        A selective cyclin-dependent kinase 4, 6 dual inhibitor, Ribociclib (LEE011) inhibits cell proliferation and induces apoptosis in aggressive thyroid cancer

        <P><B>Abstract</B></P> <P>The RB-E2F1 pathway is an important mechanism of cell-cycle control, and deregulation of this pathway is one of the key factors contributing to tumorigenesis. Cyclin-dependent kinases (CDKs) and Cyclin D have been known to increase in aggressive thyroid cancer. However, there has been no study to investigate effects of a selective CDK 4/6 inhibitor, Ribociclib (LEE011), in thyroid cancer. Performing Western blotting, we found that RB phosphorylation and the expression of Cyclin D are significantly higher in papillary thyroid cancer (PTC) cell lines as well as anaplastic thyroid cancer (ATC) cell lines, compared with normal thyroid cell line and follicular thyroid cancer cell line. LEE011 dose-dependently inhibited RB phosphorylation and also decreased the expressions of its target genes such as <I>FOXM1, Cyclin A1,</I> and <I>Myc</I> in ATC. Furthermore, LEE011 induced cell cycle arrest in G0-G1 phase and cell apoptosis, and inhibited cell proliferation in ATC. Consistently, oral administration of LEE011 to ATC xenograft models strongly inhibited tumor growth with decreased expressions of pRB, pAKT and Ki-67, and also significantly increased tumor cell apoptosis. Taken together, our data support the rationale for clinical development of the CDK4/6 inhibitor as a therapy for patients with aggressive thyroid cancer.</P> <P><B>Highlights</B></P> <P> <UL> <LI> pRB and Cyclin D were expressed high in aggressive thyroid cancer. </LI> <LI> LEE011 suppressed pRB and also decreased the expressions of its target genes in ATC. </LI> <LI> LEE011 induced cell cycle G1 arrest and apoptosis, and inhibited cell proliferation. </LI> <LI> LEE011 inhibited in vivo tumor growth with decreased expressions of pRB and Ki-67. </LI> <LI> We could explain the anticancer effects with the RB-E2F pathway. </LI> </UL> </P>

        김치유래 젖산균의 균체지방산 분석을 이용한 분류학적 연구

        표준균주를 포함한 230여개의 김치유래 젖산균에 대한 균체지방산(FAMEs)을 분석하였다. FAMEs profiles는 Euclidian Distance 17.5에 의해 7개의 Major Cluster와 1개의 Single Cluster로 나뉘어졌다. 이중 A, B, C 및 Cluster는 Leuconostoc속으로 분석되어졌고, F는 Lactobacillus속으로 분석되어졌다. 그리고 E와 G cluster는 두개의 Genus가 혼재되어 나타났으며 보충적인 연구가 필요하다. 앞으로 김치유래 젖산균의 균체지방산 분석결과를 기반으로 한 데이타베이스에 95가지 탄소원을 이용하는 수치분류학적 접근방법 및 Pyrolysis Mass Spectrometry 등의 화학적 분석 방법과 분자친화적 연구를 통한 종합적 분류정보 체계가 갖추어지면 젖산균의 신속, 정확한 동정 및 연구에 활발히 이용되어질 것이다. Two hundreds and thirty lactic acid bacteria, mostly isolated from Kimchi, including type strains were sued for analysis of cellular fatty acids. The 230 test strains were recoverd in 7 major and 1 single clusters defined a Euclidian distance of 17.5. These aggregate taxa were equivalent to the genus Leuconostoc (aggregate group A, B, C and D), and the genera Leuconostoc and Lactobacillus (aggregate group G). It is concluded as evident that FAMEs (Fatty Acid Methyl Esters) profile of cell can be used as a criterion in classification of lactic acid bacterial from kimchi. Additional comparative taxonomic studies need to be carried out on well chosen representative strains to determine the most appropriate methods of value.

        관상동맥 질환을 가진 제 IIa형 과지질단백혈증 환자에 동반된 건 황색종 1예

        We report a case of type IIa hyperlipoproteinemia with xanthoma tendinosum associated with coronary artery disease in a 51 year-old male. Multiple hard nodules appeared firstly on both achilles tendon area 30 years ago and the lesions gradually spread to the dorsa of hands and feet. On past history, he had been diagnosed as unstable angina with atherosclerosis of three vessels in coronary arteries, 2 years ago. His family history revealed nothing contributory. Serum lipid profile including lipoprotein electrophoresis showed an increase in total cholesterol and LDL-cholesterol with increased P-lipoprotein fraction, which suggested type IIa hyperlipoproteinemia. Histopathologic finding of a nodule from the dorsum of right hand showed many foamy histiocytes and cholesterol clefts in the dermis. Most of the xanthoma cells were mononuclear, but many Touton type giant cells were seen also. He died suddenly of heart failure with unstable angina. (Korean J Dermatol 2003;41(11) : 1541-1543)

        설기떡의 품질향상을 위한 부재료 첨가의 효과

        This study was carried out to investigate the effects of addition ratio(0, 5, 10 and 20%) of various minor ingredients(prosomillet, glutinous barley, oat, wheat, yellow and black soybean, safflower seed, chocolate etc.) on mechanical and sensory characteristics of Sulgiduk. In mechanical evaluation, hardness, cohesiveness and gumminess were increased with increasing the content of prosomillet, glutinous barley, oat and wheat to be added, and did not show any relationship the content of them to be added on yellow and black soybean, safflower seed and chocolate. In sensory evaluation, quality characteristics of Sulgiduk added with prosomillet, oat, yellow and black soybean and chocolate with 5∼10%, respectively was better than those of Sulgiduk without the minor ingredients. Change of hardness on Sulgiduk added with oat, yellow soybean, black soybean and chocolate was lower than those of Sulgiduk without them during storage of room temperature.

      • Zn₄SnSe_(6):Co^(2+) 단결정의 물성에 관한 연구

        In this paper, author describe the undoped and Co^(2+)-doped Zn₄SnSe_(6) single crystals were grown by the chemical transport reaction(CTR) method. For the crystal growth, the temperature gradient of the CTR furnace was kep a t 700℃ for the source aone and at 820℃ for the growth zone for 7-days. I t was found from the analysis of x-ray diffraction that undoped and Co^(2+)-doped Zn₄SnSe_(6) compounds have a monoclinic structure. The optical absorption spectra obtained near the fundamental absorption edge showed that these compounds have a direct energy gaps.

        열화된 SUS 316강의 시간-주파수 해석에 의한 비파괴평가

        지금까지 퓨리에 변환이 신호 처리법으로 가장 널리 사용되고 있다. 그러나, 이 방법은 신호의 주파수 성분이 시간에 대하여 어떻게 변화하는지를 표현하지 못한다. 따라서, 최근 이와같이 비정상신호를 표현하지 못하는 퓨리에 변환의 단점을 보완하여, 신호의 시간과 주파수에 대한 정보를 동시에 표현할 수 있는 시간-주파수 해석법들이 개발되기 시작하였다. 본 연구에서는 인공열화된 SUS 316강의 초음파 신호를 시간-주파수 해석법으로 분석하였다. 특히, 단시간 퓨리에 변화법과 위그너 빌 분포법을 이용하여 초음파 신호의 주파수 성분과 특성을 분석하였다. Fourier transform has been one of the most commonly used tools in study of frequency characteristics of signal. However, based on the Fourier transform, it is hard to tell whether a signal's frequency contents evolve in time or not. Recently, to overcome Fourier transform fault, not to represent non-stationary signal, time-frequency analysis methods are developed and those can represent informations of signal's time and frequency at the same time. In this study, we analysed ultrasonic signal for degraded SUS 316 with time-frequency analysis method. In particular, the methods such as short time Fourier(STFT) and Wigner-Ville distribution(WVD) were used to extract frequency contents and characteristics from ultrasonic signals.

        Agents are a powerful abstraction through which users may interact with a complex data universe by delegating tasks involving objects or events of interest. Digital libraries are one of the many application areas where agent technologies are used. In these architectures, digital libraries are seen as a decentralized collection of interacting self-interested agents where an agent represents the knowledge and interests of an individual user. This paper describes intelligent agents based architectures for digital libraries and suggest an intelligent agents based virtual university, a limited version of digital library.

        기호학적 입장에서의 의미 전달은 기호에 의한 의미의 교환이라 할 수 있다. 기호가 언어보다 폭넓은 인간 의식의 이해를 반영한다는 점에서, 인위적으로 설정된 상징체계 이외에 자연스러운 상황이나 이미지 자체도 하나의 기호적 의미의 범주로 해석될 수 있다. 교사와 학생의 가르치고 배우는 활동 역시 지식의 일방적인 전수가 아니라, 다양한 기호들이 상호소통함으로써 의미전달과 창출을 끊임없이 경험하는 해석적 활동이 된다. 교육활동 그 자체가 일종의 해석적 활동이기 때문에 이들 사이에 어떤 형태의 의미교환이 이루어지는가를 규명하기 위해서는 교수-학습의 기호적 역동관계를 이해하는 일이 선행되어야 한다. 따라서 교사와 학생 사이의 의미전달과 획득의 측면을 인간의 기호적 인식의 특성을 통해 밝히는 것은, 기존의 텍스트의 성격 이해를 중심으로한 해석적 연구에서 한 걸음 더 나아가 의미화 과정에서 일어나는 교육 주체들의 인식과정을 설명하는 하나의 계기가 될 것이다. The purpose of this study is to investigate the meaning of teaching-learning process in the view of Semiology. In Semiology, education is the enlarging process of meaning between teacher and students in the classroom as a field of signs. It is the hermeneutics activities to student which learn from the teacher and the text. It means students don't take the meanings not just direct way but translating way as their own meaning structures. In the same way, we can say that even teachers don't convey the meaning to students. They translate the meanings for students representing their own understanding structures. It can be said that the teaching and learning process is the relationships of hermeneutic cycle. In Semiotic view points, educational texts is not just the tools of information transmission. We can recognize the educational texts is the dynamic products from the meaningful actions between the two educational agents. These kinds of Semiotic views can give the new visions to clarify the educational communication between teachers and students in the classroom.
