        열대 적응형 조숙 다수성 단교잡종 옥수수 ‘케이엠2’

        ‘KM2’, a maize hybrid grain was developed for the Southeast Asian seed market by the maize breeding team of the Rural DevelopmentAdministration (RDA), South Korea 2018. ‘KM2’ was bred by crossing two inbred lines, ‘15VL065’ and ‘KS155’. After a advanced yieldtrial at Dong Trieu, Vietnam, for the first cropping cycle in 2016, country adaptability trials were performed twice in three different countries:Vietnam, Cambodia, and Indonesia, in 2016 and 2017. ‘KM2’, a yellow intermediate seed for food and feed, adapts well to the northern Vietnam,northwestern Cambodia, and Central Java in Indonesia. It is an early maturing high-grain yield F1 hybrid. This hybrid is resistant to downymildew, but not corn rust. The silking date of ‘KM2’ is 10 days earlier than that of ‘LVN10’, a leading cultivar in Vietnam. ‘KM2’ producedgrain yields of 811 kg/10a, 845 kg/10a, and 783 kg/10a in Vietnam, Cambodia, and Indonesia, respectively, showing even higher grain yieldthan the countries’ respective leading cultivar. ‘KM2’ produced 61%, 17%, and 14% higher grain yield in Vietnam, Cambodia, and Indonesia,respectively, than their respective control cultivars (‘LVN10’, ‘CP888’, ‘P21’) (Registration No. 8192).

        조생종 사료용 옥수수 품종의 2기작 재배 시 생산성

        This study evaluated the productivity of early maturity silage corns during continuous monocropping and the possibility of expanding forage production. Continuous monocropping of three silage corns, such as Kwangpyeongok (medium maturity), Sinhwangok (early maturity), and Sinhwangok2 (early maturity) was planted twice, in April and July from 2018 to 2019 at Suwon. The number of days from the sowing date to the silking date was 78 for the preceding crop and 52 for the succeeding crop. The number of days from the silking date to the harvesting date was 26 for the preceding crop and 46 for the succeeding crop. The sums of the temperature from the sowing date to the silking date were 1,512℃ for the preceding crop, 1,246℃ for the succeeding crop. The sums of the temperature from the sowing date to the harvesting date were 2,198℃ for the preceding crop and 1,951℃ for the succeeding crop. The dry matter yield of the preceding crop (1,637 kg/10a) was similar to that of the succeeding crop (1,565 kg/10a). The dry matter yields of Sinhwangok2 (1,673 kg/10a), Sinhwangok (1,660 kg/10a) and Kwangpyeongok (1,579 kg/10a) were similar to those of the preceding crop. The dry matter yields of Sinhwangok (1,669 kg/10a) and Kwangpyeongok (1,651 kg/10a) were similar to those of the succeeding crop and Sinhwangok2 (1,374 kg/10a) was the lowest among the three corn varieties. The total digestible nutrients (TDN) yield of the preceding crop (1,135 kg/10a) was similar to that of the succeeding crop (1,037 kg/10a). The TDN yields of Sinhwangok2 (1,183 kg/10a), Sinhwangok (1,158 kg/10a), and Kwangpyeongok (1,063 kg/10a) were similar to those of the preceding crop. The TDN yields of Sinhwangok (1,150 kg/10a) and Kwangpyeongok (1,100 kg/10a) were similar for the succeeding crop and Sinhwangok2 (970 kg/10a) was the lowest among the three corn varieties. The total dry matter yields of Sinhwangok (3,329 kg/10a) and Kwangpyeongok (3,230 kg/10a) were similar, but Sinhwangok2 (3,047 kg/10a) was the lowest among the three corn varieties. The total TDN yields of Sinhwangok (2,307 kg/10a), Kwangpyeongok (2,162 kg/10a), and Sinhwangok2 (2,152 kg/10a) were similar. It was concluded that Sinhwangok and Sinhwangok2 have high TDN yields as well as early maturity, and therefore are advantageous for direct continuous monocropping.

        종자친과 화분친을 달리한 옥수수의 품질 및 이화학 특성

        This study investigated on the chemical components, quality characteristics, antioxidant compounds, and activity of maize hybrids according to the cultivar, and breeding maize seeds crossed with seed and pollen parent. The moisture, crude fat, crude ash, crude protein, carbohydrate, and amylose contents of maize hybrids were significantly different among cultivars, and seeded and pollinated maize. The L-, a- and b-value of maize hybrids were 39.81~47.21, -0.01~0.55 and 5.85~18.47, respectively. Water binding capacity, water solubility index and swelling power were 123.29~153.32, 4.69~5.76 and 20.11~21.47%, respectively. The phenolic compounds and radical scavenging activity of maize hybrids were significantly different among cultivars, and seeded and pollinated maize. Total polyphenol and flavonoid contents of maize hybrids were 1,335.41~1,876.29 ㎍/g and 184.24~453.95 ㎍ CE/g, respectively. The DPPH and ABTS radical scavenging activities were 171.75~239.16 and 299.44~364.09 mg TE/100 g, respectively. As a result, it could be used as a basic data for cultivating phenol compounds and antioxidant activity in maize breeding.

        광평옥과 다청옥의 수이삭과 수염에서 안토시아닌 생합성 유전자 발현 분석

        본 실험은 광평옥과 다청옥 품종의 수이삭과 수염을 대상으로 안토시아닌 생합성 관련 분자생물학적 및 이화학적 특성을 검토하여 향후 기능성 옥수수 개발에 필요한 기초자료로 활용하고자 수행되었다. 1. 광평옥에서는 수이삭과 수염에서 모두 안토시아닌이 형성되지 않은 반면, 다청옥에서는 모두 형성되었다. 전체 안토시아닌 함량은 수이삭과 수염에서 모두 다청옥이 광평옥에 비해 30배 정도 높았다. 또한, 안토시아닌 성분은 다청옥의 수이삭에서 C-3-G만 측정되었고, 수염에서는 각각 C-3-G는 45.2배, Pg-3-G는 27.3배, M-3-G는 37.6배 광평옥에 비해 더 검출되었다. 2. 광평옥의 수이삭에서 F3'H, DFR, GST 유전자가 발현이 감소하였고, 수염에서는 F3'H와 DFR 유전자가 발현이 감소하였다. 반대로 다청옥에서는 수염에서 DFR 유전자만 발현이 감소되었고, 나머지 유전자들은 정상적으로 발현되었다. 또한 안토시아닌 생합성 유전자들의 발현을 조절하는 전사조절인자는 광평옥 수이삭에서는 P1이 수염에서는 R1이 관여한다는 사실을 확인하였다. 3. 광평옥의 수이삭과 수염에서 다청옥에 비해 각각 linoleic acid (C18:2)가 6.6%, 10.9% 감소하고, linolenic acid (18:3)는 8.5%, 8.5% 증가하였다. 또한, 광평옥의 수염에서는 palmitic acid (C16:0)가 4.1% 증가하고, oleic acid (C18:1)는 다청옥에 비해 2.1% 감소하였다. 그러나 stearic acid (18:0)는 수이삭과 수염에서 성분 변화가 전혀 없었다. 그리고 광평옥의 수이삭과 수염의 전체 지방산 함량은 광평옥에 비해 각각 10.3%, 30.4% 증가하였고, 각 지방산 성분과 함량 분석은 유사한 결과를 보여주었다. 그러나 phytosterol 성분 분석에서는 유의미한 결과를 확인할 수가 없었다. Anthocyanins are known to be involved in various functions such as antioxidant and antibacterial activities in plants. Although studies on anthocyanins in corn have been conducted recently, basic research related to anthocyanin biosynthesis is insufficient. In this study, we examined the molecular biological and physicochemical properties related to anthocyanin biosynthesis in the tassel and silks of Gwangpyeongok and Dacheongok cultivars. Anthocyanins were not synthesized in either the tassel or silks in Gwangpyeongok, whereas were synthesized in both in Dacheongok. The total anthocyanin content was approximately 30 times higher in the tassel and silks of Dacheongok than in those of Gwangpyeongok. In addition, C-3-G was measured only in the tassel of Dacheongok, and C-3-G, Pg-3-G, and M-3-G were 45.2 times, 27.3 times, and 37.6 times higher, respectively, in the silks of Dacheongok than of Gwangpyeongok. Expression of F3'H, DFR, and GST genes decreased in the tassel, and that of F3'H and DFR genes decreased in the silks of Gwangpyeongok. It was further confirmed that transcription factor P1 and R1 regulate the expression of anthocyanin biosynthetic genes in the tassel and silks, respectively, in Gwangpyeongok. Linoleic acid (C18:2) decreased by 6.6% and 10.9%, and linolenic acid (C18:3) increased by 8.5% and 8.5%, in the tassel and silks, respectively, of Gwangpyeongok compared to those of Dacheongok. Palmitic acid (C16:0) increased by 4.1% and oleic acid (C18:1) decreased by 2.1% in the silks of Gwangpyeongok compared to that in Dacheongok. In addition, the total fatty acid content in the tassel and silks increased by 10.3% and 30.4%, respectively, in Gwangpyeongok compared to that in Dacheongok. However, no significant results were observed in the analysis of phytosterol components. These results may be utilized as useful resources for the development of functional corn containing a large amount of anthocyanins.

        국산 옥수수 배유특성에 따른 일반성분, 색도 및 경도 비교

        This study was conducted to secure basic information for corn processing by comparing the quality characteristics according to maize cultivars and kernel types (dent, intermediate, flint-like). As a result of analyzing 15 cultivars, a range of measurements were observed: 100-kernel weight, 22.89~35.63 g; moisture, 7.57~8.42%; crude protein, 8.46~11.45%; crude lipids, 3.26~4.83%; Hunter's L-value, 83.70~86.79; a-value, 2.61~5.49; b-value, 22.01~28.15; and total carotenoids, 6.74~17.07 ㎍/g. Significance among the cultivars was shown in all quality characteristics (p<0.001), but the significance among the kernel types was found only in crude protein (p<0.005), crude fat (p<0.001), and Hunter's L-value (p<0.05). The hardness of maize was decreased proportionally to the soaking time for all maize cultivars (p<0.001). In particular, with the same soaking time for different kernel types, the hardness difference was shown in the order of flint-like > dent ≒ intermediate. It was confirmed that the decrease in the hardness of flint-like kernel of close to that of hard-type starch was slowed compare dent and intermediate kernels. So it is expected the some characteristic of kernel types will contribute to the appropriate customized use of the developed cultivars.

        SNP마커 개발을 통한 사료용 옥수수 품종판별

        옥수수 품종판별 마커 개발을 위하여 SNAP 방법을 변형하여 2bp 불일치 SNP PCR 방법을 옥수수 품종판별에 적용하였다. SNP 마커개발을 위하여 MaizeGDB 웹사이트(www.maizegdb.org)를 통해서 200 SNP 위치를 확인하였으며, 표준맵으로 알려진 B73 옥수수 게놈서열을 바탕으로 2bp 불일치 Primer을 디자인하였다. PCR 생성물은 200-500bp 사이에서 결정되었으며 SNP site가 있을시 PCR 생성물이 생성되지 않게 디자인 되었다. 선행연구에서 선발된 16개의 Primer조합을 이용해서 농촌진흥청에서 개발된 사료용 옥수수 10품종(강다옥, 광평옥, 다평옥, 안다옥, 양안옥, 신광옥, 장다옥, 청다옥, 평광옥, 평안옥)과 수입 사료용 옥수수 40품종과의 판별 가능성을 검정하였다. SNP PCR 결과를 바탕으로 한 Cluster분석에서 신광옥과 PI1395 그리고 몇몇 수입 사료용 옥수수를 제외하고는 모두 판별 가능한 것으로 검정되었다. SNP 통한 품종판별 최소조합수를 선발한 결과 강다옥은 IBM911과 IBM1798, 장다옥은 IBM440과 IBM549, 평강옥은 IBM440과 IBM1269, 평안옥은 IBM795와 IBM1601였다. 이는 SNP 마커 개발을 통해 빠르고, 손쉽게 품종판별이 가능한 마커로 활용가능하다는 것을 보여준다. Single nucleotide polymorphisms (SNP) markers allow rapid screening of crop varieties in early growth stages. We developed a modified SNP PCR procedure for assaying SNPs in maize. For SNP marker development, we chosen 200 SNP sites from MaizeGDB database, and designed two base pair mismatch primers based on putative SNP site of B73 genome sequence. PCR products size was from 200 to 500 bp or was not shown in the case of SNP site existing in Korean silage corns. Using previously discovered 16 primer sets, we investigated distinctness of 50 silage F1 hybrid corns including 10 Korean silage corns developed by RDA such as Gangdaok, Kwangpyeongok, Dapyeongok, Andaok, Yanganok, Singwangok, Jangdaok, Cheongdaok, Pyeonggangok, and Pyeonganok as well as 40 foreign commercial silage corns. From cluster analysis, we confirmed that 10 Korean silage F1 hybrid corns were clearly distinguished except for Singwangok, P1395, and several foreign commercial corns, and selected minimum SNP primer combination for Gangdaok, Jangdaok, Pyeonggangok, and Pyeonganok. Therefore, development of SNP marker sets might be faster, cheaper, and feasible breed discrimination method through simple PCR and agarose gel electrophoresis.

        The instrument-centering ability of four Nickel-Titanium instruments in simulated curved root canals

        본 연구의 목적은 새로 시판된 NRT 기구의 근관 형태 유지 능력을 기존의 3종의 기구와 비교하는 것이다. 만곡된 레진 모형 근관을 ProFile, K3, ProTaper, NRT 기구를 이용하여 형성하였고 형성 전, 후의 상을 스캔한 후 중첩시켜 이미지 분석 프로그램을 이용하여 근관 만곡도의 변화와 근관 중심축에 대한 근관의 변이 정도를 근관 중심율의 변화로 계산하여 분석하였다. 근관 변이의 유의성 검정을 위해 one-way ANOVA분석을 시행하였으며 각 기구간의 유의성 검정은 Duncan's test로 사후 분석하였다. 근첨에서 1, 2 mm 상방 위치에서의 근관 중심율은 ProFile 군이 다른 군에 비해 우수한 것으로 나타났다. (p < 0.05). NRT기구의 근관 중심율 변화를 측정한 결과 근첨으로부터 5 mm 상방에서는 Profile에 비해서만 큰 것으로 나타났고 6, 7 mm 상방에서는 다른 모든 군들에 비해 큰 것으로 나타났다 (p < 0.05). 비록 NRT기구가 근첨 부위에서는 ProFile을 제외한 다른 기구에 비해 유의차 없는 중심률 유지 능력을 보였으나 근관 입구 및 중간부위의 형성에 사용되는 스테인레스-스틸 파일로 인해 coronal flaring시 다른 기구들에 비해 더 많은 중심률의 변화를 보이는 것으로 사료된다. The aim of this study was to evaluate the ability of newly marketed NRT instruments to maintain the original root canal configuration and curvature during preparation in comparison with the three existing instruments in simulated root canals. Simulated canals in resin blocks were prepared with ProFile. K3, ProTaper and NRT instrument (n = 10 canals in each case). Pre- and post-operative images were recorded, and assessment of canal shape was completed with a computer image analysis program. The data were analyzed statistically using the One-way ANOVA followed by Duncan s test. The ability or instruments to remain centered in prepared canals at 1-, 2-mm levels was significantly better in ProFile groups than in other groups (p < 0.05). The change of centering ratio in NRT groups at 5-mm level was significantly greater than ProFile group and at 6- and 7-mm level than all other groups (p < 0.05). Although the NRT system was comparable to other systems in regards to its ability to maintain the canal configuration of apical portion, this system was more influenced by the mid-root curvature due to its stainless-steel files for coronal preflaring.

        옥수수 품종별 볶음 시간에 따른 이화학적 특성 변화

        The purpose of this study was to investigate the Maillard reaction-related physicochemical properties of three maize varieties (Kwangpyeongok, Sinhwangok2ho and Gangdaok) after roasting them for different times (0, 15, 25, 40, and 55 minutes). The Maillard reaction is a non-enzymatic browning reaction involving reducing sugars and amino compounds. The content of reducing sugar, the causative agent of the Maillard reaction, decreased as roasting time increased. Gangdaok showed the lowest reducing sugar content of 1.04 mg/g after 55 minutes of roasting. In the elapsed roasting time, chromaticity 'L' and 'b' values decreased. At 55 minutes of roasting, wherein the Maillard reaction occurred most actively, Gangdaok showed the lowest 'L' value of 56.37 and the highest 'a' value of 7.60. Gangdaok had superior conditions for inducing the Maillard reaction compared to other varieties, and it is consider that 'flint-type', an endosperm characteristic, may have been the influencing agent. This study detected a total of 52 types of volatile aroma compounds (VACs), of which 28 were produced after roasting. Of the total VACs detected, 2-Formyl-5-methylfuran and 2-Furancarboxaldehyde accounted for 43.8~45.5% and have been confirmed to be the major VACs present in roasted maize. Most of the correlations between the Maillard reaction-related characteristics showed high correlation coefficients.

        옥수수 종실 및 속대의 Phytosterol 동정과 함량 변이

        옥수수 종실(kernel)과 속대(cob)의 불검화물(ZML) 중 phytosterol의 조성과 함량의 변이를 구명하여 고품질 옥수수 신품종 육성 및 생리활성물질의 유용 소재화를 위한 기초자료로 활용하고자 본 연구를 실시하여 얻어진결과를 요약하면 다음과 같다. 1. 옥수수 종실과 속대의 포화지방산은 phytosterol 함량과 부의 상관관계의 경향이었고, 불포화지방산 중 stearic 및 linoleic acid는 정상관 경향을 보였으나, linolenic acid는 종실에서 정상관($r=0.652^*$), 속대는 부상관($r=-0.505^*$) 관계를 보였다. 2. 옥수수 종실의 불검화물을 TLC로 분리한 결과 band I (campesterol, stigmasterol, ${\beta}$-sitosterol), band II (${\Delta}^5$-avenasterol), band III (${\Delta}^7$-stigmastenol), 및 band IV (${\Delta}^7$-avenasterol)로 뚜렷하게 분리되었고, 속대는 band I~IV 이외에도 3종 이상의 band가 추가적으로 분리되었다. 3. 옥수수 종실과 속대에 함유된 phytosterol의 GC 분리 패턴을 확인한 결과 campesterol, stigmasterol 및 ${\beta}$-sitosterol의 분리능이 좋았으나, ${\Delta}^7$-avenasterol (RT 22.846), ${\Delta}^7$-stigmastenol (RT 22.852) 및 ${\Delta}^5$-avenasterol (RT 22.862)은 혼합물질 상태로는 분리가 되질 않았다. 4. 옥수수 종실의 평균 phytosterol 함량은 635.9 mg/100g, 속대는 273.0 mg/100 g으로 종실이 속대에 비해 약 2.4배 정도 phytosterol 함량이 높았다. 옥수수 종실의 phytosterol 조성은 ${\beta}$-sitosterol 80.05% > campesterol 10.5% > stigmasterol 9.46% 순이었으나, 속대는 ${\beta}$-sitosterol 59.43% > stigmasterol 31.72% > campesterol 10.98%으로 종실과 속대의 phytosterol 조성비는 다소 상이하였다. 5. 본 연구 결과를 토대로 판단할 때 옥수수 종실에 함유된 ${\beta}$-sitosterol, campesterol 및 stigmasterol의 생합성 경로에서 전구물질이 되는 ${\Delta}^7$-avenasterol, ${\Delta}^7$-stigmastenol 및 ${\Delta}^5$-avenasterol이 옥수수 속대에서 검출되는 것으로 보아 옥수수 종실의 phytosterol은 속대에서 합성되어 종실로 전이되는 물질인 것으로 추정되었다.

        본 연구는 사료용 옥수수의 내습성 검정을 위한 기초자료를 얻고자 자식 6계통을 3엽기에 30일간 습해 처리 후 옥수수의 생육 특성 및 수량 변화를 구명하고자 농촌진흥청 국립식량과학원 중부작물부 시험포장(온실)에서 수행한 결과를 요약하면 다음과 같다. 1. 습해 처리 시 모든 자식 계통이 무처리에 비하여 초장, 완전전개엽수 및 엽색도(SPAD)는 모든 계통에서 유의적으로 감소하였으며 엽색도는 KS140 계통이 높았고 KS141 계통이 낮았다. 2. 지상부와 뿌리의 건물중은 습해처리 후 모든 계통이 유의적으로 감소하였으며 자식 계통들 간에는 KS140이 가장 많았고 KS163이 가장 적었다. 부정근의 건물중은 무처리와 유의적 차이는 없었으나 KS140 계통에서 높은 경향을 보였다. 3. 수확기 옥수수의 이삭길이와 두께 및 착립이삭길이, 개체당 종실수 모두 무처리에 비하여 유의적으로 감소하였다. 무처리에 비하여 이삭 길이와 두께는 KS141 계통이 착립 이삭길와 개체당 종실수에서는 KS164 계통이 가장 적게 감소하였다. 4. 옥수수 수량은 무처리에 비하여 습해 처리한 모든 계통에서 유의적으로 감소하였으나 습해 처리 계통들 간에는 KS164 계통만 유의적으로 수량이 높아 내습성인 것으로 판단하였다. 5. 옥수수 유묘기 30일간 습해처리시 초장, 엽색도(SPAD값), 완전전개엽수가 옥수수 수량과 상관이 높았다. Waterlogging strongly affects maize (Zea mays L.) growth. It is necessary to find the screening method of waterlogging tolerant maize lines. This study was to investigate the growth characters at V3 stage of maize, when is very sensitive to waterlogging. Six Korean maize inbred lines were subjected to waterlogging at V3 stage for 30 days. The 30 days waterlogging treatment significantly reduced plant height, number of expanded leaves, and SPAD value, compared with the control plants. SPAD values were significantly different among the six inbred lines, KS140 was the highest. The dry matter accumulation of aerial and root part were significantly decreased by 30 days waterlogging. KS140 was the weightiest among inbred lines. The dry matter of adventitious root showed same trend. Waterlogging treatment significantly reduced to ear length and thickness, grains filling length, grain number per ear, and maize grain. Plant height, SPAD value, and number of fully-expanded leave showed high correlation with maize grain yield, but number of senescent leaves, dry matter of adventitious root and TR ratio did not, suggesting that the former three traits may be good indicator for evaluating 30-day waterlogging tolerance of maize inbred lines. KS164 was the highest yield by increasing of grains filling length and grain number per ear of among waterlogging inbred lines. According to the results, evaluation of maize waterlogging should be consider both early growth characteristics and resilience in the later growth stages.
