        국내 작은도서관의 열람실 규모에 따른 배치유형 특성에 관한 연구 - 광주광역시·전라남도 지역을 중심으로 -

        With the recent increase in income levels, demand for leisure, cultural life, and education has increased. Policies such as public libraries and small libraries promotion act have begun to build many libraries in the public sphere of public life. Currently, public libraries are concentrated in the metropolitan area. The size of reading rooms is growing, but the quality for visitors such as indoor environment and characteristics of visitors is insufficient. The characteristics are analyzed according to the classification of public libraries by region. We will find out the direction of space planning and indoor composition for public libraries to be built in Gwangju and Jeollanam-do later. Analyze the VGA (Visibility Graph Analysis) of the floor plan in the library reading room. The children's reading room in the city center is divided into reading seats and bookcases area. Children's reading rooms in the suburbs are organically inclusive arrangements. The type includes the general reading room in the city center. The layout of the suburbs was not common and varied.

        비자 유박 추출물이 최종당화산물과 콜라겐 교차결합에 미치는 효과

        최종당화산물은 환원당과 단백질, 지질 또는 핵산 사이의 당화반응에 의해서 생성되는 물질로 이 물질은 피부 노화 과정에 밀접하게 연관되어 있다고 보고되어 있다. 본 연구에서는 항산화 활성, 최종당화산물 생성 저해, 콜라겐과 최종당화산물의 교차결합 저해 및 억제, 엘라스타제 활성 저해 효능 시험을 통해서 비자나무 유박 추출물의 항노화 효과를 평가하였다. 시험 결과, 비자나무 유박 추출물은 폴리페놀과 플라보 노이드를 함유하고 있으며, DPPH와 ABTS 자유라디칼을 50% 소거할 수 있는 농도는 각각 16.4 μg (Dried materials, DM)/mL과 16.7 μg DM/mL 로 관찰되었다. 또한, 비자나무 유박 추출물은 농도 의존적으로 최종당화산물 생성과 엘라스타제 활성을 저해하였고, 콜라겐과 최종당화산물의 교차결합을 저해 및 억제하였다. 따라서 본 연구 결과를 바탕으로 비자나무 유박 추출물은 항노화 효능이 있는 화장품 소재로서 활용가치가 있음을 확인하였다. Advanced glycation end products (AGEs) are final products formed by glycation reaction between reducing sugars and proteins, lipids or nucleic acids. These AGEs are related to progress of skin aging. In this study, we evaluate anti-aging activity of Defatted Torreya nucifera seed extract (DTSE) through antioxidant, anti-glycation, anti-elastase and inhibitory and breaking activity on the cross-linking of AGEs to collagen assay. Results showed that DTSE contained polyphenols and flavonoids. The IC<sub>50</sub> values of 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity were 16.4 μg (Dried materials, DM)/mL and 16.7 μg DM/mL, respectively. DTSE also inhibited the formation of AGEs, elastase activity and cross-linking of AGEs to collagen as well as broke existing cross-linking of AGEs to collagen in a dose-dependent manner. Consequently, our findings suggest that DTSE could be useful as a cosmetic material with anti-aging activity.

        유암 처방에 사용되어온 한약재 복합 처방전의 항산화 및 항균활성에 관한 연구

        본 연구에서 동의보감에 기록된 유암 치료에 사용되어온 5가지 한약재 처방전인 십육미류기음, 단자청피탕, 지패산 Ⅰ, 지패산 Ⅱ와 청간해울탕의 열수와 에탄올 추출물에 대한 총 페놀 함량의 비교 분석과 항산화능 및 항균활성능을 비교 조사하였다. 열수와 70% 에탄올 추출물의 수율은 각각 지패산 Ⅱ와 단자청피탕이 가장 높았으며, 총 페놀 함량은 다른 한약재에 비해 단자청피탕이 가장 높았다. 열수 추출물의 총 페놀 함량은 지패산 Ⅰ < 청간해울탕 < 지패산 Ⅱ < 십육미류기음 < 단자청피탕 순으로 높게 나타났다. 70% 에탄올 추출물의 총 페놀 함량은 지패산 Ⅱ < 지패산 Ⅰ < 청간해울탕 < 십육미류기음 < 단자청피탕 순으로 풍부하였다. 그 중 단자청피탕과 십육미류기음의 환원력과 DPPH radical과 ABTS radical 소거능이 가장 뛰어났으며, 이들 시료의 총 페놀 함량과 그들의 항산화능과의 밀접한 상관도가 관찰되었다. 또한 Staphylococcus aureus 및 Escherichia coli에 대한 항균활성은 열수보다 70% 에탄올 추출물에서 효과가 높게 관찰되었다. 결론적으로 유암의 치료에 사용되었던 조사 대상 한약재 처방전은 항산화와 항균 활성은 그들의 총 페놀 함유량과 연관성이 높았으며, 본 연구의 결과는 각 처방전에 대한 구체적인 생리활성 비교를 위한 자료로서 활용될 것이다. Sipyukmiryuki-eum (SYMYKE), Danjacheongpi-tang (DJCPT), Jipae-san Ⅰ (JPS Ⅰ), Jipae-san Ⅱ (JPS Ⅱ), and Chungganhaeul-tang (CGHUT) are representative herb-combined remedies used in traditional Korean medicine for treating breast cancer patients, as mentioned in "Dongeuibogam." In this study, we investigated the total phenolic contents (TPCs) and the anti-oxidant and anti-microbial activities of hot water and 70% ethanol extracts of these herbal prescriptions. Among the five herb-combined remedies, the extraction yields of the hot water extracts and 70% ethanol extracts were the highest in JPS Ⅱ (34.30%) and DJCPT (30.50%), respectively. The TPCs of the hot water extracts from the herb medicines were rich in the order of JPS Ⅰ < CGHUT < JPS Ⅱ < SYMYKE < DJCPT. In addition, the 70% ethanol extracts from the herb medicines were rich in the order of JPS Ⅱ < JPS Ⅰ < CGHUT < SYMYKE < DJCPT. Among them, DJCPT and SYMYKE displayed a strong anti-oxidant capability, which was determined using ferric-reducing anti-oxidant power and scavenging of 2,2"-diphenyl-1-picrylhydrazyl and 2,2"-azinobis(3-ethylbenzothiazoline-6-sulfonate) cationic radical activity assays. In addition, anti-microbial activities against Staphylococcus aureus and Escherichia coli were stronger in the 70% ethanol extracts than in the hot water extracts. Together, these findings reveal a positive relationship between TPCs and their anti-oxidant activities.

        네 가지 처방의 항산화 및 항암 효과 비교

        Objective : In this study, we compared the antioxidant and anticancer properties of four multi-herbal formulas which were recorded in 'Dongeuibogam': Gilgyung-tang (GGT), Mokdanpi-tang (MDPT), Samso-eum (SSE), Samchulbobi-tang (SCBBT). Methods : We checked antioxidant properties of four multi-herbal formula through total phenolic content, radical scavenging activities, protective effects on genomic DNA oxidation. To investigate anticancer effects, we conducted MTT assay and analyzed morphologic change in A549 non-small lung cancer cells. Results : Total phenolic contents of four multi-herbal formulas were in a rich order of MDPT > SSE > GGT > SCBBT. Especially, MDPT revealed the highest activity than others in all antioxidant experiments. Our results indicated that treatment of those multi-herbal formulas induced growth retardation in A549 cells and MDPT also showed the highest anticancer effect ($IC_{50}=1.374mg/ml$) among them. Conclusions : Our data suggested that MDPT would be a powerful ingredient for lung cancer treatment.

        C2C12 근아세포의 산화적 손상에 대한 고삼, 감초 및 백선피 복합 추출물의 보호효과

        Objectives : Increased oxidative stress by reactive oxygen species (ROS) has been suggested as a major cause of muscle fatigue. Although several studies have demonstrated the various biological properties of Sophora flavescens Aiton, Glycyrrhiza uralensis Fischer and Dictamnus dasycarpus Turcz, but the antioxidative potentials have not been clearly demonstrated. The present study was designed to investigate the protective effects of their water and ethanol extract mixtures (medicinal herbal mixtures, MHMIXs) on hydrogen peroxide ($H_2O_2$)-induced cell damage and apoptosis in C2C12 myoblasts. Methods : Cytotoxicity was assessed by an MTT assay. Quantitative evaluation of apoptosis induction and ROS production was evaluated by flow cytometry analysis. Expression levels of apoptosis regulatory and DNA-damage proteins were detected by Western blotting. Result : The inhibition of $H_2O_2$-induced cell proliferation was effectively blocked in extracts of 3: 1: 1 (EMHMIXs-1) or 2: 2: 1 (EMHMIXs-2) of S. flavescens, G. uralensis and D. dasycarpus Turcz, ethanol extracts from various complex extracts in C2C12 myoblasts. EMHMIXs-1 and EMHMIXs-2 also effectively attenuated $H_2O_2$-induced C2C12 cell apoptosis, which was associated with the restoration of the upregulation of Bad and death receptor 4, and downregulation of XIAP and cIAP-1 induced by $H_2O_2$. In addition, these herbal mixtures significantly blocked the $H_2O_2$-induced ROS generation and phosphorylation of $p-{\gamma}H2A.X$, which suggests that they can prevent $H_2O_2$-induced cellular DNA damage. Conclusions : The results suggest that EMHMIXs-1 and EMHMIXs-2 could block the DAN damage and apoptosis of C2C12 myoblasts by oxidative stress through blocking ROS generation.

        랫드에서의 Fe-NTA 유발 산화스트레스에 대한 차전초 에탄올 추출물의 전립선보호 효과

        Plantago asiatica L. (P. asiatica) has been used as one of the popular folk medicines in Asia for human health care practices. Various activities of P. asiatica have been reported, such as anti-oxidant, anti-glycation,anti-inflammatory and hepatoprotective activity. Therefore, the potential of P. asiatica to reduce oxidative stress has been studied in several ways for over 20 years, especially at liver and kidney. However no investigation has been reported revealing its protective effect on prostate. Method : Treatment of P. asiatica leaf ethanolic extract (PLE) (1 g/kg body weight (b.w.), 2 g/kg b.w., or 4 g/kg b.w.) were given separately to animals for pretreatment once per day for 7days, and on the seventh day ferric nitrilotriacetate (Fe-NTA; 0.24 mmol Fe/kg b.w.), which is known as an oxidative stress-inducer at prostate, was administrated by i.p to negative control group. At the end of the study period, dissection was carried out for detecting the prostate protective effect of PLE. Result : Fe-NTA-treated animals produced reactive oxygen species (ROS) resulting in depletion of antioxidant biomaker, such as glutathione (GSH), glutathione reductase (GR), and glutathione s-transferase (GST) and increase of lipid peroxidation in prostate. However, PLE pretreatment resulted in an increase in the GSH, GST and GR levels concentration dependent manner and in an significant decrease in the levels of lipid peroxidation. Conclusion : Our data suggest that PLE may be effective in protecting oxidative stress-induced damage of prostate, and PLE may be an chemopreventive agent against Fe-NTA-mediated prostate oxidative damage.
