EFNA1 ligand and its receptor EphA2: potential biomarkers for hepatocellular carcinoma

<P>Novel biomarkers are needed for early detection and progression evaluation of hepatocellular carcinoma (HCC). The purpose of this study was to identify useful biomolecular markers for HCC. The 26 genes that encode membrane or secretory proteins were identified from cDNA microarray data. We further examined the expression of EFNA1 and its receptor EphA2 and determined their biological implications during the development and progression of HCC. The EFNA1 mRNA was overexpressed in most HCCs as compared with its expression in corresponding nontumor tissues (36 out of 40 cases, 90%), but EphA2 expression was noted in only half of the HCC tissues (20 of 40 cases, 50%). In most of the hepatoma cell lines, the EFNA1 protein expression was positively associated with alpha-fetoprotien (AFP) expression but inversely associated with EphA2 expression. Furthermore, EFNA1 levels were detectable in the supernatant of the cultured hepatoma cells and in the serum of patients with HCC. In contrast, EphA2 expression was prominent in highly invasive hepatoma cells, and its overexpression was significantly correlated with decreased differentiation (r = 0.0248, p < 0.010) and poor survival (p = 0.0453) for HCC patients. EFNA1 and EphA2 may be useful serum markers for the detection of HCC development and progression, respectively.</P>
Mineralization of calcium phosphate in reverse microemulsion

In the present study, reverse microemulsion was prepared to regulate the mineralization of calcium phosphate. Calcium chlorideand sodium hydrogen phosphate aqueous solution were, respectively, dropped into the mixture of Span 80, Tween 80, andn-heptaneto get two kinds of emulsions.n-Buranol was used to adjust the emulsions to transparent state. Calcium phosphate was prepared byadding phosphate microemulsion to Calcium microemulsion. Heating method was used to induce the phase separation. After beingwashed with acetone and ethanol, the mineralized deposition were studied with scanning electron microscopy (SEM), X-ray dirac-tion (XRD) and transmission electron microscopy (TEM). The results indicated that the minerals in the deposition were mainlymonetite (DCPA) and the nanosized needle-like DCPA exhibited the preferential orientation along 200 direction..

<P><B>Abstract</B></P><P>Eph receptor 2 (EphA2) overexpression is frequently accompanied by the loss of its cognate ligand during tumor progression. However, the molecular mechanism of this ligand‐independent promotion of tumor by EphA2 remains unclear in highly malignant and fatal cholangiocarcinoma (CC). We examined the biological role of EphA2 in tumor growth and metastasis in CC tissues and cells according to the degree of differentiation and we explored the downstream signaling pathways of EphA2. Growth factor‐mediated EphA2 overexpression itself leads to the activation of the mammalian target of rapamycin complex 1 (mTORC1) and extracellular signal‐regulated kinase (ERK) pathways through ligand‐independent activation of EphA2 (phosphorylation of S897). An <I>in vitro</I> soft agar assay and <I>in vivo</I> orthotopic or subcutaneous tumor model showed that EphA2 enhanced colony formation and accelerated tumor growth, and which seemed to be mainly associated with Akt (T308)/mTORC1 activation. Aberrant expression and activation of EphA2 was also associated with poorer differentiation and higher metastatic ability. Enhanced metastatic ability was also observed in an orthotopic tumor model or lung metastasis model, correlating with Pyk2(Y402)/c‐Src/ERK activation in addition to activation of the canonical Raf/MEK/ERK pathway. The mTORC1 and Raf/Pyk2 pathways also appeared to affect each other. These results suggest that growth factor‐mediated EphA2 might be involved in tumor growth and metastasis through activation of the mTORC1 and Raf/Pyk2 pathways. Therapeutic strategies that target EphA2 and its downstream effectors may be useful to control CC. (H<SMALL>EPATOLOGY</SMALL> 2013;57:2248–2260)</P>

본 연구는 중국조선족 수용자들을 대상으로 한 심층인터뷰 분석자료를 토대로, 한국드라마 수용과정에서 나타나는 중국조선족의 민족 정체성을 분석하고자 했다. 연구결과, 첫째, 한국드라마 수용 동기에서 중국 조선족 수용자들의 민족 정체성이 다양하게 나타났다. 한국드라마 수용 동기나 선호이유에 있어서는 응답자의 민족 정체성 관련 여부에 따라서 세 부류로 나뉘어졌다. 첫 번째 부류에 속해 있는 응답자들은 민족성이 강하며 한국드라마를 수용하는 동기를 민족적인 요소와 스스로 연결시켰다. 두 번째 부류의 응답자들은 한국드라마 수용 동기를 민족과 연결 짓지 않았지만 친근감을 느끼게 되어 민족 정체성의 표현으로 판단되었다. 세 번째 부류는 민족 정체성보다 한국드라마 자체의 매력 때문에 수용하는 경우이다. 이러한 구분은 또한 연령층의 차이에 따라 다르게 나타났다. 나이가 많을수록 민족 정체성이 강한 수용동기를 나타내고 나이가 어릴수록 민족적인 요소보다도 한국드라마 자체에 매력을 느끼는 경향을 알아 볼 수 있었다. 둘째, 한국드라마 수용 과정에서 중국 조선족 수용자들은 자신의 민족 정체성을 발견하고 고민하며 강화하는 과정을 거치면서 민족 정체성이 형성되기도 한다. 셋째, 중국 조선족 수용자의 민족 정체성은 한국드라마 수용동기에 영향을 미치며 모순적, 우월적, 배타적, 자아 존중적, 탈조선(모국)적 등 5가지 특성을 보여준다. This paper takes the Chinese Korean Ethnic Minority as the research object and analyzes the ethnic identity that the Chinese Korean Ethnic Minority show in the process of watching Korean dramas according to a deep interview. Here are the results of my research as follows. First of all, in terms of the motives of watching Koreandramas, the Chinese Korean Ethnic Minority`sethnicidentitypresents variously. considering the motives and preference of watching Korean dramas, the characteristics of ethnic identity can be divided into three types. The respondents who belong to the first type show a strong sense of ethnic identity and are associated their motives of watching Korean dramas with ethnic factors involuntarily. Although the respondents who belong to the second type don`t attribute the motives of watching Korean dramas to ethnic factors, their affinity with Korean dramas also can be judged as a manifestation of ethnic identity. The respondents who belong to the third type watch Korean dramas due to being attracted by the charm of Korean dramas rather than ethnic identity. What`s interesting, the intensity of ethnic identity reflected from the motives of watching Korean dramas difference according to the various age groups. It obviously shows the tendency that the senior show a stronger sense of ethnic identity while the junior are attracted by the charm of Korean dramas regardless of ethnic restrictions. secondly, in the process of watching Korean dramas, the Chinese Korean Ethnic Minority will find, contemplate and strengthen their sense of ethnic identity by themselves. the last, affected by the motives of watching Korean dramas, the Chinese Korean Ethnic Minority’s ethnic identity has several characteristics such as contradiction, superiority, exclusiveness, self-respect and so on.

The aim of this study was to evaluate if oocytes, aspirated from postovulatory ovarian follicles of superovulated rabbits 14 h post-hCG administration, could be efficiently used as ooplasm recipients for somatic cell nuclear transfer (SCNT). Within a common SCNT protocol, a comparison between oocytes recovered by direct aspiration (aspirated) from available ovarian follicles and oocytes flushed out from oviducts (flushed) was carried out. The results showed that maturation and enucleation rates of aspirated oocytes were 70.7% and 69.2%, significantly lower than 95.3% (p<0.01) and 83.6% (p<0.05), respectively, from flushed oocytes. However, following enucleation of matured oocytes as ooplasm recipients for SCNT, no difference was recorded in fusion and cleavage rates, as well as blastocyst development from cleaved embryos or hatching of blastocysts between aspirated and flushed groups. Additionally, some matured aspirated and flushed oocytes were also used for immediate parthenogenetic activation and the resulting embryo development was not significantly different. Results from this study show the following: i) the majority of oocytes aspirated from postovulatory ovarian follicles of superovulated rabbits 14 h post-hCG administration are matured and can be used directly as ooplasm recipients for SCNT; ii) the reconstructed embryos derived from these oocytes have similar in vitro developmental ability to those flushed from the oviducts.
Identification of molecular markers for the oncogenicdifferentiation of hepatocellular carcinoma

The aim of this study was to identify molecular markers associated with oncogenic differentiation in hep-atocellular carcinoma (HCC). Using an unsupervised clustering method with a cDNA microarray, HCC (T) gene expression profiles and corresponding non-tu-tal 217 genes, 72 were expressed preferentialy in HCC tissues. Among 186 diferentialy regulated genes, there were molecular chaperone and tumor sup-pressor gene clusters in the Edmondson grades I and I (GI/I) subclass compared with the liver cirrhosis (LC) subclass. The Edmondson grades III and IV (GIII/IV) subclass with a poor survival (P = 0.0133) contained 122 diferentialy regulated genes with a cluster containing pared with the GI/II subclass. Imunohistochemical analysis revealed that ANXA2, one of the 72 genes pref-erentially expressed in HCC, was over-expressed in the sinusoidal endothelium and in malignant hepatocytes in HCC. The genes identified in the HCC subclasses wil be useful molecular markers for the genesis and pro-gresion of HCC. In addition, ANXA2 might be a novel marker for tumor angiogenesis in HCC.
