The zebrafish (Danio rerio) is now the prominent vertebrate model system for clarification of the roles of specific genes and signaling pathways in development. Zebrafish embryos provide many advantages for experimental and genetic analysis of vascular development. In this study we describe the use of zebrafish model system for the experimental study of angiogenesis. We established several technical methods to define cellular and molecular alterations in vascular development; whole-mount in situ hybridization, microangiography, and alkaline phosphatase staining. We also introduced the overexpression experiments by using in vitro-transcribed synthetic mRNAs and vascular-specific gene promoters. For the gene knock-down analyses, we challenged morpholino antisense oligonucleotide system. With these efforts, the zebrafish might be used as a powerful tool for the screening of effective angiogenic factors.

최근 흡연을 시작한 젊은 성언에서 급성 호산구성 폐렴의 발생에 대한 증례가 보고되고 있다. 급성 호산구성 폐렴은 대개 발열을 동반하기 때문에 초기에는 감염성 폐렴으로 잘못 진단, 치료되는 경우가 많다. 아직 정확한 병태 생리 및 조직학적 소견은 밝혀져 있지 않지만 병력 청취 및 임상 양상, 방사선 소견의 관찰을 통해 급성 호산구성 폐렴을 의심하고, 진단을 위해 기관지 폐포 세척액 검사를 시행한다면 좀 더 쉽게 진단 내릴 수 있을 것이다. 본 저자들은 최근 처음 시작한 흡연에 의해 발생한 급성 호산구성 폐렴을 진단하고, 스테로이드를 투여하여 성공적으로 치료한 환자 2예를 경험하였기에 이를 문헌 고찰과 함께 보고하는 바이다. Acute eosinophilic pneumonia(AEP) is characterized by acute febrile respiratory illness associated with diffuse pulmonary infiltration and pulmonary eosinophilia. The specific etiology for acute eosinophilic pneumonia is elusive. By some investigators, cigarette smoking is suggested as a causative substance which can cause AEP. In recent, the authors experienced two cases of AEP following cigarette smoking. Both cases had characteristic features including age around 20 years, new onset smoking before occurance of AEP, diffuse infiltration on chest radiography, pulmonary eosinophilia based on bronchoalveolar lavage and acute improvement after steroid therapy. These clinical features are resemble with previous smoking induced AEP case reports. Base on these clinical features, cigarette smoking associated AEP could be diagnosed more easily.
Expression Pattern of Gangliosides during Co-culture of Human and Micro-pig Endothelial Cells

Ganglioside are ubiquitous membrane component in mammalian cells and suggested to play important roles in various cell functions such as cell-cell recognition, differentiation and transmembrane signaling. These compounds are localized in a glycosphingolipid-enriched microdomain on the cell surface and regulated by the glycosphingolipid composition. However, the role that gangliosides play in the adhesive interaction response by xenotransplantation is not yet clearly understood. In this study, we investigated expression patterns of gangliosides in tight contact between human umbilical vein endothelial cells (HUVECs) and micro-pig aortic endothelial cells (MPAECs). Analysis of ganglioside expression pattern showed that differential expression patterns of ganglioside in tight contact between HUVECs and MPAECs. Furthermore, we confirmed that cell adhesion molecule expression in tight contact between HUVECs and MPAECs. Taken together, these results suggested that differential expression of gangliosides in tight contact between HUVECs and MPAECs may play a role in adhesive interaction response in successful clinical xenotransplantation.

Gangliosides, sialic acid-conjugated glycosphingolipids, are involved in cell proliferation and differentiation. This study investigated the possible role of gangliosides in the dopaminergic neuron-like cells of human mesenchymal stem cells (hMSCs). We have reported the in vitro conditions for inducing hMSCs into dopaminergic neuron-like cells. hMSCs were induced in vitro to become dopaminergic neuron-like cells in 12 days by using a cocktail that includes sonic hedgehog and fibroblast growth factors. First, we investigated the characterization of hMSCs through FACS analysis. The mesenchymal stem cell specific markers, CD44 and CD105, were expressed in hMSCs while the hematopoetic markers, CD45 and CD117 were not. The cells developed a neuronal morphology expressing the neuronal markers Map2 and βⅢtubulin. RT-PCR analysis revealed the expression of dopamine specific genes such as TH, Pitx3, Nurr1 and DA transporter. Immunocytochemistry showed that hMSCs expressed GM3, GM2, GD1a, GD3 and GT1b based on tyrosine hydroxylase (TH) expression. Specifically, a significant decrease in GM3 and GT1b expression was observed during dopaminergic neuron differentiation. These results suggest that ganglioside GT1b may play a role in the dopaminergic neuron differentiation and development process of hMSCs.
Gicerin/Cd146 is involved in zebrafish cardiovascular development and tumor angiogenesis

<P>Angiogenesis plays an important role in vertebrate development and tumor growth. In this process, <I>gicerin</I>, which is known as a kind of cell adhesion molecule, has recently been reported to play an important role but its <I>in vivo</I> function is still unclear in developing vasculature. To address this issue, we used gain-of-function and loss-of-function analyses of <I>gicerin</I> in zebrafish. In the gain of function experiments using enforced expression of various domains of <I>gicerin</I> constructs, extracellular domain induced angiogenic sprouting defects, most notably in the intersegmental vessels, whereas the cytoplasmic domain of <I>gicerin</I> did not affect angiogenic sprouting. Moreover, morpholino-mediated knockdown of <I>gicerin</I> in embryos resulted in angiogenic sprouting defects in intersegmental vessels. Mechanistically, the angiogenic function of <I>gicerin</I> was found to be genetically linked to VEGF signaling in the knock-down experiments using <I>vegf-a</I> mRNA, VEGFR inhibitor and <I>gicerin</I> morpholino. In addition to the physiological angiogenesis during development, <I>gicerin</I> morphants efficiently blocked the tumor angiogenesis in zebrafish. Thus, knock-down of <I>gicerin</I> might have an important implication in controlling tumor angiogenesis.</P>
Characterization of Triterpenoid Saponins in Astragalus membranaceus using UPLC_DAD-QToF/MS

Astragalus membranaceus is a member of legume family. It has been used as a traditional medicine and reported as potential therapeutic effects including immunostimulant, antioxidative and antitumor properties. In this study, fourty triterpenoid saponins were analyzed from roots of Astragalus membranaceus using UPLC-DAD-QToF/MS. They are classified with cycloastragenol and cycloartane types. Among of them, seventeen saponins were tentatively identified by eight standards and constructed saponin library. astragaloside IV, astragaloside III, astragaloside II, astragaloside I, malonylastragaloside I were shown as major saponins, and these saponins contain common aglycone, cycloastragenol, which is substituted with different sugar moieties at the C-3 or C-6 positions. Especially, malonylastragaloside II, malonylisoastragaloside II were elucidated as new compounds in roots of Astragalus. This results provide a fundamental data that the composition of Astragalus saponins and development of functional products.

However, the role that gangliosides play in immune rejection response by xenotransplantation is not yet clearly understood. In this study, differential expression patterens of gangliosides of Micro-Pig solid organs, which are heart, kidney, liver, pancreas and spleen. We examined expression level of high-performance thin-layer chromatography (HPTLC) showed that all of solid organs contained GM3, GM2, GM1 and GD1a as major gangliosides. Especially, ganglioside GD1b expressed in liver and pancreas, not in heart, kidney and spleen. Also, GT1b expressed in liver, kidney and spleen, not in heart and pancreas. As a results suggest that differential expression patterens of gangliosides of Micro-Pig solid organs are related immune rejection response in xenotransplantation by biomarker.

Human adipose-derived stem cells (hADSCs) and dental pulp-derived stem cells (hDPSCs) have been considered alternative sources of adult stem cells because of their potential to trans-differentiate into multiple cell lineages. This study investigated the possible role of gangliosides in the osteoblast differentiation of hADSCs and hDPSCs. First, we investigated characterization of hADSCs and hDPSCs using FACS analysis. Mesenchymal stem cell specific markers, CD44 and CD105, were expressed but not hematopoetic markers, CD45 and CD117 in both of hADSCs and hDPSCs. High-performance thin-layer chromatography analysis showed that increased gangliosides were associated with differentiation of hADSCs and hDPSCs into osteoblasts. RT-PCR analysis confirmed that osteoblast specific genes, ALP, BMP-2, collagen were expressed in differentiated osteoblasts, however, the another osteoblast specific gene, osteocalcin, was not expressed. When hADSCs and hDPSCs were cultured under osteoblast- differentiation conditions, alkaline phosphatase (ALP) activity was increased in comparison to hADSCs and hDPSCs. Furthermore, specifically both ALP activity and ganglioside expression increased more in hDPSCs-derived osteoblasts than hADSCs-derived osteoblasts. These results suggest that gangliosides play a more important role in regulating the osteoblast-differentiation of hDPSCs compared to hADSCs.
