        Synergistic Effect of Bone Marrow-Derived Mesenchymal Stem Cells and Platelet-Rich Plasma in Streptozotocin-Induced Diabetic Rats

        Background: Diabetic wounds are a major clinical challenge, because minor skin wounds can lead to chronic, unhealed ulcers and ultimately result in infection, gangrene, or even amputation. Studies on bone marrow derived mesenchymal stem cells (BMSCs) and a series of growth factors have revealed their many benefits for wound healing and regeneration. Platelet-rich plasma (PRP) may improve the environment for BMSC development and differentiation. However, whether combined use of BMSCs and PRP may be more effective for accelerating diabetic ulcer healing remains unclear. Objective: We investigated the efficacy of BMSCs and PRP for the repair of refractory wound healing in a diabetic rat model. Methods: Forty-eight rats with diabetes mellitus induced by streptozotocin were divided into four groups: treatment with BMSCs plus PRP, BMSCs alone, PRP alone, phosphate buffered saline. The rate of wound closure was quantified. A histopathological study was conducted regarding wound depth and the skin edge at 7, 14, and 28 days after surgery. Results: Wound healing rates were significantly higher in the BMSC plus PRP group than in the other groups. The immunohistochemistry results showed that the expression of platelet/endothelial cell adhesion molecule 1, proliferating cell nuclear antigen, and transforming growth factor-β1 increased significantly in the BMSC plus PRP group compared to the other treatment groups. On day 7, CD68 expression increased significantly in the wounds of the BMSC plus PRP group, but decreased markedly at day 14 compared to the controls.         A Research on Mathematics Learning Burnout of Non-Mathematics College Students in China

        수학은 모든 과학 연구의 기초이며 과학 기술 진보를위한 강력한 도구입니다. 그러나 점점 더 많은 학생들이 수학 공부를 좋아하지 않으며 소진 감을 느낍니다. 이 논문은 중국의 비 수학 대학생들 사이에서 수학 학습 소진의 원인을 탐구하기 위해 2005 년부터 2019 년까지 25 개의 관련 문헌을 분석합니다. 결론은 다음과 같습니다. A. 2005 년부터 2019 년까지 수학 학습 소진에 대한 평균 기사 수는 연간 2 개 미만이었습니다. B. 자금 지원 프로젝트는 단 두 가지입니다. C. 대학생 수학 학습 소진의 영향 요인에 대한 연구는 없다. D. 대학생 그룹에 대한 연구는 거의 없습니다. E. 대학생 수학 학습 소진에 관한 문헌은 모두 저널 문헌입니다. F. 비 수학 대학생의 수학 학습 소진에 영향을 미치는 요인은 개별 요인 (캐릭터, 지능 요인, 수학 능력, 수학 기초, 심리적 요인) 및 객관적 요인 (사회적 요인, 학교 요인, 가족 요인, 수학 특성)으로 나뉩니다. ).대학생의 일반 학습 소진에 따르면 가족 요인과 학교 요인의 학업 부담이 대학생의 수학 학습 소진에 영향을 미치는 요소는 아닙니다. Mathematics is the foundation of all scientific research and a powerful tool for scientific and technological progress. However, more and more students do not like to study mathematics and feel burnout. In order to explore the causes of mathematics learning burnout among non-mathematics college students in China, this paper analyzes 25 relevant literatures from 2005 to 2019. The conclusions are as follows: A. From 2005 to 2019, the average number of articles on math learning burnout was less than 2 per year. B. Only two articles are fund-supported projects. C. There is no research on the influencing factors of college students mathematics learning burnout. D. There is very little research on the group of college students. E. The literature about college students mathematics learning burnout is all journal literature. F. The influencing factors of mathematics learning burnout of non-mathematics college students are divided into individual factors(Character, intelligence factors, mathematics ability, mathematics foundation, psychological factors) and objective factors(social factors, school factors, family factors, mathematics characteristics). According to the college students general learning burnout, family factors and the academic burden of school factors does not constitute an influencing factor for college students mathematics learning burnout.

      • Knockdown of HMGN5 Expression by RNA Interference Induces Cell Cycle Arrest in Human Lung Cancer Cells

        HMGN5 is a typical member of the HMGN (high mobility group nucleosome-binding protein) family which may function as a nucleosomal binding and transcriptional activating protein. Overexpression of HMGN5 has been observed in several human tumors but its role in tumorigenesis has not been fully clarified. To investigate its significance for human lung cancer progression, we successfully constructed a shRNA expression lentiviral vector in which sense and antisense sequences targeting the human HMGN5 were linked with a 9-nucleotide loop. Inhibitory effects of siRNA on endogenous HMGN5 gene expression and protein synthesis were demonstrated via real-time RT-PCR and western blotting. We found HMGN5 silencing to significantly inhibit A549 and H1299 cell proliferation assessed by MTT, BrdU incorporation and colony formation assays. Furthermore, flow cytometry analysis showed that specific knockdown of HMGN5 slowed down the cell cycle at the G0/G1 phase and decreased the populations of A549 and H1299 cells at the S and G2/M phases. Taken together, these results suggest that HMGN5 is directly involved in regulation cell proliferation in A549 and H1299 cells by influencing signaling pathways involved in cell cycle progression. Thus, our finding suggests that targeting HMGN5 may be an effective strategy for human lung cancer treatment.

        Effect of Shifting the Pole-shoe and Damper-bar Centerlines on the No-load Voltage Waveform of a Tubular Hydro-generator

        This study optimises the no-load voltage waveform of tubular hydro-generators by a simple design scheme. For different centerlines of the pole shoe and damper bar, the optimisation effects on the no-load voltage waveform are investigated in two tubular hydro-generators with different weighted powers (34 MW and 18 MW). The results are compared with those of the traditional stator-slots skewed design. The quality of the no-load voltage waveform was related to the shifting degree, and the different optimisation effects between the integer slot generator (q = 2) and the fractional slot generator (q = 11/2) were analysed. This research can improve the quality of the power output and no-load voltage waveform, and provide an effective reference for improving the industrial design and manufacture level of tubular hydro-generators.

        Response Modification Factor and Displacement Amplification Factor of Y-Shaped Eccentrically Braced High-Strength Steel Frames

        A Y-shaped eccentrically braced high-strength steel frame is a novel structure. For such structure to exhibit good plastic deformation ability under severe earthquake, the links are made of ordinary steel ( f y ≤ 345 MPa), whereas high-strength steel ( f y ≥ 460 MPa) is used in the frame beam and column to reduce the cross section while ensuring the elasticity of the non-energy consuming component. The new structure has good ductility and energy-dissipating capacity. The response modifi cation factor R is crucial to the performance-based seismic design. For an appropriate and economical seismic design, the R value should be reasonably selected. In the 2016 edition of China's code for Seismic Design of Buildings (GB50011- 2010), it is obviously not reasonable to hide the R concept and adopt a constant value for all structural systems. It is important to study R and C d of a Y-shaped eccentrically braced high-strength steel frame, it can not only improve the structural performance design, but also provide reference for the future, and promotes the application of the structure in seismic areas, which presents excellent seismic performance. Therefore, in this study, structures with diff erent stories (4, 8, 12, and 16 stories) and link lengths (700, 900, and 1100 mm) were designed via the performance-based seismic design method. A static elastic–plastic analysis (Pushover analysis) and an incremental dynamic analysis (IDA) were conducted and the data thus obtained was fi tted to reach the IDA performance curve of the structure. The R and C d value of each prototype were calculated using the capability spectrum method, the number of structural story N and the link length e is considered. The results of the pushover analysis and IDA were compared; the values of each performance coeffi cient obtained can serve as a reference for the performance design of the new structural systems in the future.

      • CCNG2 Suppressor Biological Effects on Thyroid Cancer Cell through Promotion of CDK2 Degradation

        This study aimed to analyze the expression and clinical significance of cyclin G2 (CCNG2) in thyroid carcinoma and the biological effects of CCNG2 overexpression in a cell line. Immunohistochemistry and Western blotting were used to analyze CCNG2 protein expression in 63 cases of thyroid cancer and normal tissues to allow the relationship with clinical factors to be assessed. CCNG2 lentiviral and empty vectors were transfected into the thyroid cancer K1 cell line. Reverse transcription-polymerase chain reaction (RT-PCR) and Western blotting were applied to detect the mRNA and protein levels of CCNG2. MTT assay and cell cycle were also conducted to assess the influence of up-regulated expression of CCNG2 on K1 cell biology. The level of CCNG2 protein expression was found to be significantly lower in thyroid cancer tissue than normal tissues (P<0.05). Western blot: The relative amount of CCNG2 protein in thyroid cancer tissue was respectively found to be significantly lower than in normal tissues (P<0.05), correlating with lymph node metastasis, clinic stage and histological grade (P<0.05), but not gender, age or tumor size (P>0.05). Loss of CCNG2 expression correlated significantly with poor overall survival time on Kaplan-Meier analysis (P<0.05). The results for biological functions showed that K1 cell transfected CCNG2 had a lower survival fraction, a greater percentage in the G0/G1 phases, and lower cyclin-dependent kinase 2 (CDK2) protein expression compared with K1 cells non-transfected with CCNG2 (P<0.05). CCNG2 expression decreased in thyroid cancer and correlated significantly lymph node metastasis, clinic stage, histological grade and poor overall survival, suggesting that CCNG2 may play important roles as a negative regulator in thyroid cancer K1 cells by promoting degradation of CDK2.

        Effects of Alpha 1- and Alpha 2-Adrenoreceptor Stimulation on Galanin mRNA Expression in Primary Cultured Superior Cervical Ganglion Neurons

        Galanin (Gal) is a 29-amino-acid neuropeptide which is expressed in superior cervical ganglion (SCG) neurons and plays a trophic role in the adult animal and acts as an inhibitory modulator of cholinergic and noradrenergic neurotransmission. Whether activation or inhibition of alpha-adrenoreceptors infl uences Gal mRNA expression in SCG neurons remains unknown. Here, we have evaluated the possible regulation of Gal mRNA expression with acute (4 h) and chronic (4 days) stimulation of alpha 1- and alpha 2-adrenoreceptor agonists or antagonists in primary cultured SCG neurons. The results showed that the amount of Gal mRNA expression in cultured SCG neurons increased significantly after chronic stimulation with alpha 2-adrenoreceptor antagonist yohimbine compared with control SCG neurons at the same time point, whereas the amount of Gal mRNA expression decreased significantly after chronic stimulation with alpha 2-adrenoreceptor agonist clonidine as compared with that in control group. All these effects were not dose-dependent on the administration of alpha 2-adrenoreceptor agonist clonidine or alpha 2-adrenoreceptor antagonist yohimbine. Alpha 1-adrenoreceptor agonist phenylephrine or antagonist prazosin chronic stimulation did not have effects on Gal mRNA expression. Acute exposure of these agents did not have effects on Gal mRNA expression. The present study showed that Gal may be regulated by activation or inhibition of alpha 2-adrenoreceptors, but not alpha 1-adrenoreceptors in sympathetic neurons.

        A Role of YlBud8 in the Regulation of Cell Separation in the Yeast Yarrowia lipolytica

        The spatial landmark protein Bud8 plays a crucial role in bipolar budding in the budding yeast Saccharomyces cerevisiae. The unconventional yeast Yarrowia lipolytica can also bud in a bipolar pattern, but is evolutionarily distant from S. cerevisiae. It encodes the protein YALI0F12738p, which shares the highest amino acid sequence homology with S. cerevisiae Bud8, sharing a conserved transmembrane domain at the C-terminus. Therefore, we named it YlBud8. Deletion of YlBud8 in Y. lipolytica causes cellular separation defects, resulting in budded cells remaining linked with one another as cell chains or multiple buds from a single cell, which suggests that YlBud8 may play an important role in cell separation, which is distinct from the function of Bud8 in S. cerevisiae. We also show that the YlBud8-GFP fusion protein is located at the cell membrane and enriched in the bud cortex, which would be consistent with a role in the regulation of cell separation. The coiled-coil domain at the Nterminus of YlBud8 is important to the correct localization and function of YlBud8, as truncated proteins that do not contain the coiled-coil domain cannot rescue the defects observed in Ylbud8Δ. This finding suggests that a new signaling pathway controlled by YlBud8 via regulation of cell separation may exist in Y. lipolytica.

        Altered mRNA Levels of MOV10, A3G, and IFN-α in Patients with Chronic Hepatitis B

        To explore the relationship of the MOV10, A3G, and IFN-αmRNA levels with chronic hepatitis B virus (HBV) infection,Blood samples from 96 patients with chronic hepatitis B(CHB) and 21 healthy individuals as control were collected. HBV DNA load and aminotransferase in the serum weretested using real time PCR and velocity methods, respectively. The MOV10, A3G, and IFN-α mRNA levels in theperipheral blood mononuclear cells (PBMC) were examinedthrough qRT-PCR. The MOV10, A3G, and IFN-α mRNAlevels in CHB group was significantly lower than those inthe control group (P<0.01, P<0.05, P<0.01, respectively). TheA3G mRNA level in the high-HBV DNA load group waslower than that in the low-HBV DNA load group (P<0.05). However, no statistical difference was found in the MOV10and IFN-α mRNA levels between the two HBV DNA loadgroups. Furthermore, the MOV10 mRNA level showed positivecorrelation with IFN-α in the control group. These resultsindicated that the expression of the innate immune factorsMOV10, A3G, and IFN-α is affected by chronic HBV infection.
