Lysimachiae christnae HANCE has long been used to induce diuresis, remove calculi and treat jaundice in oriental countries. Isoferulic acid(3-hydroxy-4-methoxycinnamic acid) is the major biological active component of Lysimachiae christinae in our experiment. Effect of isoferulic acid on the activities of free radical scavenging enzymes and glutathione-related enzymes as well as detoxiflcation mechanism of isoferulic acid against ethacrynic acid-induced hepatotoxicity was investigated in rat. Hepatic glutathione level in the isoferulic acid-treated group not effected as compared to that of the control group. Although the injection of ethacrynic acid results in the significantly decrease in the level of hepatic glutathione, glutathione S-transferase and glutathione reductase, this decreasing effect was markedly lowered in the isoferulic acid-pretreated group. But, v-glutamylcystein synthease activity was not affected. Furthemore, isoferulic acid prevented against ethacrynic acid-inducible hepatotoxicity in rat as evidenced by the decreased formation of lipid peroxide. Enzyme activity, however, in the isoferulic acid-treated rat, were affected Vmax and not affected Km value in vitro. Treatment with isoferulic acid did significantly increase glutathione-related enzymes activities such as glutatione S-transferase, and the characteristics of increase in the enzyme activity might include the induction of enzyme protein. Therefore, it can be concluded that mechanism for the observed preventive effect of isoferulic acid, active component of Lysimachiae christinae, against the ethacrynic acid-induced, hepatotoxicity was due to the induction of glutathione S-transferase which related with metabolism of these materials.
Prevalence state of canine brucellosis in South Korea during 2015 and 2016

We investigated the prevalence of canine brucellosis in companion and stray dogs between March 2015 and December 2016 and determined the disease characteristics based on the geographic distribution, sex and age of the dogs in South Korea. We conducted a large-scale survey using serological and bacteriological tests. Samples were collected from 2,394 dogs (1,825 companions and 569 strays). Thirty (1.3%) samples were positive for Brucella canis antibodies including 16 (0.9%) from companion dogs and 14 (2.5%) from stray dogs. Two (1.0%) of the 196 samples cultured from the stray dogs were positive. When compared with male dogs, the female companion and stray dogs had a significantly higher prevalence of brucellosis. Moreover, the prevalence of canine brucellosis was significantly higher in stray dogs older than 6 years and the prevalence of the disease in companion dogs was highest in Incheon (2.1%) and Jeolla (2.1%) provinces. Stray dogs from the Daejeon metropolitan area had the highest prevalence of brucellosis (7.9%). National control measures for canine brucellosis have not previously been implemented. Our findings suggest that appropriate screening tests and control measures are necessary to improve the health of dogs and to protect public health in Korea, particularly with the rapid growth of the companion animal industry.
Pathological, immunohistochemical, and bacteriological findings in dogs infected with Brucella canis

This study describes pathological, immunohistochemical, and bacteriological findings in adult dogs and fetuses naturally infected with Brucella (B.) canis. A total of 42 dogs including 40 dogs and 2 aborted fetuses were examined. The most common gross lesions in infected dogs were swelling of lymph nodes and spleen. The testes showed marked swelling with multifocal to diffuse reddish discoloration. The most significant histopathological lesions were observed in the placenta. Placental trophoblasts were markedly hypertrophied due to the accumulation of intra-cellular gram-negative bacteria. Lymphocytic inflammation of varying severity was observed in the reproductive organs such as male testis, epididymis, and prostate gland and female uterus. Strong immunolabelling was observed in the cytoplasm of most trophoblasts in the placental tissues using immunohistochemistry. However, immunohistochemical staining did not demonstrate any organisms in other organs of dogs and fetuses. B. canis isolates were most frequently obtained from the whole blood (82.5%) and superficial inguinal lymph node (77.5%) in both sexes. In addition, the isolation rate was higher in male genital organs than in those of females. Hence, management of male dogs is most important because infected dogs can play a role as carriers.
Short communication : A case of canine leishmaniasis in Korea

A 26-month-old male mixed-breed dog of Korean origin was subjected to necropsy following death after a history of decreased appetite and weight loss. Necropsy revealed generalized lymphadenopathy and splenomegaly. Histopathological examination of samples from the spleen, mandibular lymph nodes, liver, kidney, and large intestine showed granulomas with numerous macrophages containing intracytoplasmic Leishmania amastigotes. Transmission electron microscopy revealed Leishmania amastigotes in the macrophage cytoplasm. All tissues with granulomas were positive for Leishmania spp, which was confirmed to be Leishmania infantum by conventional polymerase chain reaction (PCR), real-time PCR, and in situ hybridization. To our knowledge, this is the second case of canine visceral leishmaniasis in Korea.

Mesenchymal stem cells (MSCs) have ability to differentiate into multi-lineage cells, which confer a great promise for regenerative medicine to the cells. The aim of this study was to establish a method for isolation and characterization of adipose tissue-derived MSC (pAD-MSC) and bone marrow-derived MSC (pBM-MSC) in pigs. Isolated cells from all tissues were positive for CD29, CD44, CD90 and CD105, but negative for hematopoietic stem cell associated markers, CD45. In addition, the cells expressed the transcription factors, such as Oct4, Sox2, and Nanog by RT-PCR. pAD-MSC and pBM-MSC at early passage successfully differentiated into chondrocytes, osteocytes and adipocytes. Collectively, pig AD-MSC and BM-MSC with multipotency were optimized in our study.

A case of caseous lymphadenitis (CL) was detected in a native Korean goat without manifestation of superficial nodules but with pulmonary abscesses that were characterized microscopically by granulomatous inflammation consisting of a central necrotic region, peripheral fibrous capsule, and numerous cellular infiltrates with epithelioid macrophages, giant cells, and lymphocytes. Presence of Corynebacterium pseudotuberculosis was identified by using bacterial isolation and multiplex PCR. In addition, a 43% seropositivity to C. pseudotuberculosis was observed in goats from the same farm. This case highlights the risk of exposure of farmers to the causative agent and the urgent need for national monitoring for CL, a potential zoonotic.

Heat stroke in a dog is a life-threatening syndrome characterized by a high body temperature over 41°C, by the central nervous system dysfunction, and by multiple organ dysfunction. A 11-month-old male American Pit Bull Terrier was presented to clarify the cause of death to diagnostic laboratory. This dog showed clinical signs such as high body temperature (42°C), severe tachypnea, hematochezia, epistaxis and hemoptysis after transportation at hot summer time. At necropsy, there were hemorrhages in skin, serosa of stomach and small intestine, and also dark red fluid in lumen of intestine. Histopathologic examination revealed extensive hemorrhages in stomach, muscle, skin, and tongue. In microbiology, pathologic bacteria such as Pasteurella, Boedetella, Salmonella, Clostridium, and circulating virus in Korea such as CDV, CIV, CAV, CHV, CCV, CPIV were negative except CPV. There were many gross and microscopic lesions suggesting hest stroke while pathologic agents and related lesions were not detected. Therefore we diagnosed this case as heatstroke.
