전분 분해능이 있는 담자균성 효모, Filohasidium capsuligenum의 종내 및 Saccharomyces cerevisiae와의 속간 원형질체융합을 시도하였다. F capsuligenum CBS 6122-2의 종내 융합율은 각각 $2.9\times10^{-3}$ (ade+mel trp), $8.5\times10^{-3}$(ade+met)이고, F. capsuligemum CBS 4381 (cys his)과 S.cerevisiae 262-12-1 (hom3 thr1), X2180-1A (ade thr)와의 속간 융합율은 각각 $8.8\times10^{-6}$, $9.5\times10^{-4}$이었다. DNA 정량, 핵 염색, 자외선 조사에 따른 생존력 비교 그리고 형질 분리 분석 결과 등을 통해 종내와 속간에서 핵 융합이 일어난 것을 알 수 있었다. 또한, 종내 융합체의 경우 세포당 $\alpha$-amylase와 glucoamylase 생성능이 양천형에 비해 1.6-2.1배의 증진 효과가 있었다. Intraspecific fusion frequency between Filobasidium capsuligenum CBS6122-2 ade and met trp or met strains was $2.9\times 10^{-3}$ and $8.5\times 10^{-3}$, respectively. And intergeneric fusion frequency between Folobadidium capsuligenum CBS 4318 (cys his) and Saccharomyces cerevisiae 262-12-1 (hom3 thr1) or X2180-1A (ade thr) was $8.8\times 10^{-6}$ and $9.5\times 10^{-4}$, respectively. Nuclear fusion appears to occru in fusants between intraspecies and intergenera as strongly suggested by DNA content, nuclear staining, comparison of survival rate to UV light and mitotic segregation analysis. It was also found that $\alpha$-amylase and glucoamylase activity from intraspecific hybrids was 1.6-2.1 fold increased when compared with that from thier parents.

유당함유 울질로부터 알코올을 생산하기 위해 Saccharomyces cerevisiae X 2180-1 A, Candida pseudotropicalis ATCC 8619와 CBS 607 사이에 속간 및 종내 원형질체 융합을 시도하였다. S. cerevisiae X2180-1A(ade rho)와 C. pseudotropicalis CBS 607 (his met)간의 속간 융합율은 $1.0\times 10^{-5}$ C. pseudotropicalis ATCC 8619와 CBS 607 사이의 종내 융합율은 $3.9\times 10^{-6}$ - $1.0\times 10^{-4}$ 이었다. 이는 bovme serum albumin, myoinositol 그리고 ergosterol을 처리하지 않고 얻어진 원형질체 융합율과 비교해서 약 2-3.5배의 증가를 보여 이들 화합물이 속간 및 종내 융합을 증진시킨 것으로 사료된다. DNA 함량, 핵염색, UV 조사에 따른 생존력 비교 그리고 형질분리 후 교잡형 출현 등을 통해서 속간 (S. cerevisiae와 C. pseudotropicalis)과 종내 ( C psemdotropica/is strains) 융합체에서 핵융합이 일어난 것을 알 수 있었다. 또한 양친형과 비교해서 알코올 생성능이 증가된 종내 융합체를 얻었다. Intergeneric or intraspecific protoplast fusion between Saccharomyces cerevisiae X-2180-1A, Candida pseudotropicalis ATCC 8619 and CBS 607 was attempted to produce ethanol from lactose containing materials. Teh intergeneric fusion frequency between Saccharomyces cerevisiae X-2180-1A (ade rho) and Candida pseudotropicalis CBS 607 (his met) was $1.0\times 10^{-5}$. These values exhibited approximately 2-3.5 fold increase when compared with fusion frequency obtained without the treatment of bovine serum albumin, myoinositol and ergosterol, suggesting that these compounds may improve intergeneric of intraspecific protoplast fusion. Nuclear fusion appears to occur in fusants between intergenera(S. cerevisiae+C. pseudotropicalis) and intraspecies (C. pseudotropicalis strains) as strongly suggested by DNA content, nuclear staining, comparison of survival rate to UV light and isolation of recombinants after mitotic segragation. It was also found that alcohol production from intraspecific hybrids was somewhat increased when compared with that from their parents.

The factor analysis was applied to examine the effects of enviromental conditions on microbial populations in Kwangyang Bay from February to December, 1990. For this purpose, 17 abiotic and biotic parameters were determined for samples obtained from fifteen sites. Ecosystem in Kwangyang bay was regulated by 5 factors for four season. The microbial population was influenced mainly by the inflow fresh water, nitrogenous nutrients and water temperature. The overall size of microbial population was greater than that obtained in 1982.

This experiment was conducted to investigate the distributions of free sugars in commercial liquid yogurts as well as the changes of their sugar contents during the storage at different temperatures by HPLC. The results obtained were as follows: 1. The optimal operating conditions of HPLC for the detection of free, sugars in sample were as follows: mobile phase, acetonitrile + water = 78+22 (v/v); flow rate, 2㎖/min.; the chart speed of recorder, 0.25 ㎝/minute. Under the above conditions, fructose, glucose, sucrose and lactose were not only seperated at base line within 16 minutes but also quantitated by peak height. 2. The extraction of sugars is sample by ethanol was much better than that of acetonitrile. 3. Fructose, glucose, sucrose and lactose were isolated from all products examined (A.B.C.D); among these, sucrose was largest in quantity, and the others wore is order of glucose, fructose and lactose respectively. 4. The changes in sugar content after 7 days` storage at different temperatures showed that glucose, fructose and sucrose were all decreased while lactose was almost unchanged.

Two endoglucanase(EG, 1,4-β-D-glucan glucanohydrolase, EC 3.2.1.4) were purified from culture broth of P. verruculosum by ammonium sulfate precipitation, and HPLC-phenyl, HPLC-DEAE and HPLC gel filtration column chromatography. Purified enzymes were estimated to have molecular weight of 40 kDA(EG II), and 58 kDa(EG III) by SDS-PAGE and of 120 kDa(EG II), 140 kDa(EG III) by HPLC gel filtration. The optimal pH and temperature for these enzymes were found to be 4∼5 and 50-60℃, respectively. EG II and EG III hydrolyze carboxymethylcellulose but can not hydrolyze p-Nitrophenyl-α-D-glucoside and crystalline cellulose such as cotton, Avicel, filter paper. The hydrolytic products of CMC catalyzed by the enzyme were cellobiose, glucose and oligosaccharides. From the similarity in amino acid composition and elution profiles of tryptic peptides of EG II and EG III and cross reactivity of antibodies against the enzymes, it is suggested that amino acid sequences of these two EGs are very similar.

제1보에서 보고한 Aspergillus niger CAD 1의 beta-galactosidase를 cellulose triacetate를 담체로 하여 고정화조건을 검토하였던바, 13%(v/v)의 정제된 효소와 10%(w/v)의 cellulose triacetate를 30분간, 150stroke/min으로 진탕하면서 포괄시킨 고정화 효소의 활성이 가장 좋았다. 고정화 효소의 최적온도는 45℃, 최적 pH는 4.5로서 수용성 효소의 것과 같았다. 기질로서 ONPG와 유당에 대한 Km값은 각각 8.3×10^(-3)M과 117.0×10^(-3)M, Vmax값은 각각 30.3×10^(-3)M과 5.0×10^(-3)M이었다. 본 고정화 효소는 4℃에서 90일간 저장시에 90%, 45℃에서 5회 사용후에 70%의 잔존활성을 나타냈으며 유당분해율도 좋았다. The beta-galactosidase of Aspergillus niger CAD 1 described in previous report was immobilized to cellulose triacetate. The optimal conditions for immobilization of the enzyme were<br/> obtained when 13% (v/v) of the soluble enzyme (349 unit/㎖) was entrapped in fiber prepared from 10%(w/v) cellulose triacetate in methylene chloride. The optimal temperature and pH for the activity of the immobilized enzyme were 45℃ and 4.5, respectively, which showed the same values as those of the soluble enzyme. It was found that Km of the immobilized enzyme for lacotse exhibited high value compared with that of the soluble enzyme. The immobilized enzyme showed good storage stability, reusability, and also hydrolysis of lactose when introduced into skim milk and acidic whey.
