lonicera japonica are equal to effects of leaves and branch of it. The present study was conducted to evaluate the effect of flower and whole plant of Lonicera japonica on the regulatory mechanism of cytokines, inducible nitric oxide synthase (iNOS) and cyclooxygenase (COX-2) for the immunological activities in Raw 264.7 cells. In Raw 264.7 cells stimulated with lipopolysaccharide (LPS) to mimic inflammation, flower and whole plant of Lonicera japonica water extracts inhibited nitric oxide production in a dose-dependent manner and abrogated iNOS and COX-2. Flower and whole plant of Lonicera japonica water extract did not affect on cell viability. To investigate the mechanism by which flower and whole plant of Lonicera japonica water extract inhibits iNOS and COX-2 gene expression, we examined the on phosphorylation of inhibitor ${\kappa}B{\alpha}$ and assessed production of $TNF-{\alpha}$, $interleukin-1{\beta}$ $(IL-1{\beta})$ and interleukin-6 (IL-6). Results provided evidence that flower and whole plant of Lonicera japonica inhibited the production of $IL-1{\beta}$, IL-6 and activated the phosphorylation of inhibitor ${\kappa}B{\alpha}$ in Raw 264.7 cells activated with LPS. These findings suggest that flower and whole plant of Lonicera japonica can produce anti-inflammatory effect, which may play a role in adjunctive therapy in Gram-negative bacterial infections, respectively.

        8주간의 유산소성 체조활동을 통한 뇌병변 장애인의 체력 및 염증반응지표 변화

        본 연구의 목적은 뇌병변 장애인을 대상으로 8주간의 유산소성 체조활동을 실시하여 신체조성, 체력 및 염증반응지표의 변화에 대하여 알아보는 것이다. 본 연구의 피험자는남·녀 뇌병변장애 중 대표적인 뇌졸중 장애인 17명을 대상으로 시작되었지만 9명이 중도포기하여 최종 8명이 참가하였다. 운동 프로그램 참여 전 사전 검사를 실시하였으며, 기본적 변인으로 신장, 체중, Body Mass Index(BMI), 안정시 심박수, 안정시 혈압을 측정하고, 체력 변인으로는 체간 굴곡과 신전, 견관절 굴곡과 신전의 유연성과 악력을 통한근력, 폐활량을 통한 폐기능을 측정하였다. 또한 혈액성분으로는 혈중지질, 그리고 심혈 관질환 위험인자인 염증반응지표로서 C-reactive protein(CRP), Interleukin-6(IL-6), Tumor Necrosis Factor-α(TNF-α)를 분석하였다. 8주 유산소성 체조활동 프로그램은 매회50분씩 주 2회 실시하였으며, 운동강도는 운동자각도(RPE) 수준 13으로 유지하였다. 뇌병변 장애인의 유산소성 체조활동 프로그램 참여 후 운동의 효과를 검증한 결과 전체 집단에서 안정시 수축기혈압과 이완기혈압이 유의하게 감소하였다(p<.01). 또한 체력 요인에서는 체간 굴곡과 신전(p<.05), 건측의 견관절 굴곡과 신전(p<.01), 환측의 견관절 굴곡(p<.05)이 유의하게 증가하였다. 혈액 성분의 분석 결과 혈중지질에는 유의한 변화가 나타나지 않았으며, 염증반응지표의 경우 IL-6가 유의하게 감소하였고(p<.05), 유의하지는 않으나 CRP도 감소하는 경향을 나타냈다(p<.088). 결론적으로, 지속적이고 규칙적인 유산소성 체조활동은 뇌병변 장애인의 안정시 심박수와 혈압 및 체력을 개선시키는 효과가 있을 뿐만 아니라 심혈관질환의 위험요소로 알려져 있는 염증반응지표를 개선하는 데에도 긍정적인 영향을 주는 것으로 나타났다. The purpose of this study was to analysis the effect of aerobic gymnastic exercise for eight weeks on physical fitness and inflammatory markers which are known as risk factors for cardiovascular diseases in hemiplegic disabled after cerebral stroke. For this purpose, stroke patients(n=8) participated in aerobic exercise training 2 days/week, 50 mins/day, for 8 weeks and maintained at 13 level of RPE. Body composition, resting heart rate and blood pressure were measured before physical fitness test. Trunk flexion/extension, shoulder flexion/extension, grip strength, pulmonary function were measured as physical fitness test. Blood sample of 15ml was drawn from antecubital veins for measuring inflammatory markers(c-reactive protein, interleukin-6, tumor necrosis factor-α). The result of this study is as follows: Resting blood pressure decreased significantly after 8-week aerobic exercise training in total stroke patients (p<.01). After completion of the aerobic exercise program, trunk extension/flexion(p<.05), shoulder extension/fexion(p<.01), were significantly improved in total stroke patients. There was no significant improvement on blood lipid profiles in total stroke patients. However, IL-6 was significantly improved in stroke patients after 8-week aerobic exercise(p<.05). In conclusion, These findings suggests that regular aerobic gymnastic exercise decreases resting heart rate and blood pressure, and improves physical fitness, inflammatory markers which are known as a risk factor of cardiovascular diseases in person with disability occurred due to hemiparetic stroke

      • 흉막삼출의 유무에 따른 쯔쯔가무시증 환자간의 비교

        Scrub typhus is a potentially fatal infectious disease caused by the organism Orientia tsutsugamushi. Clinical manifestations are fever, skin rash, eschar and varying degree of respiratory distress. The pleural effusion in scrub typhus is rare and secondary to the destruction of vascular endothelium. Because the respiratory symptoms are generally mild and the pleural effusion in scmb typhus is rare than in interstitial pneumonia, there are few comments about the characteristics of scmb typhus with pleural effusion. So we made the comparative study of scmb typhus patients between with pleural effusion and without pleural effusion. 연구배경 치명적일 수 있는 감염질환 중 하나인 쯔쯔가무시중(Scrub typhus)에서 드문 흉막삼출을 동반한 환자에 대한 고찰은 다른 문헌에서도 자주 언급되지 않았다. 따라서 본 저자들은 흉막삼출이 있는 경우와 없는 경우에 따른 쯔쯔가무시중 환자들을 비교하고자 하였다. 방법 2003년에서 2006년까지 조선대학교 병원 내과에 입원중인 환자 중 쯔쯔가무시중 진단을 받은 총 109명의 환자를 대상으로 하여 흉부 X선 검사 및 혈청학적 검사(CPK, LDH, ESR, CRP, AST, ALT, ADA), 혈액학적 검사(WBC, PLT) 및 백분율 검사등을 실시하였다. 결과 흉막삼출이 있는 환자들의 경우 흉막 삼출이 없는 환자들에 비해 ESR 수치가 통계학적으로 의의있게 높았다(p-vaule < 0.05). 결론 흉막삼출 유무에 따른 쯔쯔가무시중 환자간의 비교에 있어 ESR 수치가 통계학적인 의의를 가졌다.

This technique is applicable to WDM(Wavelength Division Multiplexing) technique and number of sensors in this system can be increased by using this technique with WDM technique

      • Paste법에 의한 0.1V강의 침붕처리에 관한 연구

        0.1V강의 표면경도를 향상시키기 위해서 Boronizing처리를 실시하여 Hv=1500∼2000의 높은 경도값을 나타내는 붕화물층을 얻었다. 붕화물층의 두께와 처리시간과의 사이에는 방물선관계를 나타내었으며 붕화물층 생성 겉보기 활성화에너지는 94kJ/mol이다. 또한 900˚C, 3시간 침붕처리했을 때가 가장 높은 경도분포값을 나타내었으며 그 이상 장시간 처리하면 오히려 경도가 감소하는 경향을 나타낸다. 그리고 침붕층은 20% HCl, 20% H₂SO₄용액에서는 뛰어난 내식성을 가지나, 20% HNO₃용액에 대해서는 상대적으로 내식성이 떨어졌다. 그리고 내고온산화성은 대기중 800˚C까지는 침붕처리한 시편이 우수했으나 900˚C부터는 오히려 감소했다. To improve the surface hardness of the 0.1V steel, by Boronizing Treatment, We got boride layer showing high hardness, Hv=1500∼2000. A parabola relationship was appeared between boride layer thickness and treatment time, and apparent activation energy to form the boride layer 94kJ/mol. Moreover, We obtained the most high hardness distribution value when had the boronizing treatment for 3 hours at 900˚C. On the contrary, If we did boronizing treatment above 3 hours, the hardness appeared decreasing tendency. The boride layer had a good corrosion resistance at a solution of 20% HCl and 20% H₂SO₄, but correlatively the corrosion resistance of the boride layer was declined at a solution of 20% HNO₃. And the boronized specimen had a good high temperature oxidation resistance until 800˚C in the atmosphere but the high temperature oxidation resistance go down above 900˚C.

        광섬유 산란형 센서를 이용한 사회기반시설물의 보안에 관한 연구

        광섬유 ROTDR (Rayleigh optical time domain reflectometry) 센서와 보다 긴 광섬유를 감지광섬유로 사용할 수 있는 광섬유 BOTDA (Brillouin optical time domain analysis) 센서를 구성하고, 이들 각각을 이용하여 중요보안 대상체인 사회기반시설물에 침투하는 침입자를 탐지할 수 있는 기초 연구를 수행하였다. ROTDR 센서의 감지부로는 넓은 면적을 감지할 수 있는 매설형 광섬유 센서 탐지판을 제작하고, 인가된 침입물체의 위치와 무게에 따른신호특성을 고찰하였다. ROTDR 센서는 펄스 폭이 30 ns이고, 광섬유의 길이는 10 km 이상이다. 위치탐지오차는 약 2m 이내였으며, 무게에 따른 탐지능력은 20 kgf, 40 kgf, 60 kgf, 80 kgf의 네 단계를 구분할 수 있음을 알 수 있었다. 넓은 지역에 걸친 침입자의 침투를 감시하기 위하여 수십 km의 광섬유 길이 전체를 감지부로 사용할 수 있는 광섬유 BOTDA 센서를 개발하였다. BOTDA 센서는 한 개의 레이저 다이오드와 두 개의 광전 변조기(electro-optic modulator)를 사용하여 간단하게 구성하였다. 침입자에 의한 광섬유의 변형률 벽화를 탐지하는 실험을 수행하기 위하여 광학테이블 위에 광섬유에 변형률을 인가하기 위한 실험 장치를 설치하여 실험을 수행하였다. 이 실험으로부터 시간간격 1.5 초동안 광섬유 약 4.81 km의 길이를 거리분해능 3 m로 침입자를 탐지할 수 있음을 확인하였다. We have studied the detection techniques, which can determine the location and the weight of an intruder into infrastructure, by using fiber-optic ROTDR (Rayleigh optical time domain reflectometry) sensor and fiber-optic BOTDA (Brillouin Optical time domain analysis) sensor, which can use an optical fiber longer than that of ROTDR sensor. Fiber-optic sensing plates of ROTDR sensor, which are buried in sand, were prepared to respond the intruder effects. The signal of ROTDR was analyzed to confirm the detection performance. The constructed ROTDR could be used up to 10 km at the pulse width of 30 ns. The location error was less than 2 n1 and the weight could be detected as 4 grades, such as 20 kgf, 40 kgf, 60 kgf and 80 kgf. Also, fiber optic BOTDA sensor was developed to be able to detect intrusion effect through an optical fiber of tens of kilometers longer than ROTDR sensor. Fiber-optic BOTDA sensor was constructed with 1 laser diode and 2 electro-optic modulators. The intrusion detection experiment was peformed by the strain inducing set-up installed on an optical table to simulate a11 intrusion effect. I11 the result of this experiment, the intrusion effect was well detected as the distance resolution of 3 m through the fiber length of about 4.81 km during 1.5 seconds.
