      • GC/MS를 이용한 잎담배 중 알칼로이드 함량 분석

        To obtain the optimum condition for analysis of 10 alkaloids in tobacco leaves, such as nicotine, nornicotine, anatabine, anabasine, myosmine, cotinine, 2,3'-dipyridyl, $\beta-nicotyrine,\;\beta-nornicotyrine\;and\;\beta-formylnornicotin$, 5 types of extraction method were investigated by GC-FID and GC/MS. The optimum condition of alkaloid extraction was achieved by using methanol:dichloromethane(1:3, v/v) after NaOH treatment. The use of mass selective detector (MSD) provided unambiguous nicotine related alkaloid analysis. Alkaloids in various tobacco leaves were extracted with the optimum extraction condition and quantified by GC/MS/SIM mode. Compared with concentrations of alkaloids among the various tobacco leaves, the concentration of alkaloids was generally in the order burley > flue-cured > oriental tobacco. In flue-cured tobacco leaves, the order of concentration of alkaloids was nicotine > anatabine > nornicotine > $\beta-nicotyrine\;>\;\beta-formylnornicotine\; >\;myosmine\;>\;2,3'-dipyridyl\;>\;cotinine\;>\;anabasine\;>\;\beta-nornicotyrine$. However, in the case of burley and oriental tobacco leaves, the concentration of nornicotine was higher than that of anatabine.

        간헐외사시 수술직후 발생한 과교정의 자연 회복 양상

        Purpose: To assess the factors which are related to recovery time from overcorrection that occurred immediately after surgery. Methods: We reviewed the medical records of intermittent exotropia patients who had undergone bilateral lateral rectus muscle recession from February 2002 to July 2004. Fifty patients who developed overcorrection of less than 20 PD on the immediate postoperative day were included in this study. Age at surgery, sex, refractive power, preoperative deviation angle, Worth 4-dot test, and stereopsis were examined, and we evaluated the correlation between these factors and recovery time. Results: Thirty-six patients (72%) recovered to orthoposition within 4 weeks postoperatively. Significant correlations were found between the recovery time and poor stereopsis (p=0.02), and suppression of the Worth 4-dot test (p=0.01). There was no significant relationship between the recovery time and age at surgery, sex, refractive power, and preoperative deviation angle. Conclusions: In cases of immediate postoperative overcorrection of less than 20 PD after intermittent exotropia surgery, factors related to delayed recovery were poor stereopsis and suppression of the Worth 4-dot test.

        사전재해영향성검토 시 합리적인 홍수유출 모의모형 선정에 관한 연구

        도시개발은 불투수면적의 증가를 가져오는 요인이다. 영구저류지는 도시개발 유역의 우수유출을 저감시키는 시설이다. 본 연구에서는 사전재해영향성 제도에서 합리적인 홍수조절방안을 제시하고자 하였다. 먼저 홍수량 계산에서 도달시간 채택에 관한 문제점을 고찰하였다. 홍수유출 모의모형의 적합성 검토는 전원유역과 도시유역에 대하여 수행되었다. 홍수유출 모의모형은 2009년 7월 7일과 9일의 실제 호우 시 측정자료를 통하여 검증되었다. 그 결과, 전원유역과 도시유역에 적합한 모형은 각각 운동파 모형과 SWMM인 것으로 분석되었다. 본 연구에서 수행한 홍수량 산정모형과 합리적인 홍수설계 방안은 국내의 영구저류지 설계 시 기초 자료로 활용될 수 있을 것이다. 수문학적 분석방법은 영구저류지의 합리적인 용량 및 규모산정에 활용될 것으로 사료된다. Urban development is a cause of expansion of impervious area. A permanent storage is operated as a method of reducing runoff of watershed. The purpose of study is to propose reasonable choice of simulation model for rainfall-runoff in the prior review system on disaster effect. First, we indicated problem about concentration time choice in the flood simulation. To test the adequacy of a rainfall-runoff simulation model, We analyzed characteristics of rainfall-runoff about urban and natural watersheds. A simulation model was calibrated with the storm of july 7 to July 9 in 2009. From the result, we proposed that SWMM and kinematic wave method as the flood simulation models for urban and natural watersheds. A simulation model and design method of a permanent storage for flood that is proposed in this study will be useful for practical design of flood simulation. The hydrologic analysis method of the study can be used for capacity evaluation of permanent storage plan.

        전해수 생성온도에 따른 그람양성균과 그람음성균의 살균 효과

        본 연구에서는 다양한 온도의 물로 제조한 전해수를 이용하여 그람양성균과 그람음성균에 대한 살균력을 확인하였다. 전해수의 물성은 물의 온도가 높아질수록 pH와 유효 잔류염소 농도 값이 높아졌으나, 산화환원전위 값은 감소하였다. 4, 22, 40°C의 물을 이용하여 생성한 전해수에 Escherichia coli O157:H7을 15분 동안 처리하였을 때 각각 1.18, 4.47, 5.46 log CFU/mL 감소하였으며, Staphylococcus aureus는 각각 0.72, 4.90, 5.54 log CFU/mL 감소하였다. 이를 통해 전해수를 생성하는 물의 온도가 증가할수록 살균 효과가 높아지는 것을 알 수 있다. 4, 40°C의 물을 이용하여 생성한 전해수에 균주를 처리한 경우, Listeria monocytogenes의 D 값(일정한 온도에서 90 %의 미생물이 사멸하는 데 걸리는 시간)이 각각 6.60, 1.57분으로 나타나 가장 낮았으며, 22°C 전해수에서는 Salmonella Enteritidis의 D 값이 2.92분으로 가장 낮은 값을 나타내었다. 그람양성균과 그람음성균을 비교하였을 때 모든 온도에서 D 값에 대한 유의적인 차이는 나타나지 않았다(P>0.05). 본 연구 결과는 전해수를 제조할 경우 높은 온도의 물을 이용하여 제조하는 것이 미생물 살균 효과가 높다는 것을 보여주고 있다. 따라서 전해수를 생성할 때 물의 온도를 고려하여 높은 살균력을 나타내는 전해수를 생성할 수 있으며, 이는 물의 온도 외에도 물의 경도나 물의 종류 등을 고려하여 최적의 살균 전해수를 제조함으로써 식품산업에 적용하기 위한 자료로 활용될 것으로 기대된다. Electrolyzed activated water (EAW) has been reported to exhibit strong bactericidal effects on foodborne microorganisms. However, the disinfection efficacy of EAW is affected by factors such as water source and hardness. This study investigated bactericidal effects of EAW against three gram-positive (Bacillus cereus, Listeria monocytogenes, and Staphylococcus aureus) and three gram-negative (Cronobacter sakazakii, Escherichia coli O157:H7, and Salmonella Enteritidis) foodborne pathogens. Six strains were treated with EAW prepared at different water temperatures (4, 22, and 40°C) for 15 min, and D-values were generated. The results show that the lowest D-values for Lis. monocytogenes by EAW produced at 4°C and 40°C were 6.60 and 1.57 min, respectively. The lowest D-value for Sal. Enteritidis by EAW produced at 22°C was 2.92 min. D-values of all strains treated by EAW produced at 40°C decreased significantly compared to those treated by EAW produced at 4°C (P<0.05). These results demonstrate that applying EAW produced at warm temperature is more effective for reducing foodborne pathogens for food safety.

      • 담배 중 pH와 Nicotine 함량에 의한 Free nicotine 측정

        Characterizing nicotine delivery from tobacco products is important in the understanding of their addictive potential. Most previous studies report total nicotine and have not differentiated between nicotine in its protonated or free-base form. The amount of free nicotine calculated by determining pH and nicotine contents. The pH and nicotine contents in smokeless tobacco product, tobacco products and tobacco leaves were analyzed by Health Canada-Official Method T-310 and CORESTA Recommended Method $N^{\circ}62$. The content of free nicotine was calculated according to the Henderson-Hasselbalch equation and the value of $\alpha_{fb}$(the fraction of nicotine that is in the free base form) by using a pKa value of 8.02 for nicotine. The percentage of free nicotine then was calculated by dividing the free nicotine content by total nicotine content. The pH value and percentage of free nicotine ranged from 5.01 to 5.45 and $0.10\%\;to\;0.27\%$ in cut tobacco and 5.10 to 7.10 and $0.12\%\;to\;10.73\%$ in tobacco leaves, respectively.

        초등 ICT 활용교육에의 전자 포트폴리오 평가 절차 모형 개발

        Sometimes the quantitative evaluation method does not explain students' full capacity in educational fields. In this point of view, performance assessment has been adapted widely since the assessment provides a comprehensive qualitative evaluation as well as quantitative evaluation. Portfolio is one of the most effective methods in performance assessment. However, the portfolio requires longer time in evaluation, physical space for storing data gathered from students. Electronic portfolio can overcome the above disadvantages of the traditional portfolio and increase students' ICT application capacity. The purpose of this study is to develop electronic portfolio assessment processing model and present concrete activities for each stage in the model. Our model consists of four stages, Preparation, Activity, Evaluation, and Application, respectively. Especially, our model is designed to provide different activities for students and teachers in each stage. Our model is expected to provide a good guidance to students, teachers and parents in understanding and utilizing portfolio assessment. Particularly our model will provide the comprehensive guidance to teachers in assessment process.
