        Comparison of the Antioxidant Activities of Organic Solvent Fractions of Leaf and Root Extracts of Peucedanum insolens Kitagawa

        This study compared and analyzed the antioxidant activities of various organic solvent fractions from the leaves and roots of Peucedanum insolens Kitagawa. For this study, the dried leaves and roots of P. insolens Kitagawa were first extracted using 70% ethanol. The extracts were sequentially sub-fractionated in the order of hexane, chloroform, ethyl acetate, n-butanol, and water. The results revealed that the distribution of total phenolic contents by organic solvent fractions showed the same pattern in both the leaves and roots, with the highest in the ethyl acetate fraction (101.1±1.0 mg vs 71.2±3.4 mg of GAE/mg), but the lowest content in the hexane fraction (9.5±0.2 mg vs 7.5±2.1 mg of GAE/mg). The distribution of total flavonoid content in the organic solvent fractions showed the same pattern as that of total phenolic content. The results of DPPH, ABTS, and FRAP assays showed that the leaf and root extracts exhibited free radical scavenging activity in the same pattern, particularly, the ethyl acetate fraction had the highest activity. These results indicate that not only the roots of P. insolens Kitagawa but also the leaves possess potential substances that exhibit strong antioxidant activity. Significant correlations (R=0.903, p<0.0001, DPPH radical; R=0.891, p<0.001, ABTS radical; R=0.745, p<0.05, FRAP radical) between total phenolics and radical scavenging activities, but also significant correlations (R=0.867, p<0.001, DPPH vs. ABTS radicals; R=0.882, p<0.0001, DPPH vs. FRAP radicals; R=0.973, p<0.0001, ABTS vs. FRAP radicals) between radical scavenging activities were found in the organic solvent fractions. Therefore, as in the roots of P. insolens Kitagawa, the leaves possess strong antioxidant capacity and can be used as the main antioxidant material.

        제주도 어류양식장에 대한 미생물학적 오염도 조사

        The purpose of this study was to monitor and compare the contamination levels of heterotrophic bac-teria and pathogenic bacteria (total coliforms, Escherichia coli O157:H7, Salmonella spp., Vibrio parahaemolyticus,Staphylococcus aureus and Bacillus cereus). The samples colected at fish farms located in the Jeju-island were inves-tigated from March to August in 2006. The contamination levels of S. aureus, B. cereus and Salmonella spp. ranged0-3.3×102 CFU/ml, 0-2.2×102 CFU/ml and 0-5.1×101 CFU/ml, respectively, and 85% of moist pelet feeds were con-taminated with S. aureus, B. cereus and total coliforms. But total coliforms and E. coli O157:H7 were not detected inoliver flounder (Paralichthys olivaceus). For the production of safe oliver flounder (Paralichthys olivaceustion of sanitation for employees, control of fish feed, and continuous monitoring for microorganism will be required.

        포스트텐션 경량콘크리트 일방향 슬래브의 휨 거동 및 비부착 긴장재의 응력

        Six post-tensioned lightweight concrete (LWC) one-way slabs were tested under a symmetrical one-point top loading system. According to tendon profile, the specimens were classified into two groups: S-group for slabs with straight tendons and V-groups for slabs with harped (counter-moment shaped) tendons. The span-to-tendon depth ratio($L/d_p$) varied to be 15, 35 and 55 in each group. The flexural cracking moment and ultimate moment of one-way slab tested and stress increase measured in unbonded tendons were compared with those of post-tensioned normal-weight concrete (NWC) beams compiled from the comprehensive database and predictions obtained from the design equations specified in the ACI 318-08 provision. Test results showed that the distribution and width of flexural cracks was significantly affected by the $L/d_p$, indicating that the larger the $L/d_p$, the smaller the crack width. On the other hand, tendon profile had a significant influence on deflection and flexural capacity of one-way slabs, showing a smaller deflection and a higher strength in slabs with harped tendons than in slabs with straight tendons. At the same value of reinforcing inde, the flexural cracking moment and ultimate moment of post-tensioned LWC one-way slabs were pretty comparable to those of post-tensioned NWC beams. In addition, flexural strengths and tendon stress increase of post-tensioned LWC one-way slabs could be conservatively predicted by the ACI 318-08 provision.

        Comparison of Light Emission in InGaN/GaN Light-emitting Diodes with Graded, Triangular, and Parabolic Quantum-well Structures

        The light emission properties of InGaN/GaN quantum well (QW) light-emitting diodes with non-square layers with graded, triangular, and parabolic shapes are investigated using multiband effective mass theory. These results are compared with those of conventional InGaN/GaN QW structures. The spontaneous emission peak of non-square QW structures is shown to be improved compared to a conventional QW structure. In particular, the parabolic QW structures shows a slightly larger emission peak than the graded or triangular QW structure. This can be explained by the fact that a smaller In composition in the well is needed to give a transition wavelength of 440 nm.

        The amyloid β-protein (Aβ) is the principal component of the senile plaques characteristic of Alzheimer’s disease (AD) and elicits a toxic effect on neurons in vitro and in vivo. Many environmental factors including antioxidants and proteoglycans modify Aβ toxicity. In this study, we have investigated the protective effects of organic solvent-extracts of Polygonatum odoratum fractions pre-extracted with methanol on Aβ-induced oxidative cell death in cultured rat pheochromocytoma (PC12) cells. For this study, we used MTT reduction assay for detection of protective effects of organic solvent-extracts of Polygonatum odoratum fractions pre-extracted with methanol on Aβ25-35-induced cytotoxicity to PC12 cells. We also used cell-based β-secretase assay system to investigate the inhibitory effect of organic solvent-extracts of Polygonatum odoratum on β-secretase activity. We previously reported that methanol extracts of Polygonatum odoratum strongly attenuated cytotoxicity induced by the three A fragments (Aβ25-35, Aβ1-42 Aβ1-43) to both SK-N-MC and PC12 cells. In the present study, we found that the organic solvent extracts of Polygonatum odoratum fractions pre-extracted with methanol had the strong protective effects against Aβ25-35-induced cytotoxicity to PC12 cells and inhibitory potency to β-secretase activity. These results suggest that organic solvent extracts of Polygonatum odoratum fractions pre-extracted with methanol may contain the protective component(s) against Aβ-induced cell death in PC12 cells as well as inhibitory component(s) to β-secretase activity.         제주 동부 연안 넙치 양식장의 미생물학적 오염도 조사

        본 연구는 2007년 6월부터 10월까지 제주도에서 양식장이 가장 많이 밀집되어 있는 제주도 동부 연안에 위치한 양식장 4개소를 대상으로 하여 사육 용수, 양어 사료, 양식 넙치의 미생물학적 오염도를 조사하였다. 식품병원성 미생물 중 일반세균과 총대장균군 그리고 Escherichia coli O157:H7, Salmonella spp., Vibrio parahaemolyticus, Staphylococcus aureus, Bacillus cereus를 조사 하였다. 이번 연구를 수행하는 동안 동부지역은 집중적인 호우로 인해 전년보다 400 ㎜ 이상의 많은 강우량을 나타내었다. 사육 용수의 검사결과 V. parahaemolyticus, Salmonella spp., S. aureus, B. cereus가 빈번하게 검출되었고, 양어 사료의 경우 Salmonella spp, S. aureus, B. cereus는 각각 0∼4.3×10³ CFU/ml, 0∼2.7×10⁴ CFU/ml, 0∼7.6×10³ CFU/ml 검출 되었다. 또한 양식 넙치의 미생물 검사 결과 적은 양이지만 Salmonella spp., S. aureus, B. cereus가 검출되었고, 총대장균군과 E. coli O157:H7은 검출되지 않았다. 이번 조사 결과로 보아 미생물학적 위해요소는 양식장의 작업 환경이나 주변 환경 등의 환경적 요인이 크게 작용하는 것으로 사료되며, 양식장의 위생관리에 작업자들은 주의를 기울여야 할 것으로 생각된다. 앞으로 이와 관련하여 수산물 위생에 대한 좀 더 체계적인 시스템을 도입하여 적용한다면 수산물에서 야기되는 문제점들이 해결되어 제주도의 청정 브랜드의 이미지를 더욱 부각시킬 수 있으며, 안전하고 깨끗한 수산물의 생산이 가능할 것으로 생각된다. Level of microbial contamination was examined in four fish farms of Jeju east coast for sanitary indication of bacterial contamination such as heterotrophic bacteria, Escherichia coli O157:H7, Salmonella spp., Vibrio parahaemolyticus, Staphylococcus aureus, Bacillus cereus and total coliforms. The samples were collected and investigated from June to October in 2007. Bacterial species of S. aureus, B. cereus and Salmonella spp. were frequently detected from the seawater of above fish farms. Salmonella spp., S. aureus, B. cereus were detected in the range of 0∼4.3×10³, 0∼2.7×10⁴, 0∼7.6×10³ CFU/ml, respectively, from the fish feed used in all four selected farms. Additionally, Salmonella spp., S. aureus, B. cereus were detected in oliver flounder (Paralichthys olivaceus) of these farms, however total coliforms and E. coli O157:H7 were not detected during our experimental period. For the production of microbiologically safe and healthy oliver flounder, proper quality control of feed, sanitation programs, and continuous monitoring of microorganism are essential practices, which required to include in the farm management system.
