        패턴인식법에 의한 시판 녹차의 산지 및 채엽시기 추정

        Quantitative analysis of (+)-catechin (C), (-)-epigallocatechin (EGC), (-)-epicatechin (EC), (-)-epigallocatechin gallate (EGCG), (-)-epicatechin gallate (ECG) and caffeine in commercial green tea was carried out by HPLC employing gradient elution of 0.1% acetic acid and acetonitrile on ODS column. The optimized HPLC method provided satisfactory linearity, accuracy and precision. The relationship between the concentration of the components and cultivated region of the commercial green tea was not significant, while the concentration of EGCG, ECG and caffeine decreased significantly in the later harvested green tea samples (p<0.01). Multivariate analysis of the components was performed in order to characterize and evaluate the cultivated region and harvest period-related variation. Hierarchical clustering and discriminant analysis were applied to classify the geographical and seasonal origins of the green tea samples. The classification accuracy of the cultivated region and harvest period by discriminant analysis was 95% and 91%, respectively, indicating that this method could be reliable and convenient for the quality control of herbal products with different origin.

      • 키랄컬럼 쌍을 이용한 레보도파 제제 중의 l-도파와 d-도파의 HPLC 분석

        Levodopa preparations are used for the treatment of Parkinson's disease, since it is capable of crossing the protective blood-brain barrier (BBB), whereas dopamine itself cannot. d-Dopa could be often contained to a pharmaceutical levodopa preparations as impurity, however, the Korean Pharmacopeia doesn't regulate the amount of d-dopa in levodopa preparations. A new, precise and simple HPLC method for determination of l-dopa and d-dopa from levodopa formulation was developed using a pair of chiral columns, SCA and RCA, for the purpose of quality control. The selected mobile phase was composed of methanol and water (80 : 20) containing 0.01% phosphoric acid. The flow rate was 1.0 mL/min and detection wavelength was 210 nm. The intraday precision of l-dopa and d-dopa analysed by developed method were 2.71~8.18% and 2.94~6.03%, respectively. Both dopa enantiomers were separated by SCA and RCA column, a pair of chiral columns. However, RCA column was useful for the analysis of l-dopa, while SCA column for d-dopa. The method was validated according to ICH Guidelines and the method was found to be simple, accurate, precise, economical and robust.

      • Total oxy-radical scavenging capacity assay를 이용한 taurine과 hypotaurine의 항산화 활성 측정

        Taurine, a sulfur containing non-amino acid, is one of the most abundant intracellular amino acid in humans, and hypotaurine is a precursor of the synthesis of taurine. While it is widely believed that taurine may play an important role in protecting cells against toxic injury by functioning as an antioxidant, there is a lack of evidence to support this hypothesis. In this study, total oxy-radical scavenging capacity(TOSC) assay was used to evaluate antioxidant activity of taurine and hypotaurine against peroxyl radical, hydroxyl radical and peroxynitrite. The present results show that taurine does not have oxy-radical scavenging activity and hypotaurine is comparable to glutathione, an antioxidant substance, in oxy-radical scavenging activity. The results suggest that protective effects of taurine against oxidative stress may not be associated with its direct oxy-radical scavenging activity. Difference in oxy-radical scavenging capacity between taurine and hypotaurine may be ascribed to their sulfur oxidation state.

      • 기체크로마토그래피법을 이용한 오가피 중 아칸토산과 그 이성질체의 분석

        Acanthoic acid, continentalic acid and kaurenoic acid are bioactive diterpenoids which were structural isomers isolated from Acanthopanax species. For the quality evaluation of Acanthopanax species a gas chromatographic method for the separation and quantitation of these isomers was developed. Three compounds were successfully separated from the extracts of leaves, stems and roots of Acanthopanax species. GC-MS provided the identification of components separated from the extracts. Adequate separation of the components from Acanthopanax species was achieved on a fused-silica dimethyl polysiloxane capillary column (15 m x 0.25 ㎜, 0.25 ㎛ film thickness) operated with temperature programming from 160℃ held 0.5 min, increased at 11℃/min to 290℃ held 0.5 min with the carrier gas flow-rate (helium) of 5.1 ㎖/min. Among six Acanthopanax species only two species (A. koreanum and A. trifoliatus) contained the diterpenoids indication that the two species might possess the taxonomical similarity.
