        Alu-Derived Old World Monkeys Exonization Event and Experimental Validation of the LEPR Gene

        The leptin receptor (LEPR) is a crucial regulatory protein that interacts with Leptin. In our analysis of LEPR, novel AluJb-derived alternative transcripts were identified in the genome of the rhesus monkey. In order to investigate the occurrence of AluJb-derived alternative transcripts and the mechanism underlying exonization events, we conducted analyses using a number of primate genomic DNAs and adipose RNAs of tissue and primary cells derived from the crab-eating monkey. Our results demonstrate that the AluJb element has been integrated into our common ancestor genome prior to the divergence of simians and prosimians. The lineage-specific exonization event of the LEPR gene in chimpanzees, orangutans, and Old World monkeys appear to have been accomplished via transition mutations of the 5' splicing site (second position of C to T). However, in New World monkeys and prosimians, the AluJb-related LEPR transcript should be silenced by the additional transversion mutation (fourth position of T to G). The AluJb-related transcript of human LEPR should also be silenced by a mutation of the 5' splicing site (first position of G to A) and the insertion of one nucleotide sequence (minus fourth position of A). Our data suggests that lineage-specific exonization events should be determined by the combination event of the formation of splicing sites and protection against site-specific mutation pressures. These evolutionary mechanisms could be major sources for primate diversification.

      • Chirality-induced antisymmetry in magnetic domain wall speed

        <P>In chiral magnetic materials, numerous intriguing phenomena such as built-in chiral magnetic domain walls (DWs) and skyrmions are generated by the Dzyaloshinskii-Moriya interaction (DMI). The DMI also results in asymmetric DW speeds under an in-plane magnetic field, which provides a useful scheme to measure the strength of the DMI. However, recent findings of additional asymmetries such as chiral damping have inhibited the unambiguous determination of the DMI strength, and the underlying mechanism of overall asymmetries comes under debate. Here, we experimentally investigate the nature of the additional asymmetry by extracting the DMI-induced symmetric contribution from the DW speed. Our results reveal that the additional asymmetry has a truly antisymmetric nature with the typical behavior governed by the DW chirality. In addition, the antisymmetric contribution alters the DW speed by a factor of 100, dominating the overall variation in DW speed. Thus, experimental inaccuracies can be largely removed by calibration with such antisymmetric contributions, enabling the standard DMI measurement scheme.</P>

        Korean Mistletoe (Viscum album coloratum) Extract Improves Endurance Capacity in Mice by Stimulating Mitochondrial Activity

        The beneficial effects of exercise on overall health make it desirable to identify the orally active agents that enhance the effects of exercise in an effort to cure metabolic diseases. Natural compounds such as resveratrol (RSV) are known to increase endurance by potentiating mitochondrial function. Korean mistletoe (Viscum album coloratum) extract (KME) has characteristics similar to those of RSV. In the present study, we determined whether KME could increase mitochondrial activity and exert an anti-fatigue effect. We found that KME treatment significantly increased the mitochondrial oxygen consumption rate (OCR) in L6 cells and increased the expression of peroxisome proliferator-activated receptor c coactivator (PGC)-1a and silent mating type information regulation 2 homolog 1 (SIRT1), two major regulators of mitochondria function, in C2C12 cells. In the treadmill test, KME-treated mice could run 2.5-times longer than chow-fed control mice. Additionally, plasma lactate levels of exhausted mice were significantly lower in the KME-treated group. In addition, the swimming time to exhaustion of mice treated with KME was prolonged by as much as 212% in the forced-swim test. Liver and kidney histology was similar between the KME-treated and phosphate-buffered saline-treated animals, indicating that KME was nontoxic. Taken together, our data show that KME induces mitochondrial activity, possibly by activating PGC-1a and SIRT1, and improves the endurance of mice, strongly suggesting that KME has great potential as a novel mitochondria-activating agent.

        23게이지 유리체절제술시 23게이지 주사바늘을 이용한 공막절개의 임상 결과

        Purpose: To report the clinical results of 23-gauge sclerotomy using 23-gauge needle during 23- gauge vitrectomy. Methods: A retrospective review was conducted on 331 eyes underwent 23-gauge sclerotomy using 23-gauge needle during vitrectomy from April 5th, 2011 to January 14th, 2013. All sclerotomy wounds were sutured with 8-0 vicryl at the end of the surgery. The main outcome measures included changes in best corrected visual acuity (BCVA), intraocular pressure (IOP), corneal astigmatism, and presence of intraoperative, postoperative complications after surgery. Results: The mean preoperative logMAR BCVA improved from 0.92 ± 0.76 to 0.43 ± 0.36 at 3 months after surgery (p<0.05). There was no significant difference between the mean preoperative IOP and IOP on the 3 months after surgery (p>0.05). There was no clinically significant change in the mean corneal astigmatism 2 weeks after surgery (p>0.05). Intraoperative complications had conjunctival incision due to conjunctival chemosis in 24 eyes, choroidal detachment in 2 eyes, retinal tear in 11 eyes. Postoperative complications included increased IOP in 29 eyes, hypotony in 1 eye, recurrent vitreous hemorrhage in 6 eyes, proliferative vitreoretinopathy in 1 eye, and subfoveal perfluorocarbon liquid in 1 eye. Mild foreign body sensation due to conjunctival sutures was noted, but most cases were improved at 1 to 4 weeks after surgery. However, there were no severe postoperative complications such as retinal detachment or endophthalmitis. Conclusions: 23-gauge sclerotomy using 23-gauge needle during vitrectomy is safe without serious complications postoperatively and also cost effective.

        부산, 경남 일부 지역 주민들의 생선 섭취량과 혈중 수은 농도의 관련성

        Objectives: The objective of this study was to identify the relationship between fish consumption and blood mercury levels in a sample of adult Koreans. Methods: The study subjects were 299 residents of Busan (male: 65, female: 234) and 185 residents of Namhae (male: 69, female: 116), South Korea. Demographic characteristics, current smoking, current drinking, fish consumption per week, past history of amalgam treatment, and residential district were recorded by trained interviewers in June and July, 2009. We considered a portion of fish to be equivalent to 70 gm weight, and calculated amounts of fish consumed per week. Using chi-square tests, t-tests, ANOVA, and multiple linear regression analysis, we estimated the relationships between blood mercury levels and amount of fish consumed per week and other factors. Results: The mean blood mercury level of our subjects was 6.61 ㎍/L, higher than the criterion defined by the United States Environmental Protection Agency (EPA) (5.8 ㎍/L). In multiple linear regression analysis, residential district and amount of fish consumed per week were associated with blood mercury levels. However, marital status, current drinking, and gender, and age were not associated with blood mercury levels. Conclusions: In conclusion, we suggest the implementation of systematic and periodic population-based studies to decrease the risks of mercury poisoning among South Koreans who consume fish as a regular part of the diet.

        빛간섭단층촬영기의 노후화가 측정치에 미치는 영향

        목적: 빛간섭단층촬영기의 노후화가 신호 강도에 미치는 영향과 측정치 변화에 대해 알아보고자 하였다. 대상과 방법: 2012년 1월부터 2월까지 빛간섭단층촬영을 시행한 정상인 60명 60안을 대상으로 전향적 연구를 하였다. 5년 된 빛간섭단층촬영기(Carl Zeiss Meditec, Dublin, CA, USA)와 동일 기종인 새 기계에서 동일안을 대상으로 각각 2회씩 반복 검사하여 신호강도 및 측정치의 변화를 알아보았다. 결과: 새 기계의 신호 강도는 8.6 ± 0.8, 오래된 기계는 5.0 ± 1.0이었으며 이는 통계학적으로 유의한 차이를 보였다(p<0.001). 각 기계별로 측정한 중심황반두께는 새 기계는 201.1 ± 16.1 μm, 오래된 기계는 210.3 ± 16.0 μm로 오래된 기계에서 통계학적으로 유의하게 두꺼웠다(p=0.002). 동일안을 대상으로 2회 반복 측정한 중심황반두께 값의 차이는 새 기계에서 3.2 ± 2.3, 오래된 기계에서 10.5 ± 12.6으로 유의한 차이가 있었다(p=0.021). 각 기계의 반복성을 알아보기 위해 급내상관계수를 비교하였고, 오래된 기계는 0.371-0.872로 낮은 반복성을 보였으나 새 기계는 0.806-0.947로 높은 반복성을 보였다. 결론: 빛간섭단층촬영기 검사 시 새 기계에서 오래된 기계에 비해 신호 강도가 세고, 중심황반두께가 작게 나타났다. 반복 측정 시새 기계에서 중심황반두께의 오차는 유의하게 작았으며, 새 기계와 오래된 기계에서의 신호 강도는 차이가 없었다. 따라서 빛간섭단층촬영기의 측정값 분석 시 기계의 노후화에 따른 영향도 고려해야 할 것으로 생각된다. <대한안과학회지 2016;57(7):1087-1092> Purpose: To evaluate the effect of instrument aging on optical coherence tomography (OCT) measurements. Methods: Single eyes of 60 healthy participants (60 eyes) aged 20-30 years were examined twice using a 5-year-old OCT instrument and a new OCT instrument (Carl Zeiss Meditec, Dublin, CA, USA). The measurements and changes in signal strength were investigated using both instruments. Results: The signal strengths of the new and aged instruments were 8.6 ± 0.8 and 5.0 ± 1.0, respectively, which was a statistically significant difference (p < 0.001). In addition, the central macular thicknesses (CMT) of the new and aged instruments were 201.1 ± 16.1 μm and 210.3 ± 16.0 μm, respectively. The thickness was significantly greater using the aged instrument (p < 0.001). Repeated measurements within the same eye were compared, and the difference in CMT was 3.2 using the new instrument and 10.5 using the aged instrument (p < 0.05). The intraclass correlation coefficient (ICC) was compared to evaluate the reproducibility of each instrument. The ICC values of nine areas of Early Treatment Diabetic Retinopathy Study in the aged instrument were 0.371-0.872, indicating low reproducibility. However, the new instrument showed high reproducibility with values of 0.806-0.947. Conclusions: Higher signal strength and lower CMT were observed using the new instrument compared to the aged instrument. Additionally, there were no differences in signal strength errors between the instruments. However, the error in CMT measured using the new instrument was significantly smaller compared to that using the aged instrument. Therefore, the effect of instrument aging should be accounted for in analyses of OCT measurements. J Korean Ophthalmol Soc 2016;57(7):1087-1092

Conclusions: In conclusion, we suggest the implementation of systematic and periodic population-based studies to decrease the risks of mercury poisoning among South Koreans who consume fish as a regular part of the diet. 본 연구는 직업적으로 수은에 노출되지 않는 일반 인구 군에서 생선 섭취량과 혈중 수은 농도 사이의 관련성을 파악하기 위하여 수행되었다. 이를 위하여 부산광역시 2개 동과 남해군 3개 마을에 거주하는 사람들 중 본 연구 참여에 동의한 484(남자: 134명, 여자: 350명)명을 최종 연구대상으로 하였다. 설문조사는 2009년 6월 말부터 7월초까지 연구 참여 희망자들을 대상으로 훈련된 조사자가 면접설문조사를 실시하였다. 설문 조사의 주요 내용은 성별, 연령, 학력, 결혼상태, 직업, 현재 흡연 여부, 현재 음주 여부, 일주일간 생선 섭취량, 아말감 치료, 거주지역 등이었다. 단변량 분석에서 성별, 나이, 결혼상태, 현재 음주 여부, 일주일간의 생선 섭취량, 그리고 거주지역이 혈중 수은 농도와 유의한 관련성이 있었다. 혈중 수은 농도를 종속 변수로 한 다중선형회귀분석에서 일주일간의 생선 섭취량, 그리고 거주지역이 혈중 수은 농도와 유의한 관련성이 있었다. 생선 섭취량과 혈중 수은 농도 간에는 통계적으로 유의한 관련성이 있다. 앞으로 혈중 수은 농도와 건강 장애간의 관련성을 비롯한 추가적인 연구를 통하여 생선 섭취와 연관된 수은 노출 기준, 실제적인 생선 내 수은 잔류 기준, 생선 섭취빈도와 생선 섭취량에 관한 여러 가지 권고안들을 마련하기 위한 근거 기반을 마련해야 할 것이다.
