        강원도 춘천시의 소나무(Pinus densiflora)에서분리한 내생균류의 다양성

        Red pine (Pinus densiflora) is a dominant tree species in Korean forests. The diversity ofendophytic fungi was investigated by isolating fungi from different parts of a red pinetree during the fall and spring seasons. One hundred and fifty-two strains of endophyticfungi were isolated and classified into 42 species belonging to 29 genera. Among these,74 (49%), 47 (31%) and 31 strains (20%) were isolated from branches, leaves, and stems,respectively. The most frequently isolated species were Daldinia childiae and Biscogniauxiamaritima (28.9% and 9.8%, respectively). D. childiae was the dominant endophytic funguson branches (20 strains) and leaves (19 strains). B. maritima was most frequently isolatedfrom branches. The isolation frequencies on stems of B. maritima (6 strains) and D. childiae(5 strains) were also high. Fifty-seven isolates belonging to 19 species and 17 genera wereisolated in the spring, and 95 strains belonging to 35 species and 24 genera were isolated inthe fall. D. childiae was the most frequently isolated fungus in both spring and fall (21 and 23strains, respectively). B. maritima was also frequently isolated in both spring (7 strains) andfall (8 strains).

        평생 환경학습권 확보를 위한 환경학습계좌제의 인증기준 개발

        The purpose of this study is to develop and present certification standards for the introduction of the environmental learning account system to promote individual citizens' participation in sustainable environmental learning. The feasibility was reviewed through the identification of characteristics by life stage, the development of certification standards based on this, and the typology of environmental learning through pilot registration. As a result of the study, environmental learning was classified into history, competency, participation, and practice. Here, environmental learning history includes completion history and academic background, environmental learning competency includes career and qualification, and environmental learning participation and practice include activity history. In particular, in consideration of the specificity of environmental learning, participation and practice were suggested to include various activity experiences in connection with the local community. Based on this, a feasibility review was conducted through certification criteria and pilot registration of the environmental learning account system, and a plan to expand the effectiveness of the system was presented. 본 연구는 지속가능한 환경학습에 시민 개개인의 참여를 촉진하기 위한 환경학습계좌제도 도입을 위한 인증기준을 개발 및 제시하는 데 그 목적이 있다. 생애단계별 특성파악, 이를 기반으로 한 인증기준 개발, 시범등록을 통한 환경학습의 유형론을 통해 타당성을 검토하였다. 연구 결과 환경학습은 역사, 역량, 참여, 실천으로 분류되었다. 여기서 환경학습이력은 수료이력 및 학력을 포함하고, 환경학습역량은 경력 및 자격을 포함하며, 환경학습 참여 및 실습은 활동이력을 포함한다. 특히 환경학습의 특수성을 고려하여 지역공동체와 연계한 다양한 활동경험을 포함하도록 참여와 실천을 제시하였다. 이를 바탕으로 환경학습계좌제도 인증기준과 시범 등록 등을 통한 타당성 검토를 거쳤으며 제도 실효성 확대 방안을 제시하였다.

      • 초등학생을 대상으로 클래스팅을 활용한 디지털 미디어 리터러시 교육에 관한 연구

        미디어의 발달은 미디어를 단순히 메세지를 담고 전달하는 그릇으로서 바라보던 기존의 개념을 넘어서, 인간을 둘러싸고 있는 주요한 환경으로 바라보게 하였다(이정춘, 2004). 이는 미디어 자체의 속성을 학습함으로써 환경에 대해 적극적이면서 올바른 커뮤니케이션을 수행하는 미디어 리터러시 교육이 필요함을 시사한다. 현재 각종 유해 정보의 무분별한 유포, 악의적 댓글에 의한 명예훼손, 사생활 침해 등의 디지털 미디어 상의 여러 사회 문제는 이러한 미디어 리터러시 교육이 제대로 이루어지지 않았기 때문에 발생하고 있다. 그 피해와 심각성이 점점 낮은 연령층으로 확대되고 있어 초등학생을 대상으로 하는 디지털 미디어 리터러시 교육이 시급하다. 이를 위해 본 연구는 초등학생을 대상으로 교육용 소셜 미디어인 클래스팅에 활용하여 디지털 미디어 리터러시 교육 프로그램을 구상하였다. 프로그램은 르니홉스가 제시한 디지털 미디어 리터러시의 구성요소 5가지 영역(접근, 분석 및 평가, 창조, 성찰, 행동)을 중심으로 하여 각 영역에 따른 필수 활동을 클래스팅 기능에 대응시켜 교사와 학습자의 역할을 모두 제시하였다. 이 연구는 지금까지 체계적으로 이루어지지 않았던 초등학생을 대상으로 디지털 미디어 리터러시 교육을 시도하고자 하는데 의의가 있다. The development of media makes us think media as the important environment, surrounding us, not just as a medium, containing and conveying messages.(Lee Jung-choon, 2004). It means that we should have a media literacy education, doing active communication with the environment. Various social problems, such as indiscreet circulation of harmful contents, defamation by malicious comment and invasion of privacy, occur because media literacy education has not been done well. More and more younger students have been exposed to these problems, so the digital media literacy education should be done as soon as possible for elementary students. For this, this paper formulates the digital media literacy education program by utilizing instructional social media, Classting, targeting elementary students. The program suggests roles of teachers and students together, by corresponding each required activities to the function of Classting. These required activities are formed, being based on five sections(approach, analysis, assessment, creation, introspection, behavior). The paper has a signification in carrying out a digital media literacy education for elementary students, which has not been systematically done until now.

        결명자로부터 분리된 Aurantio-obtusin의 항염증 활성

        Cassia tora L. have been used as a folk medicine in Korea. This study investigated anti-inflammatory effect of aurantio-obtusin isolated from C. tora. We isolated aurantio-obtusin from 50% ethanol extracts of C. tora L. We investigated the anti-inflammatory effects of aurantio-obtusin on the lipopolysaccharide (LPS)-stimulated inflammatory response in murine macrophage cell line (Raw 264.7). To investigate the cytotoxicity of aurantio-obtusin on RAW 264.7 cells, MTS assay was performed. RAW 264.7 cells were treated with aurantio-obtusin at different concentrations (12.5, 25, 50, 100 μM) for 30 h. The result showed that aurantio-obtusin had no cytotoxic effect in a concentration range of 12.5 – 100 μM. To determine the effect of aurantio-obtusin on LPS-induced NO production, the NO concentration measurement was performed. RAW 264.7 cells were treated with aurantio-obtusin at 12.5, 25, 50 and 100 μM for 24 h, and the results showed that the NO production of aurantio-obtusin-treated cells compared to LPS alone treated group was significantly decreased in a dose-dependent manner. Pretreatment of aurantio-obtusin inhibited LPS-induced NO production in a dose-dependent manner. To find out inhibitory mechanisms of aurantio-obtusin on inflammatory mediators, we examined the PGE2 pathways. As a result, PGE2 were decreased in a dose-dependent manner by aurantio-obtusin. The release of interleukin-1b (IL-1b) and IL-6 were also reduced. Moreover, aurantio-obtusin suppressed LPL-induced IκB-a degradation. These results suggest that the down regulation of NO, PGE2, IL-1b and IL-6 expression by aurantio-obtusin are achieved by the downregulation of NF-κB activity.

        Sprague-Dawley Rats을 이용한 결명자 물 추출물의 26주 반복 경구투여 독성시험 및 4주 회복시험

        Background: Cassia tora L., an annual or perennial plant of the Fabaceae family, is traditional medicine with various biological activities, including anti-constipation and, anti-inflammation. Chemical compounds such as anthraquinone glycoside and naphthalene derivatives have been isolated from this plant. Cassia tora L. is a common contaminant of agricultural commodities, but is toxic to cattle and poultry. Methods and Results: To investigate the potential toxicity, Cassia tora L. aqueous extract (CO) was administered orally to rats for 26 weeks at 0 (control), 300, 1,500 and 3,000㎎/㎏/day (n = 10 for male rats for each dose). The positive control comprised animals orally administered anthraquinone 100㎎/㎏/day. There was no treatment-related mortality. An increase in the kidney weight was observed at 3,000㎎/㎏/day of CO and anthraquinone 100㎎/㎏/day. Macrophage infiltration in the colon was observed at CO 1,500 and 3,000㎎/㎏/day and anthraquinone 100㎎/㎏/day, but there were no significant toxicological changes in the incidence and severity of the finding. Conclusions: The oral no-observed-adverse-effect level (NOAEL) of CO was 3,000㎎/㎏/day in male rats and no target organs were identified. In addition, 300㎎/㎏ was found to be the no-observed-effect level (NOEL) for systemic toxicity under the conditions of the study.

        Loperamide로 유도된 변비모델에서 Lactobacillus casei에 의해 발효된 볶은 결명자 물 추출물의 효과

        Background: Constipation is one of the most common functional gastrointestinal disorder. The present study examined the ability of water extract of fermented (FRC) and non-fermented (NFRC) roasted Cassia tora to improve intestinal function and reduce constipation in a rat constipation model.Methods and Results:Different concentration of FRC and NFRC were orally administered loperamide (5 ㎎/㎏; LOP) reduced the number, weight, and water content of feces, as well as intestinal transit motility. However, 24 h-(24 hour fermented roasted-Cassia tora) 300 ㎎/㎏ FRC administration increased the number, weight, and water concent of feces, compared to that seen in the LOP group, and also improve intestinal transit mitility and, the thickness of distal colon and mucous fluid.Conclusions:The results of the present study indicated that LOP-induced constipation was improved by treatment with FRC. Therefore FRC could be used to develop functional foods or natural medicine for constipation. However, further study is needed to clarify how fermentation improves the medicinal properties of roasted C. tora.

        고함량 폴리감마글루탐산(PGA) 청국장에 대한 칼슘 생체이용률 및 골다공증에 미치는 영향

        본 연구는 칼슘 생체이용률 시험에 수컷 랫드를 이용하여 고함량 폴리감마글루탐산 청국장이 기존 청국장보다 칼슘의 생체이용률을 증가시키는지 알아보고자 실시하였다. 고함량 폴리감마글루탐산(PGA) 청국장 및 기존 청국장을 투여하고 0.5, 1, 2, 3, 4, 5시간에 채혈하여 칼슘 함량을 측정한 결과, 고함량 PGA 청국장이 기존 청국장보다 투여 후 3~5시간에 칼슘 흡수율이 증가할 것으로 생각한다. 골다공증 시험에 난소를 적출하여 폐경을 유도한 후 PGA 함유량이 증가한 청국장을 12주간 경구투여 하여 골다공증에 미치는 효과를 연구하고자 하였다. 혈청 검사에서 오스테오칼신은 청국장 200 mg/kg 투여군에서 OVX 대조군보다 유의성 있는 감소가 관찰되었고, 프로게스테론은 청국장 200 mg/kg투여군에서는 OVX 대조군보다 유의성 있는 증가가 관찰되어 청국장이 골다공증 개선에 효과가 있는 것으로 생각한다. 간장 조직액의 지질과산화물은 OVX 대조군보다 대조약 및 청국장 투여군에서 모두 감소 경향이 관찰되어 청국장 투여로 지질과산화물 생성을 감소시키는 것으로 기대된다. 대퇴골에 대한 조직병리학적 검사에서 대퇴골 원위부 골간단의 해면골 감소가 대조약은 OVX 대조군보다 유의성 있는 감소가 관찰되었으며, 청국장 200 mg/kg 투여군에서도 감소가 관찰되어 청국장이 해면골 감소를 억제시키는 것으로 생각한다. 이상의 결과는 고함량 PGA 청국장이 기존 청국장보다 투여 후 3~5시간에 칼슘 흡수율을 증가시킬 것으로 생각하고, 난소적출 후에 일어나는 골격 대사의 이상에도 청국장이 골 교체율 억제, 호르몬 변화, 항산화 및 해면골 감소 억제등 유익하게 나타남으로써 임상상 폐경기에 나타나는 골다공증을 어느 정도 예방시킬 수 있음을 시사하며, 폐경 후 여성의 골대사에 유익한 효과를 줄 것으로 생각한다. Various types of fermented soybean foods are consumed in various Asian countries, including China, Indonesia, Japan, Korea, and Vietnam. Cheonggukjang is a Korean whole soybean paste fermented by Bacillus subtilis and regarded as a healthy food. The objective of this study was to investigate the Ca-bioavailability and anti-osteoporotic effects of cheonggukjang (CGJ) containing high poly-gamma glutamate (PGA) contents in an animal model. Blood samples were collected from the jugular vein at 0, 0.5, 1, 2, 3, 4, and 5 h after a single oral dose in male rats. Ca-bioavailability in CGJ containing high PGA contents was approximately 3∼5 times higher than that of natural CGJ. Female rats were either sham-operated (sham; n=5) or surgically ovariectomized (OVX; five animals per group) and then administered to OVX control, raloxifene hydrochloride (RLX) 1 mg/kg/d, or CGJ containing high contents of PGA (20 and 200 mg/kg/d) for 12 weeks. Serum osteocalcin concentration was significantly lower in the CGJ 200 mg/kg/d group compared with the OVX control group. Serum progesterone concentration was significantly higher in the CGJ 200 mg/kg/d group compared with the OVX control group. Reduction grade of the trabecular bone decreased in the RLX 1 and CGJ 200 mg/kg/d group compared with that of the OVX control group. In conclusion, CGJ 200 mg/kg/d may have inhibitory effects on osteoporosis in OVX rats, and Ca-bioavailability was improved in CGJ containing high PGA contents.
