Gonadotropins and Nitric Oxide Can Suppress the Expression of Mouse Follicular Bad and Bax Genes

the pupose of this study was to investigate the effects of gonadotropin and nitric oxide (NO) on the expression of mouse follicular bad and bax genes that are known induce apoptosis. Large and midium size follicles of immature mice were obtained at 0, 24, and 48 hours time intervals after Pregnant Mare's Serum gonadotropins(PMSG, 5 I.U.) injection. Preovulatory follicles collected at 24 hrs after PMSG injection were cultured with or without various chemicals such as gonadotropin, gonadotropin Releasing hormone(GnRH), testosterone, Sodium nitroprusside (SNP) for 24 hrs at . After 24 hrs culture, the culture media was used for nitrite assay and total RNA was extracted, subjected to RT-PCT for the analyses of bad and bax expression. We found that expression of bad and bax genes in follicles was markedly reduced before and after in vivo priming with hCG. When the preovulatory follicles were cultured for 24 hrs in culture media with PMSG and hCG, the expression of bad and bax genes was decreased. Moreover, SNP (NO generating agent) can significantly suppress the expression of bad and bax genes in follicles when apoptosis was induced by GnRH agonist and testosterone. At the same time, nitrite production of culture media was increased in GnRH agonist + SNP, testosterone + SNP and SNP treated groups than control group. These data demonstrated for the first time that peptide hormones and NO may play important roles in the regulation of mouse follicular differentiation and may prevent apoptosis via supressing the expression of bad and bax genes.

several regenerated plants were obtained from anther culture of the hybrid lines of italian ryegrass(lolium multiflorum lam.) X tall fescue(festuca arundinacea schreb). when the anthers of the hybrid plants were incubated for 20 days on callus induction medium, MS medium containing 30g/l of sucrose, 2mg/l of NAA and 1 mg/l of kinetin, their calli were induced. the mean ratio of callus induction was 11.6percent, and the mean of callus weight was 9.1 mg/callus/anther. when the calli of the hybrid plants were incubated for 50 days on plant regeneration medium, MS medium containing 30g/l of sucrose, 1 mg/l of NAA and 2 mg/l of kinetin, the hybrid plants were regenerated. the mean ratio of plant regeneration was 27.1 percent and line hyb-1 showed highest regenerability with the frequency of 30.2 percent.

소프트웨어의 개발은 생산성을 향상시키기 위해 미리 만들어진 소프트웨어의 컴포넌트를 이용한다. 컴포넌트의 재사용은 소프트웨어 개발비용을 절감하고 개발기간을 단축시킬 수 있다. 소프트웨어 개발에서 컴포넌트를 재사용함으로써 소프트웨어 설계에서 구현까지의 위험요소를 최소화시킬 수 있는 것이다. 소프트웨어를 구현하기 위한 기술로 UML을 도입하여 컴포넌트에 대한 명세를 버전으로 관리하여 재사용의 효율성을 높일 수 있는 방법을 버전 제어 시스템을 통해서 제시하고자 한다. 본 연구의 특징은 UML을 이용하여 컴포넌트의 버전 제어를 통해 소프트웨어 개발시 효율적인 재사용이 될 수 있도록 하는데 그 목적이 있다.

We obtained regenerated Italian ryegrass (Lolium multiflorum Lam) plants by anther culture. When Italian ryegrass anther was incubated for 20 days on callus induction medium. MS medium containing 20 g/l of sucrose, 2 mg/l of NAA and 1 mg/l of kinetin, its callus was induced. The ratio of callus induction was 9.2%, the mean of callus weight was 8.6 mg/callus/anther. When Italian ryegrass callus was incubated for 50 days on plant regeneration medium, MS medium containing 30 g/l of sucrose, 1 mg/l of NAA and 2 mg/l of kinetin, Italian ryegrass plant was regenerated. The ratio of plant regeneration was 26%.
MEASUREMENT OF TOXICITY OF NINE INSECTICIDES TO HYLYPHANTES GRAMINICOLA(ARANEAE : LINYPHIIDAE)

본 연구는 DIPPING METHOD에 의해 HYLYPHANTES GRAMINICOLA에 9개의 살충제의 유독성을 조사를 위해 수행되었다. 결과는 MONOSULTAP, IMIDACLOPRID 그리고 BT가 거미에 해롭지 않은 것을 보여주었다. AVERMECTIN은 거미에 가장 높은 유독성을 가지고 있었으며, 나머지 5개의 살충제의 상관적인 유독성의 순서는 다음과 같았다. (FIPRONIL>DELTAMETHRIN>FENVALERATE>PHOXIM>METHAMIDOPHOS). 결과적으로 생물학 살충제들이 거미들에 모두 안전하지는 않았던 것을 나타내었다.

The alfalfa(Medicago sativa L.) callus was induced from seeds on SH medium contained 3 mg/l of 2,4-D. Several regenerated alfalfa plants and many shoots were obtained by procedure of Kim et al. (1999); 1) incubation for 28~30 days on SH medium added 5 mg/l of NAA and 2mg/l of kinetin, 2) incubation for 3~5 days on SH medium added 11mg/l of 2,4-D and 1 mg/l of kinetin, 3) incubation for 21~25 days on SH medium added 1.6g/l of ammonium sulfate and 5.75g/l of proline. To increase of root induction ratio on plant regeneration process of alfalfa, root induction ratio was examined on 8 kinds of medium, containing different amount of hormone and SH salt. Root induction ratio was higher on SH medium contained IBA than SH basal medium. In case 1.5 mg of IBA was added in SH medium, root induction ratio was the highest to 56.0% in this study. On the other hand, root induction ratio was higher on SH medium diminished SH salt amount to half volume and addition of IBA makes high root induction ratio, too. Thus, we conclude that the medium for root induction of alfalfa may be added 1.5mg/l of IBA and diminished SH salt amount to half volume.
