        대사과정에 있어서 율속단계(律速段階) 결정에 관한 연구(Ⅰ) : 발아종자자엽(發芽種子子葉)에 있어서 산소활성에 미치는 광선의 영향 The Affect of the Light to the Enzyme Activities in the Cotyledons of the Germinating Seeds

        In order to study the rate control step in the protein metabolic course of the chlorophyll formation, the transaminase activities which are obtained freely in the extracts of cotyledons of germinating peas at the light and the dark places, are measured in Beckman mopel D.U, spectrophoto meter at 490 mu. In this case, of two enzymatic reaction products; oxalacetic acid and pyruvic acid, the former is converted to pyruvic acid by aniline citrate and after each pyruvate phenyl hydrazones are extracted by toluenes: when this is treated with strong alcoholic alkali, a colored hydrazone is formed and it is measured by above apparatus. The estimated G.O.T. and G.P.T. in the germinated cotyledons at dark and light places considerably differ in their activities; G.O.T. and G.P.T. activities which are formed at the light are more increased than at the dark and also they differ in their rates through germination, though G.O.T. activity increment is smoothly but that of G.P.T. is more sharply, and they are considered to be directly affected to the chlorophyll formation and indirectly to the growth. G.O.T. and G.P.T. in each fractions of cell in the cotyledons should be formed by dissociation of zymogens in the microsomal fractions and it seems to promoted by light. In the formation of the chlorophyll, the protein metabolism occurred mainly in the microsomal fractions and the rate determining step is found at the point where the zymogene that is able to produce G.P.T. is activated, and this activation is promoted by light as noted above.

        Present investigation deals with extraction and detection of flavoooid constituents in rice. Qualitative test for the presence of flavonoid constituents in rice obtained from the polishing processes No. 1 to No. 8 shows all positive results. Thin layer chromatography using chloroform-methanol-benzene(5 : 3 : 2) as solvent system gives nine spots, suggesting flavonoid constituents, Further investigation is continuing to separate purely the flavonoid constituents by column chromatography.

      • 韓國産 Papaya의 有機成分에 關한 硏究

        全南 麗川郡 栗村面에서 Vinyl house에 栽培되고 있는 Carica papaya의 雌性株果와 雌雄同株果의 熟期에 따른 有機成分과 遊離糖의 變化와 蛋白質의 性質에 대하여 分析 檢討하여 얻어진 結果를 要約하면 다음과 같다. 1. 粗蛋白質은 10~20%, 粗脂肪은 2~5%, 粗纖維는 1~3%의 範圍로 粗蛋白質은 雌雄同株果가, 粗脂肪은 雌性果株가 약간 높으나 熟成에 따른 變化는 兩株果가 현저한 差를 나타내지 않았다. 2. 可溶性 無窒素物은 70~75%의 범위로 雌雄同株果, 雌性株果間에는 差는 없으나 收穫期에는 조금 減少했다. 3. 遊離糖은 雌雄同株果가 雌性株果에 비해 含量이 높았으며 大部分이 還元糖으로 非還元糖은 含量이 아주 적었다. 4. TLC의 結果로는 遊離糖中 還元糖은 Glucose와 Fructos이며 非還元糖은 Raffinose인 것 같으며 兩株果間에 差는 없었다. 5. Protease 活性은 雌雄同株果가 높았으며 雌性果株는 經時的으로 減少하였으며 雌雄同株果는 收穫期에 上昇하는 傾向이었다. 6. 蛋白質의 電氣泳動類型은 兩株果에 있어서 差는 있으나 各 株果의 熟期에 따른 類型의 變化는 一定한 傾向이 없다. 7. 兩株果의 共通帶數는 10이며 2, 3, 13, 17 帶數가 一般的으로 共通性을 띠었다. 8. 一般的으로 Rm値가 낮은 側이 蛋白質 含量이 많으며 Rm値가 높은 側이 蛋白質 含量이 적었다. In order to obtain the basic information of utilization of Carica papaya which was cultivated under vinyl house at Youchon, Cbonnam province, organic constituents, free sugars, protein properties and enzyme activity depending on the ripening stages of papaya fruit, were determined. The obtained results were summarized as follows. 1. The crude protein contents, crude fat and crude fiber, was 10-20%, 2-5%, 1-3%, respectively. The crude protein in hermaphrodite fruit and crude fat in female fruit were slightly higher than the other. 2. The content of soluble nitrogen-free compound in both fruits was 70-75%. There was no difference between hermaphrodite and female fruit but it was slightly decreased in hermaphrodite fruit at harvesting stage. 3. The free sugar contents were relatively higher in hermaphrodite fruit than female fruit, and most of free sugars were reducing sugar form rather than non-reducing sugar. 4. From the thin layer chromatographic result, it seemed that free sugars in papaya fruit were grucose and fructose which are reducing sugars, and raffioose which is non-reducing sugar. 5. Protease activity of female fruit was higher than hermaphrodite but decreased in harvesting stage. It was thought that papain content was decreased in harvesting stage. 6. In the electrophoretic patterns of protein, there was some difference between hermaphrodite and female fruit. 7. Proteins were divided into 13 different protein bands by their Rm values, and the common bands in both fruits were 2, 3, 10, 13 and 17 band. 8. It seemed that the protein content was higher in low Rm value than in high Rm value.

      • RECOVERING OF ETHANOL AND METHANOL FROM THE WASTE LIQUID OF ALCOHOL DISTILLATION PART(Ⅱ) : STUDIES BY PHYSICAL METHOD 物理的 方法의 硏究

        連續蒸溜와 共沸蒸溜를 더욱 效率的으로 操作하기 爲해서 새로운 류族型 鐘泡塔을 設計하였다. 이 塔을 사용하여 實驗한 結果에 依하면 에타놀과 메타놀처럼 比揮發度가 1.73程度로 적은 알코홀 同族體는 理論的인 最小還流比의 3倍를 取해도 完全分離가 되지 못했으며 兩成分과 共沸混合物을 形成하는 溶媒(n-Hexane)를 使用했을 때도 共沸蒸溜의 效率이 적다는 것을 알 수 있다. 따라서 이와 같은 兩成分의 分溜에 있어서는 第一報¹ 에서 言及한 바와 같이 比揮發度를 增加시키는 分離劑(酸化硼素)를 使用하는 것이 有效하다는 것을 알았다. 著者들이 完全分離를 目的으로 設計한 塔이 僅少한 誤差를 나타내는 理由는 McCabe Thiele의 方程式과 Dalton의 法則을 適用한데 있다고 생각되며 其他 많은 因子들이 關與하는 蒸溜塔의 複雜性에도 原因이 있다고 推測된다. 따라서 完全分離를 위해서는 上記한 誤差에 對한 相當한 補正이 加해져야 되리라고 믿는다. 이 實驗을 위해서 著者들이 設計한 류族型 鐘泡塔의 特徵은 다음과 같다. ① 擴散平衡에 要하는 時間을 充分히 줌으로써 塔의 濃縮部와 回收部의 Time interval을 增加시킬 수 있으므로 塔效率이 높다 ② 各 段에서 Hold-up를 減少시킴으롯써 接觸面積이 擴大되어 分離效率을 增加시킨다. ③ 段의 直徑을 크게 하고 塔의 높이를 줄일 수 있으므로 塔의 建設費가 低廉하다. ④ 蒸溜 操作이 간단하여 分離效率이 높다. 끝으로 本 硏究를 위해서 硏究費를 支給하여 주신 文敎部에 感謝를 드린다.

      • RECOVERING OF ETHANOL AND METHANOL FROM THE WASTE LIQUID OF ALCOHOL DISTILLATION (PARTⅠ) : STUDIES BY CHEMICAL METHOD 化學的 方法의 硏究

        메타놀과 에타놀을 순수分離하기 위하여 分離劑로서 鹽化칼슘과 酸化硼素를 사용하였다. 메타놀과 에타놀의 混合溶液에서 形成된 鹽化칼슘 錯化物들을 共히 140℃以上에서 加熱하면, 메타놀이 約 25%의 分離效率로서 순수하게 分離되었다. 混合溶液으로 부터 兩 成分을 完全 分離하기 위해서는 上記한 操作을, 메타놀 30% 含有한 에타놀을 試料로 하는 경우 35回 되풀이하여야 한다. 鹽化칼슘 錯化物과는 對照的으로, 硼素의 메칠과 에칠에스텔사이의 比揮發度는 메타놀과 에타놀의 比揮發度에 比해서 顯著하게 增加하고 있다. 따라서 適當한 分溜操作을 適用한다면 兩成分은 容易하게 分離될 수 있을 것이다. 分離後에 加水分解와 蒸溜를 實施함으로써, 메타놀, 에타놀, 硼酸은 좋은 收率로 回收될 수 있다. 더우기 메타놀과 에타놀 同族體의 混合溶液에서 生成되는 鹽化칼슘 錯化物과 硼素에스텔의 分子組成을, 多少의 制限이 있다고 하더라도, 간단한 物理的 方法에 의하여 決定할 수 있다는 것을 알았다. 끝으로 本硏究를 위해서 硏究費를 支給하여 주신 文敎部에 感謝를 드린다.

        대사과정에 있어서 율속단계(律速段階)의 속도론적 해석 : [제1보] 수정된 효소적 방법에 대하여 [Part Ⅰ] A Modified Emzymatic Method

        The determination of rate limiting step in the metabolic process is of great importance to understand the metabolic properties. In this paper the authors propose a modified enzymatic method instead of thermodynamic method. This method is based on the assumption that the over-all rate increment would be larger than by any other steps to which the individual enzyme are added respectively, provided that the enzyme participated in the rate limiting step is added to the reactions composed of n steps of metabolic process with which n kind of enzymes are concerned. The present paper deals with analysis and discussion about some factors having influence on the proposed process, mainly about the metabolic process constituted with homogeneous steps. The results show that the determination of rate limiting step by a modified enzymatic method is feasible, provided that some restrictions arc added in any type of mechanisms.

      • 신공법에 의해 제조한 흑삼의 아토피 억제효과

        The purpose of this study was to investigate the effects of crude saponin fraction of black ginseng(SFBG) on atopic dermatitis in vitro. The house dust mite(Dermatophagoides pteronissinus, DP) plays a key role in the pathogenesis of allergic diseases, including atopic dermatitis and asthma. Monocyte chemo-attractant protein 1(MCP-1/CCL2)/interleukin-6(IL-6)/IL-8(CXCL8) plays a pivotal role in mediating the infiltration of various cells into the skin of atopic dermatitis. We found that SFBG potently suppressed the elevated production of MCP-1, IL-6 and IL-8 induced by D. pteronissinus extract(DpE) treatment in the human monocytic cell line, THP-1 cells. Also, SFBG potently suppressed the elevated production of IL-6 and IL-8 induced by DpE treatment in the human eosinophilic cell line, EoL-1 cells. These results suggest that SFBG have an anti-atopic property, and that in future further development of main effective constituents in SFBG can provide a novel therapeutic agent for atopic dermatitis.

        쿠마린 유도체의 합성과 그들의 항암효과

        A novel series of 4-senecioyloxymethyl-6,7-dimethoxycoumarin, isolated from Crinum latifolium, was prepared by reacting 4-bromomethyl or 4-bromomethyl-6,7- dimethoxycoumarin with various carboxylic acids and examined for their anti-angiogenic activities in human umbilical vein endothelial cells (HUVECs). Among them, 4e, 4f, 4g and 4i with noncyclic moiety exhibited potent anti-angiogenic activity. However, compounds with cyclic moiety such as phenyl, pyridinyl, thiophenyl and furanyl group did not exhibit any anti-angiogenic activity. Also, compounds 4f and 4g which exhibited strong anti-angiogenic activity in a dose-dependent manner showed antitumor activity.
