Characterization of proteolysis in muscle tissues of sea cucumber Stichopus japonicus

The proteolysis in muscle tissues of sea cucumber Stichopus japonicus (sjMTs) was characterized. The proteins from sjMTs were primarily myosin heavy chains (MHCs), paramyosin (Pm), and actin (Ac) having a molecular mass of approximately 200, 98, and 42 kDa, respectively. Based on SDS-PAGE analysis and quantification of trichloroacetic acid (TCA)-soluble peptides released, degradation of muscle proteins from sjMTs was favorable at pH 5 and 50°C. Proteolysis of MHCs was mostly inhibited by cysteine protease inhibitors, including trans-epoxysuccinyl-L-leucyl-amido (4- guanidino) butane (E-64) and antipain (AP). E-64 and AP completely inhibited the degradation of Pm and Ac, while iodoacetic acid showed a partially inhibitory effect. These results indicated that the proteolysis of sjMTs was mainly attributed to cysteine proteases. Avoidance of setting the tissues at 40–50oC and slightly acidic condition and inhibition of cysteine proteases are helpful for decreasing sea cucumber autolysis.
Static and dynamic characteristics of a novel moving magnet linear compressor

Linear compressors are popular in the field of household refrigeration due to their high efficiency and excellent capacity modulation capability. In this study, we proposed a novel moving magnet linear compressor and investigated its static and dynamic characteristics. Static performance included static shaft force, force constant, and magnetic spring, which were examined via finite element analysis and verified by conducting specific thrust experiments. For dynamic characteristics, resonance frequency, piston stroke, and piston offset were calculated by considering the magnetic spring effect. Compared with the results that didn’t consider the magnetic spring effect, higher accuracy was achieved under various conditions. In addition, piston offset increased with discharge pressure, but it was insensitive to changes in piston stroke. The motor efficiency of the designed linear compressor prototype can reach 0.92 under no-load condition and 0.80 under a discharge pressure of 0.6 MPa with a stroke of 9 mm under air condition.
Narratives and Spectatorships of the Northeast Chinese Online Drama: Affect and Emotion

Chinese Northeast online drama has increasingly become a phenomenon in online drama creation and viewing. However, despite its significance, scholarly attention to this special region remains limited. This paper aims to address this gap by examining the collaborative influence of affect and emotion on the popularity of the Northeast online dramas. Prior to conducting close reading of the drama, this paper firstly clarifies the distinction between affect and emotion as a response to the scholarly bracketed use of two terms and for the purpose of locating the two into their best field of film analysis. Through the case study of a well-received Northeast online drama—Good Guy Good Luck, this paper proposes that, one of the reasons for the popularity of the Northeast online dramas is that they configure the dilemmas of characters in the period of social transformation. Good Guy Good Luck succeeds in presenting a disordered Northeast atmosphere of 1990s when individuals living in anxious life in the large scale laid-off wave intend to strive for honor or wealth but end with their diminution of power. The cruel affect and unsafe society in the film elicit an uneasy emotional response from the audience, which makes them better immersed in the Northeast drama narratives.

This research explores the role of political leadership in China in response to the Covid-19 at the linguistic level. The researcher conducted a political corpus analysis on transcripts from the official website of the Ministry of Foreign Affairs, People’s Republic of China, in Covid-19, as well as transcripts from previous political speeches addressed by Chinese leaders and transcripts from other Asian countries to determine the extent to which the research subjects use effective leadership and rhetoric proofs to project its national image. Under the framework of Aristotle’s ethos and pathos, the results suggest that Chinese leaders on the global stage tend to use we/us to invent their ethos to become an in-group member of the audience in the international arena. Meanwhile, it is found out that Chinese leaders have a preference in using positive sentiment than its Asian peers, and that, compared with its pre-pandemic speeches, the language of Chinese leaders during the pandemic is actually more negative, implying the severity of this pandemic.

The calcium sulfate scale is a common scale in the industrial water system, which is difficult to beremoved by common chemical cleaning methods. In this paper, a new phosphorous free of 1,6-Diaminohexane-contained polyaminoamide dendrimers (PAMAM) scale inhibitor with a threedimensional(3D) branched structure has been developed. The scale inhibition performances of differentgenerations of PAMAM dendrimers (PAMAM-mid, PAMAM-0G, PAMAM-0.5G and PAMAM-1G) are estimatedby the static scale inhibition method. Moreover, the effects of the scale inhibitor concentrationsand solution temperatures on the CaSO4 scale inhibition efficiency are thoroughly discussed. Results indicatethat the PAMAM-mid and PAMAM-0.5G) exhibit excellent inhibition performance on CaSO4 precipitation. Especially, the CaSO4 scale inhibition efficiency of PAMAM-0.5G is above 95% when the dose is10 mg/L, and the corresponding value is almost 100% when the inhibitor dose is 20 mg/L. In addition,the influence of descaling agents on the crystal and morphology of CaSO4 scales are studied by XRDand SEM tests. The results show that the scale inhibition ability of the PAMAM molecules mainly inhibitsthe growth of these crystal planes by adsorption on the surface of the growing crystal. Molecular dynamicssimulation results find that the PAMAM-0.5G dendrimer molecule can energetically interact well withthree crystal planes of calcium sulfate dihydrate. Finally, the simulation results provide a theoreticalguidance to judge the performance of scale inhibitors and synthesize new high-efficiency scale inhibitor.

Stable infections of maternally transmitted secondary endosymbionts in aphids are frequently found in field populations; however,whether this phenomenon changes in artificial lab conditions has not been clear. To investigate, (1): we collected Sitobion avenae (Fabricius) fromsix different regions and detected the three common secondary endosymbionts (Regiella insecticola, Hamiltonella defensa and Serratia symbiotica); (2): we raised Sitobion avenae fromChuzhou, Anhui for 14 months in the laboratory, and analyzed these aphids' DNA using the long PCR protocol after the 1stmonth, 3rdmonth, 5th month, 7thmonth, 9thmonth, 11thmonth, 12thmonth, 13thmonth and 14th month. The results showed that S. symbiotica and R. insecticola could be detected in all aphid populations,while H. defense were detected in three locations. After reared in the lab, the infection frequencies of Serratia symbiotica did not change across the 14 study months, but the infection frequencies of Regiella insecticola after the 9th month and of Hamiltonella defensa after the 11th month decreased significantly. Additionally, the results show that facultative endosymbiont infections may decline in artificial lab conditions, suggesting that temporal changes of endosymbionts may occur within the host under different environmental pressures. This result may partially explain why different geographic populations harbor a diverse variety of endosymbionts. The results of this study can be helpful in understanding the stability of secondary endosymbionts in Sitobion avenae under long-termreproduction in controlled laboratory conditions.

Objective: To investigate the changes of serum vascular endothelial growth factor (VEGF), soluble interleukin-2 receptor (SIL-2R) and hepatocyte growth factor (HGF) contents in patients with primary hepatocellular carcinoma (HCC) before and after percutaneous microwave coagulation therapy (PMCT) and determine their clinical significance. Materials and Methods: Fasting venous blood (3 mL) from 81 patients with primary HCC diagnosed by pathology was collected in the mornings 1 day before PMCT, and 1 day, 7 days and 1 month after PMCT, and then the serum was separated and stored in $-70^{\circ}C$. The contents of VEGF, SIL-2R and HGF were detected by enzyme linked immunosorbent assay (ELISA). Results: The serum VEGF, SIL-2R and HGF contents in 81 patients with primary HCC had obviously dynamic changes before and after PMCT. By comparison to 1 day after PMCT with pre-operation, there was no statistical significance regarding VEGF and SIL-2R contents (P>0.05), but HGF content showed significant difference (P<0.01). Compared with pre-operation, VEGF, SIL-2R and HGF contents 7 days and 1 month after PMCT all manifested significant differences (P<0.01). By comparison to 7 days with 1 month after PMCT, there was no statistical significance regarding the VEGF content (P>0.05), whereas SIL-2R and HGF contents showed significant change (P<0.01). Conclusions: The contents of serum VEGF, SIL-2R and HGF have obviously dynamic changes in primary HCC before and after PMCT, and their joint detection is expected to be an effective hematologic evaluation index of PMCT for primary HCC.
