        Comparison of Melanoma Subtypes among Korean Patients by Morphologic Features and Ultraviolet Exposure

        Background: Genetic alterations have been identified in melanomas according to different levels of sun exposure. Whereas the conventional morphology-based classification provides a clue for tumor growth and prognosis, the new classification by genetic alterations offers a basis for targeted therapy. Objective: The purpose of this study is to demonstrate the biological behavior of melanoma subtypes and compare the two classifications in the Korean population. Methods: A retrospective chart review was performed on patients found to have malignant melanoma in Severance Hospital from 2005 to 2012. Age, sex, location of the tumor, histologic subtype, tumor depth, ulceration, lymph node invasion, visceral organ metastasis, and overall survival were evaluated. Results: Of the 206 cases, the most common type was acral melanoma (n=94, 45.6%), followed by nonchronic sun damage-induced melanoma (n=43, 20.9%), and mucosal melanoma (n=40, 19.4%). Twenty-one patients (10.2%) had the chronic sun-damaged type, whereas eight patients (3.9%) had tumors of unknown primary origin. Lentigo maligna melanoma was newly classified as the chronic sun-damaged type, and acral lentiginous melanoma as the acral type. More than half of the superficial spreading melanomas were newly grouped as nonchronic sun-damaged melanomas, whereas nodular melanoma was rather evenly distributed. Conclusion: The distribution of melanomas was largely similar in both the morphology-based and sun exposure-based classifications, and in both classifications, mucosal melanoma had the worst 5-year survival owing to its tumor thickness and advanced stage at the time of diagnosis.         The measurement system for long bolt has been proposed. As a sensing device LVDT has been adopted. Tentative result revealed that the measurement system was efffective in measuring bolt longer than 100mm in length. It has also been studied regarding the distortion of system due to its kinematic structure.

        A new device, muscle clamping system was developed to facilitate exact quatifying technique A strabismus surgery, and reduce the risk of complications. The device is composed of a lower fixing body with three jaws and an upper supporting body. They are used to clip an extraocular muscle and fix it to the sclera with a single bite. Superior rectus recession on 16 rabbit eyes were performed with this new device. Conjunctival injection, muscle adhesion strength, and light microscopic findings were examined at 1, 2, 4 and 8 weeks postoperatively. The Conjunctival injection were minimal, adhesion power ranged from 420 to 600 gram gravity, which is sufficient in withstand the normal pull of human extraocular muscle. In microscopic exam, some Inflammatory cells and fibrosis were found. The new device was technically easy, fast, and accurate, so it may be useful in stabismus surgery.

        中學 科學敎育의 探究學習에 대한 評價問項 開發 (Ⅱ)

        This study was accomplished to develope the evaluation items for inquiry learning in the 2nd grade Middle School Science for the consecutive study of the evaluation items for inquiry learning in the 1st grade Middle School Science(U-Hang Ki et al, 1984). In this study, paper and pencil test items and performance test items are made by analyzing the abilities of inquiry according to the contents and four basic experiments from each unit in the 2nd grade Middle school science. These evaluation items were applied to tke five classes of the 2nd grade of middle school to test their validity. It is desirable that performance test schuld be used for the evaluation for the abilities of inquiry which can not be evaluated by paper and pencil test. In the evaluation methods of performance test, tester evaluation, peer evaluation and self-evaluation can be applied to the science class in a multi-student class. In higher grade, however tester evaluation is more desirable than peer and self evaluation. It is found that peer evaluation and self-evaluation make possible the perfect study by feedback.

      • Vibrio parahemolyticus bc-7가 생산하는 Protease에 관한 연구

        목 적 : 세포외 효소인 protease는 몇몇 병원균, 즉 pseudomonase aeruginosa, Serratia marcescens, Vibrio cholerae 등에 의해 야기되는 질병에 있어서 주요한 독성 인자로 작용하며, 균의 독력과 관계된 요소이다. 비브리오 속의 경우, protease 생산능이 결여된 변이주에서는 독력이 현저히 감소된다는 사실이 여러 연구자에 의해 밝혀졌다. 그러나 protease 생산능과 병원성과와 관련성을 명백히 입증하기 위해서는 효소의 정제 및 특이 항혈청을 포함하는 실험적 근거가 필요하다. 따라서 1997년 7월에서 10월 사이에 부산시내의 해수 및 해산물로부터 비브리오속 100균주를 분리하여, protease를 정제하여 물리화학적 특성을 밝히고자 하였다. 방 법 : 분리된 비브리오속 균주들의 protease생산 여부를 알아보기 위하여 1% 젤라틴이 함유된 tryptic soyagar 배지에 전배양액을 접종하고, 30℃에서 18시간 배양하여 12.5% HCl 용액을 부어서 투명환이 생산되는 지의 여부로 판정하였다. 또한 casein에 대한 protease의 활성을 측정하고, 활성이 가장 높은 균주를 대상으로 pH, 온도 및 NaCl 농도가 protease 생산에 미치는 영향을 조사하고, 최적조건하에서 생산된 protease를 유안침전, DEAE-Sephadex A-50 및 Phenyl-Toyopearl 650M column chromatography를 실시하여 정제하고, 정제된 효소의 분자량, 온도 안정성 및 pH 안정성 등 물리화학적 성상을 규명하였다. 결 과 : 분리된 100균주를 대상으로 protease 생산능 및 활성을 측정한 결과, V. parahemolyticus bc-7이 protease 생산능이 가장 우수하였고, 이 공시균주는 초발 pH 8.0에서 pH 9.0, NaCl 농도가 3%, 배양온도 30℃에서 protease 생산이 가장 우수한 것으로 나타났다. V. parahemolyticus bc-7이 생산하는 protease는 배양 9시간째 부터 그 활성이 나타나기 시작하여 대수증식기 말기 및 정지기에 이르러 최대로 되었으며, 그 이후 점차 감소하였다. 정제과정을 거치는 동안 비활성은 1896.7units/ml로 배양상등액에 비해 1459.0배 정제되었다. 수율은85%로 나타났다. 정제된 V. parahemolyticus bc-7가 생산한 protease의 자외선 영역에서의 흡광도는 단백질의 전형적인 양상을 나타내었고 280nm에서 최대흡광도를 나타내었으며, SDS-PAGE를 실시한 결과 정제된 protease의 분자량은 65KD 이었다. 또한 정제된 protease는 60℃까지 활성이 비교적 안정되게 유지되었고 최적반응 온도는 50℃였으며, pH 5에서 pH 10범위에서 비교적 안정하였으나 pH 4.0이하에서는 활성이 손실되었으며, 최적 pH는 7.0이었다. 결 론 : 비브리오 속이 생산하는 세포외 효소인 protease에 관한 다양한 연구들이 진행되어 왔지만 아직까지 그 병리기전 및 독성에 관해서는 정확하게 밝혀지지 않고 있다. 따라서 계속적인 연구를 통해 실험동물 및 어류에 대한 독성을 밝히는 한편, 분리균주가 숙주내로 침입하여 질병을 유발하는 병리기전을 분자적 수준에서 연구되어야 할 것이며, protease가 작용하는 표적기관에 대해서도 더욱 심도있게 연구되어야 할 것으로 사료된다. Background : Bacterial extracellular protease has been implicated as an important virulence factor in several infections. In Vibrio sp., protease involvement has been demonstrated by the decreased virulence of protease-dificient mutants. We characterized the protease from V. parahemolyticus be-7 with purification. Methods : 100 Vibrio isolates from sea water and sea products were studied. The bacteria grown in Brain Heart Infusion broth were screened for protease production by incubating on tryptic soy agar plate containing gelatin and subsequently flooding the plate with 12.5% ?? HCl. A clear zone indicated protease activity. Optival conditions for protease production were determined by pH, temperature and NaCl concentration. Protease was purified from culture filtrates and its biological analysis was carried out. Results : Out of Vibrio isolates studied, V. parahemolyticus be-7 produced protease in the highest titer. Maximal yields of the protease was obtained when the bacteria were grown in Brain Heart Infusion broth with initial pH of 8.0, 3% NaCl at 30℃. Production of the protease was optimal during the late exponential phase. Homogeneity of the purified protease was demonstrated by revealing a 65kDa band on SDS-PAGE. The protease was stable at 50℃ and 70% of the activity was lost by heating at 60℃ for 30 min. The protease was stable at a pH between 5∼10, but under pH 4.0, the activity was lost completely. Conclusion : General and serological studies are warranted to clarify the diffences and further characterize the protease as a virulence factor in Vibrio sp.
