        성성숙 후 거세가 한우 거세우의 성장 및 도체형질에 미치는 효과

        8 steers and 22 bulls were employed to investigate the effects of castration after puberty on the growth performance and carcass traits of Hanwoo (Korean cattle). Even though the steers castrated at 16 months of age had a 12.9% reduction and 15.9% reduction in average daily gain (ADG) compared to bulls during the period of 16 to 25 months of age, the steers only had a 3.4% reduction in gain (P = 0.26) from birth to 25 months of age, and there was no difference in ADG during this period. Longissimus dorsi area and dressing percentage of bulls were larger and higher than those of steers (P$lt;0.05). Steers showed a superior (P$lt;0.01) marbling score to bulls (Steers : 4.8±0.32, Bulls : 1.8 ± 0.19). Therefore the castration after puberty improves meat quality and has little effect on growth performance.

        The purpose of this study was to monitor the status of pedigree structure and to estimate the additive relationship and inbreeding coefficients of active Hanwoo proven bulls. A total 3,760 Pedigree records of proven and young bulls and heifers from 1983 to 2008 were used in this study. Intel Visual Fortran Compiler (Window Ver. 11.1) was used to compute the numerator relationship matrix(A) and to estimate the inbreeding coefficients using the self-made fortran program included three subroutines(MATCH, SHIFT, NRM). The ratio of relatedness among the active Hanwoo proven bulls ranged from 20.75 to 94.34% and the inbreeding coefficients of 4 proven bulls were estimated 0.78, 3.13, 0.05 and 1.56%, respectively. The estimated inbreeding coefficients were less than other researches. However, the ratio of relatedness among the active Hanwoo proven bulls was estimated very high. Because of these results, the probability of inbred animals will be increased. Therefore, cattle breeders will be considered the active selection index consisting of economic weight as well as relationship and inbreeding coefficients of animals when the proven bulls will be selected.

        영양수준 , 연령 및 체중이 한우 정액성상에 미치는 영향

        Thirty proven Hanwoo sires, ranging from 5 to 9 years old, were used to investigate the effects of dietary nutrition level, age and body weight on semen characteristics which were analyzed using CASA(Computer-aided sperm analyzer) and to determine the proper nutritive level, age and body weight for production of high quality semen. The groups fed a diet meeting the NRC requirements for maintenance(NRC) had more semen volume than the groups fed at additional 10% level(NRC + 10%). However, tendencies were the opposite in sperm concentration and motion characteristics. Sperm concentrations of 9 years old sires had lower than those of others and the sires below 800㎏ body weight were better than the sires over 800㎏ body weight in sperm motion characteristics[curvilinear velocity(VCL), straight-line velocity(VSL), beat-cross frequency(BCF)] and viability. Although the interaction of age and the nutrition level had not be significant on semen characteristics, VCL, VSL and ALH(amplitude of lateral head displacement) of the groups fed at NRC + 10% were better than those of the grops fed at NRC in every age. The sperm concentration of 9 years old sires with below 800kg of body weight were the lowest. The sperm motion characteristics of 7-8 and 9 years old were best in below 700㎏ and over 700㎏ of body weight, respectively. But individual difference of sperm motion characteristics were large in 9 years old sires. In every body weight except 700∼800㎏, the semen characteristics of groups fed at NRC + 10% were better than the those of groups fed at NRC. But the semen characteristics of the group of 700∼800㎏ body weight fed at NRC was the best among the those of others. Therefore, the proper age and body weight of Hanwoo sire for production of high quality semen would be 5∼8 years old with below 800㎏ of body weight. Feeding plane of sire below and over 700㎏ body weight would be fed at NRC + 10% level and NRC, respectively.

        거세가 한우의 도체특성과 혈청 대사물질에 미치는 영향

        Twenty steers and 20 bulls averaging 7 Mo. of age were employed to investigate the effects of casrration on the carcass characteristics, serum metabolites and hormones of Korean cattle from 7 Mo. to 25 Mo. of age. Dressing percent(P$lt;0.05), cold carcass weight(P$lt;0.01) and longissimus dorsi area(P$lt;0.01) of bulls and steers were 57.6±0.44; 58.6±0.36%, 306.9±0.97; 344.9±0.51Kg and 74.3±0.97; 84.7±0.15㎠, respectively. Steers had more marbling score than bulls(4.4±0.11 vs 1.5±0.22; P$lt;0.01). Differences between bulls and steers in serum insulin-like growth factor-1(IGF-1) concentrations and testosterone, stimulate protein synthesis and growth, were large(P$lt;0.01) after 15 Mo. and 9 Mo. of age, respectively. Serum cortisol concentrations had increased rapidly at 19 Mo. of age both in bulls and steers. Steers had higher cortisol concentrations, stimulate lipid metabolism and deposit of intermuscular lipid, than bulls. These results suggested that castration would be extended after 15 Mo. of age to increase beef quality and to conpensate the loss of weight gain due to castration.

        농후사료 급여수준이 비거세 한우의 증체와 부위별 지방조직의 지방산 조성에 미치는 효과

        The study was conducted with 63 Hanwoo bull calves of 193 days old for 19 months(up to 26 month of age) to examine the effect of feeding level of concentrate on the body weight gain, feed requirements and fatty acid composition of adipose tissues of various locations(subcutaneous, abdominal, kidney, intermuscular and intramuscular fats). The calves were allotted into 3 treatments according to the feeding level of concentrate(85, 100 and 115%), and fed concentrates of Grower(7∼10 month of age), Finisher I(11∼16 month of age), Finisher II (17∼20 month of age) and Finisher III(21∼26 month of age). The feeding levels of concentrates for the cattle of 85% and 115% were decreased or increased by 15%, respectively, up to 15 month of age based on the similar body weight to the cattle of 100%. The feeding levels of the concentrates for the cattle of 85% and 100% were continuously increased for the next 2 months, to make same levels as for the cattle of I15%, thereafter amount of concentrate were continuously increased based on the rate of body gain for all the cattle. The cattle were fed in a individual feeding system through the experimental period. The cattle were slaughtered at the ages of 6, 14, 18 and 24 month and adipose tissues were collected to analyze long chain fatty acids. Results observed from the study were summarized as follows Overall mean dry matter intake and body gain per day and feed requirements of Hanwoo bulls during the 19 months of feeding from 193 day old calves were 6.69㎏, 0.95㎏ and 7.06, respectively. Body gain of the Hanwoo bulls during experimental period was greatly affected by the feeding level of concentrate, but feed requirement was improved up to 20 month of age with the decreased feeding level of concentrate. Fatty acids of C_(16:0) C_(18:0) and C_(18:1) in the adipose tissues at various locations of Hanwoo bulls were dominated. The feeding level of concentrate did not affect the fatty acid composition except for C_(16:1) and C_(18:1) in subcutaneous fat, and C_(18:1) and C_(18:2) in intramuscular fat of the cattle that were fed at the level of 85% at the age of 14 month(P$lt;.05). No differences were observed in fatty acid composition of adipose tissues of each location for the cattle of 18 and 24 month of age among treatments. There were tendencies of greater C_(16:1) and C_(18:1) in subcutaneous and intramuscular fats while higher C_(18:0) in abdominal fat. There were also trends of greater unsaturated fatty acid compositions in subcutaneous and intramuscular fats and higher saturated fatty acids in abdominal, kidney and intermuscular fats. Percent C_(18:0) tended to be decreased while C_(16:1) C_(18:1) and C_(18:2) increased with the age of Hanwoo bulls.

        제라놀(Zeranol) 처리가 한우의 성장 및 혈청대사물질에 미치는 영향

        Twenty calves averaging 7Mo. of age were used to study the effect of zeranol implants(ZI) on growth, carcass traits and concentrations of hormone and serum metabolites in bulls and steers of Korean cattle. The animal were treated with none or three implants of zeranol at average ages of 7, 10 and 13Mo. There were significant(P$lt;0.05) differences between bulls and steers of control group in live weight, back fat thickness(BF), cold carcass weight and yield grade index. In steers, ZI increased serum IGF-1(Insulin-like growth factor-1) concentration, and resulted in greater live weight(10.5%) and BF(25.4%) compared to those of control. However, it did'nt affect marbling scores(P$gt;0.05). ZI in bullls tended to increase cortisol concentration, marbling scores and BF. The results indicate that zeranol implants had more advantage in steers than in bulls.

        Albumin 밀도 경사를 이용한 정자분리가 정자운동특성 및 수태율에 미치는 영향

        The 16 ejaculates from 3 Hanwoo sires were used to study whether the sperm separation by 6, 8 and 10% of bovine serum albumin (BSA) gradients increase the motional characteristics relating sperm fertility and pregnancy rate in Hanwoo. The motional characteristics of sperm were analysed by Computer-assisted sperm analyser (CASA). The sperm separated by each 6, 8, 10% of BSA gradients showed significantly hid value in motility, curvillinear velocity, straight-line velocity, average-path velocity, and beat-cross frequency as compared with control sperm(P$lt; 0.05). Artificial insemination was performed with sperm separated by 6% BSA gradients, which was revealed the highest sperm recovery. The pregnancy rate of sperm separated by 6% BSA gradients was 15.4% higher than that of the control sperm (P = 0.22). Therefore, the sperm separation by BSA gradients may be used to increase the motional characteristics of sperm relating the fertility of sperm and the pregnancy rate of artificial insemination in Hanwoo.

        엘크사슴의 냉동정액 생산과 인공수정

        To establish the system for frozen-semen production, oestrus synchronization, and artificial insemination (AI) in Elk deer, the characteristics of frozen-semen and conception rate by artificial insemination were observed. Semen were collected from 2 heads of elk (Cervus Elaphus canadensis) sire by an electroejaculator and diluted to 0.9- 1.1 X 10^9 sperms/straw with the extender containing glycerol, hydroxymethyl amino methane, citric acid, fructose, and 20% egg yolk. The diluted semen were chilled at 4℃ for 3-4 hours, prefrozen at -140㎏ for 30-40 minutes, and preserved at liquid nitrogen (-196℃). The motility, viability, and abnormality of frozen-thawed semen were 62.4-70.5%, 73.7-75.3% and 6.8-7.3%, respectively. Oestrus was synchronized in 48 heads of hind with progesterone- impregnated intravaginal devices (CIDRs). The CIDRs were removed on Day 14, and 10-250 IU pregnant mare's serum gonadotrophin (PMSG) were given to each hind. Oestrus exhibition rate was 95.8% (46/48). Forty-six hinds, showing oestrus, were artificially inseminated with frozen-thawed semen 60-62 hours after PMSG injection. Pregnancy was detected by ultrasonography at 40 days after Al. The overall conception rate was 78.3% (36/46). These results indicate that the system for frozen-semen production and AI established in this study may be adapted to deer industry.

        PCR 기법을 이용한 한우의 κ-casein 과 β-lactoglobulin 유전자의 다형성

        This study was performed to find genetic polymorphisms of κ-casein and β-lactoglobulin loci in Hanwoo using the PCR-RFLP. Genomic DNA was prepared from the blood of 327 Hanwoo. Amplified products of 530bp and 262bp fragments were obtained from the amplification of K-casein and β-lactoglobulin loci by PCR. The K -casein AA genotype showed a 530bp fragment in amplified products of K -casein loci with the restricted enzyme digestion of Taq I. The κ-casein AB genotype showed 530bp, 3446p and 186bp fragments and BB genotype showed 3446p and 186bp fragments. The β-lactoglobulin AA genotype showed 153bp and 109bp fragments in amplified products of β-lactoglobulin loci with the restricted enzyme digestion of Hae III. The β-lactoglobulin AB genotype showed 153bp, 109bp, 79bp and 74bp fragments and BB genotype showed 109bp, 79bp and 74bp fragments. The κ-casein genotype of Hanwoo gave frequencies of 45.9%, 37.7% and 16.4% for AA, AB and BB types. The rc-casein A and B alleles were 0.647 and 0.353, respectively. The β-lactoglobulin genotype of Hanwoo gave frequencies of 6.4%, 28.8% and 64.8% for AA, AB and BB types. The β-lactoglobulin A and B alleles were 0.208 and 0.792, respectively.
