        Association between Melatonin Receptor 1A Gene and Expression of Reproductive Seasonality in Sheep

        To determine whether a link exists between reproductive seasonality and the structure of the melatonin receptor 1A (MTNR1A) gene, the latter was studied in nonseasonal estrous breeds (Small Tail Han and Hu ewes) and seasonal estrous breeds (Dorset, Suffolk and German Mutton Merino ewes). A large fragment of the exon 2 of the MTNR1A gene was amplified and a uniform fragment of 824 bp was obtained in 239 ewes of five breeds. The 824 bp PCR product was digested with restriction endonucleases Mnl I and Rsa I, and checked for the presence of restriction sites. The presence (allele M) or absence (allele m) of an Mnl I site at base position 605 led to three genotypes MM (236 bp/236 bp), Mm (236 bp/303 bp) and mm (303 bp/303 bp) in five sheep breeds. The presence (allele R) or absence (allele r) of a Rsa I site at base position 604 led to three genotypes RR (267 bp/267 bp), Rr (267 bp/290 bp) and rr (290 bp/290 bp) in five sheep breeds. Frequencies of MM and RR genotypes were obviously higher, and frequencies of mm and rr genotypes were obviously lower in nonseasonal estrous sheep breeds than in seasonal estrous sheep breeds. Sequencing revealed four mutations (G453T, G612A, G706A, C891T) in mm genotype compared to MM genotype and one mutation (C606T) in rr genotype compared to RR genotype. For polymorphic Mnl I and Rsa I cleavage sites, the differences of genotype distributions were very highly significant (p<0.01) between Small Tail Han ewes and seasonal estrous sheep breeds. In each group, no significant difference (p>0.05) was detected. These results preliminarily showed an association between MM, RR genotypes and nonseasonal estrus in ewes and an association between mm, rr genotypes and seasonal estrus in ewes.

        Responses of Blood Glucose, Insulin, Glucagon, and Fatty Acids to Intraruminal Infusion of Propionate in Hanwoo

        This study was carried out to investigate the effects of intraruminal infusion of propionate on ruminal fermentation characteristics and blood hormones and metabolites in Hanwoo (Korean cattle) steers. Four Hanwoo steers (average body wt. 270 kg, 13 month of age) equipped with rumen cannula were infused into rumens with 0.0 M (Water, C), 0.5 M (37 g/L, T1), 1.0 M (74 g/L, T2) and 1.5 M (111 g/L, T3) of propionate for 1 hour per day and allotted by 4×4 Latin square design. On the 5th day of infusion, samples of rumen and blood were collected at 0, 60, 120, 180, and 300 min after intraruminal infusion of propionate. The concentrations of serum glucose and plasma glucagon were not affected (p>0.05) by intraruminal infusion of propionate. The serum insulin concentration at 60 min after infusion was significantly (p<0.05) higher in T3 than in C, while the concentration of non-esterified fatty acid (NEFA) at 60 and 180 min after infusion was significantly (p<0.05) lower in the propionate treatments than in C. Hence, intraruminal infusion of propionate stimulates the secretion of insulin, and decreases serum NEFA concentration rather than the change of serum glucose concentration.

      • 枸杞子 體細肥雜種 合成을 위한 生物工學的 基礎硏究

        The present study was carried out in order to develop techniques for rapid mass propagation and afford the basic information for somatic hybrid of Lycium chinense, used commonly as landscape purpose and medicinal resource. Through biotechnology, bud culture, callus culture, protoplast isolation and culture, fusion and gene transformation were examined. Results obtained were summarized as follows ; 1. Plantlets were developed from axillary buds attached to the stem of the current year growth collected in early June and cultured on MS media supplemented with 1.0㎎/l IAA and 2.0㎎/l Kinetin by 56% as the highest yield rate. 2. The highest rate of shoot induction was resulted at 0.5㎎/l kinetin, root induction at MS + 1.0㎎/l IAA + 2.0㎎/l kinetin (56%). 3. The rate of plantlets formation supplemented with 1.0㎎/l IAA and 2.0㎎/l kinetin was the highest and WPM (60%), MS·SH (50%), B_5 (40%) and GD (30%) media gave the result in descending order. 4. Plantlets (10-12 ㎝ of length) survived without fail after being transferred to pots with a mixture of perlite : vermiculite : peat (1:1:1 v/v/v). 5. The leaf collected from in vitro cultured plants cultured on MS media without growth regulators supply gave 60% callus formation and 40% shoot induction. 6. The highest protoplast yield and viability were resulted in the treatment plot of 3.0% macerozyme, 1% cellulase R-10, 0.4M mannitol, pH 6.2 and incubation for 6 hours. And the more effective was at the more juvenile leaf tissue. When protoplasts were cultured at 5x10^4/ml density in MSP_19M medium, the first budding occured at 18 days after culture and 28 days later cell colony was formed. 7. The highest cell fusion yield was 29.6% between leaf cells and 5.1% between leaf and root cell, using 30% (W/V) polyethlene glycol (PEG) concentration, 10 minutes treated and 2x10^5/ml protoplasts density from seedlings in virto. 8. When leaf was inoculated with Agrobacterium tumefaciens ATCC 11157, tumour was formed and rooting was initiated 8 days after inoculation and the growth was continued for 3 weeks. And the rate of tumour formation was 10%. The genetic transformation was confirmed through the opine analysis by the electrophoration.

        Machining of the KSTAR TF coil structure

        The KSTAR Toroidal Field (TF) magnet system consists of 16 TF structures. Each structure must be machined for precise assembly and reducing error field. Dimensions of the structure are a height of 4.5 m and a width of 3.2 m. The allowable tolerance of the structure is ±1 2 mm, and that of the bolt hole for Poloidal Field (PF) coil basement is ±0.5 mm. The final machining of the structure is also important for assembly of the interface structures such as toroidal ring, PF coil structures, and Central Solenoid (CS) structure. Before coil encasing, the inside surface of the structure is machined. Most of the outer surface of the structure, except for the surface of the intercoil structure (ICS), is machined after final welding. In particular, the side surface of an inboard leg, the connection plate of the ICS, the toroidal ring basement, the PF structure basements, shear key holes, and conical bolt holes are precisely machined. Fabrication and assembly tolerances are absorbed by the thickness of the electrical insulation, the spacer of the shear key, and the taper ring of the conical bolt. Six TF coil structures have been assembled and two structures are waiting for assembly. Another three structures are under final machining. A laser tracker is used for three dimensional survey and measurement of the structure.

        Cryptococcus性 腦髓膜炎 1例

        A case of cryptococcal meningitis is reported, which occured in a 25-year old female whose chief compaints ware severe headache, vomiting and double vision lasting one month. Neurological examination on admission (September 7, 1973) showed no significant abnormalities except for the bilateral papilledema and subjective horizontal diplopia. Simple skull and chest films including results of specific diagnostic procedures were grossly intact. Cerebrospinal fluid examination disclosed pleocytosis, increased protein, and decreased sugar and chloride, levels which was the finding compatible with those of tuberculous meningitis etc. The subject was treated under the diagnosis of tuberculous meningitis. Two weeks after the admisson, direct smear of the spinal fluid showed numerous yeast like cells, and cryptococcus neoformans was confirmed by India ink preparation and culture in Sabouraud's glucose agar. Amphotericin B was administered by slow intravenous drip over a span of several hours starting with 0.25㎎/㎏ of body weight and increasing gradually up to 1.25㎎/㎏ of body weight daily. Clinical improvements of the patient's condition along with disappearance of cryptococcus neoformans in CSF examination were obtained by the treatment with Amphoteric in B injection, although a fall in hemoglobin, hypopotassemia, moderate degree of fever and occasional vomiting were observed as side effects during the course of treatment. Cryptococcus is highly fatal infection caused by cryptococcus neoformans which is a special predilections for the central nervous system. Crytococcosis may also involve the lungs, bones and skin, and occurs with increased frequency in patients with leukemia or lymphomas. This infection is most common in male adult, and the protal entry is the respiratory tract.

      • Radiation-induced angiogenic signaling pathway in endothelial cells obtained from normal and cancer tissue of human breast

        Despite strong possibility that endothelial cells (ECs) of tumors and normal tissues may differ in various aspects, most previous studies on ECs have used normal cells. Here, we purified ECs from tumorous and normal human breast tissues, and studied the effect of radiation on angiogenesis and relevant molecular mechanisms in these cells. We found that in normal tissue-derived ECs (NECs), 4 Gy irradiation increased tube formation, matrix metalloproteinase 2 (MMP-2) expression and extracellular signal-regulated kinase (ERK) pathway activation. In cancer-derived ECs (CECs), however, 4 Gy irradiation significantly reduced tube formation, increased the production of angiostatin and interleukin-6 (IL-6), and upregulated AKT and c-Jun N-terminal kinase (JNK) pathway activation. Knockdown experiments showed that siMMP-2 efficiently inhibited tube formation by irradiated NECs, whereas siPlasminogen effectively attenuated the radiation-induced suppression of tube formation and the upregulation of angiostatin in CECs. Moreover, siIL-6 clearly inhibited the radiation-induced generation of angiostatin in CECs. Inhibition of ERK with a pharmacological inhibitor or small interfering RNAs (siRNAs) markedly suppressed the radiation-induced tube formation and MMP-2 upregulation in NECs, whereas the inhibition of either AKT or JNK with pharmacological inhibitor or siRNA treatment of CECs markedly attenuated the inhibition of tube formation and the upregulation of angiostatin and IL-6 caused by 4 Gy irradiation. These observations collectively demonstrate that there are distinct differences in the radiation responses of NECs and CECs, and might provide important clues for improving the efficacy of radiation therapy.
