        온도 및 염분 등의 환경요인이 참돔(Pagrus major)의 Interleukin-1 Receptor Accessory Protein 발현에 미치는 영향

        Interleukin-1 (IL-1) is one of the proinflammatory cytokines, after IL-1 binds to IL-1RI, IL-1RacP (interleukin-1 receptor accessory protein) joins with IL-1/IL-1RI to form a complex, and leading to cell activation. IL-1RAcP is involved in immune response, stress and apoptosis. The purpose of this study was to investigate the gene expression of IL-1RAcP in red seabream (Pagrus major) exposure to low water temperature (8℃, 33 psu) and low salinity (20℃, 10 psu). Results showed that, the expression of IL-1RAcP was significantly increased in the experiment groups, such as low water temperature (8℃, 33 psu), and low salinity (20℃, 10 psu). These results suggest that IL-1RAcP was played roles in biomarker gene on the environmental stress such as low water temperature and low salinity.

        대서양 연어(Salmo salar)의 수온 스트레스에 의한 Hsp90 및 CYP1A 발현 양상 비교

        수온의 변화는 어류의 거의 모든 생리학적 부분에 영향을 미친다. 기후 변화로 인한 수온의 상승은 어류에게 물리적 피해를 줄 수 있다. 이 연구는 최적의 수온(15℃)보다 높은 수온(20℃)에서의 대서양 연어의 건강상태를 평가하기 위해 수행하였다. 간 조직은 열 적응에 중요한 대사기능을 발휘하기에 본 연구에 간 조직을 사용하였다. 생체지표유전자의 개발을 위한 분석 방법으로는 NGS RNAseq 방법을 사용하였고, 생체지표유전자의 발현 양상을 관찰하기 위한 분석 방법으로는 RT-qPCR을 사용하였다. NGS RNAseq 분석을 통해 1,366개의 차별적 발현 유전자를 확인하였으며, 그 중에서 880개의 증가하는 유전자와 486개의 감소하는 유전자를 확인하였다. 생체지표유전자로는 heat shock protein 90 alpha (Hsp90α), heat shock protein 90 beta (Hsp90β) 및 cytochrome P450 1A (CYP1A)을 선정하였는데 이들 유전자는 NGS RNAseq 분석에서 수온의 변화에 민감하게 반응하는 유전자들이었다. 이들 유전자의 RT-qPCR을 통한 발현 양상은 NGS RNAseq 분석과 유사하게 나타났다. 이 연구의 결과는 다른 어종에도 적용할 수 있으며, 산업적으로도 유용하다고 생각된다.

      • 생식소 자극 호르몬과 스테로이드 호르몬에 의한 생쥐 여포의 bad와 bax 유전자 발현

        Abstract The purpose of this study was to investigate the effects of gonadotropin and steroid hormone on the expression of mouse follicular bad and box genes that are known to induce apoptosis. Ovary of immature mice were obtained at 0, 24, 48 and 72 hours time intervals after Pregnant Mare's Serum Gonadotropins (PMSG, 5 I.U.) injection. Large and preovulatory follicles collected at 24hrs after PMSG injection were cultured with or without various chemicals such as gonadotropin, progesterone, estrogen, testosterone for 24hrs at 37℃. After 24hrs culture, the follicles was used for total RNA extracted, subjected to RT-PCR for the analysis of bad and bax expression. We found that expressions of bad and bax genes in ovary was markedly reduced before and after PMSG injections. When the preovulatory follicles were cultured for 24 hrs in culture media with progesterone and estrogen, the expression of bad and bax genes was decreased. However testosterone significantly induce the expressions of bad and bax genes in follicles. These data demonstrated that gonadotropin and sex steroid hormone may play an important roles in the regulation of mouse follicular differentiation. PMSG, progesterone and estrogen may prevent apoptosis via suppressing the expressions of bad and bax genes but testosterone may accelerate apoptosis via inducing the bad and bax gene expressions.         Differential Expressions of Adhesion Receptor Genes in the Rat Uterus Associated with Ovarian Steroid Hormone

        본 연구는 생식주기 중의 흰쥐 자궁에서 발현되는 유전자들 중 adhesion수용체 유전자들의 발현이 프로게스테론(P)에 의하여 차별적으로 조절되는 실험을 하였다. 첫째 실험군은 난소절제 흰쥐와 배란기 흰쥐를 사용하였고(OVX/estrus), 둘째 실험군은 난소절제 흰쥐와 난소절제 후 P4를 주사한 흰쥐를 사용하였다(OVX/OVX+P). 적출한 자궁조직에서 total RNA를 추출, [P]-dATP로 probe를 제작한 후 Rat Atlas away 1. This report aimed at investigating the differential gene expressions of the adhesion receptors between ovariectomized (OVX) and estrus stage rat uteri (OVX vs. estrus pair) using the cDNA expression away analysis. In addition, this report aimed at confirming of the differential gene expressions of the adhesion receptors between OVX and progesterone (P) injected OVX rat uteri (OVX vs. OVX+P pair). RNA samples were extracted from the uterus and reverse-transcribed in the presence of [ P]-dATP. Membrane sets of Rat Atlas array 1.2 II (Clontech) were hybridized with CDNA probe sets. RT-PCR was employed to validate the relative gene expression patterns obtained by the cDNA array. The results were well consistent to cDNA array analysis data except the fold changes of gene expression. Among a total of 1176 cDNAs, 5 genes of adhesion receotor including embigin protein, activated leukocyte cell adhesion molecule, afadin, neuroligin 2, semaphorin Z showed significant (more than 2-fold) changes in the OVX vs. late estrus pair. All of these genes were up regulated in estrus stage than OVX rat uterus. In the OVX vs. OVX+P pair, 4 genes including osteonectin, afadin, neuroligin 2, semaphorin Z showed significant changes. All of these genes were also up regulated in OVX+P injected rat uterus than OVX control. Three genes including afadin, neuroligin 2, semaphorin Z which were up regulated in estrus and OVX+P injected rat uteri of both experimental pairs than OVX rat uteri. These genes seem to be under the control of P.

      • 흰쥐 발정주기에 따른 자궁에서의 Ras 유전자 발현에 관한 연구

        It is now widely accepted that the product of ras gene serves as essential transducer of physiological signals. The expression of ras gene may be changed during rat estrus cycle. The present study aims to determine whether the levels of ras mRNA are changed in uterus during estrus cycle and in ovariectomized ras. Adult female ras exhibiting at least two consecutive 4-day estrus cycles were divided into pro-estrus(P), estrus(E), metestrus(M) and diestrus(D) stages by checking the vaginal smear sample. Ovariectomized ras treated with various steroid hormones were used in this study. All sample tissue were dissected at each stage of estrus cycle and ovariectomized ras were treated with steroids and then the expression level of mRNA was determined and quantified using RT-PCR. The results are as follows. 1. The levels of ras mRNA reached a maximum level in endo, myometrium at metestrus stage and then gradually decreased. 2. Estrogen induced the syntheses of ras mRNA in endometrium tissue of ovariectomized rat. But in myometrium tissue the synthesis of ras mRNA is induced by progesterone.         Study on the Regulation of KAP3 Gene Involved in the Brain Sexual Differentiation by DDT during the Critical Period of Fetal and Neonatal Age

        환경에 방출되어 있는 많은 내분비교란물질들은 사람과 동물의 내분비계에 교란을 일으킬 수 있는 잠재력을 가진다. 뇌의 성분화는 생식소 호르몬 영향하에 비가역적으로 진행되며 흰쥐의 경우 이 시기는 임신말기에서 생후 7∼10일 가량이다. 최근에 본 연구진은 횐쥐의 뇌 성 분화의 결정적인 시기에 발현되는 KAP3유전자를 클로닝하였다 (Choi & Lee, 1999). KAP3의 기능은 신경세포를 포함한 세포에서 aronal tansport를 조절하는 것으로 알 A large number of man-made chemicals that have been released into the environment have the potential to disrupt the endocrine system of animals and humans. There is a critical developmental period during which sexual brain differentiation proceeds irreversibly under the influence of gonadal hormone. Recently we identified KAP3 gene expressed during the critical period of rat brain sexual differentiation. KAP3 functions as a microtubule-based motor that transports membranous organelles anterogradely in cells, including neurons. In the present study, we aimed to investigate the effect of endocrine disrupter, Dichlorodiphenyl trichloroethane (DDT), on the KAP3 gene expression during critical period of rat brain development. Maternal exposure to DDT increased the level of KAP3 mRNA in male and female fetus brains when examined on the gestational day 17 (GDl7). In postnatal day 6, DDT suppressed the expression of KAP3 gene in male and female rat brain. Also, the body weight and fertilization rate were decreased in the DDT exposured rats. These results showed that endocrine disrupter, DDT, can affect the transcriptional level of brain sexual differentiation related gene, KAP3, in the prenatal and the neonatal rat brain and that maternal exposure to endocrine disruptors may lead to a toxic response in embryonic differentiation of brain. And so KAP3 gene may be used a gene maker to analyse the molecular mechanism for toxic response in animal nerve tissues exposed to endocrine disruptors.

        왕우럭조개 (Tresus keenae) 의 산란 및 유생의 발생

        This study interested on spawning and early embryo development in surf clam, Tresus keenae. For spawning induction, we performed natural spawning, physical stimulus (temperature, air dry), freshwater stimulus and chemical stimulus (sperm suspension). In addition, the sperm amounts were differently treated during artificial insemination and were investigated optimal amount of sperm through fertilization rate and hatching rate. The effects of water temperature and salinity on early embryo development were also investigated. Result that, the physical stimulation method was the most effective method with 35% efficiency in inducing spawning. The fertilization rate and hatching rate were best when 50 × 104 inds./egg sperm were used for artificial insemination. The effects of water temperature and salinity on reaction velocity of embryogenesis were investigated. The fast velocity of embryogenesis had the water temperature was higher and salinity concentration was close to natural condition.

        개인의무기록 기반 헬스케어 어플리케이션을 통한 건강관리 가이드 제공 및 서비스 만족도 평가 연구 : 응급의료현장 근로자의 직무・환경・ 건강 정보 활용을 중심으로

        The market of the health care service based on individual medical records is growing 45.1% every year from about 3 trillion won in 2019. It is expected to reach about 14 trillion won by 2023. Among various industries, the emergency medical field is directly related to the lives and health care of the people and has very strong publicity. Due to the recent increase in medical demand, an increase in adult diseases, and an aging population, emergency medical services are the core of the domestic medical delivery system, which is used by more than 10 million people every year. This study was conducted for the purpose of empirical analysis to collect job information, work environment information, and health information of workers in special job environments such as emergency medical sites for the purpose of building big data. Since the area of big data construction of personal medical records is the process of collecting individual sensitive information, problems will inevitably arise from the information provision process if appropriate services are not performed after providing information. The results of this study derived service value that can continuously provide sensitive information. Among the sensitive information, job characteristics, physical health, and psychological health were found to have a high intention to provide information, and information on the working environment was found to have little influence on the value of health care services. Through this study, positive factors have been prepared to solve the problems of low utilization of healthcare applications in previous studies, and we hope that it will contribute greatly to the provision of health care guides and services for workers in the industry in the future.

        왕우럭 (Tresus keenae) 에서 분리한 장내미생물 Bacillus species를 이용한 유생 사육

        In this study, we conducted a larval breeding study of Tresus keenae using 5 Bacillus genera, which are excellent in the ability to decompose high molecular weight organic matter and the antibacterial activity of pathogenic bacteria, among the 65 Bacillus genera. Research results that the relationship between larval growth and viability was high in the TKI42 and microbial mixed strains of TKI02 + TKI14 + TKI26 + TKI32 + TKI42. The growth and viability of larvae at the amount of the mixed microbial strain (TKI02 + TKI14 + TKI26 + TKI32 + TKI42) was 1.0×106 CFU/ml, which was the best growth, and 1.0×107 CFU/ml, which was the lowest. And the survival rate was the best at 1.0×105 CFU/ml. In addition, the growth and survival rate were high when the mixed microbial strain (1.0×106 CFU/ml) with a water temperature of 25°C was treated. In this study, when the growth and survival of larval breeding was investigated based on five Bacillus strains, the prediction of the application to the breeding of organisms in the aquaculture industry based on the special ability of a single strain is uncertain. Rather than developing and using only a single strain, it is thought that the effect can be expected to using mixed strains.
