The reproducibility as the standard deviation or standard error of analytical data is used to estimate the performance ability of the isotope ratio mass spectrometer (IRMS). The standard deviation of □^13C value of a sample is not supposed to be the same on a wide range of sample weights. In order to determine the suitable sample amounts required for high precision and accuracy and the factors required for optimal equipment conditions, certified reference materials were repeatedly measured with various weight ranges using an elemental analyzerisotope ratio mass spectrometer (EA-IRMS). The measured □^13C values of the standards EMA-P1 and EMAP2 fell within the certified value ±2σ with the minimum carbon contents of 102.107 μg and 116.879 μg, respectively. In addition, for the two standards, the peak height ratios calculated by the CO_2(^12C^16O^16O) peak height of the sample to the working standard were 62.8% and 66.8%, respectively. The isotopic composition change was also investigated by the fractionation factor □applied and did not show a large difference in a wide range of sample weight for the two standards. We suggest that carbon isotope composition needs to be determined by considering several factors such as element contents, standard deviation of □^13C value, peak height ratio(or peak height) and fractionation factor □applied.
대장(大腸)과 관련(關聯)된 경혈(經穴)들의 신경해부학적(神經解剖學的) 연구(硏究)

The purpose of this morphological studies was to investigate the relation between the meridian, acupoints and viscera using neuroanatomical tracers. The common locations of the spinal ganglia, sympathetic chain ganglia, spinal cord and brain projecting to the large intestine meridian were observed following injection of transganglionic tracer, WGA-HRP and transsynaptic neurotropic virus, pseudorabies virus(PRV), Bartha strain(Ba) and PRV-Ba-Gal (Galactosidase)) into the the large intestine(cecum, colon and rectum), ST37 and LI4. After survival times of 96 hours following injection into the thirty rats with WGA-HRP, PRV-Ba and PRV-Ba-Gal. They were perfused, and their spinal ganglia, sympathetic chain ganglia, spinal cord and brain were frozen sectioned($30{\mu}m$). These sections were stained by HRP and X-gal histochemical and PRV immunohistochemical staining method, and observed with a light microscope. The results were as follows : 1. WGA-HRP labeled neurons innervating the large intestine were observed bilaterally within the T13-L4 sympathetic chain ganglia, and T9-11 spinal ganglia. WGA-HRP labeled neurons innervating ST37 were observed within the L3-5 sympathetic chain ganglia, and L2-4 spinal ganglia. WGA-HRP labeled neurons innervating LI4 were observed in the middle cervical ganglion and stellate ganglion, and C5-8 spinal ganglia. 2. In spinal cord, PRV-Ba labeled neurons projecting to the large intestine, ST37 and LI4 were found in thoracic, lumbar and sacral spinal segments. Densely labeled areas of each spinal cord segment were founded in lamina N, V, VII(intermediolateral nucleus), Ⅸ, X and dorsal nucleus. 3. In medulla oblongata, PRV-Ba and PRV-Ba-Gal labeled neurons projecting to the large intestine, ST37 and LI4 were commonly found in the A1 noradrenalin cells/C1 adrenalin cells/caudoventrolateral reticular nucleus, dorsal motor nucleus of vagus nerve, nucleus tractus solitarius, raphe obscurus nucleus, raphe pallidus nucleus, raphe magnus nucleus and gigantocellular nucleus. 4. In pons, PRV-Ba and PRV-Ba-Gal labeled neurons were commonly found in locus coeruleus, Kolliker-Fuse nucieus and A5 cell group. 5. In midbrain, PRV-Ba and PRV-Ba-Gal labeled neurons were commonly found in central gray matter. 6. In diencephalon, PRV-Ba and PRV-Ba-Gal labeled neurons were commonly found in paraventricular hypothalamic nucleus. These results suggest that PRV-Ba and PRV-Ba-Gal labeled common areas projecting to the large intestine may be correlated to that of the large intestine meridian, ST37 and LI4. Especially, These morphological results provide that interrelationship of meridian-acupoints -viscera may be related to the central autonomic pathways.

Analytical conditions of amino acids in ground-water by using UFLC-MS/MS(ESI) were optimized for prompt and accurate analysis. Under the Edmen`s method, derivatization of amino acids with PITC (phenylisothiocyanate) was performed and chromatographic separation was achieved on ODS (octadodesylsulfonate)- column with 3,400 psi. Within 10 minutes (6 min for MS collection), 21 types of amino acids were simultaneously analyzed by UFLC-MS/MS (ESI) after the selection of qualitative and quantitative ions. This method was precise with percent relative standard deviation of 2.6~13.8 % on 7 replicate analysis and accurate with percent recovery of 81.6~115.3 %. Confidence interval (CI, 3.53~4.25 mg/L). coefficient of determination (r2, 0.970562 (TRP)~0.999329 (VAL)), limits of detection (0.29(PRO)~1.88(ASN) mg/L (mean. 0.91 mg/L)), and limits of quantification (0.99(PRO)~6.28(ASN) mg/L (mean, 3.03 mg/L)) were obtained and proven to be a reliable method. Derivatives was relatively stable when it occurred for more than three hours under 45oC based on optimum tests. Also, it was notable that m/z 164.15 ion, reagent of derivatization under MS/MS, was identified with phenylalanine in the same time frame, indicating excessive quantity. However, when the quantitative ion was changed from m/z 164.15 to m/z 221.10, relevant result was achieved.

This study was conducted to analyze the composition of amino acids to develop of a method for tracing contamination sources in groundwater which is polluted by livestock wastewater or leachate from carcass burial sites. In this study, the FALCON (Fingerprinting Analysis of Leachate Contaminants) procedure was used to find unique characteristics in the samples of livestock wastewater and carcass leachate. For the livestock wastewater samples, Lysine accounted for the largest portion (23.39%), followed by alanine (21.93%) and glycine (6.94%). As for the composition of amino acids in carcass leachate, on the other hand, valine made up the highest percentage (28.72%), followed by leucine (19.25%) and isoleucine (15.40%), To identify distinct characteristics between livestock wastewater and carcass leachate in terms of their amino acid composition, individual data sets were estimated for the statistical reproducibility/comparability of fingerprint patterns, and consequently six amino acids such as valine, isoleucine, leucine, lysine, methionine and tryptophan were selected to develop the indicators. To statistically estimate the suitability of these six amino acids, the hierarchical clustering method was used. As a result, all samples except one livestock wastewater sample were clearly divided into two groups: livestock wastewater and carcass leachate. The similarity was 0.665 among the seven samples of livestock wastewater and 0.500 among the seven samples of carcass leachate, which shows that there are similar distribution patterns of amino acids among the samples of each group. On the other hand, the similarity between the two groups turned out to be 0.121, which indicates that livestock wastewater and carcass leachate have significantly different characteristics from each other in terms of amino acid concentration. Therefore, this method of analyzing the composition of amino acids may be useful to trace contamination sources at monitoring locations with pollution fingerprints.

The Caesalpinia sappan is widely used in the traditional oriental herbal medicine for anti-inflammatory, antioxidant effects. The effects of water extract of C. sappan on the cell viability and induction of apoptosis were investigated in human lung cancer cell line A549. The water extract of C. sappan produced apoptotic cell death and DNA fragmentation and nucleus chromatin condensation in A549 cells. The enzyme activity of caspase-3 and protein level of actived caspase-3 were markedly increased in A549 cells treated with the water extract of C. sappan. In addition, the extract of C. sappan induced cleavage of Poly (ADP-ribose) polymerase (PARP), a known substrate for caspase-3, and dropped in cellular ATP levels. These results suggest that the extract of C. sappan exerts anticancer activity by induction of apoptosis via activation of caspase-3, cleavage of PARP protein, and depletion of cellular ATP levels in A549 cells.
