박테리오 파아지 T7 RNA 중합효소는 어떤 전사인자도 관여하지 않는 간단한 시스템으로 파아지 RNA 중합효소와 전사촉진제만의 단백질-DNA 상호작용에 의해 전사가 진행된다. T7 파아지의 숙주 세포의 파괴에 관여하는 T7 파아지 lysozyme은 전사를 억제하고, 전사종결에 영향을 미친다. 따라서 T7 파아지 lysozyme 유전자를 대장균 발현 벡터에 삽입하여 pT7lys를 얻었고, 발현시켜 Ni-NTA column chromatography로 순수 분리하였다. T7 파아지 lysozyme은 SDS-gel에서 단일 밴드로 확인하였으며, amidase 활성 역시 확인하였다. 염기서열 특이적 전사 종결에 미치는 T7 파아지 lysozyme의 역할을 알아보기 위하여, rrnB T1 전사종결 인자 부근에서의 전사연장 복합체 제조에 미치는 T7 파아지 lysozyme의 영향력을 조사하였다. 이 결과 T7 파아지 lysozyme 존재 하에 형성되는 전사연장 복합체는 불안정함을 알 수 있었다. T7 RNA polymerase is a single subunit RNA polymerase able to accomplish whole transcription process without auxiliary factors. T7 phage lysozyme involcing in destruction of host cell wall repress T7 transcription and affects transcription termination process. Therefore expression vector pT7lys containing T7 phage lysozyme gene was constructed and expressed. T7 phage lysozyme protein was purified to homogeneity by Ni-NTA column chromatography. Also amidase activity of the purified lysozyme was identified. In order to understand the effect of the lysozyme on the sequence specific transcription termination. T7 transcription elongation complexes at the site rrnB T1 transcription termination signal were made in the presence the lysozyme. The results shows that the transcription elongation complexes are unstable in the presence of T7 phage lysozyme.

Ordered nanostructure materials have received attention due to their unique physical properties and potential applications in electronics, mechanics and optical devices. To actualize most of the proposed applications, it is quite important to obtain highly ordered nanostructure arrays. The well-aligned nanostructure can be achieved by synthesizing nanostructure material in the highly ordered template. To get well-aligned pore array and reduce process time, rapid thermal anneal by an IR lamp was employed in vacuum state at $500^{\circ}C$ for 2 hour. The pore array is comparable to a template annealed in vacuum furnace at $500^{\circ}C$ for 30 hours. The well-fabricated AAO template has the mean pore diameter of 70 nm, the barrier layer thickness of 25 nm, the pore depth of $9{\mu}m$, and the pore density of higher than $1.2{\times}10^{10}cm^{-2}$.

The purpose of this study is to develop a port simulation program for Inchon Port. The arrival and departure data from Inchon Port is analyzed and the berthing rule based on formal materials and experiences is constructed. A Port-Simulation program to analyze the effects of the changes in ship arrivals, service capacities upon the demurrage is constructed and several scenarios are analyzed. It is expected that we can use the simulation results in order to prepare proper service levels and plan appropriate investment strategies.

In these days many people have studied on the berthing problem. The operation rules differ from port so port and the problem is highly dependent on the environment of port. The purpose of this study is to develop a decision support system decision makers of the berthing problem for Inchon Port. The system is developed with graphic user interface(GUI) using user-interactive approach and some general and specific rules for Inchon Prot are considered. The system is composed of the following four parts ; the input/output part, the automatic berthing part by the system using rules, the manual berthing part by user, and the part for modifying results or handling exceptional events. The system is designed to assign ship to berths by matching the characteristics with environmental and operational constraints of Inchon Port. We expect that this system can provide decision makers with an efficient and fast way to berthing and can reduce wastes of time, space, and manpower in port operations.

19 years old girl presented with bilateral 1, 4th brachymetatarsia. We have used an autograft interposition technique to lengthen the brachymetatarsia involving first and fourth metatarsal. The technique was to graft the bone fragment from the relatively long second and third metatarsal bone to be used as an autograft to the short first and fourth metatarsal bone. The method is superior in not having the necessity of a long term external fixator which is needed using callotasis method and in also avoiding the inconvenience of performing an allograft. Absence of donor site complication during autograft from iliac bone is also an advantage to be mentioned. Our technique can therefore be ascertained as a successful method in both cosmetic results and improvement of symptoms including reduction in length of recovery.

Idiopathic hypertrophic cranial pachymeningitis (ICHP) is diffuse inflammatory process of the dura mater. ICHP can produce similar presentation with Tolosa-Hunt syndrome (THS) if it involves cavernous sinus. A-29-year old male with persistent headache and no definite neurologic dysfunction was noted. Two weeks later, he complained of ophthalmoplegia, and his symptoms were thought to be manifestations of THS. Brain magnetic resonance images revealed diffuse thickened, enhanced pachymeninges in left tentorium. The patient was diagnosed with IHCP. We report a IHCP patient who showed very similar presentation as THS.

Objectives: The aim of this study is to investigate cell viability, anti-inflammatory, antioxidant, immunoenhancing activity using various extracts of Yeonsan Ogye. Methods: In order to evaluate cytotoxicity, MTT assay was performed. We investigated production levels of pro-inflammatory cytokines such as tumor necrosis factor (TNF)-α and interleukin (IL)-6, and nitric oxide(NO) in LPS-induced RAW 264.7 cells. NO production in RAW 264.7 cells was measured by using Griess reagent. Cytokines including IL-6 and TNF-α were measured by Luminex and ROS was measured by Flow cytometry. Results: No cytotoxicity of various extracts of Yeonsan Ogye was observed in RAW 264.7 cells. Productions of ROS in RAW 264.7 cells were increased from extraction of bones and decreased from extraction of skin. Also, productions of NO in RAW 264.7 cells were increased to bone extract and decreased at skin extract. In addition, productions of pro-inflammatory cytokines (IL-6 and TNF-α) in LPS-induced RAW 264.7 cells were decreased at skin, meat extracts, respectively. Finally, the levels of immune-related cytokines (IL-6 and TNF-α) were increased compared to those of the normal group. Conclusions: It is concluded that Yeonsan Ogye extracts seem to have significant biological activities likes anti-inflammatory, antioxidant, immuno-enhancing etc. These results may be developed as a raw material for new health food and new therapeutics to ease the symptoms related with inflammatory and oxidative stress. In terms of oriental traditional medicine, we expect that it contribute to building of EBM (Evidence-Based Medicine) from the this result.
해빈류 벡터 장미도를 통한 해운대 해수욕장의 이안류 민감도 분석

Rip current forecasts, based on intensity, are marked in four levels-notice, watch, warning, and danger. However, numerical results are represented by current vectors, whose magnitudes are then converted into predictive levels. In the present study, the rose diagram is adapted as a determinative forecasting index and examined for the case of an ideal rip channel consisting of surface, bottom, and averaged currents. Further, it is employed in the sensitivity analysis of wave-induced currents generated by wave conditions at the Haeundae Beach. The simulation of surface onshore and bottom undertow currents is accomplished by including a mass flux term in the wave-averaged continuity equation.
장기간 운동이 비만 여중생의 신체구성 및 혈중 호르몬에 미치는 영향

The purpose of this study was to evaluate the effect of aerobic exercise and resistance exercise on the body composition and blood hormone. The subjects for this study were thirty obesity girl students and divided into 3 groups : Control group(n =7), aerobic exercise(running) group(n=7) and resistance exercise(dumbbell) group(n=7) for 10 weeks. The aerobic exercise group performed running 30-40 min in a bout, four times a week at the intensity of 50-70% HRmax(1~5 week ; 50~60% HRmax, 6~10 week ; 61~70% HRmax). The resistance exercise group performed dumbbell exercise consisting 12 factor using 1-2kg weight(1~5 weeks ; 1kg, 6~10 weeks ; 2kg). Body composition(Body weight, %Fat, fat mass, WHR) and the change of Thyroxine(TSH), Testosterone(T), Cortisol(COR), and Insulin have been measured before, by post exercise 5 weeks and 10 weeks respectively. Statistical techniques for data analysis were One-way ANOVA exercise duration(before, 5 weeks, 10 weeks) and repeated measure ANOVA to determine the difference between the aerobic exercise group and the resistance exercise group. The 5% level of significance was utilized as the critical level for acceptance of hypotheses for the study. The following results were obtained from this study. Body composition- Body weight, %Fat, fat mass, WHR- were decreased significantly after long-term exercise in aerobic and resistance exercise group except control group. The change of TSH in the aerobic exercise group was decreased after 10 weeks of exercise. T and Insulin were decreased significantly after 10 weeks but COR was increased significantly after 10 weeks. The change of TSH and Insulin in the resistance exercise group were decreased significantly after 10 weeks of exercise. T was decreased after 10 weeks but COR was increased significantly after 10 weeks.
