        Effects of Horse Riding Simulator on Pain, Oswestry Disability Index and Balance in Adults with Nonspecific Chronic Low Back Pain

        PURPOSE: The purpose of this study was to investigate effects of Horse riding simulator exercise on pain, dysfunction and dynamic balance in adults with nonspecific chronic low back pain. METHODS: In this study, total 19 college students usually complain of low back pain who were randomly divided into the horse riding simulator exercise group (n=10), lumbar strengthening exercise group (n=9) were recruited. Each group carried out for 30 minutes exercise three times a week for 4 weeks. Horse riding simulator exercise group carried out 15 minutes horse riding simulator exercise and 15 minutes lumbar strengthening exercise. Lumbar strengthening exercise group carried out 30 minutes lumbar strengthening exercise. Visual analogue scale (VAS) were measured for evaluation back pain. Korean oswestry disability index (KODI) were measured for dysfunction. Limits of stability (LOS) were measured for dynamic balance. RESULTS: VAS, KODI, LOS results showed a significant change within both horse riding simulator exercise group, lumbar strengthening exercise group. CONCLUSION: Present study suggested that the horse riding simulator exercise can improve back pain, dysfunction, dynamic balance. Horse riding simulator exercise provides more convenience, interest and motivation than conversional therapy and it could be a possible approach to adults with nonspecific chronic low back pain.

        Causal Relations between Exposome and Stroke: A Mendelian Randomization Study

        Background and Purpose To explore the causal relationships of elements of the exposome with ischemic stroke and its subtypes at the omics level and to provide evidence for stroke prevention. Methods We conducted a Mendelian randomization study between exposure and any ischemic stroke (AIS) and its subtypes (large-artery atherosclerotic disease [LAD], cardioembolic stroke [CE], and small vessel disease [SVD]). The exposure dataset was the UK Biobank involving 361,194 subjects, and the outcome dataset was the MEGASTROKE consortium including 52,000 participants. Results We found that higher blood pressure (BP) (systolic BP: odds ratio [OR], 1.02; 95% confidence interval [CI], 1.01 to 1.04; diastolic BP: OR, 1.03; 95% CI, 1.01 to 1.05; pulse pressure: OR, 1.03; 95% CI, 1.00 to 1.06), atrial fibrillation (OR, 1.18; 95% CI, 1.13 to 1.25), and diabetes (OR, 1.13; 95% CI, 1.07 to 1.18) were significantly associated with ischemic stroke. Importantly, higher education (OR, 0.69; 95% CI, 0.60 to 0.79) decreased the risk of ischemic stroke. Higher systolic BP (OR, 1.06; 95% CI, 1.02 to 1.10), pulse pressure (OR, 1.08; 95% CI, 1.02 to 1.14), diabetes (OR, 1.28; 95% CI, 1.13 to 1.45), and coronary artery disease (OR, 1.58; 95% CI, 1.25 to 2.00) could cause LAD. Atrial fibrillation could cause CE (OR, 1.90; 95% CI, 1.71 to 2.11). For SVD, higher systolic BP (OR, 1.04; 95% CI, 1.00 to 1.07), diastolic BP (OR, 1.06; 95% CI, 1.01 to 1.12), and diabetes (OR, 1.22; 95% CI, 1.10 to 1.36) were causal factors. Conclusions The study revealed elements of the exposome causally linked to ischemic stroke and its subtypes, including conventional causal risk factors and novel protective factors such as higher education.

        IRS-2 Partially Compensates for the Insulin Signal Defects in IRS-1−/−Mice Mediated by miR-33

        Insulin signaling is coordinated by insulin receptor substrates (IRSs). Many insulin responses, especially for blood glucose metabolism, are mediated primarily through Irs-1 and Irs-2. Irs-1 knockout mice show growth retardation and insulin signaling defects, which can be compensated by other IRSs in vivo; however, the underlying mechanism is not clear. Here, we presented an Irs-1 truncated mutated mouse (Irs-1−/−) with growth retardation and subcutaneous adipocyte atrophy. Irs-1−/− mice exhibited mild insulin resistance, as demonstrat-ed by the insulin tolerance test. Phosphatidylino-sitol 3-kinase (PI3K) activity and phosphorylated Protein Kinase B (PKB/AKT) expression were elevated in liver, skeletal muscle, and subcu-taneous adipocytes in Irs-1 deficiency. In addition, the expression of IRS-2 and its phosphorylated version were clearly elevated in liver and skeletal muscle. With miRNA microarray analysis, we found miR-33 was down-regulated in bone marrow stromal cells (BMSCs) of Irs-1−/− mice, while its target gene Irs-2 was up-regulated in vitro studies. In addition, miR-33 was down-regulated in the presence of Irs-1 and which was up-regulated in fasting status. What’s more, miR-33 restored its expression in re-feeding status. Meanwhile, miR-33 levels decreased and Irs-2 levels increased in liver, skeletal muscle, and subcutaneous adipocytes of Irs-1−/− mice. In primary cultured liver cells transfected with an miR-33 inhibitor, the expression of IRS-2, PI3K, and phosphorylated-AKT (p-AKT) increased while the opposite results were observed in the presence of an miR-33 mimic. Therefore, decreased miR-33 levels can up-regulate IRS-2 expression, which appears to compensate for the defects of the insulin signaling pathway in Irs-1 deficient mice.

        Development and characterization of a potential diagnostic monoclonal antibody against capsid protein VP1 of the chicken anemia virus

        Chicken anemia virus (CAV) is an important viral pathogen that causes anemia and severe immunodeficiency syndrome in chickens worldwide. In this study, a potential diagnostic monoclonal antibody against the CAV VP1 protein was developed which can precisely recognize the CAV antigen for diagnostic and virus recovery purposes. The VP1 gene of CAV encoding the N-terminus-deleted VP1 protein, VP1Nd129, was cloned into an Escherichia (E.) coli expression vector. After isopropyl-b-D-thiogalactopyronoside induction, VP1Nd129 protein was shown to be successfully expressed in the E. coli. By performing an enzyme-linked immunoabsorbent assay using two coating antigens, purified VP1Nd129 and CAV-infected liver tissue lysate, E3 monoclonal antibody (mAb) was found to have higher reactivity against VP1 protein than the other positive clones according to the result of limiting dilution method from 64 clones. Using immunohistochemistry, the presence of the VP1-specific mAb, E3, was confirmed using CAV- infected liver and thymus tissues as positive-infected samples. Additionally, CAV particle purification was also performed using an immunoaffinity column containing E3 mAb. The monoclonal E3 mAb developed in this study will not only be very useful for detecting CAV infection and performing histopathology studies of infected chickens, but may also be used to purify CAV particles in the future.

        Safety and Efficacy of Adalimumab for Patients With Moderate to Severe Crohn`s Disease: The Taiwan Society of Inflammatory Bowel Disease (TSIBD) Study

        Background/Aims: Only moderate to severe Crohn`s Disease (CD) patients without a satisfactory conventional therapy effect are eligible to get reimbursement from the National Health Insurance of Taiwan for using adalimumab. These are more stringent criteria than in many Western countries and Japan and Korea. We aim to explore the efficacy of using adalimumab in CD patients under such stringent criteria. Methods: A retrospective analysis was conducted in nine medical centers in Taiwan and we collected the results of CD patients receiving adalimumab from Sep 2009 to Mar 2014. The clinical characteristics, response measured by CDAI (Crohn`s Disease Activity Index), adverse events and survival status were recorded and analyzed. CR-70, CR-100, and CR-150 were defined as attaining a CDAI decrease of 70, 100 or 150 points compared with baseline. Results: A total of 103 CD patient records were used in this study. Sixty percent of these patients received combination therapy of adalimumab together with immunomodulators. CR-70 was 68.7%, 74.5% and 88.4% after week 4, 8 and 12 of treatment, respectively. The steroid-free rate, complications and survival were 47.6%, 9.7% and 99% of patients, respectively. In considering the mucosal healing, only 25% patients achieve mucosal healing after treatment for 6 to12 months. Surgery was still needed in 16.5% of patients. Combination treatment of adalimumab with immunomodulators further decreased the level of CDAI at week 8 when compared with the monotherapy. Conclusions: Even under the stringent criteria for using adalimumab, the response rate was comparable to those without stringent criteria. (Intest Res 2014;12:287-292)

      • Polypoid Lesions of the Gallbladder: A Long-Term Follow-Up of 1204 Patients

        Aims: Polypoid lesions of the gallbladder (PLG) are common, and most of them are benign. Few lesions are found malignant, but cannot be distinguished preoperatively by using common image modalities. Therefore, we compared characteristics of benign and malignant PLGs. Methods: We enrolled 1204 consecutive patients diagnosed with PLG at Taipei Veterans General Hospital during January 2004 to December 2013. Patients underwent either surgery or regular follow-up with various imaging modalities for at least 24 months. The mean follow-up duration was 72 ± 32 months. Results: Amoung our 1204 patients, 194 underwent surgical treatment and 1010 underwent regular follow-up. In addition, 73% patients were asymptomatic. The mean PLG size was 6.9 ± 7.7 (range, 0.8-129) mm; the PLGs of 337 patients (28%) grew during their follow-up periods. The majority of PLGs (90.4%) were single lesions, and 10.5% of patients had associated gallstones. The PLGs of 20.1% of surgical patients were malignant. Malignant PLGs were found in 32.4% of patients ≥50 years old and in 4.7% of those <50 years old (p < 0.001). Right quadrant abdominal pain, epigastric pain, and body weight loss were the three most common symptoms associated with malignancy. Malignant PLGs were significantly larger than benign lesions (means: 27.5 ± 18.4 mm vs. 12.3 ± 12.3 mm respectively, p < 0.001). Notably, the size of 5% of malignant PLGs was 3-5 mm, and that of 8% was 5-10 mm. The negative predictive value for gallbladder malignancy was 92.8% based on a size ≥10 mm and 100% based on a size ≥3 mm. Conclusions: Our study reassesses the PLG size that warrants more aggressive intervention. Cholecystectomy remains mandatory for PLGs >10 mm, but should also be considered a definitive diagnostic and treatment modality for PLGs with diameters of 3-10 mm.

        Zebrafish Dnd protein binds to 3`UTR of geminin mRNA and regulates its expression

        Dnd (dead end) gene encodes an RNA binding protein and is specifically expressed in primordial germ cells (PGCs) as a vertebrate-specific component of the germ plasma throughout embryogenesis. By utilizing a technique of specific nucleic acids associated with proteins (SNAAP), 13 potential target mRNAs of zebrafish Dnd (ZDnd) protein were identified from 8-cell embryo, and 8 target mRNAs have been confirmed using an RT-PCR analysis. Of the target mRNAs, the present study is focused on the regulation of geminin, which is an inhibitor of DNA replication. Using electrophoretic mobility shift assay (EMSA), we demonstrated that ZDND protein bound the 67-nucleotide region from 864 to 931 in the 3`UTR of geminin mRNA, a sequence containing 60.29% of uridine. Results from a dual-luciferase assay in HEK293 cells showed that ZDND increases the translation of geminin. Taken together, the identification of target mRNA for ZDnd will be helpful to further explore the biological function of Dnd in zebrafish germ-line development as well as in cancer cells.       • Bisorption of Organic Compounds by Spirulina Subsalsa

        In this paper, the biosorption of 13 organic compounds was investigated by using biomass of nonliving, dried algae Spirulina subsalsa as sorbent. The results showed that: the biosorption of `13 organic compounds could be described by the Freundlich isoterm. The isoterm binding constants suggested that the biosorption capacity series was phenlo > o-dihydroxy benzene > p-dihydroxy benzene > o-aminophenol > chlorophenol > 2,4-dichlorophenol for 6 phenolic compounds and alkaline blue BO > active blue KN-R > acidic red 4BS > black G > brown MM > blue 5B > black ATT for 7 dyes. The biosorption constants (lgk) of above 6 phenolic compounds showed a good linear correlation to their molecular connectivity index.         SAFE TECHNOLOGY WITH A NOVEL REAR COLLISION AVOIDANCE SYSTEM OF VEHICLES

        This study aims to reduce the severity of injuries caused by and the frequency of rear-end vehicle collisions. A better collision prediction method is proposed by means of millimeter-wave radar sensors able to rapidly detect potential collisions and alert drivers. Implementation of the system would bring about a reduction in accidents and potentially reduce the severity of the accidents that cannot be avoided. The proposed collision avoidance system adopted millimeter-wave radar sensors to collect information on vehicle’s rear surroundings. The designed experimental device has several unique features. The device has a wide monitoring range, incorporates a continuous detection process, and contains a rapid warning system. A series of real-world experiments were performed on cars (Model: Tercel 1.5, Toyota) to demonstrate the monitoring efficiency and warning frequency of this device under several operating conditions. This study furthered the development of a safe technology for avoidance of rear-end collisions, and this offers a novel system which can be used in a variety of vehicles to reduce the frequency of rear-end vehicle collisions.

        Surfactant Effect on Formation of CaWO<sub>4</sub>:Eu<sup>3+</sup> Crystals with Distinguished Morphologies in Hydrothermal Ambient

        <P>Metal tungstates, expressed by the general formula of MWO4, have important properties and applications in photoluminescence, microwave applications, optical fibers, scintillator materials, humidity sensors, magnetic properties, and catalysts. In this paper, we report a successful synthesis of CaWO4: Eu3+ crystals with various morphologies in mild hydrothermal conditions with surfacntant including sodium citrate, CTAB, PEG and citrate acid (CA). The formation of the crystals are strongly dependent on the employment of surfactant. The surfactant concentration has been found significant influence in the resulting morphologies due to different properties of each one. Extensive characterization have been performed by using X-ray diffraction (XRD), field emission scanning electron microscope (FE-SEM) in search of the formation mechanism of multi-morphological CaWO4: Eu3+ crystals. The growth mechanism of monodispersed CaWO4: Eu3+ crystal are proposed. And the photoluminescence properties were investigated.</P>
