        ZNF488 Enhances the Invasion and Tumorigenesis in Nasopharyngeal Carcinoma Via the Wnt Signaling Pathway Involving Epithelial Mesenchymal Transition

        Purpose The purpose of this study was to investigate the function of Zinc finger protein 488 (ZNF488) in nasopharyngeal carcinoma (NPC). Materials and Methods The endogenous expression of ZNF488 in NPC tissues, normal nasopharyngeal epithelium tissues and NPC cell lines were detected by quantitative reverse transcription polymerase chain reaction. ZNF488 over-expressing and knock-down NPC cell line models were estab- lished through retroviral vector pMSCV mediated over-expression and small interfering RNA (siRNA) mediated knock-down. The invasion and migration capacities were evaluated by wound healing and transwell invasion assays in ZNF488 over-expressing and control cell lines. Soft-agar colony formation and a xenograft experiment were performed to study tumorigenic ability in vitro and in vivo. Immunofluorescence and western blotting analysis were used to examine protein changes followed by ZNF488 over-expression. Microarray analysis was performed to explore gene expression profilings, while luciferase reporter assay to evaluate the transcriptive activity of Tcf/Lef. Results ZNF488 was over-expressed in NPC tissues compared with normal tissues, especially higher in 5-8F and S18, which are well-established high metastatic NPC clones. Functional studies indicate that over-expression of ZNF488 provokes invasion, whereas knock-down of ZNF488 alleviates invasive capability. Moreover, over-expression of ZNF488 promotes NPC tumor growth both in vitro and in vivo. Our data further show that over-expression of ZNF488 induces epithelial mesenchymal transition (EMT) by activating the WNT/β -catenin signaling pathway. Conclusion Our data strongly suggest that ZNF488 acts as an oncogene, promoting invasion and tumorigenesis by activating the Wnt/β -catenin pathway to induce EMT in NPC.

      • 전시참관객의 유형에 따른 전시회 효과성에 대한 인식이 구매행동에 미치는 영향

        본 연구는 선행연구를 바탕으로 하여 참관객의 참관목적 요인을 도출하고, 그에 따른 참관객 유형을 분류하며, 분류된 유형에 따른 전시회의 효과성에 대한 인식이 참관객의 실제 구매행동에 어떠한 영향을 미치는지를 파악하는 것을 목적으로 하고 있다. 연구의 결과 전시참관목적에 따라 "집중적 참관객", "특정 목적 참관객", "순회자", "저 관심 참관객" 등 4개의 참관객 유형이 도출되었고, 각 유형의 특성을 파악하였다. 그리고 탐색적 요인 분석을 통해 "상호작용 효과성 요인", "전문정보 획득 효과성 요인", "현장 프로모션 효과성 요인", "업무정보 획득 요인" 등 4개의 전시회 효과성 요인이 도출되었고, "계획구매 요인"과 "비 계획구매 요인" 등 2개의 구매행동 요인이 도출되었다. 전시참관객이 인식한 전시회 효과성과 구매행동 간의 영향 관계를 검증한 결과, 계획구매에 영향을 미치는 효과성 요인은 현장 프로모션 효과성, 전문정보획득 효과성, 업무정보획득 효과성의 순으로 나타났으며, 비 계획구매에 영향을 미치는 효과성 요인은 전문정보획득 효과성, 상호작용 효과성, 현장 프로모션 효과성의 순으로 나타났다. 이는 앞으로 전시참가업체나 주최자가 더 효율적으로 마케팅 전략을 수립하는 데 도움이 될 수 있다. The purpose of this study is to classify trade show visitors based on their purpose of visiting the trade show and then to examine whether there were any differences between each type of trade show visitors on perception of trade show effectiveness and purchase behavior. Also the other purpose of this study is to explore the effect of visitors` perception of trade show effectiveness on visitors` purchase intention. As a result, 4 types of trade show visitors such as "intensive user", "special purpose user", "stroller" and "pragmatist" were clustered based on the purpose of visiting the trade show, and each type of visitors` characteristics were identified in details. 4 factors such as "overall information acquisition effectiveness factor", "specialized information acquisition factor", "interaction effectiveness factor", and "on-site promotion effectiveness factor" were deducted as determinant factors of trade show effectiveness, and 2 factors such as "planned purchase" and "unplanned purchase" were deducted as determinant factors of visitors` purchase intention. The results also show that "on-site promotion effectiveness factor", "specialized information acquisition factor", and "overall information acquisition effectiveness factor" positively affects visitors` planned purchase behavior, and "specialized information acquisition factor", "interaction effectiveness factor" and "on-site promotion effectiveness factor" have a positive effect on visitors` unplanned purchase behavior. This study employing a theoretical framework to evaluate the visitors` purpose of visiting the trade show and trade show effectiveness from the perspective of visitors. By viewing such results, it can help trade show exhibitors and organizers to make more effective marketing strategies in the future.

        The influence factors on CeSn0.8W0.6Ox/TiO2 for catalytic removals of NO, CO and C3H8

        Series of CeSn0.8W0.6Ox/TiO2 catalysts were tested for selective catalytic reduction of NO and forsynergistic catalytic removals of CO and C3H8 from diesel engine exhaust. Results revealed that catalyst12%-CeSn0.8W0.6Ox/TiO2 calcined at 500 C exhibited the optimal catalytic performance for NH3-SCR ofNO. The catalyst obtained more than 90% NO conversion at a wide temperature range of 252–456 C. BothCO and C3H8 could be oxidized into CO2 by the optimized catalyst. Moreover, excellent redox property,rich surface acidity and big specific surface area were the promotional factors for good catalyticperformance in catalytic removals of NO, CO and C3H8.

        Expression of lipid metabolism genes provides new insights into intramuscular fat deposition in Laiwu pigs

        Objective: The objective of this study was to measure the special expression pattern of lipid metabolism genes and investigate the molecular mechanisms underlying intramuscular fat (IMF) deposition in Longissimus dorsi muscle of Laiwu pigs. Methods: Thirty-six pigs (Laiwu n = 18; Duroc×Landrace×Yorkshire n = 18) were used for the measurement of the backfat thickness, marbling score, IMF content, and expression of lipid metabolism genes. Results: Significant correlations were found between IMF content and the mRNA expression of lipid metabolism genes. Of the 14 fat deposition genes measured, fatty acid synthase (FASN) showed the strongest correlation (r = 0.75, p = 0.001) with IMF content, and of the 6 fat removal genes, carnitine palmitoyl transferase 1B (CPT1B) exhibited the greatest negative correlation (r = -0.66, p = 0.003) with IMF content in Laiwu pig. Multiple regression analysis showed that CPT1B, FASN, solute carrier family 27 member 1 (SLC27A1), and fatty acid binding protein 3 (FABP3) contributed 38% of the prediction value for IMF content in Laiwu pigs. Of these four variables, CPT1B had the greatest contribution to IMF content (14%) followed by FASN (11%), SLC27A1 (9%), and FABP3 (4%). Conclusion: Our results indicate that the combined effects of an upregulation in fat deposition genes and downregulation in fat removal genes promotes IMF deposition in Laiwu pigs.

        Serum lipocalin-2 is a potential biomarker for the clinical diagnosis of nonalcoholic steatohepatitis

        Background/Aims: Nonalcoholic steatohepatitis (NASH) is a progressive form of nonalcoholic fatty liver disease (NAFLD) characterized by hepatic steatosis, inflammation, hepatocellular injury, and fibrosis. We aimed to investigate the usefulness of a key biomarker, lipocalin-2 (LCN2), for the detection of NASH progression. Methods: A mouse NASH model was established using a high-fat diet and a high-sugar drinking water. Gene expression profile of the NASH model was analyzed using RNA sequencing. Moreover, 360 NAFLD patients (steatosis, 83; NASH, 277), 40 healthy individuals, and 87 patients with alcoholic fatty liver disease were recruited. Results: Inflammatory infiltration, focal necrosis in the leaflets, steatosis, and fibrosis were documented in the mouse liver. In total, 504 genes were differentially expressed in the livers of NASH mice, and showed significant functional enrichment in the inflammation-related category. Upregulated liver LCN2 was found to be significantly interactive with various interleukins and toll-like receptors. Serum LCN2 levels were significantly increased in NAFLD patients. Serum LCN2 levels were correlated with steatosis, intralobular inflammation, semiquantitative fibrosis score, and nonalcoholic fatty liver disease activity score. The area under the curve of serum LCN2 was 0.987 with a specificity of 100% and a sensitivity of 93.5% for NASH diagnosis, and 0.977 with almost the same specificity and sensitivity for steatosis. Conclusions: LCN2 might be involved in the transition from NAFL to NASH by mediating inflammation. Serum LCN2 levels might be a novel biomarker for the diagnosis of NASH.         QTL mapping and identification of candidate genes for cold tolerance at the germination stage in wild rice

        Background Cold damage stress significantly affects rice growth (germination and seedling) and causes serious losses in yield in temperate and high-altitude areas around the globe. Objective This study aimed to explore the cold tolerance (CT) locus of rice and create new cold-tolerant germplasm. We constructed a chromosome segment substitution line (CSSL) with strong CT and fine mapped quantitative trait loci (QTLs) associated with CT by performing the whole-genome resequencing of CSSL with phenotypes under cold treatment. Methods A chromosome CSSL, including 271 lines from a cross between the cold-tolerant wild rice Y11 (Oryza rufipogon Griff.) and the cold-sensitive rice variety GH998, was developed to map QTLs conferring CT at the germination stage. The whole-genome resequencing was performed on CSSL for mapping QTLs of associated with CT at the germination stage. Results A high-density linkage map of the CSSLs was developed using the whole-genome resequencing of 1484 bins. The QTL analysis using 615,466 single-nucleotide polymorphisms (SNPs) led to the identification of 2 QTLs related to germination rate at low-temperature on chromosome 8 (qCTG-8) and chromosome 11 (qCTG-11). The qCTG-8 and qCTG-11 explained 14.55% and 14.31% of the total phenotypic variation, respectively. We narrowed down qCTG-8 and qCTG-11 to 195.5 and 78.83-kb regions, respectively. The expression patterns of important candidate genes in different tissues, and of RNA-sequencing (RNA-seq) in CSSLs, were identified based on gene sequences in qCTG-8 and qCTG-11 cold-induced expression analysis. LOC_Os08g01120 and LOC_Os08g01390 were identified as candidate genes in qCTG-8, and LOC_Os11g32880 was identified as a candidate gene in qCTG-11. Conclusions This study demonstrated a general method that could be used to identify useful loci and genes in wild rice and aid in the future cloning of candidate genes of qCTG-8 and qCTG-11. The CSSLs with strong CT were supported for breeding cold-tolerant rice varieties.
