        An Efficient Method for Production of Extracellular Human Tissue Factor in Escherichia coli

        인간조직인자는 혈액응고인자 factor VII 과 복합체를 형성하며 연속적인 혈액응고 연쇄반응을 촉매하는 효소 활성체이다. 복합체 형성에 필수적인 이 조직인자의 세포 외 부분이, 기존의 융합 단백질 및 히스티딘 말단이 없는 새로운 발현 벡터에 의해 대장균 내에서 과량 발현 되었다. 봉입체 형태로 발현된 재조합 인간조직인자는 DEAE-Sephacel 크로마토그라피 기술을 적용하여 분리, 정제 및 구조적 복원이 동시에 시도 되었다. 정제된 재조합 단백질은 SDS-PAGE 분석에서 순수한 형태로 나타났으며, 생물학적 활성도 또한 기존의 조직인자와 거의 동등함을 보였다. 본 연구의 발현 및 정제 시스템은 이전의 보고에서 보여진 방법들에 비해 단백질 분해효소를 사용하지 않아 추가적인 크로마토그라피 과정이 필요 없어 좀 더 효율적이기 때문에 기존의 발현 시스템에 대해 대체할 수 있는 매우 유용한 방법으로 제공된다. Human Tissue factor is an essential enzyme activator that forms a catalytic complex with factor VII/VIIa, and catalyzes both the extrinsic and intrinsic blood coagulation cascades. The extracellular domain of human tissue factor is responsible for association with the biological partner. The efficient procedures for preparing biologically active human tissue factor are essential for the preclinical and clinical studies with coaguligands. An expression vector in Escherichia coli has been constructed to direct the production of extracellular human tissue factor without a fusion protein or a His? at the N-terminus. The recombinant human tissue factor was expressed in large amounts as a non-native state in E. coli. The recombinant protein was simply renatured during the DEAE-sephacel chromatographic purification procedure. Our expression and purification system does not require a protease treatment or an additional chromatographic step to remove a fusion contaminant, which provides a very useful alternative to conventional expression systems for the production of human tissue factor.

        충북지역산업의 가치사슬 확장을 위한 혁신클러스터 구축 조건에 관한 연구

        Purpose - This study studies the conditions for building an innovation cluster for the expansion of the value chain of local industries. Design/Methodology/Approach - Based on the spatial model of Krugman & Baldwin(2005), the analytical framework of this study is set up. Findings - The agglomeration of human capital strengthens the value chain by increasing both the forward and backward linkage effects. In addition, it is shown that the effect of human capital on the value chain strengthens as the preference for innovative products increases. A key condition for cluster formation holds when the transaction cost between regions falls below a certain level, and the core-periphery equilibrium is stable. When transaction costs between regions are eliminated, the core-periphery equilibrium becomes unstable, and a symmetrical equilibrium that prevents clusters from being agglomerated into one region becomes possible. Research Implications - First, there is a policy to keep transaction costs at a certain level. For example, a policy to increase the proportion of local university graduates can be considered for public enterprises or public institutions relocating to the provinces. The second is a policy that completely lowers the level of transaction costs. In this case, the act of being located in the capital region or located in the region itself becomes indifferent. Since there are no transaction costs, the firm’s profit is the same no matter where it is located. Therefore, it is implicated that local governments need to provide an incentive policy that attracts firms to locate in the region.

        [중소기업/중견기업/소상공인/지역기업] : 중소기업 영업직근로자들의 직무향상프로그램과 업무환경이 개인성과에 미치는 영향관계에서의 학습전이의 매개역할

        본 연구는 중소기업 직무수행자들이 학습한 내용에 대해 얼마나 전이를 인식하고 있는지, 본인의 직무와 성과에는 얼마나 도움을 주는 지에 대한관심에서 출발했다. 구체적으로 중소기업 직무능력향상 교육훈련과정과 업무환경이 학습전이를 매개로 하여 개인성과에 미치는 영향을 측정하고자 하였다. 연구결과, 중소기업 직무능력향상과정 교육프로그램의 교육내용 타당성, 전이설계, 강사능력 중에서 유일하게 전이설계만이 중소기업 영업직무자들의 개인성과에 미치는 영향관계에서 학습전이는 직무역량과 개인매출에 매개역할을 하는 것으로 나타났다. 또한, 기존 선행연구들과 달리 중소기업 업무환경이 중소기업 영업직무자들의 개인성과에 미치는 영향관계에서 학습전이는 모두 매개역할을 하지 못하는 것으로 나타났다. 본 연구 결과는 중소기업 재직자들의 교육훈련의 효율성 증대와 개인성과에 긍정적 영향을 주기 위해서는 중소기업이라는 특성을 감안할 때 사업주의 적극적인 지원 및 관심이 필요하고 매출에 직접적으로 여향을 주는 현장 학습전이가 높은 직무교육 훈련프로그램 설계와 이를 위한 설계자의 육성이 매우 필요하다는 것을 시사한다. This study began from an interest about how much small and medium business employees are aware of transferring what they have learned and to what extent learning transfer contribute to their jobs and attainments. The purpose of this study was to have an overall understanding about the relationships among learning transfer, individual performance and other variables when a certain amount of time has past since receiving a small and medium business job performance improvement educational training, and thereby provide basic data that will help educational training designed for employees at small and medium businesses contribute to the advancement of learning transfer and individual performance. To this end, the present study utilized research models proposed by Baldwin and Ford (1998), Holton (2005), and Holton and Baldwin (2003). These studies were employed since the proposed models have been frequently cited as exemplary models explaining the learning transfer process of educational training within an organization, and also because they have been received well as preceding studies helpful for an overall understanding of learning transfer process of business education due to their specific selection of subfactors. In order to verify these models, this study measured the mediation effect of learning transfer in terms of the effects the validity, transfer design, and instructor`s ability of small and medium business job performance improvement educational programs and work environment have on individual performance. The subjects of this study included 210 employees employed at a small and medium-sized business with less than 300 employees and who participated in a small and medium business job performance improvement course. Out of the 210 course participants, a total of 79 subjects responded to the research survey; survey respondents were composed of 93.7% male employess, 6.3% female employees, 67.1% of employees in administrative posts or higher class, and 332.9% of employees in worker level positions. For empirical verification, the present study conducted the survey related to the educational program immediately after the completion of the small and medium business job performance improvement program, and then, collected the questionnaires related to work environment, learning transfer, and individual performance six months later. Data from these two surveys were analyzed using SPSS 19.0 In order to raise the empirical validity of the study outcomes and to carry out feasibility assessment of questionnaire items, factor analysis was conducted using the principal component analysis (PCA). In doing so, nine questions that indicated factor loading of less than 0.6 were eliminated. Cronbach`s a was confined to verify the reliability of measurement tools. As a result, Cronbach`s a values on the research variables ranged between 0.834 and 0.984, indicating good reliability. In addition, the three-step regression analysis for mediation analysis proposed by Baron and Kenny (1986) was employed to verify the mediation effect of learning transfer in the effects small and medium job performance improvement program and work environment have on individual performance. According to the study results, it was found that learning transfer played some mediating role toward the effects small and medium job performance improvement program and work environment have on small and medium business sales workers` individual performance when demographic characteristics are held constant as control variables. Study results showed that learning transfer partially mediated the relationship between transfer design and job competency and that there is full mediation by learning transfer in the relationship between transfer design and individual sales. On the other hand, after regulating the demographic characteristics as control variables in the same manner, it was found that learning transfer played no mediating role in the effects small and medium business work environment have on small and medium business work environment have on small and medium business sales workers` individual performance. With respect to the study by Baldwin and Ford(1998) that suggests that learning transfer mediates the effects on performance by educational training design and work environment that serve as factors of educational input, the present study results support the part about educational training design. However, the present study differs from existing studies in that its results do not support the part about work environment such as organizational support, supervisor support, and restrictions by supervisors, which were identified as major influencing factors in most preceding international and domestic studies. Holton(1998) states that self-efficiency which affects learning transfer has to do with the leamer`s conviction that he/she has the ability to apply the contents learned from training to one`s job. Kim Youngmin (2005) views that there is a greater need for advises from surrounding people and support from the organization and supervisors in order to have employees experience learning transfer in their jobs. Kim also shows that, in the case of employees. With positions higher than managers, their experiences and awareness for their job performing abilities are more strongly related to learning transfer than the support from the organization and supervisors. With these points taken into consideration, it is surmised in this study that learning transfer has no mediating effect in how work environment influences job performance, since the subjects, with 67.1% of them having managerial positions or higher, were less influenced by factors such as organizational support, supervisor support, and restrictions by supervisors, but rather by the factor of personal volition. Considering that each learner is the person in charge of a given job at a small and medium business who can be the representative of that relevant job given the nature of small and medium business, this study has the following implications that will positively influence future learning transfer and job performance improvement at small and medium business. In the future. Third, due to the nature of small and medium business, the business proprieter holds the rights and the keys to developing human resources of the company. Without the business owner`s will, it is impossible to have the employees either actively participate in the educational training or apply what they have learned into practice after the training. Hence, studies covering various fields and ranges and efforts to verify the effectiveness of more educational programs should be carried out in the future in order to have business owners recognize that educational training have a positive effect on the performance of an organization and of each member of that organization.

        Objective : This study is to report the effect that taken by using oriental treatment on the patient with Fibromyalgia Syndrome(FMS). Methods : We have observed this case of patient treated by Acupuncture therapy, pharmacopuncture therapy and herbal medication. Results : There was improvement in condition of the patient who had Fibromyalgia Syndrome(FMS) after treated by using oriental treatment. Conclusion : Oriental treatments such as Acupuncture therapy, Pharmacopuncture therapy and herbal medication can be effective for the Fibromyalgia Syndrome(FMS).

        성공적인 컨설팅 프로젝트 완수를 위한 암묵적 지식의 융합

        본 연구는 중소기업의 성공적인 컨설팅 프로젝트 완수를 위한 방안을 제시하고자 컨설턴트의 몰입이나 기업의 흡수역량과 같은 암묵적 지식의 융합을 강조하고 있다. 컨설팅 수행 경험이 있는 중소,중견기업의 설문표본을 대상으로 모형분석 및 가설검증을 실시했으며 다음과 같은 결과를 도출하였다. 첫째, 컨설턴트의 몰입은 컨설팅 프로젝트 완수에 가장 높은 영향력을 주고 있으며 기업의 흡수역량을 통한 간접효과 역시 다른 변수들에 비해 상대적으로 높은 영향력을 주는 것으로 나타났다. 둘째, 수진기업 컨설팅 환경의 경우 경영자의 지원은 흡수역량에 직접효과를 주지 못하는 반면 제도적인 여건은 흡수역량과 높은 상관관계를 가지고 있는 것으로 나타났다. 셋째, 경영자의 지원은 흡수역량에 직접영향을 주지 못하나 제도적 여건을 통해 간접영향을 주고 있으며 해당 경로를 통해 컨설팅프로젝트 완수에도 영향을 주는 것으로 나타났다. 마지막으로, 컨설턴트 몰입의 총 효과는 흡수역량을 통한 수진기업 컨설팅환경의 총효과 보다 높게 나타났으며 두 요소의 통합효과는 컨설팅 프로젝트완성도를 높이는데 긍정적인 영향을 주는 것으로 나타났다. In this study, we focus on the importance of the convergence of tacit knowledge, such as commitment of consultants and absorptive capacity of consulting client firms, for the completion of successful consulting projects. In order to examine effects of the commitment of consultants and the absorptive capacity of consulting client firms, we establish the empirical models and test the research hypotheses by using the survey data of SMEs, in which those firms have been consulting experiences. First, the findings from the empirical test results show that the commitment of consultants has a higher impact on the completion of consulting projects than other variables. In the indirect effect through the absorptive capacity, by the same token, it has a relatively higher impact than other variables. Second, the test results indicate that in the consulting environment of the client firms, while the CEO’s support, which takes into account the understanding of consulting purposes and etc., is statistically insignificant to the absorptive capacity, the institutional conditions for the consulting projects are highly statistically significant at 0.1 percent level.Third, although the CEO’s support is insignificant to the absorptive capacity, it can affect indirectly to the absorptive capacity through the path of institutional conditions, and then eventually, the CEO’s support has a positive impact on the completion of consulting projects through those pathway. Finally, the total effect,sum of all possible indirect and direct effect, of consultants’ commitment is relatively higher than the absorptive capacity in the consulting client firms, but the combination between these variables makes it possible to increase the level of the completion of consulting projects.

        초기 말초성 안면신경마비에 대한 단무지 안추 신전법 병행 치료 효능의 임상적 고찰

        Objectives : The aim of this study is to investigate the effectiveness of combining "Danmuji Anchu Traction technique" on acute peripheral facial paralysis. Methods : Clinical studies were done 36 patients who were treated acute peripheral facial paralysis to Dept. of Acupuncture and Moxibustion, of Oriental Medicine Hospital Dongeui University from July 10, 2009 to September 15, 2010. subjects were randomly divided into 2 groups. : Complex oriental medical treatment without "Danmuji Anchu Traction technique" treated group (Group A, n=18), Complex oriental medical treatment with "Danmuji Anchu Traction technique" treated group (Group B, n=18). All process of treatment were performed by double blinding method. To compare the effectiveness of treatment applied for two groups, we used Yanagihara's unweighed grading system at before treatment, after 1week and 2weeks of treatment. Results: The Yanagihara's Scores of Group B and Improvement Indexes of Group B were higher than those of Group A, and showed significant difference statistically. "Danmuji Anchu Traction technique" can be available for relieving symptoms related with Acute Peripheral Facial Paralysis. And there were significant differences statistically between Complex oriental medical treatment without "Danmuji Anchu Traction technique" and Complex oriental medical treatment with "Danmuji Anchu Traction technique" on Acute Peripheral Facial Paralysis. Conclusions: These results suggested that "Danmuji Anchu Traction technique" effected for Acute Peripheral Facial Paralysis.

        차별적 무역정책의 정치경제학

        본 연구는 오늘날의 미국 및 EU 등 주요국의 차별적인 대외무역정책을 결정과정의 정치경제학적 특성을 분석한다. 정치경제적 의사결정과정에 대해 기업들은 로비라는 수단으로 유권자들은 투표라는 수단을 통해 참여할 수 있고 각 정당은 선거승리확률을 극대화하기 위한 대외무역정책을 선택한다. 주요 분석결과는 다음과 같다. 첫째, 정당간 정치철학이 동일한 경우 정당의 대외무역정책은 수렴하지만, 정당의 정치철학이 다른 경우에는 수렴하지 않는다. 둘째, 각 정당은 사회후생극대화보다 높은 극단적 수준의 대외무역정책을 제시한다. 셋째, 대외무역정책을 통해 달성할 수 있는 정치경제학 경우의 사회후생수준은 사회후생극대화 경우의 사회후생수준보다 항상 낮다. This study explores the political economy of the decision-making process for the discriminatory foreign trade policies of the United States and the European Union. In the political and economic decision-making process, firms can participate through lobbying, voters can participate through voting, and each political party selects the trade policy to maximize the partys probability of winning in the election. The results of the analysis are as follows. First, when the political philosophy of the political parties is the same, the trade policy of the political parties converges, but when the political philosophy of the political parties is different, they do not converge. Second, each political party proposes an extreme level of the trade policy. Third, the level of social welfare in the case of political economy is always lower than the social welfare in the case of benevolent government.

        노화과정의 생쥐 간과 신장에서 paraquat 에 의한 superoxide dismutases 와 metallothionein 의 유도에 관한 연구

        N/A Background: A number of theories have been proposed to explain the nature of aging process. Unfortunately, there is so far no theory that can completely explain all aging processes. In the present study, to investigate roles of inducibility of defense mechanisms by oxidative stress, cellular mRNA level of super oxide dismutases (SODs) and metallothionein (MT) as well as their inducibility by paraquat, an intracellular super oxide generator , was examined in the liver and kidney of the mice of aging process. Methods: The steady-state levels of SODs and MT mRNA and their induction by paraquat were determined by the RT-PCR assay in male mice of 4 ages, 1, 4, 8, and 12 months. Results: In the liver, the steady-state levels of Mn-SOD, Cu/Zn-SOD and MT mRNA increased until 8 months with age and decreased significantly at 12 months. Cu/Zn- SOD and MT mRNA were induced well by paraquat at all ages but Mn-SOD mRNA not at 12 months. In the kidney, their mRNA levels of Mn-SOD, Cu/Zn-SOD and MT increased with age. Mn-SOD mRNA was induced by paraquat only at 1 month but Cu/Zn-SOD mRNA not at all ages. On the other hand, MT mRNA was significantly induced by paraquat at all ages. Conclusion: These results suggest that SODs and MT are differentially expressed and induced according to the age and organs . In addition, it is thought that the lack of induction of Mn-SOD by oxidative stress in both the liver and kidney may be one of causative factors in the aging process while Cu/Zn-SOD and MT in the liver and MT in the kidney may play protective roles in the aging process. It is therefore implicated that the tissue antioxidant/prooxidant balance could be one of determinants of mean life span. (Korean J Med 61:430-438, 2001)

        종합격투기 스포츠의 마케팅

        연구목적: 스포츠 산업의 발전에 따라, 스포츠를 통한 마케팅뿐만 아니라, 스포츠의 마케팅도 점점 중요성이 높아지고 있다. 신생 스포츠단체인 UFC가 어떻게 치열한 경쟁에서 승리하여 세계 최대 종합격투기 스포츠 리그로 성장했는지에 대한 성공 요인 분석과 국제화 및 한국 시장에서의 마케팅 전략 분석을 통해서 '스포츠의 마케팅'에 대한 시사점을 찾아보았다. 연구 설계 및 방법론: 본 연구는 UFC에 대한 사례분석 방법을 이용하였다. 스포츠를 마케팅 대상으로 하는 종목/단체의 수가 한정되어 있고, 마케팅 전략 관점의 연구가 필요했기 때문이었다. 사례 연구는 2차 자료 조사와 전문가 인터뷰 등의 방법을 사용하였다. 분석 및 연구 결과: UFC의 성공 요인으로는 적극적 M&A를 통한 브랜드의 집중, TUF 프로그램, 선수를 활용한 스타 마케팅 및 애국심 마케팅 등을 들 수 있다. 그러나 이러한 UFC의 성공 요인들이 한국 시장에서의 마케팅 성공으로 이어지기 위해서는, 한국 시장 환경에 맞는 마케팅 전략과 사업모델에 대한 고려가 필요하다. 시사점 및 연구의 한계점: 본 연구는 UFC라는 신생 스포츠 단체의 마케팅 성공 사례 분석을 통해, '스포츠의 마케팅'에 있어서 브랜드 전략과 마케팅 커뮤니케이션, 글로벌 마케팅 등의 시사점을 제시한다. 그러나 사례 분석 중심의 탐색적 연구 방법론의 한계를 갖는다. 향후 연구방향: 향후 성과 분석을 포함한 실증 연구와 다른 스포츠와의 비교 분석 등을 통해 의미있는 결과를 도출할 수 있을 것으로 기대한다. 공헌점: 스포츠 산업의 성장에 따라, 스포츠간의 경쟁이 점점 치열해지고 있다. 본 연구를 통해 스포츠 종목/단체 등에 마케팅의 필요성을 인식시켜 주고, 스포츠의 글로벌 마케팅의 필요성, 각 팀의 성적보다 스포츠라는 생태계의 발전의 중요성 등의 시사점을 도출하였다. Purpose: With the rapid growth of the sports industry, the importance of 'marketing of sport' as well as 'marketing through sport' has greatly increased. The goal of this study is to explore the implications of 'marketing of sport' through the case of UFC (Ultimate Fighting Championship), a major MMA (Mixed Martial Arts) league. Design/methodology/approach: This study adopts a case study methodology to analyze the success factors of UFC along with a comprehensive marketing strategy in Korean market. Secondary data analysis and in-depth interviews are used to develop the case. Results/findings: Success factors of UFC can be summarized as aggressive M&A and brand unification, TUF(The Ultimate Fighter) program as an effective marketing communication tool, star marketing and patriotism marketing. However, these success factors are not sufficient conditions in Korean market because the UFC requires a more specific marketing program and business model in Korean market. Research implications/limitations: This study proposes several implications to sport-related organizations in order to conduct 'marketing of sport' through the success case of UFC such as branding, marketing communication and global marketing. However, this study is an exploratory study, which adopts a case study methodology that has several inherent limitations. Future work/research: The future direction of this study should include an empirical work or comparative study with other sports, hereby providing more meaningful results. Originality/value: The growth of the sports industry also arouses a strong competition among sports related organizations. This study suggest implications to the 'marketing of sport'. In addition, this study suggests the importance of global marketing of sports and sports as an ecosystem.
